Our basic understanding of how to combat fungal infections has not kept pace with the recent sharp rise in life-threatening cases found particularly among immuno-compromised individuals. Current investigations for new potential antifungal agents have focused on antimicrobial peptides, which are used as a cell-free defense mechanism in all organisms. Unfortunately, despite their high antibacterial activity, most of them are not active toward fungi, the reason of which is not clear. Here, we present a new approach to modify an antibacterial peptide, a magainin analogue, to display antifungal activity by its conjugation with lipophilic acids. This approach has the advantage of producing well-defined changes in hydrophobicity, secondary structure, and self-association. These modifications were characterized in solution at physiological concentrations using CD spectroscopy, tryptophan fluorescence, and analytical ultracentrifugation. In order of increasing hydrophobicity, the attachment to the magainin-2 analogue of (i) heptanoic acid results in a monomeric, unordered structure, (ii) undecanoic acid yields concentration-dependent oligomers of alpha helices, and (iii) palmitic acid yields concentration-independent alpha-helical monomers, a novel lipopeptide structure, which is resistant to proteolytic digestion. Membrane-lipopeptide interactions and the membrane-bound structures were studied using fluorescence and ATR-FTIR in PC/PE/PI/ergosterol (5/2.5/2.5/1, w/w) SUV, which constitute the major components of Candida albicans bilayers. A direct correlation was found between oligomerization of the lipopeptides in solution and potent antifungal activity. These results provide insight to a new approach of modulating hydrophobicity and self-assembly of antimicrobial peptides in solution, without altering the sequence of the peptidic chain. These studies also provide a general means of developing a new group of lipopeptide candidates as therapeutic agents against fungal infections.

Notre compr&eacute;hension de base de la lutte contre les infections fongiques n&#39;a pas suivi le rythme avec la r&eacute;cente flamb&eacute;e cas mortelle trouv&eacute; particuli&egrave;rement chez les individus immunod&eacute;prim&eacute;s. Enqu&ecirc;tes en cours pour les nouveaux agents antifongiques potentiels ont mis l&#39;accent sur les peptides antimicrobiens, qui sont utilis&eacute;s comme un m&eacute;canisme de d&eacute;fense acellulaire dans tous les organismes. Malheureusement, malgr&eacute; leur forte activit&eacute; antibact&eacute;rienne, la plupart d&#39;entre eux n&#39;est pas active vers les champignons, la raison qui n&#39;est ne pas clair. Nous pr&eacute;sentons ici une nouvelle approche pour modifier un peptide antibact&eacute;rien, un analogue de la magainine, pour afficher une activit&eacute; antifongique par sa conjugaison avec les acides lipophiles. Cette approche a l&#39;avantage de produire des changements bien d&eacute;finis dans l&#39;hydrophobie, structure secondaire et auto-association. Ces modifications ont &eacute;t&eacute; caract&eacute;ris&eacute;es en solution, &agrave; des concentrations physiologiques &agrave; l&#39;aide de la spectroscopie, la fluorescence du tryptophane et l&#39;ultracentrifugation analytique de CD. Par ordre croissant d&#39;hydrophobie, l&#39;attachement &agrave; l&#39;analogue de la magainine-2 de l&#39;acide heptano&iuml;que (i) se traduit par une structure monom&eacute;rique, non tri&eacute;e, acide und&eacute;cano&iuml;que (ii) les rendements des oligom&egrave;res de concentration-d&eacute;pendante des h&eacute;lices alpha et (iii) l&#39;acide palmitique donne la concentration ind&eacute;pendant monom&egrave;res alpha-h&eacute;lico&iuml;dales, une structure nouvelle lipopeptide, qui r&eacute;siste &agrave; la digestion prot&eacute;olytique. Membrane-lipopeptide les interactions et les structures membranaires ont &eacute;t&eacute; &eacute;tudi&eacute;es par fluorescence et IRTF-en PC/PE/PI/ergost&eacute;rol (5/2.5/2.5/1, p/p) SUV, qui constituent les principales composantes des bicouches de Candida albicans. Une corr&eacute;lation directe entre l&#39;oligom&eacute;risation des lipopeptides en solution et puissante activit&eacute; antifongique. Ces r&eacute;sultats fournissent un aper&ccedil;u d&#39;une nouvelle approche permettant de moduler l&#39;hydrophobie et autoassemblage de peptides antimicrobiens en solution, sans modifier la s&eacute;quence de la cha&icirc;ne peptidique. Ces &eacute;tudes fournissent &eacute;galement qu&#39;un g&eacute;n&eacute;ral signifie de mettre au point un nouveau groupe de candidats lipopeptide comme agents th&eacute;rapeutiques contre les infections fongiques.

Unser Grundverst&auml;ndnis von wie Pilzinfektionen bek&auml;mpft hat mit den j&uuml;ngsten Anstieg der lebensbedrohlichen F&auml;llen, insbesondere bei immunsupprimierten Individuen gefunden nicht Schritt gehalten. Aktuelle Untersuchungen bei potenziellen antimykotischen Wirkstoffen konzentrierten sich auf antimikrobielle Peptide, die als Abwehrmechanismus zellfreien in allen Organismen verwendet werden. Leider sind die meisten von ihnen trotz ihrer hohe antibakterielle Aktivit&auml;t, nicht aktiv gegen Pilze, die Grund davon ist nicht klar. Hier pr&auml;sentieren wir ein neues Konzept um eine antibakterielle Peptid, ein Magainin Analogon, antimykotische Aktivit&auml;t durch die Konjugation mit lipophilen S&auml;uren anzeigen zu &auml;ndern. Dieser Ansatz hat den Vorteil des Produzierens der klar definierten &Auml;nderungen in Hydrophobie, Sekund&auml;rstruktur und Self-association. Diese &Auml;nderungen waren L&ouml;sung bei physiologischen Konzentration mit CD-Spektroskopie, Tryptophan Fluoreszenz und analytische Ultrazentrifugation gekennzeichnet. Im Auftrag der zunehmenden Hydrophobie die Anlage auf dem Magainin-2-Analog (i) Heptanoic S&auml;ure ergibt ein Monomer, ungeordnete Struktur (Ii) C11H22O2 S&auml;ure ergibt Konzentration abh&auml;ngige Oligomeren von Alpha-Helices und (Iii) Palmitins&auml;ure ergibt Konzentration-unabh&auml;ngige Alpha-Kegelrad Monomere, eine neuartige Lipopeptid-Struktur, die proteolytische Verdauung resistent ist. Membran-Lipopeptid-Interaktionen und die Membrane-springen-Strukturen wurden untersucht mit Fluoreszenz und ATR-FTIR in PC/PE/PI/Ergosterin (5/2.5/2.5/1, w/w)-Gel&auml;ndewagen, die Hauptbestandteile des Candida Albicans Doppelschichten zu bilden. Eine direkte Korrelation wurde zwischen Oligomerisierung von der Lipopeptides in L&ouml;sung und potente antimykotische Aktivit&auml;t gefunden. Diese Ergebnisse liefern Einblicke in ein neues Konzept des modulierenden Hydrophobie und Selbstmontage antimikrobielle Peptide in L&ouml;sung, ohne dabei die Sequenz der Peptid-Kette. La nostra comprensione di base di come combattere le infezioni fungine non ha tenuto il passo con il recente forte aumento nei casi letali trovato particolarmente fra gli individui immunocompromessi. Indagini corrente per i nuovi potenziali agenti antifungini sono concentrati su peptidi antimicrobici, che vengono utilizzati come meccanismo di difesa cellulare gratis in tutti gli organismi. Purtroppo, nonostante la loro elevata attivit&agrave; antibatterica, la maggior parte di loro non sono attiva verso i funghi, il motivo per cui non &egrave; chiaro. Qui, vi presentiamo un nuovo approccio per modificare un peptide antibatterico, un analogo di magainin, per visualizzare l&#39;attivit&agrave; antifungina sua coniugazione con acidi lipofili. Questo approccio ha il vantaggio di produrre cambiamenti ben definiti di idrofobicit&agrave;, struttura secondaria e moltiplicava. In ordine crescente di idrofobicit&agrave;, l&#39;attaccamento per l&#39;analogo di magainin-2 dell&#39;acido eptanoico (i) risultati in una struttura monomerica, non ordinata, (ii) undecanoic acid rendimenti oligomeri di concentrazione-dipendente di alfa eliche e (iii) acido palmitico rese indipendente dalla concentrazione di monomeri alfa-elicoidale, una struttura lipopeptide romanzo, che &egrave; resistente alla digestione proteolitica. Membrana-lipopeptide interazioni e le strutture di membrana-limiti sono state studiate mediante fluorescenza e ATR-FTIR in SUV PC/PE/PI/ergosterolo (5/2.5/2.5/1, w/w), che costituiscono i componenti principali del bilayer di Candida albicans. Tra oligomerizzazione di lipopeptides in soluzione e potente attivit&agrave; antimicotica &egrave; stata trovata una correlazione diretta. Questi risultati forniscono informazioni a un nuovo approccio di modulante idrofobicit&agrave; e auto-assemblaggio di peptidi antimicrobici nella soluzione, senza alterare la sequenza della catena peptidici. Nuestra comprensi&oacute;n b&aacute;sica de c&oacute;mo combatir las infecciones mic&oacute;ticas no ha seguido el ritmo con el fuerte aumento reciente en casos mortales, especialmente entre individuos inmunodeficientes. Las investigaciones actuales para nuevos antif&uacute;ngicos potenciales se han centrado en p&eacute;ptidos antimicrobianos, que se utilizan como un mecanismo de defensa celular libre en todos los organismos. Desafortunadamente, a pesar de su alta actividad antibacteriana, la mayor&iacute;a de ellos no son activa hacia hongos, la raz&oacute;n de que no est&aacute; claro. Aqu&iacute;, presentamos un nuevo enfoque para modificar un p&eacute;ptido antibacteriano, un an&aacute;logo de la magainin, para mostrar la actividad antif&uacute;ngica por su conjugaci&oacute;n con &aacute;cidos lipof&iacute;licos. Este enfoque tiene la ventaja de producir cambios bien definidos de hidrofobicidad, estructura secundaria y autoasociaci&oacute;n. Estas modificaciones se caracterizaron en soluci&oacute;n a concentraciones fisiol&oacute;gicas usando Espectroscopia CD, fluorescencia de tript&oacute;fano y ultracentrifugaci&oacute;n. En orden creciente de hidrofobicidad, el accesorio para el an&aacute;logo de magainin-2 del &aacute;cido heptanoico (i) da lugar a una estructura monom&eacute;rica, desordenada, &Aacute;cido undecanoico (ii) rinde olig&oacute;meros dependiente de la concentraci&oacute;n de alfa h&eacute;lices, y (iii) &aacute;cido palm&iacute;tico produce mon&oacute;meros alfa helicoidales de concentraci&oacute;n-independiente, una estructura Lipop&eacute;ptido novela, que es resistente a la digesti&oacute;n proteol&iacute;tica. Membrana-Lipop&eacute;ptido interacciones y las estructuras de membrana-limitan se estudiaron mediante fluorescencia y ATR-FTIR en SUV de PC/PE/PI/ergosterol (5/2.5/2.5/1, w/w), que constituyen los principales componentes de bicapas de Candida albicans. The interaction of many lytic cationic antimicrobial peptides with their target cells involves electrostatic interactions, hydrophobic effects, and the formation of amphipathic secondary structures, such as alpha helices or beta sheets. We have shown in previous studies that incorporating approximately 30%d-amino acids into a short alpha helical lytic peptide composed of leucine and lysine preserved the antimicrobial activity of the parent peptide, while the hemolytic activity was abolished. However, the mechanisms underlying the unique structural features induced by incorporating d-amino acids that enable short diastereomeric antimicrobial peptides to preserve membrane binding and lytic capabilities remain unknown. In this study, we analyze in detail the structures of a model amphipathic alpha helical cytolytic peptide KLLLKWLL KLLK-NH2 and its diastereomeric analog and their interactions with zwitterionic and negatively charged membranes. Calculations based on high-resolution NMR experiments in dodecylphosphocholine (DPCho) and sodium dodecyl sulfate (SDS) micelles yield three-dimensional structures of both peptides. Structural analysis reveals that the peptides have an amphipathic organization within both membranes. Specifically, the alpha helical structure of the L-type peptide causes orientation of the hydrophobic and polar amino acids onto separate surfaces, allowing interactions with both the hydrophobic core of the membrane and the polar head group region. Significantly, despite the absence of helical structures, the diastereomer peptide analog exhibits similar segregation between the polar and hydrophobic surfaces. Further insight into the membrane-binding properties of the peptides and their depth of penetration into the lipid bilayer has been obtained through tryptophan quenching experiments using brominated phospholipids and the recently developed lipid/polydiacetylene (PDA) colorimetric assay. The combined NMR, FTIR, fluorescence, and colorimetric studies shed light on the importance of segregation between the positive charges and the hydrophobic moieties on opposite surfaces within the peptides for facilitating membrane binding and disruption, compared to the formation of alpha helical or beta sheet structures.

Les calculs bas&eacute;s sur des exp&eacute;riences de RMN &agrave; haute r&eacute;solution dans la dod&eacute;cylphosphocholine (DPCho) et les micelles de sodium dod&eacute;cyl sulfate (SDS) produisent des structures tridimensionnelles des deux peptides. Analyse structurale r&eacute;v&egrave;le que les peptides ont une organisation amphipathiques dans les deux membranes. Berechnungen basierend auf hochaufl&ouml;sende NMR-Experimente in Dodecylphosphocholine (DPCho) und Natrium Natriumdodecylsulfat-Sulfat (SDS) Micellen Ausbeute dreidimensionale Strukturen der beiden Peptide. Strukturelle Analyse zeigt, dass die Peptide AMPHIPHILE Organisationen mit beiden Membranen haben. Insbesondere bewirkt die alpha spiralf&ouml;rmige Struktur des Peptids Typ-L Orientierung der hydrophoben und polaren Aminos&auml;uren auf eigenen Fl&auml;chen, so dass Interaktionen mit beiden hydrophoben Kern der Membran und der polaren Kopf Gruppe-Region. Deutlich, trotz des Fehlens von spiralf&ouml;rmigen Strukturen Exponate das Diastereomer Peptid analog &auml;hnliche Trennung zwischen den polaren und hydrophoben Oberfl&auml;chen. Weitere Einblicke in die Membran-Bindungseigenschaften der Peptide und ihre Tiefe des Eindringens in die Lipid-Bilayer wurde durch Tryptophan abschrecken Experimente mit bromhaltigen Phospholipide und kolorimetrische Assays k&uuml;rzlich entwickelten Lipid/Polydiacetylene (PDA) erteilt. Los c&aacute;lculos basados en alta resoluci&oacute;n NMR experimentos en dodecylphosphocholine (DPCho) y rendimiento de micelas de sodio dodecil sulfato (SDS) estructuras tridimensionales de ambos p&eacute;ptidos. An&aacute;lisis estructural revelan que los p&eacute;ptidos tienen una organizaci&oacute;n anfip&aacute;ticos dentro de ambas membranas. Espec&iacute;ficamente, la estructura helicoidal alfa del p&eacute;ptido tipo L provoca la orientaci&oacute;n de los amino&aacute;cidos hidrof&oacute;bicos y polares en superficies separadas, permitiendo interacciones con ambos el n&uacute;cleo hidrof&oacute;bico de la membrana y la regi&oacute;n del grupo de cabeza polar. Significativamente, a pesar de la ausencia de estructuras helicoidales, el p&eacute;ptido Diastereois&oacute;mero anal&oacute;gico exhibe segregaci&oacute;n similar entre las superficies hidrof&oacute;bicas y polares. M&aacute;s informaci&oacute;n sobre las propiedades de uni&oacute;n a la membrana de los p&eacute;ptidos y la profundidad de penetraci&oacute;n en la bicapa lip&iacute;dica se ha obtenido a trav&eacute;s de tript&oacute;fano Temple experimentos utilizando fosfol&iacute;pidos bromados y el ensayo colorim&eacute;trico recientemente desarrollados l&iacute;pido/polydiacetylene (PDA). El combinado NMR, FTIR, fluorescencia y estudios colorim&eacute;tricos arrojar luz sobre la importancia de la segregaci&oacute;n entre las cargas positivas y los bandos hidrof&oacute;bicos en superficies opuestas dentro de los p&eacute;ptidos para facilitar la Uni&oacute;n de la membrana y la interrupci&oacute;n, compararon con la formaci&oacute;n de helicoidal de alfa o beta las estructuras de la hoja.

The dramatically increased frequency of opportunistic fungal infections has prompted research to diversify the arsenal of antifungal agents. Antimicrobial peptides constitute a promising family for future antibiotics with a new mode of action. However, only a few are effective against fungal pathogens because of their ability to self-assemble. Recently, we showed that the conjugation of fatty acids to the potent antibacterial peptide magainin endowed it with antifungal activity concomitant with an increase in its oligomeric state in solution. To investigate whether a high potency of the parental peptide is prerequisite for antifungal activity, we conjugated undecanoic acid (UA) and palmitic acid (PA) to inactive diastereomers of magainin containing four d-amino acids ([D]-4-magainin), as well as to a weakly active diastereomeric lytic peptide containing Lys and Leu ([D]-K(5)L(7)). All lipopeptides gained potent activity toward Cryptococcus neoformans. Most importantly, [D]-K(5)L(7)-UA was highly potent against all microorganisms tested, including bacteria, yeast, and opportunistic fungi. All lipopeptides increased the permeability of Escherichia coli spheroplasts and intact C. neoformans, as well as their corresponding membranes, phosphatidylethanol (PE)/phosphatidylglycerol (PG) and phosphatidylcholine (PC)/PE/phosphatidylinositol (PI)/ergosterol, respectively. The extent of membrane-permeating activity correlated with their biological function, suggesting that the plasma membrane was one of their major targets. Circular dichroism (CD) and attenuated total reflectance Fourier transform infrared (ATR-FTIR) spectroscopy revealed that their mode of oligomerization in solution, structure, and organization in membranes have important roles regarding their antibacterial and antifungal activities. Together with the advantage of using diastereomers versus all l-amino acid peptides, this study paves the way to the design of a new group of potent antifungal peptides urgently needed to combat opportunistic fungal infection.

Am wichtigsten ist, [D]-K (5) L 7-UA war hochwirksames gegen alle Mikroorganismen getestet, wie Bakterien, Hefe und opportunistische Pilze. Alle Lipopeptides erh&ouml;ht die Durchl&auml;ssigkeit der Escherichia coli Spheroplasts sowie intakte C. Neoformans und ihre entsprechenden Membranen, Phosphatidylethanol (PE) / Phosphatidylglycerol (PG) und Phosphatidylcholin (PC) / PE/Phosphatidylinositol (PI) / Ergosterin, beziehungsweise. Das Ausma&szlig; der Membran durchdringt Aktivit&auml;t korreliert mit ihrer biologischen Funktion, was darauf hindeutet, dass die Plasmamembran einer ihrer wichtigsten Ziele war. We report on the synthesis, biological function, and a plausible mode of action of a new group of lipopeptides with potent antifungal and antibacterial activities. These lipopeptides are derived from positively charged peptides containing d- and l-amino acids (diastereomers) that are palmitoylated (PA) at their N terminus. The peptides investigated have the sequence K(4)X(7)W, where X designates Gly, Ala, Val, or Leu (designated d-X peptides). The data revealed that PA-d-G and PA-d-A gained potent antibacterial and antifungal activity despite the fact that both parental peptides were completely devoid of any activity toward microorganisms and model phospholipid membranes. In contrast, PA-d-L lost the potent antibacterial activity of the parental peptide but gained and preserved partial antifungal activity. Interestingly, both d-V and its palmitoylated analog were inactive toward bacteria, and only the palmitoylated peptide was highly potent toward yeast. Both PA-d-L and PA-d-V lipopeptides were also endowed with hemolytic activity. Mode of action studies were performed by using tryptophan fluorescence and attenuated total reflectance Fourier transform infrared and circular dichroism spectroscopy as well as transmembrane depolarization assays with bacteria and fungi. The data suggest that the lipopeptides act by increasing the permeability of the cell membrane and that differences in their potency and target specificity are the result of differences in their oligomeric state and ability to dissociate and insert into the cytoplasmic membrane. These results provide insight regarding a new approach of modulating hydrophobicity and the self-assembly of non-membrane interacting peptides in order to endow them with both antibacterial and antifungal activities urgently needed to combat bacterial and fungal infections.

Diese Lipopeptides sind aus positiv geladenen Peptide mit d - und l-Aminos&auml;uren (Diastereomere), die Palmitoylated (PA) ihre N-Terminus sind abgeleitet. Die Peptide, die untersucht haben die Sequenz K (4) X (7) W, wobei X Gly, Ala, Val oder Leu (bezeichneten d-X-Peptide) bezeichnet. I peptidi indagati hanno la sequenza K (4) X (7) W, dove X indica Gly, Ala, Val o Leu (peptidi designati d-X). I dati hanno rivelato che PA-d-G e PA-d-A guadagnato potente attivit&agrave; antibatterica e antimicotica, nonostante il fatto che entrambi peptidi parentale erano completamente privo di qualsiasi attivit&agrave; verso microrganismi e modello fosfolipide membrane. Divulgamos sobre la s&iacute;ntesis, funci&oacute;n biol&oacute;gica y un modo plausible de acci&oacute;n de un nuevo grupo de lipop&eacute;ptidos con potentes actividades antihongos y antibacterias. Estos lipop&eacute;ptidos derivan de p&eacute;ptidos cargados positivamente que contiene d - y l-amino&aacute;cidos (diastereois&oacute;meros) que son palmitoylated (PA) en su extremo N terminal. Los p&eacute;ptidos investigados tienen la secuencia K (4) X (7) W, donde X se&ntilde;ala Gly, Ala, Val o Leu (designados p&eacute;ptidos de d-X). Los datos revelaron que el PA-d-G y PA-d-A ganaron potente actividad antibacteriana y antif&uacute;ngica, pese a que ambos padres p&eacute;ptidos eran totalmente desprovisto de cualquier actividad hacia microorganismos y membranas de fosfol&iacute;pidos de modelo. Por el contrario, PA-d-L perdi&oacute; la potente actividad antibacteriana del p&eacute;ptido parental pero gan&oacute; y hab&iacute;a conservada parcial actividad antif&uacute;ngica. Curiosamente, tanto d-V y su palmitoylated anal&oacute;gico eran inactivos hacia las bacterias, y s&oacute;lo el p&eacute;ptido palmitoylated era altamente potente hacia la levadura. Host-defense cationic antimicrobial peptides ( approximately 12-50 aa long) play an essential protective role in the innate immune system of all organisms. Lipopeptides, however, are produced only in bacteria and fungi during cultivation, and they are composed of specific lipophilic moieties attached to anionic peptides (six to seven amino acids). Here we report the following. (i) The attachment of an aliphatic chain to otherwise inert, cationic D,L tetrapeptides endows them with potent activity against various microorganisms including antibiotic resistance strains. (ii) Cell specificity is determined by the sequence of the short peptidic chain and the length of the aliphatic moiety. (iii) Despite the fact that the peptidic chains are very short, their mode of action involves permeation and disintegration of membranes, similar to that of many long antimicrobial peptides. Besides adding important information on the parameters necessary for host-defense lipopeptides to kill microorganisms, the simple composition of these lipopeptides and their diverse specificities should make them economically available, innate immunity-mimicking antimicrobial and antifungal compounds for various applications.

Endogenous peptide antibiotics (termed also host-defense or antimicrobial peptides) are known as evolutionarily old components of innate immunity. They were found initially in invertebrates, but later on also in vertebrates, including humans. This secondary, chemical immune system provides organisms with a repertoire of small peptides that act against invasion (for both offensive and defensive purposes) by occasional and obligate pathogens. Each antimicrobial peptide has a broad but not identical spectrum of antimicrobial activity, predominantly against bacteria, providing the host maximum coverage against a rather broad spectrum of microbial organisms. Many of these peptides interact with the target cell membranes and increase their permeability, which results in cell lysis. A second important family includes lipopeptides. They are produced in bacteria and fungi during cultivation on various carbon sources, and possess a strong antifungal activity. Unfortunately, native lipopeptides are non-cell selective and therefore extremely toxic to mammalian cells. Whereas extensive studies have emerged on the requirements for a peptide to be antibacterial, very little is known concerning the parameters that contribute to antifungal activity. This review summarizes recent studies aimed to understand how antimicrobial peptides and lipopeptides select their target cell. This includes a new group of lipopeptides highly potent against pathogenic fungi and yeast. They are composed of inert cationic peptides conjugated to aliphatic acids with different lengths. Deep understanding of the molecular mechanisms underlying the differential cells specificity of these families of host defense molecule is required to meet the challenges imposed by the life-threatening infections.

Antibi&oacute;ticos de p&eacute;ptidos end&oacute;genos (llamados tambi&eacute;n p&eacute;ptidos de defensa del hu&eacute;sped o antimicrobiano) se conocen como evolucionariamente antiguos componentes de la inmunidad innata. Se encontraron inicialmente en invertebrados, pero m&aacute;s tarde tambi&eacute;n en los vertebrados, incluidos a los seres humanos. Este sistema inmune secundaria, qu&iacute;mica proporciona organismos con un repertorio de peque&ntilde;os p&eacute;ptidos que act&uacute;an contra la invasi&oacute;n (con fines defensivos y ofensivos) por pat&oacute;genos ocasionales y obligados.
