A parallel screening method has been developed to rapidly evaluate discrete library substrates of biomaterials using cell-based assays. The biomaterials used in these studies were surface-erodible polyanhydrides based on sebacic acid (SA), 1,6-bis(p-carboxyphenoxy)hexane (CPH), and 1,8-bis(p-carboxyphenoxy)-3,6-dioxaoctane (CPTEG) that have been previously studied as carriers for drugs, proteins, and vaccines. Linearly varying compositional libraries of 25 different polyanhydride random copolymers (based on CPH:SA and CPTEG:CPH) were designed, fabricated, and synthesized using discrete (organic solvent-resistant) multi-sample substrates created using a novel rapid prototyping method. The combinatorial libraries were characterized at high throughput using infrared microscopy and validated using 1H NMR and size exclusion chromatography. The discrete libraries were rapidly screened for biocompatibility using standard SP2/0 myeloma, CHO and L929 fibroblasts, and J774 macrophage cell lines. At a concentration of 2.8 mg/mL, there was no appreciable cytotoxic effect on any of the four cell lines evaluated by any of the CPH:SA or CPTEG:CPH compositions. Furthermore, the activation of J774 macrophages was evaluated by incubating the cells with the polyanhydride libraries and quantifying the secreted cytokines (IL-6, IL-10, IL-12, and TNFalpha). The results indicated that copolymer compositions containing at least 50% CPH induced elevated amounts of TNFalpha. In summary, the results indicated that the methodologies described herein are amenable to the high throughput analysis of synthesized biomaterials and will facilitate the rapid and rational design of materials for use in biomedical applications.

Die diskreten Bibliotheken wurden rasch zum Biokompatibilit&auml;t mit standard SP2/0-Myelom, CHO und L929 Fibroblasten und Makrophagen-Zelllinien J774 &uuml;berpr&uuml;ft. Bei einer Konzentration von 2,8 mg/mL gab es keine nennenswerten zytotoxische Wirkung auf jedem der vier Zelllinien, die von keinem der CPH:SA oder CPTEG:CPH Kompositionen ausgewertet. Dar&uuml;ber hinaus wurde die Aktivierung der Makrophagen J774 ausgewertet durch Inkubation der Zellen mit den Polyanhydride-Bibliotheken und Quantifizierung der sezernierte Zytokine (IL-6, IL-10, IL-12 und TNFalpha). Die Ergebnisse zeigten, dass Copolymer Kompositionen mit mindestens 50 % CPH induzierte erh&ouml;hte Mengen von TNFalpha. Zusammenfassend l&auml;sst sich sagen erkl&auml;rt die Ergebnisse, dass die hier beschriebenen Methoden empf&auml;nglich f&uuml;r die Hochdurchsatz-Analyse der synthetisierten Biomaterialien sind und erleichtern die schnelle und rationelle Gestaltung der Materialien f&uuml;r Biomedizinische Anwendungen.

Hochdurchsatz-zellbasierte-Screening der biologisch abbaubare Polyanhydride-Bibliotheken.

Un metodo di screening di parallelo &egrave; stato sviluppato per valutare rapidamente substrati libreria discreti di biomateriali usando cell-based assays. I biomateriali utilizzati in questi studi sono stati polyanhydrides superficie-erodibili basata su acido sebacico (SA), bis(p-carboxyphenoxy)-1,6 esano (CPH) e 1,8-bis(p-carboxyphenoxy)-3,6-dioxaoctane (CPTEG) che sono stati in precedenza studiati come vettori per farmaci, proteine e vaccini. Un m&eacute;todo de detecci&oacute;n de paralelo ha sido desarrollado para evaluar r&aacute;pidamente sustratos discreta biblioteca de biomateriales usando an&aacute;lisis basadas en c&eacute;lulas. Los biomateriales utilizados en estos estudios fueron polyanhydrides superficie-erosionables basado en &aacute;cido seb&aacute;cico (SA), 1,6-bis(p-carboxyphenoxy) hexano (CPH) y 1, 8-bis(p-carboxyphenoxy)-3, 6-dioxaoctane (CPTEG) que han sido previamente estudiados como portadores para vacunas, medicamentos y prote&iacute;nas. Linealmente diferentes bibliotecas compositivas de 25 diferentes polyanhydride de copol&iacute;meros random (basados en CPH:SA y CPTEG:CPH) fueron dise&ntilde;adas, fabricados y sintetizados usando discretos (org&aacute;nico resistentes a los disolventes) sustratos multi-muestra creados usando un m&eacute;todo novedoso de prototipado r&aacute;pido. Las librer&iacute;as combinatorias fueron caracterizadas en alto rendimiento usando microscopia infrarroja y validado mediante 1 H NMR y cromatograf&iacute;a de exclusi&oacute;n de tama&ntilde;o. Las bibliotecas discretas r&aacute;pidamente fueron defendidas para biocompatibilidad con mieloma est&aacute;ndar de SP2/0, CHO y eluci&oacute;n de L929 fibroblastos y l&iacute;neas de c&eacute;lulas macr&oacute;fagos J774. En una concentraci&oacute;n de 2,8 mg/mL, no hubo ning&uacute;n efecto citot&oacute;xico apreciable en cualquiera de las l&iacute;neas de cuatro c&eacute;lulas evaluadas por cualquiera de las composiciones de CPH:SA o CPTEG:CPH. Adem&aacute;s, la activaci&oacute;n de los macr&oacute;fagos J774 se evalu&oacute; por incubaci&oacute;n de las c&eacute;lulas con las bibliotecas de polyanhydride y cuantificaci&oacute;n de las citoquinas secretadas (IL-6, IL-10, IL-12 y TNFalpha). Los resultados indicaron que copol&iacute;mero composiciones que contienen por lo menos 50% CPH inducida elevan cantidades de TNFalpha. Polyanhydrides are a promising class of biomaterials for use as vaccine adjuvants and as multi-component implants. Their properties can be tailored for such applications as controlled drug release, drug stability, and/or immune regulation (adjuvant effect). Understanding the induction of immunomodulatory mechanisms of this polymer system is important for the design and development of efficacious vaccines and tissue compatible multi-component implantable devices using this polymer system. This study describes the development of a rapid multiplexed method for the investigation of the adjuvanticity of polyanhydride nanospheres and films using murine dendritic cells (DCs). To assess the immune response, cell surface markers including MHC II, CD86, CD40, and CD209 and cytokines including IL-6, IL-12p40, and IL-10 were measured. The DCs incubated with nanospheres displayed enhanced expression of all the surface markers and the production of IL-12p40 compared to DCs incubated with polymer films in a chemistry-dependent manner. This suggests that polyanhydrides of various chemistries and device geometries can be tailored to achieve desired levels of immune cell activation for specific applications. The observed biocompatibility and activation of DCs by polyanhydride devices supports their inclusion in vaccine delivery devices as well as in multi-component medical implants.

Polyanhydrides sont une classe prometteuse de biomat&eacute;riaux pour utilisation comme adjuvants des vaccins et comme implants multi-composants. Leurs propri&eacute;t&eacute;s peuvent &ecirc;tre adapt&eacute;es pour des applications telles que la distribution d&#39;une drogue contr&ocirc;l&eacute;e, stabilit&eacute; de la drogue et/ou r&eacute;gulation immunitaire (effet adjuvant). Il est important pour la conception et le d&eacute;veloppement de vaccins efficaces et tissus multicomposants implantable dispositifs compatibles avec ce syst&egrave;me de polym&egrave;re de comprendre l&#39;induction de m&eacute;canismes immunomodulateurs de ce syst&egrave;me de polym&egrave;re. Combinatorial methods have been developed to study the phase behavior of biodegradable polyanhydrides for drug delivery applications. The polyanhydrides of interest are poly[1,6-bis(p-carboxyphenoxy) hexane] (CPH) and poly[sebacic anhydride] (SA). Both continuous and discrete polymer blend libraries were fabricated by using a combination of solution-based gradient deposition and rapid prototyping. Blend compositions were characterized via a high throughput transmission Fourier transform infrared (FTIR) sampling technique and compared against theoretical mass balance predictions. To obtain phase diagrams of CPH/SA, the effect of blend composition and annealing temperature on the miscibility of the blend was studied. This gradient library was observed with optical microscopy in order to determine cloud points. These results were compared with a theoretical phase diagram obtained from Flory-Huggins theory and with atomic force microscopy (AFM) experiments on blend libraries and the agreement between the methods was very good. The high throughput method demonstrates that the CPH/SA system exhibits upper critical solution temperature behavior. These libraries are amenable to other high throughput applications in biomaterials science including cell viability, cell activation, and protein/biomaterial interactions.

M&eacute;todos combinatorios se han desarrollado para estudiar el comportamiento de la fase de polyanhydrides biodegradable para solicitudes de entrega de medicamentos. Las polyanhydrides de inter&eacute;s son poly[1,6-bis(p-carboxyphenoxy) hexano] (CPH) y poly [seb&aacute;cico anh&iacute;drido] (SA). Ambas bibliotecas de mezcla de pol&iacute;mero continuas y discretas fueron fabricados usando una combinaci&oacute;n de deposici&oacute;n de gradiente basadas en soluciones y prototipado r&aacute;pido. Composiciones de mezcla se caracterizaron mediante una transmisi&oacute;n de alto rendimiento Fourier transforma la t&eacute;cnica de muestreo de infrarrojo (FTIR) y comparan con las predicciones te&oacute;ricas de balance de masa. Para obtener los diagramas de fases de CPH/SA, se estudi&oacute; el efecto de la composici&oacute;n de la mezcla y recocido de temperatura en la miscibilidad de la mezcla. Esta biblioteca gradiente fue observada con microscop&iacute;a &oacute;ptica para determinar puntos de la nube. Estos resultados fueron comparados con un diagrama de fase te&oacute;rica Obtenido de la teor&iacute;a de Flory-Huggins y con fuerza at&oacute;mica microscop&iacute;a (AFM) experimentos en las bibliotecas de la mezcla y el acuerdo entre los m&eacute;todos fue muy bueno. This work demonstrates that amphiphilic polyanhydride microparticles based on co-polymers of 1,6-bis(p-carboxyphenoxy)hexane (CPH) and 1,6-bis(p-carboxyphenoxy)-3,6-dioxaoctane (CPTEG) provide stabilizing environments for proteins. A cryogenic atomization method was used to fabricate protein-loaded polyanhydride microparticles. These microparticles were tested for their ability to provide controlled delivery of lipocalin 2 (Lcn2) and to maintain its structure and function. Lcn2 is an acute-phase protein suspected to play a role in cell migration and tissue repair. The in vitro release kinetics of Lcn2 from the microparticles were a function of the chemistry of the polymer carrier. The biological activity of Lcn2 released from polyanhydride microparticles was investigated by its ability to stimulate migration of human colon epithelial cells (HCT116). Lcn2 released from 50:50 and 20:80 CPTEG/CPH microparticles maintained its biological activity as demonstrated by the increased rate of cell migration. In addition, the Lcn2-loaded 50:50 and 20:80 CPTEG/CPH microparticles promoted cell migration over that of the Lcn2 administered alone. This was interpreted as the ability of the amphiphilic microparticles to stabilize the encapsulated protein and release it in a controlled manner over a period of time. This work demonstrates the potential for therapeutic use of amphiphilic polyanhydride microparticles as protein/drug carriers.

Several challenges currently exist for rational design of functional tissue engineering constructs within the host, which include appropriate cellular integration, avoidance of bacterial infections, and low inflammatory stimulation. This work describes a novel class of biodegradable, amphiphilic polyanhydrides with many desirable protein-material and cell-material attributes capable of confronting these challenges. The biocompatible amphiphilic polymer films were shown to release laminin in a stable and controlled manner, promote neural cell adhesion and differentiation, and evade inflammatory responses of the immune system. Using high-throughput approaches, it was shown that polymer chemistry plays an integral role in controlling cell-film interactions, which suggests that these polyanhydrides can be tailored to achieve the desired cell adhesion and differentiation while minimizing immune recognition. These findings have important implications for development of engineered constructs to regulate differentiation and target the growth of transplanted cells in stem cell-based therapies to treat nervous system disorders.

Techniques in materials design, immunophenotyping, and informatics can be valuable tools for using a molecular based approach to design vaccine adjuvants capable of inducing protective immunity that mimics a natural infection but without the toxic side effects. This work describes the molecular design of amphiphilic polyanhydride nanoparticles that activate antigen presenting cells in a pathogen-mimicking manner. Biodegradable polyanhydrides are well suited as vaccine delivery vehicles due to their adjuvant-like ability to: 1) enhance the immune response, 2) preserve protein structure, and 3) control protein release. The results of these studies indicate that amphiphilic nanoparticles possess pathogen-mimicking properties as evidenced by their ability to activate dendritic cells similarly to LPS. Specific molecular descriptors responsible for this behavior were identified using informatics analyses, including the number of backbone oxygen moieties, percent of hydroxyl end groups, polymer hydrophobicity, and number of alkyl ethers. Additional findings from this work suggest that the molecular characteristics mediating APC activation are not limited to hydrophobicity but vary in complexity (e.g., presentation of oxygen-rich molecular patterns to cells) and elicit unique patterns of cellular activation. The approach outlined herein demonstrates the ability to rationally design pathogen-mimicking nanoparticle adjuvants for use in next-generation vaccines against emerging and re-emerging diseases.

An opportunity exists today for cross-cutting research utilizing advances in materials science, immunology, microbial pathogenesis, and computational analysis to effectively design the next generation of adjuvants and vaccines. This study integrates these advances into a bottom-up approach for the molecular design of nanoadjuvants capable of mimicking the immune response induced by a natural infection but without the toxic side effects. Biodegradable amphiphilic polyanhydrides possess the unique ability to mimic pathogens and pathogen associated molecular patterns with respect to persisting within and activating immune cells, respectively. The molecular properties responsible for the pathogen-mimicking abilities of these materials have been identified. The value of using polyanhydride nanovaccines was demonstrated by the induction of long-lived protection against a lethal challenge of Yersinia pestis following a single administration ten months earlier. This approach has the tantalizing potential to catalyze the development of next generation vaccines against diseases caused by emerging and re-emerging pathogens.

Die molekularen Eigenschaften verantwortlich f&uuml;r die Nachahmung von Pathogen-F&auml;higkeiten dieser Materialien wurden identifiziert. Der Wert der Verwendung Polyanhydride Nanovaccines zeigte die Induktion von langlebigen Schutz gegen eine t&ouml;dliche Herausforderung von Yersinia Pestis nach eine einzige Gabe zehn Monate zuvor. Dieser Ansatz hat das verf&uuml;hrerische Potenzial, die Entwicklung der n&auml;chsten Generation-Impfstoffe gegen Erkrankungen durch Schwellen- und ersteht jetzt wieder Krankheitserreger zu katalysieren.
