Vibrio fischeri is the exclusive symbiont residing in the light organ of the squid Euprymna scolopes. To understand the genetic requirements for this association, we searched a library of V. fischeri transposon insertion mutants for those that failed to colonize E. scolopes. We identified four mutants that exhibited severe defects in initiating colonization. Sequence analysis revealed that the strains contained insertions in four different members of a cluster of 21 genes oriented in the same direction. The predicted gene products are similar to proteins involved in capsule, exopolysaccharide or lipopolysaccharide biosynthesis, including six putative glycosyltransferases. We constructed mutations in five additional genes and found that they also were required for symbiosis. Therefore, we have termed this region syp, for symbiosis polysaccharide. Homologous clusters also exist in Vibrio parahaemolyticus and Vibrio vulnificus, and thus these genes may represent a common mechanism for promoting bacteria-host interactions. Using lacZ reporter fusions, we observed that transcription of the syp genes did not occur under standard laboratory conditions, but could be induced by multicopy expression of sypG, which encodes a response regulator with a predicted sigma54 interaction domain. This induction depended on sigma54, as a mutation in rpoN abolished syp transcription. Primer extension analysis supported the use of putative sigma54 binding sites upstream of sypA, sypI and sypM as promoters. Finally, we found that multicopy expression of sypG resulted in robust biofilm formation. This work thus reveals a novel group of genes that V. fischeri controls through a sigma54-dependent response regulator and uses to promote symbiotic colonization.

A novel, conserved cluster of genes promotes symbiotic colonization and sigma-dependent biofilm formation by Vibrio fischeri.

Vibrio fischeri est le symbiote exclusif r&eacute;sidant dans l&#39;orgue lumineux de la seiche Euprymna scolopes. Pour comprendre les g&eacute;n&eacute;tiques requis par cette association, nous avons recherch&eacute; une biblioth&egrave;que de V. fischeri mutants d&#39;insertion de transposon pour ceux qui ont &eacute;chou&eacute; &agrave; coloniser scolopes E.. Nous avons identifi&eacute; quatre mutants qui pr&eacute;sentaient de graves d&eacute;fauts dans le d&eacute;clenchement de la colonisation. Le s&eacute;quen&ccedil;age a r&eacute;v&eacute;l&eacute; que les souches contenaient des insertions dans quatre diff&eacute;rents membres d&#39;un groupe de 21 g&egrave;nes orient&eacute;s dans la m&ecirc;me direction. Les produits des g&egrave;nes pr&eacute;dits sont similaires &agrave; des prot&eacute;ines impliqu&eacute;es dans la biosynth&egrave;se de capsule, exopolysaccharide ou lipopolysaccharide, y compris six glycosyltransf&eacute;rases putatifs. Homologe Cluster existieren auch in Vibrio Parahaemolyticus und Vibrio Vulnificus und somit k&ouml;nnen diese Gene einen gemeinsamen Mechanismus f&uuml;r die F&ouml;rderung von Bakterien-Host Interaktionen vertreten. Mit LacZ Reporter Fusionen, beobachteten wir, dass Transkription der Syp Gene nicht unter Laborbedingungen Norm auftreten, aber konnte durch multicopy Ausdruck des SypG, das eine Antwort-Regler mit einer prognostizierten sigma54 Interaktion Dom&auml;ne codiert induziert werden. Diese Induktion hing von sigma54, als eine Mutation in RpoN abgeschafft Syp Transkription. Grundierung Erweiterung Analyse unterst&uuml;tzt die Verwendung von vermeintlichen sigma54 Sites stromaufw&auml;rts von SypA, SypI und SypM als Veranstalter verbindlich. Schlie&szlig;lich fanden wir, dass eine robuste Biofilm-Bildung multicopy Ausdruck des SypG f&uuml;hrte. Diese Arbeit zeigt somit eine neuartige Gruppe von Genen, die V. Vibrio fischeri &egrave; il simbionte esclusivo che risiedono nell&#39;organo leggero di calamari Euprymna scolopes. Per comprendere i requisiti genetici per questa associazione, abbiamo cercato una libreria di V. fischeri mutanti di inserimento trasposone per coloro che non &egrave; riuscito a colonizzare E. scolopes. Abbiamo individuato quattro mutanti che esposte gravi difetti in avvio di colonizzazione. Analisi di sequenza ha rivelato che i ceppi contenevano inserimenti in quattro diversi membri di un cluster di 21 geni orientato nella stessa direzione. Mediante fusioni reporter lacZ, abbiamo osservato che trascrizione di syp geni non si &egrave; verificato in condizioni standard di laboratorio, ma potrebbero essere indotta da modo espressione di sypG, che codifica per un regolatore di risposta con un dominio di interazione sigma54 predetto. Questa induzione dipendeva da sigma54, come una mutazione nella trascrizione syp del rpoN abolito. Analisi di estensione dell&#39;iniettore supportato l&#39;uso di sigma54 putativi siti a monte del sypA, sypI e sypM come promotori di legame. Infine, abbiamo trovato che gestisce espressione di sypG ha portato a formazione di biofilm robusto. Two-component signal transduction systems are utilized by prokaryotic and eukaryotic cells to sense and respond to environmental stimuli, both to maintain homeostasis and to rapidly adapt to changing conditions. Studies have begun to emerge that utilize a large-scale mutagenesis approach to analyzing these systems in prokaryotic organisms. Due to the recent availability of its genome sequence, such a global approach is now possible for the marine bioluminescent bacterium Vibrio fischeri, which exists either in a free-living state or as a mutualistic symbiont within a host organism such as the Hawaiian squid species Euprymna scolopes. In this work, we identified 40 putative two-component response regulators encoded within the V. fischeri genome. Based on the type of effector domain present, we classified six as NarL type, 13 as OmpR type, and six as NtrC type; the remaining 15 lacked a predicted DNA-binding domain. We subsequently mutated 35 of these genes via a vector integration approach and analyzed the resulting mutants for roles in bioluminescence, motility, and competitive colonization of squid. Through these assays, we identified three novel regulators of V. fischeri luminescence and seven regulators that altered motility. Furthermore, we found 11 regulators with a previously undescribed effect on competitive colonization of the host squid. Interestingly, five of the newly characterized regulators each affected two or more of the phenotypes examined, strongly suggesting interconnectivity among systems. This work represents the first large-scale mutagenesis of a class of genes in V. fischeri using a genomic approach and emphasizes the importance of two-component signal transduction in bacterium-host interactions.

Zwei-Komponenten-Signal-Transduktion-Systeme sind durch prokaryotischen und eukaryotischen Zellen Sinn verwendet und reagieren auf Umwelt Reize, Hom&ouml;ostase aufrechtzuerhalten und sich schnell an ver&auml;nderte Bedingungen anzupassen. Studien haben begonnen, entstehen zu lassen, die einen gro&szlig; angelegten Mutagenese-Ansatz zur Analyse dieser Systeme in prokaryotischen Organismen nutzen. Aufgrund der j&uuml;ngsten Verf&uuml;gbarkeit seiner Genom-Sequenz kann solch ein globales Konzept jetzt f&uuml;r die marine biolumineszierende Bakterium Vibrio Fischeri, die entweder in einem freilebenden Zustand oder als ein mutualistischen Symbiont in einen Wirtsorganismus, z. Gli studi hanno cominciato ad emergere che utilizzano un approccio di mutagenesi su larga scala all&#39;analisi di questi sistemi negli organismi eucarioti. Grazie alla recente disponibilit&agrave; della sua sequenza del genoma, un tale approccio globale &egrave; ora possibile per il batterio bioluminescente marino Vibrio fischeri, che esiste in uno stato libero-vivente o come un simbionti mutualistici all&#39;interno di un organismo ospite come le specie di calamaro hawaiano Euprymna scolopes. Sistemas de transducci&oacute;n de se&ntilde;al de dos componentes son utilizados por las c&eacute;lulas procariotas y eucariotas sentido y responden a los est&iacute;mulos ambientales, tanto para mantener la homeostasis y adaptarse r&aacute;pidamente a las condiciones cambiantes. Estudios han comenzado a surgir que utilizan un enfoque de mutag&eacute;nesis a gran escala para el an&aacute;lisis de estos sistemas en organismos procari&oacute;ticos. Debido a la reciente disponibilidad de su secuencia de genoma, un enfoque ahora es posible para la Marina bioluminiscente bacteria Vibrio fischeri, que existe en un estado de vida libre o como un simbionte mutualista dentro de un organismo hu&eacute;sped tales como la especie de calamar hawaiano Euprymna scolopes. En este trabajo, se identificaron 40 reguladores de la supuesta respuesta de dos componentes codificados en el genoma V. fischeri. Basado en el tipo de dominio efector presente, seis como tipo NarL, 13 como tipo de OmpR, clasificamos y seis como NtrC tipo; los 15 restantes carec&iacute;an de un dominio de uni&oacute;n al ADN previsto. Posteriormente se mut&oacute; 35 de estos genes mediante un enfoque de integraci&oacute;n del vector y analizaron a los mutantes resultantes para papeles en bioluminiscencia, motilidad y colonizaci&oacute;n competitiva de calamar. A trav&eacute;s de estos ensayos, se identificaron tres nuevos reguladores de V. fischeri luminiscencia y siete reguladores que alteraci&oacute;n la motilidad. Adem&aacute;s, se encontraron 11 reguladores con un efecto de Sines en colonizaci&oacute;n competitiva del calamar anfitri&oacute;n. Curiosamente, cinco de los reguladores recientemente caracterizados cada afectados dos o m&aacute;s de los fenotipos examinados, sugiriendo fuertemente la interconectividad entre sistemas. Este trabajo representa la primera mutag&eacute;nesis a gran escala de una clase de genes en V. fischeri utilizando un enfoque gen&oacute;mico y subraya la importancia de la transducci&oacute;n de la se&ntilde;al de dos componentes en las interacciones de la bacteria hu&eacute;sped.

Two-component signal transduction systems, composed of sensor kinase (SK) and response regulator (RR) proteins, allow bacterial cells to adapt to changes such as environmental flux or the presence of a host. RscS is an SK required for Vibrio fischeri to initiate a symbiotic partnership with the Hawaiian squid Euprymna scolopes, likely due to its role in controlling the symbiosis polysaccharide (syp) genes and thus biofilm formation. To determine which RR(s) functions downstream of RscS, we performed epistasis experiments with a library of 35 RR mutants. We found that several RRs contributed to RscS-mediated biofilm formation in V. fischeri. However, only the syp-encoded symbiosis regulator SypG was required for both biofilm phenotypes and syp transcription induced by RscS. These data support the hypothesis that RscS functions upstream of SypG to induce biofilm formation. In addition, this work also revealed a role for the syp-encoded RR SypE in biofilm formation. To our knowledge, no other study has used a large-scale epistasis approach to elucidate two-component signaling pathways. Therefore, this work both contributes to our understanding of regulatory pathways important for symbiotic colonization by V. fischeri and establishes a paradigm for evaluating two-component pathways in the genomics era.

RscS functions upstream of SypG to control the syp locus and biofilm formation in Vibrio fischeri.

Syst&egrave;mes de transduction de signal bi-composants, compos&eacute;s de kinase de capteur (SK) et de prot&eacute;ines de r&eacute;ponse de r&eacute;gulateur (RR), permettent &agrave; des cellules bact&eacute;riennes &agrave; s&#39;adapter aux changements tels que les flux environnementaux ou la pr&eacute;sence d&#39;un h&ocirc;te. Cependant, il fallait seulement le r&eacute;gulateur de la symbiose cod&eacute; syp SypG pour les ph&eacute;notypes de biofilm et transcription syp induite par la CTF. Ces donn&eacute;es supportent l&#39;hypoth&egrave;se que les fonctions RscS en amont de SypG pour induire la formation de biofilm. En outre, ce travail a &eacute;galement r&eacute;v&eacute;l&eacute; un r&ocirc;le pour le codage syp RR SypE dans la formation de biofilm. &Agrave; notre connaissance, aucune autre &eacute;tude n&#39;a utilis&eacute; une approche &agrave; grande &eacute;chelle d&#39;&eacute;pistasie pour &eacute;lucider les voies de signalisation bi-composants. RscS ist ein SK f&uuml;r Vibrio Fischeri erforderlich, eine symbiotische Partnerschaft mit der hawaiischen Tintenfisch Euprymna Scolopes, wahrscheinlich wegen seiner Rolle bei der Kontrolle der Symbiose Polysaccharid (Syp) Gene und somit Biofilm-Bildung zu initiieren. Um festzustellen, welche RR(s) Funktionen stromabw&auml;rts des RscS, f&uuml;hrten wir Epistase Experimente mit einer Bibliothek von 35 RR-Mutanten. Wir fanden, dass mehrere RRs RscS-vermittelte Biofilm Formation in V. Fischeri beigetragen. Allerdings war nur der Syp-codierte Symbiose-Regler SypG f&uuml;r Biofilm Ph&auml;notypen und Syp Transkription induziert durch RscS erforderlich. Diese Daten unterst&uuml;tzen der Hypothese, RscS Funktionen oberhalb des SypG Biofilm-Bildung zu veranlassen. Diese Arbeit zeigte dar&uuml;ber hinaus auch eine Rolle f&uuml;r die Syp-codierte RR-Skype im Biofilm-Bildung. Unseres Wissens hat keine andere Studie einen gro&szlig; angelegte Epistase-Ansatz verwendet, um zwei-Komponenten-Signalwege zu erhellen. Daher diese Arbeit sowohl tr&auml;gt zu unserem Verst&auml;ndnis der Regulationsmechanismen wichtig f&uuml;r symbiotische Kolonisierung durch V. RscS &egrave; un SK necessaria per Vibrio fischeri di avviare una collaborazione simbiotica con il calamaro hawaiano Euprymna scolopes, probabilmente a causa del suo ruolo nel controllo della simbiosi polisaccaride (syp) geni e quindi formazione di biofilm. Sistemas de transducci&oacute;n de la se&ntilde;al de dos componentes, compuestos de sensor cinasa (SK) y prote&iacute;nas del regulador (RR) de respuesta, permiten a las c&eacute;lulas bacterianas para adaptarse a los cambios tales como flujo ambiental o la presencia de un host. RSC es un SK para Vibrio fischeri para iniciar una asociaci&oacute;n simbi&oacute;tica con el calamar hawaiano Euprymna scolopes, probablemente debido a su papel en el control de los genes de polisac&aacute;rido (syp) de simbiosis y as&iacute; la formaci&oacute;n de biopel&iacute;culas. Para determinar qu&eacute; funciones de RR(s) aguas abajo de la RSC, realizamos experimentos de epistasis con una biblioteca de 35 mutantes de RR. Encontramos que varios RRs contribuyeron a la formaci&oacute;n de biofilm mediada por RSC en V. fischeri. Colonization of the Hawaiian squid Euprymna scolopes by the marine bacterium Vibrio fischeri requires the symbiosis polysaccharide (syp) gene cluster, which contributes to symbiotic initiation by promoting biofilm formation on the surface of the symbiotic organ. We previously described roles for the syp-encoded response regulator SypG and an unlinked gene encoding the sensor kinase RscS in controlling syp transcription and inducing syp-dependent cell-cell aggregation phenotypes. Here, we report the involvement of an additional syp-encoded regulator, the putative sensor kinase SypF, in promoting biofilm formation. Through the isolation of an increased activity allele, sypF1, we determined that SypF can function to induce syp transcription as well as a variety of biofilm phenotypes, including wrinkled colony formation, adherence to glass, and pellicle formation. SypF1-mediated transcription of the syp cluster was entirely dependent on SypG. However, the biofilm phenotypes were reduced, not eliminated, in the sypG mutant. These phenotypes were also reduced in a mutant deleted for sypE, another syp-encoded response regulator. However, SypF1 still induced phenotypes in a sypG sypE double mutant, suggesting that SypF1 might activate another regulator(s). Our subsequent work revealed that the residual SypF1-induced biofilm formation depended on VpsR, a putative response regulator, and cellulose biosynthesis. These data support a model in which a network of regulators and at least two polysaccharide loci contribute to biofilm formation in V. fischeri.

Microbial symbioses are essential for the normal development and growth of animals. Often, symbionts must be acquired from the environment during each generation, and identification of the relevant symbiotic partner against a myriad of unwanted relationships is a formidable task. Although examples of this specificity are well-documented, the genetic mechanisms governing it are poorly characterized. Here we show that the two-component sensor kinase RscS is necessary and sufficient for conferring efficient colonization of Euprymna scolopes squid by bioluminescent Vibrio fischeri from the North Pacific Ocean. In the squid symbiont V. fischeri ES114, RscS controls light-organ colonization by inducing the Syp exopolysaccharide, a mediator of biofilm formation during initial infection. A genome-level comparison revealed that rscS, although present in squid symbionts, is absent from the fish symbiont V. fischeri MJ11. We found that heterologous expression of RscS in strain MJ11 conferred the ability to colonize E. scolopes in a manner comparable to that of natural squid isolates. Furthermore, phylogenetic analyses support an important role for rscS in the evolution of the squid symbiosis. Our results demonstrate that a regulatory gene can alter the host range of animal-associated bacteria. We show that, by encoding a regulator and not an effector that interacts directly with the host, a single gene can contribute to the evolution of host specificity by switching 'on' pre-existing capabilities for interaction with animal tissue.

Mikrobielle Symbiosen sind essentiell f&uuml;r die normale Entwicklung und das Wachstum der Tiere. Oft m&uuml;ssen Symbionten aus der Umgebung erworben werden, w&auml;hrend jeder Generation und Identifizierung der relevanten symbiotische Partner gegen eine Vielzahl von unerw&uuml;nschten Beziehungen ist eine gewaltige Aufgabe. Beispiele f&uuml;r diese Besonderheit gut dokumentiert sind, zeichnen sich die genetischen Mechanismen, die EZB es schlecht. Hier zeigen wir, dass das zwei-Komponenten-Sensor-Kinase RscS notwendig und hinreichend f&uuml;r die Verleihung von effizienten Kolonisation Euprymna Scolopes Tintenfisch durch phosphoreszierende Vibrio Fischeri aus n&ouml;rdlichen Pazifik. In der Squid-Symbionten V. Fischeri ES114 steuert RscS Licht-Orgel Kolonisation durch induzieren die Syp Exopolysaccharide, ein Vermittler von Biofilm-Bildung bei der Erstinfektion. Ein Vergleich der Genom-Ebene offenbart, dass RscS, obwohl im Tintenfisch Symbionten, fehlt aus der Fisch-Symbionten V. Fischeri MJ11. Wir fanden, dass heterologe Ausdruck des RscS Belastung MJ11 die F&auml;higkeit, E. Scolopes in gewissem Sinne vergleichbar mit der nat&uuml;rlichen Tintenfisch-Isolate zu kolonisieren verliehen. Phylogenetische Analysen unterst&uuml;tzen dar&uuml;ber hinaus eine wichtige Rolle f&uuml;r RscS in der Evolution der Tintenfisch-Symbiose. Unsere Ergebnisse zeigen, dass ein Gen Regulatory Wirtsbereich von Tier-assoziierte Bakterien ver&auml;ndern k&ouml;nnen. Vibrios are natural inhabitants of aquatic environments and form symbiotic or pathogenic relationships with eukaryotic hosts. Recent studies reveal that the ability of vibrios to form biofilms (i.e. matrix-enclosed, surface-associated communities) depends upon specific structural genes (flagella, pili and exopolysaccharide biosynthesis) and regulatory processes (two-component regulators, quorum sensing and c-di-GMP signaling). Here, we compare and contrast mechanisms and regulation of biofilm formation by Vibrio species, with a focus on Vibrio cholerae, Vibrio parahaemolyticus, Vibrio vulnificus and Vibrio fischeri. Although many aspects are the same, others differ dramatically. Crucial questions that remain to be answered regarding the molecular underpinnings of Vibrio biofilm formation are also discussed.

Los vibrios son habitantes naturales de los ambientes acu&#xE1;ticos y forman relaciones simbi&#xF3;ticas o pat&#xF3;genas con hu&#xE9;spedes eucariotas. Estudios recientes revelan que la capacidad de los vibrios para formar biopel&#xED;culas (es decir, la matriz cerrados, superficie asociada a las comunidades) depende de determinados genes estructurales (flagelos, pili y la bios&#xED;ntesis de exopolisac&#xE1;ridos) y los procesos reguladores de dos componentes (reguladores, la detecci&#xF3;n de qu&#xF3;rum y de c-di -GMP de se&#xF1;alizaci&#xF3;n). En este sentido, comparar y contrastar los mecanismos y la regulaci&#xF3;n de la formaci&#xF3;n de biopel&#xED;culas por especies de Vibrio, con un enfoque en el Vibrio cholerae, Vibrio parahaemolyticus, Vibrio vulnificus y Vibrio fischeri. Aunque muchos aspectos son los mismos, otros difieren dram&#xE1;ticamente. Las cuestiones cruciales que quedan por responder en relaci&#xF3;n con los fundamentos moleculares de la formaci&#xF3;n de biopel&#xED;culas Vibrio tambi&#xE9;n se discuten.
