Haemophilus parasuis is a swine pathogen of significant industry concern, but little is known about how the organism causes disease. A related human pathogen, Haemophilus influenzae, has been better studied, and many of its virulence factors have been identified. Two of these, outer membrane proteins P2 and P5, are known to have important virulence properties. The goals of this study were to identify, analyze, and compare the genetic relatedness of orthologous genes encoding P2 and P5 proteins in a diverse group of 35 H. parasuis strains. Genes encoding P2 and P5 proteins were detected in all H. parasuis strains evaluated. The predicted amino acid sequences for both P2 and P5 proteins exhibit considerable heterogeneity, particularly in regions corresponding to predicted extracellular loops. Twenty-five variants of P2 and 17 variants of P5 were identified. The P2 proteins of seven strains were predicted to contain a highly conserved additional extracellular loop compared to the remaining strains and to H. influenzae P2. Antigenic-site predictions coincided with predicted extracellular loop regions of both P2 and P5. Neighbor-joining trees constructed using P2 and P5 sequences predicted divergent evolutionary histories distinct from those predicted by a multilocus sequence typing phylogeny based on partial sequencing of seven housekeeping genes. Real-time reverse transcription-PCR indicated that both genes are expressed in all of the strains.

The design of biodegradable polymeric delivery systems based on polyanhydrides that would provide for improved structural integrity of Yersinia pestis antigens was the main goal of this study. Accordingly, the full-length Y. pestis fusion protein (F1-V) or a recombinant Y. pestis fusion protein (F1(B2T1)-V10) was encapsulated and released from microparticles based on 1,6-bis(p-carboxyphenoxy)hexane (CPH) and sebacic acid (SA) copolymers and 1,8-bis(p-carboxyphenoxy)-3,6-dioxaoctane (CPTEG) and CPH copolymers fabricated by cryogenic atomization. An enzyme-linked immunosorbent assay was used to measure changes in the antigenicity of the released proteins. The recombinant F1(B2T1)-V10 was unstable upon release from the hydrophobic CPH:SA microparticles, but maintained its structure and antigenicity in the amphiphilic CPTEG:CPH system. The full-length F1-V was stably released by both CPH:SA and CPTEG:CPH microparticles. In order to determine the effect of the anhydride monomers on the protein structure, changes in the primary, secondary, and tertiary structure, as well as the antigenicity of both Y. pestis antigens, were measured after incubation in the presence of saturated solutions of SA, CPH, and CPTEG anhydride monomers. The results indicated that the amphiphilic environment provided by the CPTEG monomer was important to preserve the structure and antigenicity of both proteins. These studies offer an approach by which a thorough understanding of the mechanisms governing antigenic instability can be elucidated in order to optimize the in vivo performance of biodegradable delivery devices as protein carriers and/or vaccine adjuvants.

Entsprechend der in Y. Pestis-Fusionsprotein (F1-V) oder ein rekombinantes Y. Pestis-Fusionsprotein (F1(B2T1)-V10) war gekapselt und Mikropartikel basierend auf 1,6-Bis(p-carboxyphenoxy) Hexan (CPH) und CPH-Copolymere hergestellt von kryogenen Atomisierung, Sebacins&auml;ure (SA)-Copolymere und 1,8-Bis(p-carboxyphenoxy)-3, 6-Dioxaoctane (Westwood) entbunden. Amphiphilic polyanhydride &#xB4A4;&#xC5D0; &#xCEA1;&#xC290;&#xD654; &#xC548;&#xC815;&#xD654; Yersinia pestis &#xD56D; &#xC6D0;.

El dise&ntilde;o de sistemas polim&eacute;ricos biodegradables basado en polyanhydrides que proporcionen la integridad estructural mejorada de ant&iacute;genos de Yersinia pestis fue el objetivo principal de este estudio. Por consiguiente, la prote&iacute;na integral Y. pestis fusi&oacute;n (F1-V) o una Y. pestis fusi&oacute;n prote&iacute;na recombinante (F1(B2T1)-V10) fue encapsulado y de micropart&iacute;culas basado en copol&iacute;meros de &aacute;cido seb&aacute;cico (SA), 1,6-bis(p-carboxyphenoxy) hexano (CPH) y copol&iacute;meros CPH fabricados por atomizaci&oacute;n criog&eacute;nica y 1,8-bis(p-carboxyphenoxy)-3, 6-dioxaoctane (CPTEG). Un enzyme-linked immunosorbent assay fue utilizado para medir los cambios en la antigenicidad de las prote&iacute;nas liberadas. La F1 recombinante (B2T1)-V10 era inestable sobre el lanzamiento de las micropart&iacute;culas CPH:SA hidrof&oacute;bico, pero mantuvo su estructura y la antigenicidad de la anfif&iacute;licas CPTEG:CPH sistema. El largometraje F1-V estable fue lanzado por CPH:SA y micropart&iacute;culas de cpteg:CPH. Con el fin de determinar el efecto de los monomers del anh&iacute;drido en la estructura de la prote&iacute;na, cambios en la primaria, secundaria, y la estructura terciaria, as&iacute; como la antigenicidad de ambos ant&iacute;genos de Y. pestis, fueron medida despu&eacute;s de la incubaci&oacute;n en presencia de soluciones saturadas de mon&oacute;meros de anh&iacute;drido SA, CPH y CPTEG. Los resultados indicaron que el entorno de anfif&iacute;licas proporcionado por el mon&oacute;mero CPTEG importante para preservar la estructura y la antigenicidad de ambas prote&iacute;nas. Estos estudios ofrecen un enfoque mediante el cual una profunda comprensi&oacute;n de los mecanismos que regulan la inestabilidad antig&eacute;nica puede ser aclarada con el fin de optimizar el rendimiento in vivo de dispositivos de entrega biodegradables como portadores de la prote&iacute;na o adyuvantes de la vacuna.
