The semi-experimental approach to approximating physicochemical data relevant to environmental distribution (vapor pressure and gas-octanol distribution) by correlation with gas chromatography (GC) retention data has been extended to the determination of Kow values. We estimated Kow values >10(4) for polychlorinated biphenyls (PCB), which are often derived by liquid chromatography, by correlation with gas chromatographic retention data. Selecting a set of reference compounds with known Kow values for relative retention time (RRT) correlation enables easy and accurate semi-empirical calculation of further Kow values for a given group of congeners. The RRT/log Kow correlation is validated in this paper with regard to the following gas chromatographic conditions: (1) isothermal versus temperature-programmed elution, (2) the possible effect of the polarity of the stationary phase, and (3) the effect of the format of the standardized GC retention data. The advantages of our Kow(GC) method can be summarized as follows: complex mixtures can be analyzed, only amounts in the nanogram-range or less are required, Kow values of isomers can be determined and the exact structure of compounds need not be known. Normalized GC retention data of persistent organic pollutants are readily available. The quality of the Kow values obtained by the GC method compares well with that for other Kow estimation methods. It depends mainly on the accuracy of the Kow data of the structurally correlated compounds used as standards for the correlation cohort. The Kow(GC) data for all 209 PCB congeners are given.

Der semi-experimental Ansatz zur Angleichung von physikalisch-chemischen Daten in Bezug auf &ouml;kologische Verteilung (Dampfdruck und Gas-Octanol-Verteilung) durch Korrelation mit Gaschromatografie (GC) aufzubewahrenden Daten wurde f&uuml;r die Bestimmung der Kow-Werte erweitert. Wir gesch&auml;tzte Kow-Werte &gt; 10 Absatz 4 f&uuml;r Polychlorierte Biphenyle (PCB), die oft durch fl&uuml;ssige Chromatographie, durch Korrelation mit Gas-Chromatographische aufzubewahrenden Daten abgeleitet werden. Auswahl einer Reihe von Referenzverbindungen mit bekannten Kow-Werte f&uuml;r die relative Aufbewahrung Zeit (RRT) Korrelation erm&ouml;glicht einfaches und genaues Halbempirische Berechnung der weiteren Kow-Werte f&uuml;r eine bestimmte Gruppe von Kongenere. Die RRT/Log Kow-Korrelation wird in diesem Papier in Bezug auf die folgenden Gas-Chromatographische Bedingungen validiert: (1) Isotherme gegen Temperatur-programmierte Elution, (2) die m&ouml;gliche Wirkung von der Polarit&auml;t der station&auml;ren Phase und (3) die Wirkung des Formats der standardisierten GC Aufbewahrung Daten. Die Vorteile unserer Kow(GC)-Methode k&ouml;nnen wie folgt zusammengefasst werden: komplexe Mischungen k&ouml;nnen analysiert werden, nur im Nanogramm-Bereich oder weniger Betr&auml;ge erforderlich sind, Kow-Werte der Isomere bestimmt werden k&ouml;nnen und die genaue Struktur der Verbindungen nicht bekannt sein muss. Normalisierte GC aufzubewahrenden Daten von persistenten organischen Schadstoffen sind leicht verf&uuml;gbar. Die Qualit&auml;t der Kow-Werte, die von der GC-Methode vergleicht gut mit, die f&uuml;r andere Sch&auml;tzmethoden Kow. Es h&auml;ngt haupts&auml;chlich von der Richtigkeit der Kow der strukturell korrelierten Verbindungen als Normen f&uuml;r die Korrelation-Kohorte verwendet. Die Kow(GC)-Daten f&uuml;r alle 209 PCB-Kongenere erhalten.

Proteases play prominent roles in many physiological processes and the pathogenesis of various diseases, which makes them interesting drug targets. To fully understand the functional role of proteases in these processes, it is necessary to characterize the target specificity of the enzymes, identify endogenous substrates and cleavage products as well as protease activators and inhibitors. The complexity of these proteolytic networks presents a considerable analytic challenge. To comprehensively characterize these systems, quantitative methods that capture the spatial and temporal distributions of the network members are needed. Recently, activity-based workflows have come to the forefront to tackle the dynamic aspects of proteolytic processing networks in vitro, ex vivo and in vivo. In this review, we will discuss how mass spectrometry-based approaches can be used to gain new insights into protease biology by determining substrate specificities, profiling the activity-states of proteases, monitoring proteolysis in vivo, measuring reaction kinetics and defining in vitro and in vivo proteolytic events. In addition, examples of future aspects of protease research that go beyond mass spectrometry-based applications are given.

Proteasas juegan un papel prominente en muchos procesos fisiol&oacute;gicos y la patogenia de varias enfermedades, que hace de ellos objetivos interesantes de la droga. Para entender completamente el papel funcional de las proteasas en estos procesos, es necesario caracterizar la especificidad del objetivo de las enzimas, identificar sustratos end&oacute;genos y productos de escote as&iacute; como activadores de la proteasa e inhibidores. La complejidad de estas redes proteol&iacute;ticas presenta un considerable desaf&iacute;o anal&iacute;tico. Para caracterizar exhaustivamente estos sistemas, se necesitan m&eacute;todos cuantitativos que captan las distribuciones espaciales y temporales de los miembros de la red. Recientemente, los flujos de trabajo basados en actividad han llegado a la vanguardia para abordar los aspectos din&aacute;micos de las redes de procesamiento proteol&iacute;tico in vitro, ex vivo e in vivo. En esta revisi&oacute;n, se discutir&aacute; c&oacute;mo masa enfoques basados en la espectrometr&iacute;a pueden utilizarse para obtener nuevos conocimientos sobre la biolog&iacute;a de la proteasa por determinar la especificidad de sustrato, perfiles de los Estados de actividad de las proteasas, monitoreo prote&oacute;lisis en vivo, cin&eacute;tica de la reacci&oacute;n de medici&oacute;n y definir eventos proteol&iacute;ticos in vitro e in vivo.
