Apolipoprotein A-V (apoA-V), the newest member of the plasma apolipoprotein family, was recently discovered by comparison of the mouse and human genomes. Studies in rodents and population surveys of human apoA-V polymorphisms have noted a strong effect of apoA-V on plasma triglyceride levels. Toward the elucidation of the biologic function of apoA-V, we used spectroscopic and surface chemistry techniques to probe its structure and interfacial activity. Computer-assisted sequence analysis of apoA-V predicts that it is very hydrophobic, contains a significant amount of alpha-helical secondary structure, and probably is composed of discrete structural regions with varying degrees of lipid affinity. Fluorescence spectroscopy of recombinant human apoA-V provided evidence of tertiary folding, and light scattering studies indicated that apoA-V transforms dimyristoylphosphatidylcholine vesicles into discoidal complexes with an efficiency similar to that of apoA-I. Surface chemistry techniques revealed that apoA-V displays high affinity, low elasticity, and slow binding kinetics at hydrophobic interfaces, properties we propose may retard triglyceride-rich particle assembly. Metabolic labeling and immunofluorescence studies of COS-1 cells transfected with human apoA-V demonstrated that apoA-V is poorly secreted, remains associated with the endoplasmic reticulum, and does not traffic to the Golgi. Given that overexpression of the apoA-V gene lowers plasma triglycerides in mice, these data together suggest that apoA-V may function intracellularly to modulate hepatic VLDL synthesis and/or secretion.

To examine the role of apolipoprotein A-IV (apoA-IV) in the intracellular trafficking and secretion of apoB, COS cells were cotransfected with microsomal triglyceride transfer protein (MTP), apoB-41 (amino terminal 41% of apoB), and either native apoA-IV or apoA-IV modified with the carboxy-terminal endoplasmic reticulum (ER) retention signal, KDEL (apoA-IV-KDEL). As expected, apoA-IV-KDEL was inefficiently secreted relative to native apoA-IV. Coexpression of apoB-41 with apoA-IV-KDEL reduced the secretion of apoB-41 by approximately 80%. The apoA-IV-KDEL effect was specific, as neither KDEL-modified forms of human serum albumin or apoA-I affected apoB-41 secretion. Similar results were observed in McA-RH7777 rat hepatoma cells, which express endogenous MTP. The full inhibitory effect of apoA-IV-KDEL on apoB secretion was observed only for forms of apoB containing a minimum of the amino-terminal 25% of the protein (apoB-25). However, apoA-IV-KDEL inhibited the secretion of both lipid-associated and lipid-poor forms of apoB-25. Dual-label immunofluorescence microscopy of cells transfected with native apoA-IV and apoB-25 revealed that both apolipoproteins were localized to the ER and Golgi, as expected. However, when apoA-IV-KDEL was cotransfected with apoB-25, both proteins localized primarily to the ER. These data suggest that apoA-IV may physically interact with apoB in the secretory pathway, perhaps reflecting a role in modulating the process of triglyceride-rich lipoprotein assembly and secretion.

Para examinar el papel de apolipoprote&iacute;na A-IV (apoA-IV) en el tr&aacute;fico intracelular y la secreci&oacute;n de apoB, c&eacute;lulas COS fueron cotransfected con prote&iacute;na de transferencia de triglic&eacute;ridos micros&oacute;micos (MTP), apoB-41 (amino terminal 41% de apoB), apoA nativo-IV o apoA-IV modificado con la se&ntilde;al de retenci&oacute;n de carboxi-terminal ret&iacute;culo endopl&aacute;smico (ER), KDEL (apoA-IV-KDEL). Como era de esperar, apoA-IV-KDEL ineficazmente fue secretada en relaci&oacute;n con los nativo apoA-IV. Coexpresi&oacute;n de apoB-41 con apoA-IV-KDEL reduce la secreci&oacute;n de apoB-41 por aproximadamente el 80%. D. (2002) j Biol Chem. 277, 31929-31937). Per studiare il meccanismo di questo miglioramento, IPEC-1 cellule erano transfected con un sistema di espressione regolabile tetraciclina (Tet-On). In cellule incubate con acido oleico, &egrave; stata osservata una relazione di risposta dose tra concentrazione medio doxiciclina e basolaterale apoA-IV e secrezione di TG. Utilizzando lo stesso sistema di espressione, full-length umano apoA-IV (376 aminoacidi); un &quot;maiale-like&quot; umano apoA-IV, manca il C-terminale EQQQ ripete (361 amminoacidi); e un &quot;pollo-come&quot; apoA-IV, ulteriormente troncato a 343 gli amminoacidi, sono stati espressi nelle cellule IPEC-1. Concludiamo che sovraespressione della suina apoA-IV migliora la secrezione basolaterale TG in modo dose-dipendente aumentando le dimensioni delle lipoproteine secrete. Intestinal apolipoprotein A-IV expression is highly regulated by dietary lipid in newborn swine, suggesting a role in lipid absorption. Constitutive overexpression of apoA-IV in newborn swine enterocytes enhances basolateral secretion of triacylglycerol (TG) in TG-rich lipoproteins 4.9-fold (Lu, S., Yao, Y., Meng, S., Cheng, X., and Black, D. D. (2002) J. Biol. Chem. 277, 31929-31937). To investigate the mechanism of this enhancement, IPEC-1 cells were transfected with a tetracycline-regulatable expression system (Tet-On). In cells incubated with oleic acid, a dose response relationship was observed between medium doxycycline concentration and basolateral apoA-IV and TG secretion. Similarly regulated expression of apoA-I did not enhance lipid secretion. The mean diameter of TG-rich lipoproteins secreted from doxycycline-treated cells was larger than from untreated cells (87.0 nm versus 53.4 nm). Basolateral apoB secretion decreased. Using the same expression system, full-length human apoA-IV (376 amino acids); a "pig-like" human apoA-IV, lacking the C-terminal EQQQ repeats (361 amino acids); and a "chicken-like" apoA-IV, further truncated to 343 amino acids, were expressed in IPEC-1 cells. With increasing protein secretion, cells expressing the full-length human apoA-IV displayed a 2-fold increase in TG secretion; in sharp contrast, cells expressing the pig-like human apoA-IV displayed a 25-fold increase in TG secretion and a 27-fold increase in lipoprotein diameter. When human apoA-IV was further truncated to yield a chicken-like protein, TG secretion was inhibited. We conclude that overexpression of swine apoA-IV enhances basolateral TG secretion in a dose-dependent manner by increasing the size of secreted lipoproteins. These data suggest that the region in the human apoA-IV protein from residues 344 to 354 is critical to its ability to enhance lipid secretion, perhaps by enabling the packaging of additional core TG into chylomicron particles. The EQQQ-rich region may play an inhibitory or modulatory role in chylomicron packaging in humans.

Intestinal apolipoprotein expresi&oacute;n A-IV est&aacute; altamente regulada por l&iacute;pidos diet&eacute;ticos en los cerdos reci&eacute;n nacidos, sugiriendo un papel en la absorci&oacute;n de los l&iacute;pidos. Sobreexpresi&oacute;n constitutiva de apoA-IV en enterocitos de cerdos reci&eacute;n nacidos mejora secreci&oacute;n basolateral de triacilglicerol (TG) en lipoprote&iacute;nas ricas en TG 4.9-fold (Lu, S., Yao, Y., Meng, S., Cheng, x. y negro, D. D. (2002) J. Biol Chem 277, 31929-31937). Para investigar el mecanismo de esta mejora, las c&eacute;lulas IPEC-1 fueron transfectadas con un sistema de expresi&oacute;n tetraciclina-regulable (Tet-On). In a previous study we demonstrated a requirement for activation of mTORC1 in the stimulation of eIF2Bepsilon mRNA translation in skeletal muscle in response to resistance exercise. Although that study established the necessity of mTORC1 activation, the experimental model used did not lend itself readily to address the question of whether or not mTORC1 activation was sufficient to produce the response. Therefore, the present study was designed to address the sufficiency of mTORC1 activation, using cultures of Rat2 fibroblasts in which mTORC1 signaling was repressed by serum/leucine-depletion and stimulated by repletion of leucine and/or IGF-1. Repletion with leucine and IGF-1 caused a shift of eIF2Bepsilon mRNA into actively translating polysomes and a stimulation of new eIF2Bepsilon protein synthesis, but had no effect on mRNAs encoding the other four eIF2B subunits. Stimulation of eIF2Bepsilon translation was reversed by pre-treatment with the mTORC1 inhibitor rapamycin. Exogenous overexpression of FLAG-Rheb, a proximal activator of mTORC1, also caused a re-distribution of eIF2Bepsilon mRNA into polysomes and a stimulation of eIF2Bepsilon protein synthesis. The stimulation of eIF2Bepsilon mRNA translation occurred in the absence of any effect on eIF2Bepsilon mRNA abundance. RNAi-mediated knockdown of eIF2Bepsilon resulted in reduced cellular proliferation, a result that phenocopied the known cytostatic effect of mTORC1 repression. Overall the results demonstrate that activation of mTORC1 is both necessary and sufficient to stimulate eIF2Bepsilon mRNA translation and that this response may represent a novel mechanism through which mTORC1 can affect mRNA translation initiation, rates of protein synthesis, and cellular growth/proliferation.

En un estudio anterior hemos demostrado un requisito para la activaci&oacute;n de mTORC1 en la estimulaci&oacute;n de la traducci&oacute;n de ARNm de eIF2Bepsilon en m&uacute;sculo esquel&eacute;tico en respuesta al ejercicio de resistencia. Aunque ese estudio estableci&oacute; la necesidad de la activaci&oacute;n de la mTORC1, el modelo experimental utilizado no presta f&aacute;cilmente para abordar la cuesti&oacute;n de si o no la activaci&oacute;n de mTORC1 fue suficiente para producir la respuesta. Por lo tanto, el presente estudio fue dise&ntilde;ado para hacer frente a la suficiencia de la activaci&oacute;n de mTORC1, en cultivos de fibroblastos de Rat2 que mTORC1 se&ntilde;alizaci&oacute;n fue reprimida por suero/leucina-agotamiento y estimulada por la repleci&oacute;n de leucina o IGF-1. Repleci&oacute;n con leucina y IGF-1 caus&oacute; un cambio de eIF2Bepsilon mRNA en traducir activamente polisomas y una estimulaci&oacute;n de la s&iacute;ntesis de prote&iacute;nas de eIF2Bepsilon nuevo, pero no tuvo efecto sobre mRNAs que codifican las subunidades de eIF2B otras cuatro. Estimulaci&oacute;n de la traducci&oacute;n de eIF2Bepsilon fue invertida por el tratamiento previo con el mTORC1 inhibidor de la rapamicina. Ex&oacute;gena sobreexpresi&oacute;n de bandera-Rheb, un activador proximal de mTORC1, tambi&eacute;n caus&oacute; una redistribuci&oacute;n del mRNA de la eIF2Bepsilon en polisomas y una estimulaci&oacute;n de la s&iacute;ntesis de prote&iacute;nas eIF2Bepsilon. La estimulaci&oacute;n de la traducci&oacute;n de ARNm de eIF2Bepsilon ocurri&oacute; en ausencia de cualquier efecto sobre la abundancia de ARNm de eIF2Bepsilon. Ca&iacute;da de silenciaci&oacute;n de eIF2Bepsilon dio lugar a la proliferaci&oacute;n celular reducida, un resultado que phenocopied el conocido efecto citost&aacute;tico de represi&oacute;n mTORC1. En general, los resultados demuestran que la activaci&oacute;n de mTORC1 es necesario y suficiente para estimular la traducci&oacute;n del mRNA de eIF2Bepsilon y que esta respuesta puede representar un nuevo mecanismo a trav&eacute;s del cual mTORC1 puede afectar iniciaci&oacute;n de traducci&oacute;n de mRNA, tasas de crecimiento/proliferaci&oacute;n celular y la s&iacute;ntesis de prote&iacute;nas.

Activaci&oacute;n de la blanco mam&iacute;fero de rapamicina complejo 1 es necesario y suficiente para estimular la traducci&oacute;n de ARNm eucari&oacute;tico de iniciaci&oacute;n factor 2Bvarepsilon y s&iacute;ntesis de prote&iacute;nas.

Eukaryotic initiation factor 2B (eIF2B), a five-subunit guanine nucleotide exchange factor, plays a key role in the regulation of mRNA translation. Expression of its epsilon-subunit is specifically up-regulated in certain conditions associated with increased cell growth. Therefore, the purpose of the present study was to examine the effect of repressing eIF2Bepsilon expression on growth rate, protein synthesis, and other characteristics of two tumorigenic cell lines that display up-regulated expression of the epsilon-subunit. Experiments were designed to compare spontaneously transformed fibroblasts to transformed mouse embryonic fibroblasts infected with a lentivirus containing a short hairpin RNA directed against eIF2Bepsilon. Cells expressing the short hairpin RNA displayed a reduction in eIF2Bepsilon abundance to 30% of the value observed in uninfected transformed mouse embryonic fibroblasts, with no change in the expression of any of the other four subunits. The repression of eIF2Bepsilon expression was accompanied by reductions in guanine nucleotide exchange factor activity and global rates of protein synthesis. Moreover, repressed eIF2Bepsilon expression led to marked reductions in cell growth rate in culture, colony formation in soft agar, and tumor progression in nude mice. Similar results were obtained in MCF-7 human breast cancer cells in which eIF2Bepsilon expression was repressed through transient transfection with a small interfering RNA directed against the epsilon-subunit. Overall, the results support a role for eIF2Bepsilon in the regulation of cell growth and suggest that it might represent a therapeutic target for the treatment of human cancer.

Eucaryotes initiation factor 2 b (eIF2B), un facteur d&#39;&eacute;change de nucl&eacute;otide de guanine cinq sous-unit&eacute;s, joue un r&ocirc;le cl&eacute; dans la r&eacute;gulation de la traduction de l&#39;ARNm. Expression de la sous-unit&eacute; epsilon est r&eacute;gul&eacute;e sp&eacute;cifiquement dans certaines conditions associ&eacute;es &agrave; la croissance cellulaire accrue. Ausdruck der Epsilon-Untereinheit ist spezifisch oben-geregelt in bestimmten Bedingungen im Zusammenhang mit erh&ouml;hten Zellwachstum. Eucari&oacute;tico de iniciaci&oacute;n factor 2B (eIF2B), un factor de cambio cinco-subunidad de guanina nucle&oacute;tido, desempe&ntilde;a un papel clave en la regulaci&oacute;n de la traducci&oacute;n del mRNA. Expresi&oacute;n de su &eacute;psilon-subunidad est&aacute; espec&iacute;ficamente regulado, hasta en ciertas condiciones asociadas con el crecimiento de la c&eacute;lula mayor. Por tanto, el prop&oacute;sito del presente estudio fue analizar el efecto de reprimir la expresi&oacute;n de eIF2Bepsilon en la tasa de crecimiento, la s&iacute;ntesis de prote&iacute;nas y otras caracter&iacute;sticas de dos l&iacute;neas de celulares tumor&iacute;geno que muestran expresi&oacute;n regulada por arriba de la &eacute;psilon-subunidad. Experimentos fueron dise&ntilde;ados para comparar fibroblastos espont&aacute;neamente transformados a fibroblastos embrionarias de rat&oacute;n transformada infectados con un lentivirus que contiene un short hairpin RNA dirigida contra eIF2Bepsilon. Las c&eacute;lulas que expresan el short hairpin RNA muestran una reducci&oacute;n en la abundancia de eIF2Bepsilon al 30% del valor observado en fibroblastos embrionarios de rat&oacute;n transformada no infectada, sin cambios en la expresi&oacute;n de cualquiera de las otras cuatro subunidades. Although the evidence linking apoA-IV expression and triglyceride (TG)-rich lipoprotein assembly and secretion is compelling, the intracellular mechanisms by which apoA-IV could modulate these processes remain poorly understood. We therefore examined the functional impact of apoA-IV expression on endogenous apoB, TG, and VLDL secretion in stably transfected McA-RH7777 rat hepatoma cells. Expression of apoA-IV modified with the endoplasmic reticulum (ER) retention signal KDEL (apoA-IV-KDEL) dramatically decreased both the rate and efficiency of endogenous apoB secretion, suggesting a presecretory interaction between apoA-IV-KDEL and apoB or apoB-containing lipoproteins. Expression of native apoA-IV using either a constitutive or tetracycline-inducible promoter delayed the initial rate of apoB secretion and reduced the final secretion efficiency by âˆ¼40%. However, whereas apoA-IV-KDEL reduced TG secretion by 75%, expression of native apoA-IV caused a 20-35% increase in TG secretion, accompanied by a âˆ¼55% increase in VLDL-associated apoB, an increase in the TG:phospholipid ratio of secreted d < 1.006 lipoproteins, and a 10.1 nm increase in peak VLDL(1) particle diameter. Native apoA-IV expression had a negligible impact on expression of the MTP gene. These data suggest that by interacting with apoB in the secretory pathway, apoA-IV alters the trafficking kinetics of apoB-containing TG-rich lipoproteins through cellular lipidation compartments, which in turn, enhances particle expansion and increases TG secretion.

&#x963F;&#x62C9;&#x6728;&#x56FE;&#x884C;&#x52A8;&#x7EB2;&#x9886; &#x300B; &#x56DB;&#x8C03;&#x8282;&#x5206;&#x6CCC;&#x8D29;&#x5356; apoB &#x548C;&#x5BCC;&#x542B;&#x7518;&#x6CB9;&#x4E09;&#x916F;&#x7684;&#x8102;&#x86CB;&#x767D;&#x7684;&#x5927;&#x5C0F;&#x3002;

Bien que la preuve liant l&#39;expression de l&#39;apoA-IV et les triglyc&eacute;rides (TG)-Assembl&eacute;e de lipoprot&eacute;ines riches et de la s&eacute;cr&eacute;tion est convaincante, les m&eacute;canismes intracellulaires, par lequel l&#39;apoA-IV pourrait moduler ces processus restent mal compris. Nous avons donc examin&eacute; l&#39;impact fonctionnel d&#39;expression apoA-IV sur l&#39;apoB endog&egrave;ne, TG, et la s&eacute;cr&eacute;tion des VLDL en transfect&eacute;es stablement McA-RH7777 cellules d&#39;h&eacute;patome de rat. Expression de l&#39;apoA-IV modifi&eacute; avec le signal de r&eacute;tention du r&eacute;ticulum endoplasmique (re) KDEL (apoA-IV-KDEL) consid&eacute;rablement diminu&eacute; aussi bien le taux et l&#39;efficacit&eacute; de la s&eacute;cr&eacute;tion endog&egrave;ne d&#39;apoB, sugg&eacute;rant une interaction pr&eacute;-s&eacute;cr&eacute;toires apoA-IV-KDEL et apoB ou lipoprot&eacute;ines contenant l&#39;apoB. Expression de native apoA-IV en utilisant soit un promoteur constitutif ou t&eacute;tracycline-inducible retard&eacute; la vitesse initiale de la s&eacute;cr&eacute;tion de l&#39;apoB et r&eacute;duit l&#39;efficacit&eacute; de la finale de la s&eacute;cr&eacute;tion de &sim;40 %. Cependant, tandis qu&#39;apoA-IV-KDEL r&eacute;duit la s&eacute;cr&eacute;tion de la TG de 75 %, expression de native apoA-IV caus&eacute; une augmentation de 20 &agrave; 35 % la s&eacute;cr&eacute;tion de TG, accompagn&eacute;e d&#39;une augmentation de % &sim;55 apoB associ&eacute;es aux VLDL, une augmentation du ratio de TG:phospholipid des lipoprot&eacute;ines s&eacute;cr&eacute;t&eacute;es d &lt; 1.006 et une augmentation de 10,1 nm de diam&egrave;tre de particules VLDL(1) PIC. Expression Native apoA-IV a eu un impact n&eacute;gligeable sur l&#39;expression du g&egrave;ne MTP. Ces donn&eacute;es sugg&egrave;rent que, en interagissant avec l&#39;apoB dans la voie de s&eacute;cr&eacute;tion, apoA-IV modifie la cin&eacute;tique traite de l&#39;apoB contenant des lipoprot&eacute;ines riches en TG &agrave; travers des compartiments cellulaires lipidation, qui &agrave; son tour, am&eacute;liore la dilatation de la particule et augmente la s&eacute;cr&eacute;tion de TG.

Jedoch w&auml;hrend ApoA-IV-KDEL TG Sekretion um 75 % reduziert, verursachte Ausdruck des systemeigenen ApoA-IV eine Erh&ouml;hung der TG-Sekretion, begleitet von einem &sim;55 % Anstieg der VLDL-assoziierte ApoB, eine Erh&ouml;hung des TG:phospholipid Verh&auml;ltnisses der sezernierte d &lt; 1.006 Lipoproteine, 20-35 % und eine Erh&ouml;hung der 10,1 nm Spitze VLDL(1) Partikeldurchmesser. Native ApoA-IV Ausdruck hatte einen vernachl&auml;ssigbaren Einfluss auf Expression der MTP-Gens. ApoA IV &#xBCC0;&#xC870; Apob&#xC640; triglyceride &#xD48D;&#xBD80;&#xD55C; &#xC9C0; &#xB2E8;&#xBC31;&#xC9C8;&#xC758; &#xD06C;&#xAE30;&#xC758; &#xBD84; &#xC778;&#xC2E0; &#xB9E4;&#xB9E4;.

Aunque la evidencia que vincule la expresi&oacute;n de apoA-IV y triglic&eacute;ridos (TG)-Asamblea de lipoprote&iacute;na rica y secreci&oacute;n es convincente, los mecanismos intracelulares que apoA-IV podr&iacute;a modular estos procesos siguen siendo mal entendidos. Por lo tanto, examinamos el impacto funcional de la expresi&oacute;n de apoA-IV en apoB end&oacute;geno, TG, y secreci&oacute;n de VLDL en estable transfectadas McA-RH7777 c&eacute;lulas de hepatoma de rata. Expresi&oacute;n de apoA-IV nativa utilizando un promotor constitutivo o tetraciclina-inducible retrasa la tasa inicial de secreci&oacute;n de apoB y reducirse la eficacia de la final de la secreci&oacute;n de &sim;40%.
