Silica optical microcavity sensors show great promise in the kinetic evaluation of binding pairs, fundamental in understanding biomolecular interactions. Here, we develop and demonstrate a novel platform, based on bioconjugated silica microsphere resonators, to study the binding kinetics of the biotin-streptavidin system. We characterize the optical performance, verify the covalent attachment of biotin to the surface, and perform streptavidin detection experiments. We perform preliminary kinetic analysis of the detection data which shows the potential of whispering gallery mode resonators in the determination of the dissociation constant of the binding pair, which is in good agreement with previously published values.

Capteurs de silice microcavit&#xE9; optique sont tr&#xE8;s prometteurs dans l&#x26;#39;&#xE9;valuation cin&#xE9;tique de paires de liaison, fondamentale dans la compr&#xE9;hension des interactions biomol&#xE9;culaires. Ici, nous d&#xE9;velopper et de d&#xE9;montrer une nouvelle plateforme, bas&#xE9;e sur bioconjugated r&#xE9;sonateurs microsph&#xE8;res de silice, d&#x26;#39;&#xE9;tudier la cin&#xE9;tique de liaison du syst&#xE8;me biotine-streptavidine. On caract&#xE9;rise la performance optique, v&#xE9;rifier la fixation covalente de la biotine &#xE0; la surface, et r&#xE9;aliser des exp&#xE9;riences de d&#xE9;tection streptavidine. Nous effectuons une analyse pr&#xE9;liminaire de cin&#xE9;tique des donn&#xE9;es de d&#xE9;tection qui montre le potentiel de chuchoter r&#xE9;sonateurs en mode galerie dans la d&#xE9;termination de la constante de dissociation de la paire de liaison, qui est en bon accord avec les valeurs publi&#xE9;es ant&#xE9;rieurement.

Kiesels&auml;ure-optische Microcavity-Sensoren zeigen viel versprechend in der kinetischen Auswertung der Bindung-Paare, grundlegende Biomolekulare Wechselwirkungen zu verstehen. Hier entwickeln wir und zeigen eine neuartige Plattform, basierend auf Bioconjugated Kiesels&auml;ure Microsphere Resonatoren, die verbindliche Kinetik der Biotin-Streptavidin-Systems zu studieren. Wir charakterisieren die optische Leistung, &uuml;berpr&uuml;fen die kovalente Anlage von Biotin an die Oberfl&auml;che und Streptavidin Erkennung Experimente durchf&uuml;hren. Wir f&uuml;hren erste kinetische Analyse der Erkennungsdaten, die das Potenzial der Fl&uuml;sternde Galerie Modus Resonatoren bei der Ermittlung des der Dissoziation des Paares Bindung, Konstante zeigt, die in guter &Uuml;bereinstimmung mit den zuvor ver&ouml;ffentlichten Werten ist.

Sensores &#xF3;pticos de s&#xED;lice microcavidad muestran una gran promesa en la evaluaci&#xF3;n cin&#xE9;tica de los pares de uni&#xF3;n, fundamental en la comprensi&#xF3;n de las interacciones biomoleculares. En este sentido, desarrollar y demostrar una novedosa plataforma, sobre la base de microesferas de s&#xED;lice bioconjugated resonadores, para estudiar la cin&#xE9;tica de uni&#xF3;n del sistema de biotina-estreptavidina. Nos caracterizar el rendimiento &#xF3;ptico, verificar la uni&#xF3;n covalente de biotina a la superficie, y realizar experimentos de detecci&#xF3;n estreptavidina. Llevamos a cabo an&#xE1;lisis preliminar cin&#xE9;tica de los datos de detecci&#xF3;n que muestra el potencial de susurrante resonadores galer&#xED;a modo en la determinaci&#xF3;n de la constante de disociaci&#xF3;n del par de uni&#xF3;n, que est&#xE1; en buen acuerdo con los valores publicados anteriormente.
