TRPV4 C1 &#x7F1D;&#x9699;&#x901A;&#x9053;&#x7535;&#x751F;&#x7406;&#x7279;&#x6027;&#x3002;

We previously reported that TRPV4 and TRPC1 can co-assemble to form heteromeric TRPV4-C1 channels [12]. In the present study, we characterized some basic electrophysiological properties of heteromeric TRPV4-C1 channels. 4Î±-Phorbol 12,13-didecanoate (4Î±-PDD, a TRPV4 agonist) activated a single channel current in HEK293 cells co-expressing TRPV4 and TRPC1. The activity of the channels was abrogated by a TRPC1-targeting blocking antibody T1E3. Conductance of the channels was ~95pS for outward currents and ~83pS for inward currents. The channels with similar conductance were also recorded in cells expressing TRPV4-C1 concatamers, in which assembled channels were expected to be mostly 2V4:2C1. Fluorescence Resonance Energy Transfer (FRET) experiments confirmed the formation of a protein complex with 2V4:2C1 stoichiometry while suggesting an unlikeliness of 3V4:1C1 or 1V4:3C1 stoichiometry. Monovalent cation permeability profiles were compared between heteromeric TRPV4-C1 and homomeric TRPV4 channels. For heteromeric TRPV4-C1 channels, their permeation profile was found to fit to Eisenman sequence VI, indicative of a strong field strength cation binding site, whereas for homomeric TRPV4 channels, their permeation profile corresponded to Eisenman sequence IV for a weak field strength binding site. Compared to homomeric TRPV4 channels, heteromeric TRPV4-C1 channels were slightly more permeable to Ca2+ and had a reduced sensitivity to extracellular Ca2+ inhibition. In summary, we found that, when TRPV4 and TRPC1 were co-expressed in HEK293 cells, the predominate assembly type was 2V4:2C1. The heteromeric TRPV4-C1 channels display distinct electrophysiological properties different from those of homomeric TRPV4 channels.

Pour les canaux h&eacute;t&eacute;rom&eacute;rique TRPV4-C1, leur profil de perm&eacute;ation s&#39;est av&eacute;r&eacute; apte &agrave; Eisenman s&eacute;quence VI, indicatif d&#39;un cation fort champ de force binding site, tandis que pour les canaux homom&egrave;res TRPV4, leur profil de perm&eacute;ation correspondait &agrave; Eisenman s&eacute;quence IV pour un site de liaison de la force de champ faible. Par rapport aux canaux homom&egrave;res TRPV4, h&eacute;t&eacute;rom&eacute;rique TRPV4-C1 cha&icirc;nes &eacute;taient l&eacute;g&egrave;rement plus perm&eacute;ables au Ca2 + et avaient une sensibilit&eacute; r&eacute;duite &agrave; l&#39;inhibition de Ca2 + extracellulaire. En r&eacute;sum&eacute;, nous avons constat&eacute; que, quand TRPV4 et TRPC1 ont &eacute;t&eacute; co-exprim&eacute;e dans les cellules HEK293, le type de montage pr&eacute;dominant &eacute;tait 2V4:2 C 1. Zuvor berichteten wir, dass TRPV4 und TRPC1 Form heteromeren TRPV4-C1 Kan&auml;le [12] co-assemble k&ouml;nnen. Fluoreszenz Resonanz Energie Transfer (FRET) Experimente best&auml;tigt die Bildung eines Proteins Komplex mit 2V4:2 C 1 St&ouml;chiometrie zwar vorgeschlagen, eine Unlikeliness der 3V4:1 C 1 oder 1V4:3 C 1 St&ouml;chiometrie. Monovalenten kation Durchl&auml;ssigkeit Profile wurden zwischen heteromeren TRPV4-C1 und Homomeric TRPV4 Kan&auml;le verglichen. F&uuml;r heteromeren TRPV4-C1-Kan&auml;le fand ihre Permeation-Profil passen zu Eisenman Sequenz VI, bezeichnend f&uuml;r ein starkes Feld-St&auml;rke-kation, Website, verbindlich, w&auml;hrend f&uuml;r Homomeric TRPV4 Kan&auml;le, ihr Profil Permeation Eisenman Sequenz IV f&uuml;r ein schwaches Feld-St&auml;rke-Bindungsstelle entsprach. Im Vergleich zu Homomeric TRPV4 Kan&auml;le, heteromeren TRPV4-C1-Kan&auml;le waren etwas durchl&auml;ssiger zu Ca2 + und eine geringere Empfindlichkeit gegen&uuml;ber extrazellul&auml;re Ca2 + Hemmung. Zusammenfassend l&auml;sst sich sagen fanden wir, dass ich bei TRPV4 und TRPC1 co-expressed in HEK293 Zellen waren die Montageart Gregorianische 2V4:2 C 1. Die heteromeren TRPV4-C1-Kan&auml;le zeigen verschiedene elektrophysiologische Eigenschaften von denen des Homomeric TRPV4 Kan&auml;le.

Elektrophysiologische Eigenschaften des heteromeren TRPV4-C1-Kan&auml;le.

Abbiamo segnalato in precedenza che TRPV4 e TRPC1 pu&ograve; co-assemble a canali di forma eteromerico TRPV4-C1 [12]. Divulgamos previamente que TRPV4 y TRPC1 pueden co-assemble a los canales de TRPV4-C1 forma heterom&eacute;ricos [12]. En el presente estudio, hemos caracterizado algunas propiedades electrofisiol&oacute;gicas b&aacute;sicas de canales heterom&eacute;ricos TRPV4-C1. 4&Alpha;-forbol 12, 13-didecanoate (4&alpha;-PDD, un agonista TRPV4) activa un solo canal actual en c&eacute;lulas HEK293 co-expressing TRPV4 y TRPC1. La actividad de los canales fue abrogada por un anticuerpo bloqueo metas de TRPC1 T1E3. Conductancia de los canales fue ~ 95pS para corrientes hacia afuera y ~ 83pS por corrientes hacia adentro. Tambi&eacute;n se registraron los canales con conductancia similar en las c&eacute;lulas que expresan TRPV4-C1 concatamers, en que canales montados deb&iacute;an ser en su mayor&iacute;a 2V4:2 C 1. Experimentos de transferencia de energ&iacute;a de resonancia (FRET) fluorescencia confirmaron la formaci&oacute;n de una prote&iacute;na compleja con 2V4:2 C 1 estequiometr&iacute;a mientras sugiriendo un unlikeliness de 3V4:1 C 1 o 1V4:3 C 1 estequiometr&iacute;a. Perfiles de permeabilidad de cationes monovalentes se compararon entre heterom&eacute;ricos TRPV4-C1 y los canales homomeric TRPV4. Canales heterom&eacute;ricos TRPV4-C1, su perfil de impregnaci&oacute;n se han encontrado para ajustarse a Eisenman secuencia VI, indicativo de un cati&oacute;n de campo fuerte fuerza vinculante sitio, mientras que para los canales homomeric TRPV4, su perfil de impregnaci&oacute;n correspondi&oacute; a Eisenman secuencia IV para un sitio de uni&oacute;n de fuerza de campo d&eacute;bil. En comparaci&oacute;n con los canales homomeric TRPV4, heterom&eacute;ricos TRPV4-C1 canales fueron ligeramente m&aacute;s permeables a Ca2 + y tuvieron una disminuci&oacute;n de la sensibilidad a la inhibici&oacute;n de Ca2 + extracelular. En Resumen, encontramos que, cuando TRPV4 y TRPC1 eran co-expressed en c&eacute;lulas HEK293, el tipo de Asamblea predominante era 2V4:2 C 1.
