Methylation of cytosines in CpG motifs is an important mechanism for epigenetic regulation of gene expression in mammalian cells. The initiating event(s) for de novo methylation in mammalian cells, particularly in cancer, is unknown. In plants, short RNAs homologous to DNA sequences are known to initiate de novo methylation. To investigate whether short hairpin RNAs (shRNAs) may also serve as initiators for de novo methylation in human cells we have expressed short hairpin RNAs complementary to the CpG island including the promoter and early transcribed regions of the human RASSF1A gene. RASSF1A encodes a putative tumor suppressor that is hypermethylated in a variety of human cancers, whereas in some human cell lines, such as HeLa, RASSF1A is unmethylated and transcriptionally active. We demonstrate that shRNAs complementary to the RASSF1A promoter or early transcribed regions can direct low levels of de novo DNA methylation and partial gene silencing in HeLa cells. In contrast, an shRNA harboring four central mismatches with the target cannot direct such methylation. The results presented suggest provocative potential mechanisms for transcriptional gene silencing via DNA methylation in cancer cells.

La m&#xE9;thylation de cytosines dans des motifs CpG est un m&#xE9;canisme important pour la r&#xE9;gulation &#xE9;pig&#xE9;n&#xE9;tique de l&#x26;#39;expression des g&#xE8;nes dans les cellules de mammif&#xE8;res. L&#x26;#39;&#xE9;v&#xE9;nement initiateur (s) pour la m&#xE9;thylation de novo dans les cellules de mammif&#xE8;res, en particulier dans le cancer, est inconnue. Chez les plantes, les ARN courts homologues &#xE0; des s&#xE9;quences d&#x26;#39;ADN sont connus pour d&#xE9;clencher la m&#xE9;thylation de novo. La metilazione di cytosines in motivi CpG &egrave; un importante meccanismo per la regolazione epigenetica dell&#39;espressione genica in cellule di mammifero. Gli eventi di avvio per de novo metilazione nelle cellule dei mammiferi, in particolare nel cancro, sono sconosciuto. Nelle piante, breve RNAs omologhe alle sequenze del DNA sono noti per avviare de novo metilazione. Per indagare se short hairpin RNAs (shRNAs) pu&ograve; anche fungere da iniziatori per de novo metilazione nelle cellule umane abbiamo espresso short hairpin RNAs complementari all&#39;isola CpG compreso il promotore e primi regioni trascritti del gene umano RASSF1A. RASSF1A codifica un soppressore tumorale putativo che &egrave; hypermethylated in una variet&agrave; di tumori umani, considerando che in alcune linee cellulari umane, come ad esempio HeLa, RASSF1A &egrave; transcriptionally active e non metilato. Dimostriamo che shRNAs complementari al promotore di RASSF1A o prime regioni trascritti pu&ograve; dirigere bassi livelli di metilazione del DNA di de novo e silenziamento genico parziale in cellule HeLa. Al contrario, un shRNA harboring quattro disadattamento centrale con l&#39;obiettivo non pu&ograve; dirigere tale metilazione. I risultati presentati suggeriscono provocatori possibili meccanismi per silenziamento genico transcriptional tramite metilazione del DNA nelle cellule tumorali.

RNA interference occurs when cytoplasmic small interfering RNAs (siRNAs) enter the RNA-induced silencing complex and one strand guides cleavage of the target RNA by the Argonaute 2 protein. A significant concern when applying siRNAs or expressing small hairpin RNAs (shRNAs) in human cells is activation of the interferon (IFN) response. Synthetic siRNAs harboring certain motifs can induce an immune response when delivered to mouse and human immune cells such as peripheral blood mononuclear cells, monocytes, plasmacytoid dendritic cells (pDCs) and nonplasmacytoid dendritic cells (mDCs). In the present study we have tested the immunostimulatory effects of lipid-delivered siRNAs versus Pol III promoter-expressed shRNAs in primary CD34+ progenitor-derived hematopoietic cells. We show that in this system, lipid-delivered siRNAs are potent inducers of IFNalpha and type I IFN gene expression, whereas the same sequences when expressed endogenously are nonimmunostimulatory.

Stable expression of shRNAs in human CD34+ progenitor cells can avoid induction of interferon responses to siRNAs in vitro.

Combinatorial therapies for the treatment of HIV infection have changed the course of the AIDS epidemic in developed nations where the antiviral drug combinations are readily available. Despite this progress, there are many problems associated with chemotherapy for AIDS including toxicities and emergence of viral mutants resistant to the drugs. Our goal has been the development of a hematopoietic gene therapy treatment for HIV infection. Like chemotherapy, gene therapy for treatment of HIV infection should be used combinatorially. We have thus combined three different inhibitory genes for treatment of HIV infection into a single lentiviral vector backbone. The inhibitory agents engage RNAi via a short hairpin RNA targeting HIV tat/rev mRNAs, a nucleolar localizing decoy that binds and sequesters the HIV Tat protein, and a ribozyme that cleaves and downregulates the CCR5 chemokine receptor used by HIV for cellular entry. This triple combination has proven to be highly effective for inhibiting HIV replication in primary hematopoietic cells, and is currently on track for human clinical application.

Unser Ziel war die Entwicklung einer h&auml;matopoetischen gen-Therapie-Behandlung f&uuml;r HIV-Infektion. Wie Chemotherapie sollten combinatorially Gentherapie zur Behandlung der HIV-Infektion verwendet werden. Wir haben also drei verschiedene hemmende Gene zur Behandlung der HIV-Infektion in einem einzigen Lentiviralen Vektor-Backbone kombiniert. Die hemmenden Agenten engagieren RNAi &uuml;ber eine kurze Haarnadel-RNA, HIV Tat/Umdrehung mRNAs, ein nukleol&auml;res Lokalisierung von K&ouml;der, der bindet und das HIV-Tat-Protein, Sequester und ein Ribozym, das spaltet und Downregulates den CCR5-Chemokin-Rezeptor von HIV f&uuml;r zellul&auml;re Eintrag verwendet. Lentiviral vectors are able to transduce non-dividing cells and maintain sustained long-term expression of the transgenes. Many cell types including brain, liver, muscle, and hematopoietic stem cells have been successfully transduced with lentiviral vectors carrying a variety of genes. These properties make lentiviral vectors attractive vehicles for delivering small interfering RNA (siRNA) genes into mammalian cells. RNA polymerase III (Pol III) promoters are most commonly used for expressing siRNAs from lentiviral vectors. Pol III promoters are relatively small, have high activity, and use simple termination signals of short stretches of U. It is possible to include several Pol III expression cassettes in a single lentiviral vector backbone to express different siRNAs or to combine siRNAs with other transgenes. This chapter describes the delivery of Pol III promoted siRNAs by HIV-based lentiviral vectors and covers vector design, production, and verification of siRNA expression and function. This chapter should be useful for establishing a lentiviral vector-based delivery of siRNAs in experiments that require long-term gene knockdown or developing siRNA-based approaches for gene therapy applications.

The anti-multiple myeloma (MM) efficacy of bortezomib has led to the development of other proteasome inhibitors (PI), including CEP-18770 which has shown anti-MM effects in preclinical studies. However, the efficacy of orally (PO) or intravenously (IV) administered CEP-18770 in multiple MM models and in combination with conventional anti-MM therapies has not been evaluated. Herein, we show that CEP-18770 combined with melphalan or bortezomib induces synergistic inhibition of MM cell viability in vitro. In MM xenograft models, the addition of CEP-18770 IV to melphalan completely prevented the growth of both melphalan-sensitive and melphalan-resistant tumours. The combination of CEP-18770 IV and bortezomib induced complete regression of bortezomib-sensitive tumours and markedly delayed progression of bortezomib-resistant tumours compared to treatment with either agent alone. Single agent CEP-18770 PO also showed marked anti-MM effects in these xenograft models. These studies provide strong preclinical rationale for further development of this novel PI in the treatment of MM as a monotherapy as well as combined with either melphalan or bortezomib.

The gene archaerhodopsin-3 (Arch) from Halorubrum sodomense enables near-100% silencing of neurons in the awake brain when virally expressed in the mouse cortex and illuminated with yellow light. Arch mediates currents of several hundred picoamps at low light powers, and supports neural silencing currents approaching 900 pA at light powers easily achievable in vivo. Furthermore, Arch spontaneously recovers from light-dependent inactivation, unlike light-driven chloride pumps that enter long-lasting inactive states in response to light. These properties of Arch are appropriate to mediate the optical silencing of significant brain volumes over behaviourally relevant timescales. Arch function in neurons is well tolerated because pH excursions created by Arch illumination are minimized by self-limiting mechanisms to levels comparable to those mediated by channelrhodopsins or natural spike firing. To highlight how proton pump ecological and genomic diversity may support new innovation, we show that the blue-green light-drivable proton pump from the fungus Leptosphaeria maculans (Mac) can, when expressed in neurons, enable neural silencing by blue light, thus enabling alongside other developed reagents the potential for independent silencing of two neural populations by blue versus red light. Light-driven proton pumps thus represent a high-performance and extremely versatile class of 'optogenetic' voltage and ion modulator, which will broadly enable new neuroscientific, biological, neurological and psychiatric investigations.

AIDS patients who develop lymphoma are often treated with transplanted hematopoietic progenitor cells. As a first step in developing a hematopoietic cell-based gene therapy treatment, four patients undergoing treatment with these transplanted cells were also given gene-modified peripheral blood-derived (CD34(+)) hematopoietic progenitor cells expressing three RNA-based anti-HIV moieties (tat/rev short hairpin RNA, TAR decoy, and CCR5 ribozyme). In vitro analysis of these gene-modified cells showed no differences in their hematopoietic potential compared with nontransduced cells. In vitro estimates of successful expression of the anti-HIV moieties were initially as high as 22% but declined to approximately 1% over 4 weeks of culture. Ethical study design required that patients be transplanted with both gene-modified and unmanipulated hematopoietic progenitor cells obtained from the patient by apheresis. Transfected cells were successfully engrafted in all four infused patients by day 11, and there were no unexpected infusion-related toxicities. Persistent vector expression in multiple cell lineages was observed at low levels for up to 24 months, as was expression of the introduced small interfering RNA and ribozyme. Therefore, we have demonstrated stable vector expression in human blood cells after transplantation of autologous gene-modified hematopoietic progenitor cells. These results support the development of an RNA-based cell therapy platform for HIV.

Als ersten Schritt bei der Entwicklung einer h&auml;matopoetischen Zellen basierender gen-Therapie erhielten vier Patienten die Behandlung mit diesen transplantierten Zellen auch Gen-modifizierte periphere Blut abgeleitet (CD34(+)) h&auml;matopoetischen Vorl&auml;uferzellen drei Anti-HIV-RNA-basierte Adsorptionseigenschaften (Tat/u kurze Haarnadel RNA, TAR K&ouml;der und CCR5 Ribozym) zum Ausdruck zu bringen. In-vitro-Analyse dieser gen ver&auml;nderten Zellen zeigte keine Unterschiede in ihrem h&auml;matopoetischen Potential im Vergleich zu nontransduced Zellen. In-vitro-Sch&auml;tzungen der Ausdruck von der Anti-HIV-Adsorptionseigenschaften erfolgreich waren anf&auml;nglich immerhin 22 % sank auf etwa 1 % &uuml;ber 4 Wochen Kultur Ethische Studiendesign ben&ouml;tigt, dass Patienten mit beiden gen-modifiziert und Zellebene h&auml;matopoetischen Vorl&auml;uferzellen, die die vom Patienten durch Apherese transplantiert werden. Transfected Zellen wurden erfolgreich bei allen vier Patienten Infusion von Tag 11 Abbau, und es gab keine unerwarteten Infusion-bezogene Toxizit&auml;ten. Persistente Vektor Ausdruck in mehrere &Uuml;bertragungslinien der Zelle wurde auf niedrigem Niveau f&uuml;r bis zu 24 Monaten beobachtet, als Ausdruck der eingef&uuml;hrten small interfering RNA und Ribozym war. RNA-basierten Gentherapie f&uuml;r HIV mit Lentiviralen Vektor-modifizierte CD34(+) Zellen bei Patienten Transplantation zur AIDS-Bek&auml;mpfung-Lymphoms.

Pazienti affetti da AIDS che sviluppano il linfoma sono spesso trattati con le cellule progenitrici ematopoietiche trapiantate. Come primo passo nello sviluppo di un trattamento di terapia genica basate sulle cellule ematopoietiche, quattro pazienti sottoposti a trattamento con queste cellule trapiantate erano anche dato gene modificato derivato dal sangue (CD34(+)) progenitrici ematopoietiche cellule periferiche che esprimono tre moiety anti-HIV RNA-based (tat/rev short hairpin RNA decoy TAR e ribozima CCR5). Analisi in vitro di queste cellule modificate gene ha mostrato alcuna differenza nella loro potenziale ematopoietico confrontato con cellule nontransduced. Stime in vitro di successo expression le moiety anti-HIV sono stati inizialmente alte come il 22% ma &egrave; diminuita di circa l&#39;1% oltre 4 settimane della cultura. Disegno dello studio etico richiesto che i pazienti da trapiantare con entrambe le cellule progenitrici ematopoietiche gene modificato e manipolate ottenute dal paziente mediante aferesi. Celle transfected sono stata trapiantate con successo in tutti e quattro i pazienti infusi di giorno 11, e non c&#39;erano nessun imprevisto tossicit&agrave; correlati a infusione. Epigenetic mechanisms regulating leukemia stem cells (LSCs) are an attractive target for therapy of blood cancers. Here, we report that conditional knockout of the DNA methyltransferase Dnmt1 blocked development of leukemia, and haploinsufficiency of Dnmt1 was sufficient to delay progression of leukemogenesis and impair LSC self-renewal without altering normal hematopoiesis. Haploinsufficiency of Dnmt1 resulted in tumor suppressor gene derepression associated with reduced DNA methylation and bivalent chromatin marks. These results suggest that LSCs depend on not only active expression of leukemogenic programs, but also DNA methylation-mediated silencing of bivalent domains to enforce transcriptional repression.
