Cardiovascular complications such as hypertension are a continuous concern in patients with autosomal dominant polycystic kidney disease (ADPKD). The PKD2 encoding for polycystin-2 is mutated in approximately 15% of ADPKD patients. Here, we show that polycystin-2 is localized to the cilia of mouse and human vascular endothelial cells. We demonstrate that the normal expression level and localization of polycystin-2 to cilia is required for the endothelial cilia to sense fluid shear stress through a complex biochemical cascade, involving calcium, calmodulin, Akt/PKB, and protein kinase C. In response to fluid shear stress, mouse endothelial cells with knockdown or knockout of Pkd2 lose the ability to generate nitric oxide (NO). Consistent with mouse data, endothelial cells generated from ADPKD patients do not show polycystin-2 in the cilia and are unable to sense fluid flow. In the isolated artery, we further show that ciliary polycystin-2 responds specifically to shear stress and not to mechanical stretch, a pressurized biomechanical force that involves purinergic receptor activation. We propose a new role for polycystin-2 in transmitting extracellular shear stress to intracellular NO biosynthesis. Thus, aberrant expression or localization of polycystin-2 to cilia could promote high blood pressure because of inability to synthesize NO in response to an increase in shear stress (blood flow).

Les complications cardiovasculaires comme l&#x26;#39;hypertension sont une pr&#xE9;occupation constante chez les patients atteints r&#xE9;nale autosomique dominante polykystique (PKRAD). Nous proposons un nouveau r&#xF4;le pour la polycystine-2 dans la transmission de la contrainte de cisaillement extracellulaire intracellulaire NO biosynth&#xE8;se. Ainsi, l&#x26;#39;expression aberrante ou la localisation de la polycystine-2 &#xE0; cils pourrait favoriser l&#x26;#39;hypertension art&#xE9;rielle en raison de l&#x26;#39;incapacit&#xE9; de faire la synth&#xE8;se de NO en r&#xE9;ponse &#xE0; une augmentation de la contrainte de cisaillement (circulation sanguine).

Das PKD2-Codierung f&uuml;r Polycystin-2 ist in etwa 15 % der ADPKD-Patienten mutiert. Hier zeigen wir, dass diese Polycystin-2 auf die Flimmerh&auml;rchen Maus und menschlichen vaskul&auml;re Endothelzellen lokalisiert ist. Wir zeigen, dass die normalen Ausdrucksebene und Lokalisierung von Polycystin-2 bis Zilien ist erforderlich f&uuml;r die endothelialen Zilien Sinn fl&uuml;ssig Shear Stress durch eine komplexe biochemische Kaskade mit Kalzium, Calmodulin, Akt/PKB und Proteinkinase C. Reaktion auf fl&uuml;ssige Schubspannung verlieren Maus Endothelzellen mit Niederschlag oder Pkd2-ko die M&ouml;glichkeit zum Generieren von Stickstoffmonoxid (NO). Cos&igrave;, espressione aberrante o localizzazione di polycystin-2 per ciglia potrebbe promuovere la pressione arteriosa alta a causa della incapacit&agrave; di sintetizzare NO in risposta ad un aumento della sollecitazione di taglio (flusso sanguigno).

Autosomal-dominant polycystic kidney disease (ADPKD) is the most common hereditary and systemic disorder associated with various cardiovascular complications. It has been implicated with dysfunction in primary cilia. We and others have shown that the immediate function of endothelial cilia is to sense extracellular signal. The long-term function of cilia is hypothesized to regulate cell cycle. Here, we show that ciliary function (polycystins) and structure (polaris) are required for proper cellular division. Cilia mutant cells undergo abnormal cell division with apparent defects in mitotic spindle formation, cellular spindle assembly checkpoint and centrosome amplification. Down-regulation of the chromosomal passenger survivin contributes to these abnormalities, which further result in cell polyploidy. Re-expression of survivin restores a competent spindle assembly checkpoint and reduces polyploidy. Aged animals show a more severe phenotype in cellular division, consistent with progression of cardiovascular complications seen in older ADPKD patients. For the first time, we show that structure and function of mechanosensory cilia are crucial in maintaining proper cellular proliferation. Furthermore, developmental aging plays a crucial role in the progression of these abnormal cellular phenotypes. We propose that abnormal function or structure of primary cilia not only causes failure to transmit extracellular signals, but also is associated with cytokinesis defects in both mice and humans with polycystic kidney disease.

The zebrafish (Danio rerio) has been used as a model for studying vertebrate development in the cardiovascular system. In order to monitor heart contraction and cytosolic calcium oscillations, fish were either embedded in methylcellulose or anesthetized with tricaine. Using high-resolution differential interference contrast and calcium imaging microscopy, we here show that dopamine and verapamil alter calcium signaling and muscle contraction in anesthetized zebrafish, but not in embedded zebrafish. In anesthetized fish, dopamine increases the amplitude of cytosolic calcium oscillation with a subsequent increase in heart contraction, whereas verapamil decreases the frequency of calcium oscillation and heart rate. Interestingly, verapamil also increases myocardial contraction. Our data further indicate that verapamil can increase myocardial calcium sensitivity in anesthetized fish. Taken together, our data reinforce in vivo cardiac responses to dopamine and verapamil. Furthermore, effects of dopamine and verapamil on myocardial calcium and contraction are greater in anesthetized than embedded fish. We suggest that while the zebrafish is an excellent model for a cardiovascular imaging study, the cardio-pharmacological profiles are very different between anesthetized and embedded fish.

A Comparative Study of Embedded and Anesthetized Zebrafish in vivo on Myocardiac Calcium Oscillation and Heart Muscle Contraction.

Le poisson-z&#xE8;bre (Danio rerio) a &#xE9;t&#xE9; utilis&#xE9; comme mod&#xE8;le pour &#xE9;tudier le d&#xE9;veloppement des vert&#xE9;br&#xE9;s dans le syst&#xE8;me cardio-vasculaire. Afin de surveiller la contraction du c&#x153;ur et des oscillations de calcium cytosolique, les poissons ont &#xE9;t&#xE9; soit noy&#xE9;es dans de la m&#xE9;thylcellulose ou anesth&#xE9;si&#xE9;s avec trica&#xEF;ne. Utilisation &#xE0; haute r&#xE9;solution de contraste interf&#xE9;rentiel diff&#xE9;rentiel et de la microscopie imagerie calcique, nous avons ici montrent que la dopamine et de la signalisation calcique v&#xE9;rapamil alter et la contraction musculaire chez le poisson z&#xE8;bre anesth&#xE9;si&#xE9;s, mais pas chez le poisson z&#xE8;bre int&#xE9;gr&#xE9;. Chez les poissons anesth&#xE9;si&#xE9;s, la dopamine augmente l&#x26;#39;amplitude d&#x26;#39;oscillation de calcium cytosolique avec une augmentation subs&#xE9;quente de la contraction du coeur, tandis que le v&#xE9;rapamil diminue la fr&#xE9;quence d&#x26;#39;oscillation de calcium et le rythme cardiaque. Fait int&#xE9;ressant, le v&#xE9;rapamil augmente &#xE9;galement la contraction du myocarde. Nos donn&#xE9;es indiquent en outre que le v&#xE9;rapamil peut augmenter la sensibilit&#xE9; au calcium du myocarde chez les poissons anesth&#xE9;si&#xE9;s. Pris dans leur ensemble, nos donn&#xE9;es renforcent in vivo des r&#xE9;ponses cardiaques &#xE0; la dopamine et le v&#xE9;rapamil. Um Herzen Kontraktion und cytosolic Kalzium Schwingungen zu &uuml;berwachen, wurden Fische entweder in Methylzellulose eingebettet oder mit Tricaine bet&auml;ubt. Mit hochaufl&ouml;senden differenzielle Interferenz-Kontrast und Kalzium imaging Mikroskopie, zeigen wir hier, dass Dopamin und Verapamil Kalzium Signalisierung und Muskel Kontraktion im narkotisierten Zebrafisch, aber nicht in eingebettete Zebrafisch ver&auml;ndern. El pez cebra (Danio rerio) se ha utilizado como modelo para el estudio de desarrollo de los vertebrados en el sistema cardiovascular. A fin de controlar la contracci&#xF3;n del coraz&#xF3;n y las oscilaciones de calcio citos&#xF3;lico, los peces fueron incorporados, ya sea en metilcelulosa o anestesiados con trica&#xED;na. Uso de alta resoluci&#xF3;n de contraste de interferencia diferencial y de im&#xE1;genes de microscop&#xED;a de calcio, que aqu&#xED; muestran que la dopamina y la se&#xF1;alizaci&#xF3;n del calcio verapamilo altar y la contracci&#xF3;n muscular en el pez cebra anestesiado, pero no en el pez cebra incrustado. En los peces anestesiados, la dopamina aumenta la amplitud de oscilaci&#xF3;n de calcio en el citosol con el consiguiente aumento en la contracci&#xF3;n del coraz&#xF3;n, mientras que el verapamilo disminuye la frecuencia de oscilaci&#xF3;n de calcio y frecuencia card&#xED;aca. Curiosamente, verapamilo tambi&#xE9;n aumenta la contracci&#xF3;n del miocardio. Nuestros datos indican adem&#xE1;s que el verapamilo puede aumentar la sensibilidad al calcio de miocardio en los peces anestesiados. En conjunto, nuestros datos refuerzan in vivo las respuestas card&#xED;acas a la dopamina y verapamilo. Adem&#xE1;s, los efectos de la dopamina y verapamilo en calcio y contracci&#xF3;n del miocardio son mayores en anestesiados que el pescado incrustado. Se sugiere que mientras que el pez cebra es un modelo excelente para un estudio de imagen cardiovascular, los perfiles de cardio-farmacol&#xF3;gicos son muy diferentes entre los peces anestesiados y embebidos.

Cilia are mechanosensing organelles that communicate extracellular signals into intracellular responses. Altered functions of primary cilia play a key role in the development of various diseases including polycystic kidney disease. Here, we show that endothelial cells from the oak ridge polycystic kidney (Tg737(orpk/orpk) ) mouse, with impaired cilia assembly, exhibit a reduction in the actin stress fibers and focal adhesions compared to wild-type (WT). In contrast, endothelial cells from polycystin-1 deficient mice (pkd1(null/null) ), with impaired cilia function, display robust stress fibers, and focal adhesion assembly. We found that the Tg737(orpk/orpk) cells exhibit impaired directional migration and endothelial cell monolayer permeability compared to the WT and pkd1(null/null) cells. Finally, we found that the expression of heat shock protein 27 (hsp27) and the phosphorylation of focal adhesion kinase (FAK) are downregulated in the Tg737(orpk/orpk) cells and overexpression of hsp27 restored both FAK phosphorylation and cell migration. Taken together, these results demonstrate that disruption of the primary cilia structure or function compromises the endothelium through the suppression of hsp27 dependent actin organization and focal adhesion formation, which may contribute to the vascular dysfunction in ciliopathies.

Ici, nous montrons que les cellules endoth&#xE9;liales du rein polykystique Oak Ridge (Tg737 (orpk / orpk)) de la souris, avec l&#x26;#39;assemblage des cils avec facult&#xE9;s affaiblies, pr&#xE9;sentent une r&#xE9;duction des fibres de stress d&#x26;#39;actine et des adh&#xE9;rences focales par rapport au type sauvage (WT). Ver&auml;nderte Funktionen der prim&auml;re Zilien spielen eine Schl&uuml;sselrolle bei der Entwicklung von verschiedenen Krankheiten einschlie&szlig;lich der polyzystischen Nierenerkrankung. Hier zeigen wir, dass Endothelzellen aus der Oak Ridge polyzystische Nieren (Tg737(orpk/orpk)) Maus, mit gest&ouml;rter Zilien Montage, eine Verringerung des Aktin-Spannung-Fasern und fokalen Adh&auml;sionen im Vergleich zu Wildtyp (WT) aufweisen. Dagegen zeigen Endothelzellen von Polycystin-1 unzul&auml;nglichen M&auml;usen (pkd1(null/null)), mit gest&ouml;rter Zilien-Funktion, robuste Spannung Fasern und fokale Adh&auml;sion Assembly. Wir fanden, dass die Tg737(orpk/orpk)-Zellen gest&ouml;rter gerichtete Migration weisen und Endothelzellen Monolage Durchl&auml;ssigkeit im Vergleich zu den WT und pkd1(null/null) Zellen. Schlie&szlig;lich fanden wir, dass der Ausdruck von Hitze-Schock-Protein 27 (hsp27) und die Phosphorylierung von fokale Adh&auml;sion Kinase (FAK) Downregulated in den Tg737(orpk/orpk) Zellen sind und eine &Uuml;berexpression von hsp27, FAK-Phosphorylierung und Zellmigration wiederhergestellt. Zusammengenommen zeigen diese Ergebnisse, dass die St&ouml;rung im prim&auml;ren Zilien Struktur oder Funktion beeintr&auml;chtigt das Endothel durch die Unterdr&uuml;ckung von hsp27 abh&auml;ngige Aktin Organisation und fokale Adh&auml;sion-Formation, die zu die vaskul&auml;re Dysfunktion in Ziliopathien beitragen kann.

Los cilios son org&#xE1;nulos que se comunican mechanosensing se&#xF1;ales extracelulares en respuestas intracelulares. Alteraci&#xF3;n de funciones de los cilios primarios desempe&#xF1;an un papel clave en el desarrollo de varias enfermedades, incluyendo enfermedad renal poliqu&#xED;stica. En este sentido, muestran que las c&#xE9;lulas endoteliales de los ri&#xF1;ones poliqu&#xED;sticos Oak Ridge (Tg737 (orpk / orpk)) del rat&#xF3;n, con el conjunto de los cilios deteriorada, muestran una reducci&#xF3;n en las fibras de actina estr&#xE9;s y adhesiones focales en comparaci&#xF3;n con los de tipo salvaje (WT). Primary cilia are sensory organelles that transmit extracellular signals into intracellular biochemical responses. Structural and functional defects in primary cilia are associated with a group of human diseases, known as ciliopathies, with phenotypes ranging from cystic kidney and obesity to blindness and mental retardation. Primary cilia mediate mechano- and chemosensation in many cell types. The mechanosensory function of the primary cilia requires the atypical G-protein-coupled receptor polycystin-1 and the calcium-permeable nonselective cation channel polycystin-2. Mechanical stimulations such as fluid-shear stress of the primary cilia initiate intracellular calcium rise, nitric oxide release, and protein modifications. In this review, we describe a set of protocols for cell culture to promote ciliation, mechanical stimulations of the primary cilia, and measurements of calcium rise and nitric oxide release induced by fluid shear stress.

Non-motile primary cilia as fluid shear stress mechanosensors.

Cystic kidneys and vascular aneurysms are clinical manifestations seen in patients with polycystic kidney disease, a cilia-associated pathology (ciliopathy). Survivin overexpression is associated with cancer, but the clinical pathology associated with survivin downregulation or knockout has never been studied before. The present studies aim to examine whether and how cilia function (Pkd1 or Pkd2) and structure (Tg737) play a role in cystic kidney and aneurysm through survivin downregulation.

Survivin-induced abnormal ploidy contributes to cystic kidney and aneurysm formation.

Dopamine plays a number of important physiological roles. However, activation of dopamine receptor type-5 (DR5) and its effect in renal epithelial cells have not been studied. Here, we show for the first time that DR5 is localized to primary cilia of LLCPK kidney cells. Renal epithelial cilia are mechanosensory organelles that sense and respond to tubular fluid-flow in the kidney. To determine the roles of DR5 and sensory cilia, we used dopamine to non-selectively and fenoldopam to selectively activate ciliary DR5. Compared to mock treatment, dopamine treated cells significantly increases the length of cilia. Fenoldopam further increases the length of cilia compared to dopamine treated cells. The increase in cilia length also increases the sensitivity of the cells in response to fluid-shear stress. The graded responses to dopamine- and fenoldopam-induced increase in cilia length further show that sensitivity to fluid-shear stress correlates to the length of cilia. Together, our studies suggest for the first time that dopamine or fenoldopam is an exciting agent that enhances structure and function of primary cilia. We further propose that dopaminergic agents can be used in "cilio-therapy" to treat diseases associated with abnormal cilia structure and/or function.

Roles of dopamine receptor on chemosensory and mechanosensory primary cilia in renal epithelial cells.

Cell-based perfusion studies have provided great insight into fluid-sensing mechanisms, such as primary cilia in the renal and vascular systems. However, the intrinsic limitations of in vitro cell culture, such as the inability to reflect cellular organization within tissues, has distanced observed paradigms from possible clinical developments. Here we describe a protocol that applies ex vivo artery perfusion and calcium imaging to observe real-time cellular responses to fluid-shear stress.

Real-time vascular mechanosensation through ex vivo artery perfusion.

Dysfunction of many ciliary proteins has been linked to a list of diseases, from cystic kidney to obesity and from hypertension to mental retardation. We previously proposed that primary cilia are unique communication organelles that function as microsensory compartments that house mechanosensory molecules. Here we report that primary cilia exhibit membrane swellings or ciliary bulbs, which based on their unique ultrastructure and motility, could be mechanically regulated by fluid-shear stress. Together with the ultrastructure analysis of the swelling, which contains monosialodihexosylganglioside (GM3), our results show that ciliary bulb has a distinctive set of functional proteins, including GM3 synthase (GM3S), bicaudal-c1 (Bicc1), and polycystin-2 (PC2). In fact, results from our cilia isolation demonstrated for the first time that GM3S and Bicc1 are members of the primary cilia proteins. Although these proteins are not required for ciliary membrane swelling formation under static condition, fluid-shear stress induced swelling formation is partially modulated by GM3S. We therefore propose that the ciliary bulb exhibits a sensory function within the mechano-ciliary structure. Overall, our studies provided an important step towards understanding the ciliary bulb function and structure.
