&#x8C03;&#x67E5;&#x5728;&#x53CC;&#x5C42;&#x8868;&#x9762;&#x4F7F;&#x7528;&#x7535;&#x5B50;&#x5438;&#x6536;&#x8C31;&#x548C;&#x8367;&#x5149;&#x5171;&#x632F;&#x80FD;&#x91CF;&#x8F6C;&#x79FB;&#x7684;&#x914D;&#x4F53;-&#x53D7;&#x4F53;&#x76F8;&#x4E92;&#x4F5C;&#x7528;&#x3002;

We investigate interactions between receptors and ligands at bilayer surface of polydiacetylene (PDA) liposomal nanoparticles using changes in electronic absorption spectroscopy and fluorescence resonance energy transfer (FRET). We study the effect of mode of linkage (covalent versus noncovalent) between the receptor and liposome bilayer. We also examine the effect of size-dependent interactions between liposome and analyte through electronic absorption and FRET responses. Glucose (receptor) molecules were either covalently or noncovalently attached at the bilayer of nanoparticles, and they provided selectivity for molecular interactions between glucose and glycoprotein ligands of E. coli. These interactions induced stress on conjugated PDA chain which resulted in changes (blue to red) in the absorption spectrum of PDA. The changes in electronic absorbance also led to changes in FRET efficiency between conjugated PDA chains (acceptor) and fluorophores (Sulphorhodamine-101) (donor) attached to the bilayer surface. Interestingly, we did not find significant differences in UV-vis and FRET responses for covalently and noncovalently bound glucose to liposomes following their interactions with E. coli. We attributed these results to close proximity of glucose receptor molecules to the liposome bilayer surface such that induced stress were similar in both the cases. We also found that PDA emission from direct excitation mechanism was âˆ¼2-10 times larger than that of the FRET-based response. These differences in emission signals were attributed to three major reasons: nonspecific interactions between E. coli and liposomes, size differences between analyte and liposomes, and a much higher PDA concentration with respect to sulforhodamine (SR-101). We have proposed a model to explain our experimental observations. Our fundamental studies reported here will help in enhancing our knowledge regarding interactions involved between soft particles at molecular levels.

Investigating Ligand-Receptor Interactions at Bilayer Surface Using Electronic Absorption Spectroscopy and Fluorescence Resonance Energy Transfer.

Nous &eacute;tudions les interactions entre les r&eacute;cepteurs et ligands &agrave; la surface de la bicouche de nanoparticules liposomale polydiacetylene (PDA), &agrave; l&#39;aide de changements dans l&#39;absorption &eacute;lectronique spectroscopie et fluorescence resonance transfert d&#39;&eacute;nergie (FRET). Nous &eacute;tudions l&#39;effet du mode de liaison (covalente versus non covalentes) entre le receveur et le liposome bicouche. Nous examinons &eacute;galement l&#39;effet des interactions de la fonction de la taille entre liposome et analyte par absorption &eacute;lectronique et les r&eacute;ponses de la frette. Mol&eacute;cules de glucose (r&eacute;cepteur) &eacute;taient soit en lien covalent soit noncovalent attach&eacute;s &agrave; la bicouche de nanoparticules, et ils ont fourni la s&eacute;lectivit&eacute; pour les interactions mol&eacute;culaires entre les ligands de glucose et de la glycoprot&eacute;ine d&#39;e. coli. Ces interactions induites par le stress conjugu&eacute; cha&icirc;ne PDA qui a entra&icirc;n&eacute; des changements dans le spectre d&#39;absorption de PDA (bleu au rouge). Fait int&eacute;ressant, nous n&#39;avons trouv&eacute; des diff&eacute;rences significatives dans l&#39;UV-vis et frette r&eacute;ponses pour le glucose li&eacute; par covalence et de fa&ccedil;on non covalente aux liposomes suite &agrave; leurs interactions avec les e. coli. Nous avons attribu&eacute; ces r&eacute;sultats &agrave; proximit&eacute; de mol&eacute;cules de glucose du r&eacute;cepteur &agrave; la surface de la bicouche de liposome tel que le stress induit ont &eacute;t&eacute; similaires dans les deux cas. Nous avons &eacute;galement constat&eacute; que les &eacute;missions de PDA de m&eacute;canisme d&#39;excitation directe &eacute;taient &sim;2-10 fois sup&eacute;rieure &agrave; celle de l&#39;intervention ax&eacute;e sur la frette. Ces diff&eacute;rences dans les signaux des &eacute;missions ont &eacute;t&eacute; attribu&eacute;es &agrave; trois raisons principales : des interactions entre e. coli et des liposomes, des diff&eacute;rences de taille entre l&#39;analyte et liposomes et une concentration beaucoup plus &eacute;lev&eacute;e de PDA avec respectent &agrave; sulforhodamine (SR-101). Nous avons propos&eacute; un mod&egrave;le pour expliquer nos observations exp&eacute;rimentales. Nos &eacute;tudes fondamentales rapport&eacute;s ici aideront &agrave; am&eacute;liorer nos connaissances au sujet de la cause des interactions entre particules molles au niveau mol&eacute;culaire.

Wir untersuchen die Wechselwirkungen zwischen Rezeptoren und Liganden an Bilayer Oberfl&auml;che des Polydiacetylene (PDA) liposomalen Nanopartikel mit &Auml;nderungen im elektronischen Resorption Spektroskopie und Fluoreszenz Resonanz Energietransfer (FRET). Wir untersuchen die Auswirkungen der Art der Verbindung, die zwischen den Rezeptor und Liposomen Bilayer (kovalente gegen&uuml;ber). Wir untersuchen auch das gr&ouml;&szlig;enabh&auml;ngigen Wechselwirkungen zwischen Liposomen und Analyten durch elektronische Absorption und FRET Antworten. Interessanterweise fand wir nicht signifikante Unterschiede im UV-Vis und FRET Antworten f&uuml;r noncovalently und kovalent gebundene Glucose zu Liposomen, die nach ihrer Interaktionen mit E. Coli. Wir zugeschrieben diese Ergebnisse um N&auml;he Glukose Rezeptor Molek&uuml;le zu den Liposomen Bilayer Oberfl&auml;che zu schlie&szlig;en, so dass die induzierte Spannung waren in beiden F&auml;llen &auml;hnlich. Wir fanden auch, dass PDA-Emission aus direkte Anregung Mechanismus &sim;2-10-mal gr&ouml;&szlig;er als die der FRET-basierten Antwort war. Diese Unterschiede in der Emission Signale wurden drei Gr&uuml;nden zugeschrieben: unspezifische Interaktionen zwischen Escherichia Coli und Liposomen, Gr&ouml;&szlig;enunterschiede zwischen Analyt und Liposomen und eine viel h&ouml;here Konzentration der PDA mit Bezug auf Sulforhodamin (SR-101). Wir haben ein Modell, um unsere experimentellen Beobachtungen erkl&auml;ren vorgeschlagen. Unsere grundlegenden Studien berichtet hier hilft bei der Verbesserung unseres Wissens &uuml;ber Interaktionen, die zwischen weichen Teilchen auf molekularer Ebene beteiligt.

Indaghiamo le interazioni tra recettori e ligandi alla superficie del doppio strato di nanoparticelle liposomiale polydiacetylene (PDA), usando le modifiche di assorbimento elettronico fluorescenza e spettroscopia resonance energy transfer (FRET). Studiamo l&#39;effetto della modalit&agrave; di legame (covalente versus non covalenti) tra il recettore e liposomi doppio strato. Esaminiamo anche l&#39;effetto del dipendente dalla dimensione interazioni tra liposomi e analita tramite assorbimento elettronico e risposte FRET. Molecole di glucosio (recettore) sono stati fissati o covalenza o noncovalently presso il doppio strato di nanoparticelle, e hanno fornito la selettivit&agrave; per le interazioni molecolari tra glucosio e glicoproteina ligandi di e. coli. Queste interazioni indotta da stress su coniugati catena PDA che ha provocato cambiamenti (blu al rosso) nello spettro di assorbimento del PDA. Le variazioni di assorbanza elettronico anche portarono cambiamenti in FRET efficienza tra fluorofori (Sulphorhodamine-101) (donatore) attaccato alla superficie del doppio strato e coniugate PDA catene (accettore). &Egrave; interessante notare che, non abbiamo trovato differenze significative nelle risposte UV-vis e FRET per glucosio legato covalentemente e noncovalently ai liposomi seguendo le loro interazioni con e. coli. Abbiamo attribuito questi risultati per prossimit&agrave; di molecole di glucosio recettore alla superficie del doppio strato liposoma tale che lo stress indotto erano simili in entrambi i casi. Abbiamo anche trovato che emissione PDA dal meccanismo di eccitazione diretta era &sim;2-10 volte pi&ugrave; grande di quello della risposta basata su FRET. Queste differenze nei segnali di emissione sono state attribuite a tre motivi principali: interazioni aspecifiche tra e. coli e liposomi, differenze di dimensione tra analita e liposomi e una concentrazione molto pi&ugrave; elevata di PDA con rispettano a cutaneo (SR-101). Abbiamo proposto un modello per spiegare le osservazioni sperimentali. I nostri studi fondamentali segnalati qui aiuter&agrave; a migliorare la nostra conoscenza per quanto riguarda le interazioni coinvolte tra particelle morbidi, a livello molecolare.

Investigamos las interacciones entre receptores y ligandos en la superficie de la bicapa de nanopart&iacute;culas liposomal polydiacetylene (PDA), con cambios en la absorci&oacute;n electr&oacute;nica espectroscopia y fluorescencia resonancia transferencia de energ&iacute;a (traste). Estudiamos el efecto del modo de vinculaci&oacute;n (covalente y no covalente) entre el receptor y liposomas bicapa. Tambi&eacute;n examinamos el efecto de interacci&oacute;n dependiente de tama&ntilde;o liposoma analito a trav&eacute;s de la absorci&oacute;n electr&oacute;nica y respuestas de traste. Mol&eacute;culas de glucosa (receptor) se unen covalentemente o subunidades en la bicapa de las nanopart&iacute;culas, y previeron las interacciones moleculares entre ligandos glucosa y glicoprote&iacute;na de e. coli selectividad. Estas interacciones inducida por el estr&eacute;s de conjugado cadena de PDA, resultando en cambios (de azul a rojo) en el espectro de absorci&oacute;n del PDA. Los cambios en la absorbencia electr&oacute;nico tambi&eacute;n llevaron a cambios en eficiencia de traste entre cadenas de PDA conjugadas (Aceptador) y fluor&oacute;foros (Sulphorhodamine-101) (donante) adjunto a la superficie de la bicapa. Curiosamente, no encontramos diferencias significativas en las respuestas de UV-vis y traste para glucosa enlazada covalentemente y subunidades a liposomas siguiendo sus interacciones con e. coli. Atribuimos estos resultados para cerrar cerca de mol&eacute;culas de glucosa del receptor de la superficie de la bicapa de liposomas que estr&eacute;s inducido fueron similares en ambos casos. Tambi&eacute;n encontramos que la emisi&oacute;n de PDA de mecanismo de excitaci&oacute;n directa era &sim;2-10 veces m&aacute;s grandes que el de la respuesta basadas en el traste. Estas diferencias en las se&ntilde;ales de emisi&oacute;n fueron atribuidas a tres razones principales: interacciones no espec&iacute;ficas de e. coli y liposomas, las diferencias de tama&ntilde;o entre el analito y liposomas y una mayor concentraci&oacute;n de PDA con respetan a sulforodamina (SR-101). Hemos propuesto un modelo para explicar nuestras observaciones experimentales. Nuestros estudios fundamentales divulgados aqu&iacute; le ayudar&aacute; a mejorar nuestro conocimiento sobre las interacciones implicadas entre part&iacute;culas suaves a nivel molecular.
