Biofilms are dynamic microbial communities in which transitions between planktonic and sessile modes of growth occur interchangeably in response to different environmental cues. In the last decade, early events associated with C. albicans biofilm formation have received considerable attention. However, very little is known about C. albicans biofilm dispersion or the mechanisms and signals that trigger it. This is important because it is precisely C. albicans cells dispersed from biofilms that are the main culprits associated with candidemia and establishment of disseminated invasive disease, two of the gravest forms of candidiasis. Using a simple flow biofilm model recently developed by our group, we have performed initial investigations into the phenomenon of C. albicans biofilm dispersion, as well as the phenotypic characteristics associated with dispersed cells. Our results indicate that C. albicans biofilm dispersion is dependent on growing conditions, including carbon source and pH of the media used for biofilm development. C. albicans dispersed cells are mostly in the yeast form and display distinct phenotypic properties compared to their planktonic counterparts, including enhanced adherence, filamentation, biofilm formation and, perhaps most importantly, increased pathogenicity in a murine model of hematogenously disseminated candidiasis, thus indicating that dispersed cells are armed with a complete arsenal of "virulence factors" important for seeding and establishing new foci of infection. In addition, utilizing genetically engineered strains of C. albicans (tetO-UME6 and tetO-PES1) we demonstrate that C. albicans biofilm dispersion can be regulated by manipulating levels of expression of these key genes, further supporting the evidence for a strong link between biofilms and morphogenetic conversions at different stages of the C. albicans biofilm developmental cycle. Overall, our results offer novel and important insight into the phenomenon of C. albicans biofilm dispersion, a key part of the biofilm developmental cycle, and provide the basis for its more detailed analysis.

Les biofilms sont des dynamiques communaut&#xE9;s microbiennes dans lequel les transitions entre planctoniques et sessiles modes de croissance se produisent de fa&#xE7;on interchangeable en r&#xE9;ponse &#xE0; diff&#xE9;rents signaux de l&#x26;#39;environnement. Dans la derni&#xE8;re d&#xE9;cennie, les &#xE9;v&#xE9;nements pr&#xE9;coces associ&#xE9;s &#xE0; C. albicans la formation de biofilm ont re&#xE7;u une attention consid&#xE9;rable. Cependant, tr&#xE8;s peu est connu sur la dispersion C. albicans biofilm ou les m&#xE9;canismes et les signaux qui le d&#xE9;clenchent. Albicans Biofilm Dispersion oder die Mechanismen und die Signale, die es ausl&ouml;sen. Dies ist wichtig, denn es ist genau C. Albicans Zellen verteilt von Biofilmen, die Hauptschuldigen geschw&auml;chtes und Einrichtung verbreitet invasive Erkrankung, zwei der schwersten Formen der Candidose zugeordnet sind. Verwenden eine einfache Flussmodell Biofilm k&uuml;rzlich von unserer Fraktion, wir haben erste Untersuchungen &uuml;ber das Ph&auml;nomen von C. Albicans Biofilm Dispersion sowie die zugeordneten verteilter Zellen ph&auml;notypische Merkmale durchgef&uuml;hrt. Unsere Ergebnisse zeigen, dass C. Albicans Biofilm Dispersion Anbaubedingungen, einschlie&szlig;lich Kohlenstoffquelle und pH-Wert des Biofilms Entwicklung verwendeten Mediums abh&auml;ngig ist. C. Albicans verstreut Zellen werden haupts&auml;chlich in Form von Hefe und zeigen unterschiedliche ph&auml;notypische Eigenschaften im Vergleich zu ihren Plankton Pendants, einschlie&szlig;lich verbesserte Einhaltung, Filamentation, Biofilm-Bildung und, vielleicht am wichtigsten, erh&ouml;ht Pathogenit&auml;t in einem murinen Modell hematogenously disseminierte Candidiasis, so, dass verteilte Zellen mit ein komplettes Arsenal an &quot;Virulenzfaktoren&quot; bewaffnet sind wichtig f&uuml;r das seeding und zur Gr&uuml;ndung neuer Herde der Infektion. Dar&uuml;ber hinaus zeigen unter Verwendung gentechnisch ver&auml;nderter St&auml;mme von C. Albicans (TetO-UME6 und TetO-PES1) wir, dass C. Albicans Biofilm Dispersion geregelt werden kann, durch Manipulation der Ebenen des Ausdrucks dieser wichtigen Gene weiter die Belege f&uuml;r eine starke Verbindung zwischen Biofilmen und morphogenetische Konvertierungen in den verschiedenen Phasen des C. Albicans Biofilm-Entwicklungs-Zyklus. Insgesamt ist unsere Ergebnisse bieten neuartige und wichtige Einblicke in das Ph&auml;nomen der C. Albicans Biofilm-Dispersion, ein wichtiger Teil des Biofilms Entwicklungsst&ouml;rungen Zyklus, und bilden die Basis f&uuml;r die genauere Analyse.

Nell&#39;ultimo decennio, primi eventi associati alla formazione di biofilm di c. albicans hanno ricevuto una notevole attenzione. Tuttavia, molto poco si sa circa la c. albicans biofilm dispersione o i meccanismi e i segnali che si attivano. Questo &egrave; importante perch&eacute; &egrave; precisamente le cellule di c. albicans dislocate dal biofilm che sono i principali colpevoli associati candidemia e istituzione della diffusa malattia invasiva, due delle pi&ugrave; gravi forme di candidosi. Las biopel&#xED;culas son din&#xE1;micas comunidades microbianas en el que las transiciones entre planct&#xF3;nicos y modos s&#xE9;siles de crecimiento se producen de manera intercambiable en respuesta a diferentes est&#xED;mulos ambientales. En la &#xFA;ltima d&#xE9;cada, los primeros eventos relacionados con la formaci&#xF3;n de biopel&#xED;culas de C. albicans han recibido considerable atenci&#xF3;n. Sin embargo, se sabe muy poco sobre la dispersi&#xF3;n de C. albicans biofilm o los mecanismos y las se&#xF1;ales de que la desencadenan. Esto es importante porque es, precisamente, las c&#xE9;lulas de C. albicans dispersa de las biopel&#xED;culas, que son los principales culpables asociados con candidemia y el establecimiento de la enfermedad diseminada invasiva, dos de las formas m&#xE1;s graves de la candidiasis. El uso de un modelo de flujo de biofilm sencilla desarrollado recientemente por nuestro grupo, hemos realizado investigaciones iniciales sobre el fen&#xF3;meno de la dispersi&#xF3;n de biopel&#xED;culas de C. albicans, as&#xED; como las caracter&#xED;sticas fenot&#xED;picas asociadas a las c&#xE9;lulas dispersas. Nuestros resultados indican que C. albicans dispersi&#xF3;n biopel&#xED;cula es dependiente de las condiciones de crecimiento, incluyendo la fuente de carbono y el pH de los medios utilizados para el desarrollo del biofilm. Biofilms displayed high levels of resistance to fluconazole, and this antifungal exerted minor effects on dispersion levels. Amphotericin B proved effective in reducing viability of cells within the biofilms and dispersion, but only at high concentrations. Under flow conditions, caspofungin exhibited potent activity against biofilms and drastically reduced biofilm dispersion.

A robotic microarrayer was used to print yeast cells of C. albicans encapsulated in a collagen matrix at a volume as low as 50 nL onto surface-modified microscope slides. Upon incubation, the cells grow into fully formed "nano-biofilms". The morphological and architectural complexity of these biofilms were evaluated by scanning electron and confocal scanning laser microscopy. The extent of biofilm formation was determined using a microarray scanner from changes in fluorescence intensities due to FUN 1 metabolic processing. This staining technique was also adapted for antifungal susceptibility testing, which demonstrated that, similar to regular biofilms, cells within the on-chip biofilms displayed elevated levels of resistance against antifungal agents (fluconazole and amphotericin B). Thus, results from structural analyses and antifungal susceptibility testing indicated that despite miniaturization, these biofilms display the typical phenotypic properties associated with the biofilm mode of growth. In its final format, the C. albicans biofilm chip (CaBChip) is composed of 768 equivalent and spatially distinct nano-biofilms on a single slide; multiple chips can be printed and processed simultaneously. Compared to current methods for the formation of microbial biofilms, namely the 96-well microtiter plate model, this fungal biofilm chip has advantages in terms of miniaturization and automation, which combine to cut reagent use and analysis time, minimize labor intensive steps, and dramatically reduce assay costs. Such a chip should accelerate the antifungal drug discovery process by enabling rapid, convenient and inexpensive screening of hundreds-to-thousands of compounds simultaneously.

&#xB192;&#xC740; &#xCC98;&#xB9AC;&#xB7C9; Candida albicans biofilm &#xCE69;&#xC758; &#xAC1C;&#xBC1C;.

Hemos desarrollado una plataforma de microarrays de alta densidad de nano-biopel&iacute;culas de Candida albicans. Un microarrayer rob&oacute;tico fue utilizada para imprimir las c&eacute;lulas de la levadura de C. albicans encapsulados en una matriz de col&aacute;geno en un volumen tan bajo como 50 nL en portaobjetos de superficie modificada. Tras la incubaci&oacute;n, las c&eacute;lulas crecen en completamente formados &quot;nano-biofilms&quot;. La complejidad morfol&oacute;gica y arquitect&oacute;nica de estos biofilms se evaluaron mediante el escaneo de electrones y la microscopia confocal de barrido l&aacute;ser. El grado de formaci&oacute;n de biopel&iacute;culas se determin&oacute; utilizando un esc&aacute;ner de microarray de cambios en la intensidad de la fluorescencia debido a proceso metab&oacute;lico diversi&oacute;n 1. Esta t&eacute;cnica de tinci&oacute;n se ha adaptado tambi&eacute;n para pruebas de sensibilidad antif&uacute;ngica, que demostr&oacute; que, similar a los biofilms regulares, c&eacute;lulas dentro de las en-viruta biofilms muestran niveles elevados de resistencia contra agentes antimic&oacute;ticos (fluconazol y anfotericina B). As&iacute;, los resultados de an&aacute;lisis estructurales y pruebas de sensibilidad antif&uacute;ngica indicaron que a pesar de la miniaturizaci&oacute;n, estos biofilms Mostrar las propiedades fenot&iacute;picas t&iacute;picas asociadas con el modo de biofilm de crecimiento. En su formato final, el chip del biofilm de C. albicans (CaBChip) est&aacute; compuesto por 768 equivalente y nano-biofilms espacialmente distintos en una sola diapositiva; fichas m&uacute;ltiples se pueden imprimir y procesa simult&aacute;neamente. En comparaci&oacute;n con los m&eacute;todos actuales para la formaci&oacute;n de biofilms microbianos, es decir, el modelo de placa de 96 pocillos de microtitulaci&oacute;n, este chip de biofilm hongos tiene ventajas en t&eacute;rminos de la miniaturizaci&oacute;n y automatizaci&oacute;n, que se combinan para reducir tiempo de uso y an&aacute;lisis de reactivo, minimizar pasos intensivo de mano de obra y reducir dr&aacute;sticamente los costes de ensayo. Tal un chip debe acelerar el proceso de descubrimiento de antimic&oacute;ticos al permitir la detecci&oacute;n r&aacute;pida, conveniente y barata de cientos a miles de compuestos simult&aacute;neamente.
