Treatment with inhaled carbon monoxide (CO) has been shown to ameliorate bowel dysmotility caused by surgical manipulation of the gut in experimental animals. We hypothesized that administration of CO dissolved in lactated Ringer's solution (CO-LR) might provide similar protection to that observed with the inhaled gas while obviating some of its inherent problems. Postoperative gut dysmotility (ileus) was induced in mice by surgical manipulation of the small intestine. Some mice were treated with a single intraperitoneal dose of CO-LR immediately after the surgical procedure, whereas other mice received only the LR vehicle. Twenty-four hours later, intestinal transit of a nonabsorbable marker (70-kDa fluorescein isothiocyanate-labeled dextran) was delayed in mice subjected to intestinal manipulation but not the sham procedure. Gut manipulation also was associated with increased expression within the muscularis propria of transcripts for interleukin-1beta, cyclooxygenase-2, inducible nitric-oxide synthase, intracellular adhesion molecule-1, and Toll-like receptor-4, as well as infiltration of the muscularis propria with polymorphonuclear leukocytes and activation of mitogen-activated protein kinases and nuclear factor-kappaB. All of these effects were attenuated by treatment with CO-LR. The salutary effect of CO-LR on gut motility, as well as many of the anti-inflammatory effects of CO-LR, was diminished by treatment with a soluble guanylyl cyclase (sGC) inhibitor, suggesting that the effects of CO are mediated via activation of sGC. These data support the view that a single intraperitoneal dose of CO-LR ameliorates postoperative ileus in mice by inhibiting the inflammatory response in the gut wall induced by surgical manipulation, possibly in a sGC-dependent fashion.

In hemorrhagic shock and trauma, patients are prone to develop systemic inflammation with remote organ dysfunction, which is thought to be caused by pro-inflammatory mediators. This study investigates the role of the immuno-modulatory cytokine IL-10 in the development of organ dysfunction following hemorrhagic shock. Male C57/BL6 and IL-10 KO mice were subjected to volume controlled hemorrhagic shock for 3h followed by resuscitation. Animals were either sacrificed 3 or 24h after resuscitation. To assess systemic inflammation, serum IL-6, IL-10, KC, and MCP-1 concentrations were measured with the Luminex multiplexing platform; acute lung injury (ALI) was assessed by pulmonary myeloperoxidase (MPO) activity and lung histology and acute liver injury was assessed by hepatic MPO activity, hepatic IL-6 levels, and serum ALT levels. There was a trend towards increased IL-6 and KC serum levels 3h after resuscitation in IL-10 KO as compared to C57/BL6 mice; however this did not reach statistical significance. Serum MCP-1 levels were significantly increased in IL-10 KO mice 3 and 24 h following resuscitation as compared to C57/BL6 mice. In IL-10 KO mice, pulmonary MPO activity was significantly increased 3 h following resuscitation and after 24 h histological signs of acute lung injury were more apparent than in C57/BL6 mice. In contrast, no significant differences in any liver parameters were detected between IL-10 KO and C57/BL6 mice. Our data indicate that an endogenous IL-10 deficiency augments acute lung but not liver injury following hemorrhagic shock.

In shock emorragico e traumi, i pazienti sono inclini a sviluppare l&#39;infiammazione sistemica con disfunzione di organo remoto, che &egrave; pensata per essere causato da mediatori pro-infiammatori. Livelli sierici di MCP-1 sono stati notevolmente aumentati di IL-10 KO topi 3 e 24 h dopo rianimazione rispetto ai topi C57/BL6. Nei topi IL-10 KO, polmonare MPO attivit&agrave; era significativamente aumentata 3 h dopo rianimazione e dopo 24 h erano pi&ugrave; apparenti che nei topi C57/BL6 segni istologici di danno polmonare acuto. Al contrario, differenze significative nei parametri epatici sono state rilevate tra IL-10 KO e C57/BL6 topi. I nostri dati indicano che un deficit di IL-10 endogeno aumenta polmonare acuto ma lesioni epatiche non seguito shock emorragico.

En shock hemorr&aacute;gico y trauma, los pacientes son propensos a desarrollar inflamaci&oacute;n sist&eacute;mica con la disfunci&oacute;n del &oacute;rgano de control remoto, que se cree para ser causado por mediadores proinflamatorios. Para evaluar la inflamaci&oacute;n sist&eacute;mica, suero IL-6, se midieron concentraciones de MCP-1, IL-10 y KC con el Luminex multiplexaci&oacute;n plataforma; lesi&oacute;n pulmonar aguda (LPA) se evalu&oacute; mediante histolog&iacute;a pulmonar mieloperoxidasa (MPO) de actividad y del pulm&oacute;n y lesi&oacute;n del higado aguda evaluaron la actividad hep&aacute;tica de la MPO, hep&aacute;ticos niveles de IL-6 y niveles de ALT. Hubo una tendencia hacia el aumento de IL-6 y los niveles de suero KC 3 h despu&eacute;s de la resucitaci&oacute;n en IL-10 KO en comparaci&oacute;n con ratones C57/BL6; sin embargo esto no alcanz&oacute; significaci&oacute;n estad&iacute;stica. MCP-1 s&eacute;rico se incrementaron significativamente en IL-10 KO ratones 3 y 24 h despu&eacute;s de la resucitaci&oacute;n en comparaci&oacute;n con ratones C57/BL6. En ratones KO de IL-10, actividad MPO pulmonar fue significativamente mayor 3 h despu&eacute;s de la resucitaci&oacute;n y despu&eacute;s de 24 horas fueron m&aacute;s evidentes que en ratones C57/BL6 muestras histol&oacute;gicas de la lesi&oacute;n pulmonar aguda. Por el contrario, se detectaron diferencias significativas en los par&aacute;metros de h&iacute;gado entre ratones KO de IL-10 y C57/BL6. Significant improvements of graft and patient survival have been achieved over the past 20 years in the field of intestinal transplantation. Tacrolimus monotherapy with corticosteroids, or in combination with sirolimus is the most commonly used immunosuppressive regimen. Early (24h) after experimental allogenic small bowel transplantation in rats, sirolimus reduces the cellular and molecular inflammatory response with subsequent graft dysmotility more efficiently than tacrolimus, with contrary effects at 7 days after transplantation. This study evaluates three immunosuppressive strategies in the post-acute phase after intestinal transplantation - tacrolimus or sirolimus monotherapy and the combination therapy of tacrolimus with infliximab. After orthotopic intestinal transplantation between Brown Norway and Lewis rats, animals received 14 days of immunosuppressive treatment. Histology, infiltration of neutrophils, monocytes and macrophages, cytokine and mediator mRNA expression (real time RT-PCR) and smooth muscle function in a standard organ bath were assessed at 14 days after transplantation in all treatment groups and isogenic controls. Allogenic transplanted rats without immunosuppressive therapy and non-transplanted animals served as further control. Tacrolimus prevented acute rejection and graft dysmotility more effectively (p â‰¥ 0.05) than sirolimus. Reduced mRNA expression levels of CD4, IFN-Î³, IL-6, IL-10, iNOS, NFÎºB, TNF-Î± and MCP-1 (p â‰¤ 0.05) were observed in tacrolimus treated animals compared to sirolimus. Additional infliximab application did not influence the cellular and molecular inflammatory response in the post-acute phase after transplantation. In conclusion, the severe cellular and molecular inflammatory response in allogenic transplanted grafts without immunosuppressive therapy is ameliorated by all three immunosuppressive regimens, but tacrolimus was found to be more efficient than sirolimus at 14 days after transplantation. Our findings do not rule out the usage of sirolimus as single immunosuppressive therapy, but indicate and confirm the potency and effectivity of tacrolimus as basis immunosuppressant in the field of intestinal transplantation.

Fr&uuml;he (24 Stunden) nach der experimentellen allogene D&uuml;nndarm-Transplantation bei Ratten, reduziert Sirolimus die zellul&auml;re und Molekulare entz&uuml;ndliche Reaktion mit nachfolgenden Transplantat Dysmotility effizienter als Tacrolimus, mit gegenteilige Wirkungen bei 7 Tage nach der Transplantation. Diese Studie wertet drei immunsuppressive Strategien in der post-acute Phase nach der Darm-Transplantation - Tacrolimus oder Sirolimus Monotherapie und Kombinationstherapie von Tacrolimus mit Infliximab. Nach Orthotopic intestinale Transplantation zwischen Brown Norwegen und Lewis-Ratten erhielt Tiere 14 Tagen immunsuppressive Behandlung. Histologie, Infiltration von Neutrophilen, Monozyten und Makrophagen, Cytokine und Mediator mRNA Ausdruck (Real-Time RT-PCR) und glatten Muskel-Funktion in einem standard-Orgel-Bad wurden 14 Tage nach der Transplantation in allen Behandlungsgruppen und isogene Kontrollen beurteilt. Primi (24 h) dopo trapianto di intestino tenue allogenico sperimentale nei ratti, sirolimus riduce la risposta infiammatoria cellulare e molecolare con innesto successiva alterazione della motilit&agrave; in modo pi&ugrave; efficiente di tacrolimus, con effetti contrari a 7 giorni dopo il trapianto. Questo studio valuta tre strategie immunosoppressive in fase acuta dopo trapianto intestinale - tacrolimus o sirolimus monoterapia e la terapia di combinazione di tacrolimus con infliximab. Dopo il trapianto ortotopico intestinale tra Norvegia Brown e ratti Lewis, animali ha ricevuto 14 giorni di trattamento immunosoppressivo. Tacrolimus impedito acuta alterazione della motilit&agrave; rifiuto e innesto in modo pi&ugrave; efficace (p &ge; 0.05) rispetto a sirolimus. Ridotti livelli di espressione di mRNA di CD4, IFN-&gamma; IFN-&gamma;-6, IL-10, iNOS, NF&kappa;B, TNF-&alpha; e MCP-1 (p &le; 0,05) sono stati osservati in tacrolimus animali trattati rispetto al sirolimus. Applicazione aggiuntiva infliximab non ha influenzato la risposta infiammatoria cellulare e molecolare in fase acuta dopo trapianto. In conclusione, la severa risposta infiammatoria cellulare e molecolare nel trapianto allogenico innesti trapiantati senza terapia immunosoppressiva &egrave; migliorata da tutti i tre regimi immunosoppressivi, ma tacrolimus &egrave; stato trovato per essere pi&ugrave; efficiente di sirolimus a 14 giorni dopo il trapianto. I nostri risultati non esclude l&#39;uso di sirolimus come singola terapia immunosoppressiva, ma indicare e confermare la potenza e l&#39;efficacia del tacrolimus come immunosoppressore di base nel campo del trapianto intestinale.

Se han logrado mejoras significativas de injerto y supervivencia de los pacientes en los &uacute;ltimos 20 a&ntilde;os en el campo del trasplante intestinal. Monoterapia con tacrolimus con corticosteroides, o en combinaci&oacute;n con sirolimus es el r&eacute;gimen inmunosupresor m&aacute;s utilizado. Temprana (24 horas) despu&eacute;s del trasplante de intestino al&oacute;geno experimental en ratas, sirolimus reduce la respuesta inflamatoria celular y molecular con dismotilidad posterior injerto m&aacute;s eficientemente que el tacrolimus, con efectos contrarios a los 7 d&iacute;as despu&eacute;s del trasplante. Este estudio eval&uacute;a tres estrategias inmunosupresoras en la fase post aguda despu&eacute;s del trasplante intestinal - monoterapia con tacrolimus o sirolimus y la terapia de combinaci&oacute;n de tacrolimus con infliximab. Despu&eacute;s del trasplante intestinal de orthotopic entre marr&oacute;n Noruega y las ratas de Lewis, los animales recibieron 14 d&iacute;as del tratamiento inmunosupresor. Histolog&iacute;a, infiltraci&oacute;n de neutr&oacute;filos, monocitos y macr&oacute;fagos, citocinas y expresi&oacute;n del mRNA de mediador (RT-PCR en tiempo real) y funci&oacute;n del m&uacute;sculo liso en un ba&ntilde;o de &oacute;rgano est&aacute;ndar se evaluaron a los 14 d&iacute;as despu&eacute;s del trasplante en todos los grupos de tratamiento y controles isog&eacute;nicas. Ratas trasplantadas al&oacute;genos sin terapia inmunosupresiva y animales trasplantados no sirven como control. Tacrolimus prevenido dismotilidad injerto y rechazo agudo m&aacute;s con eficacia (p &ge; 0,05) que sirolimus. Reduce los niveles de expresi&oacute;n de mRNA de CD4, IFN-&gamma; IFN-&gamma;-6, IL-10, iNOS, NF&kappa;B, TNF-&alpha; y MCP-1 (p &le; 0.05) fueron observados en tacrolimus animales tratados en comparaci&oacute;n con sirolimus. Aplicaci&oacute;n de infliximab adicional no influy&oacute; en la respuesta inflamatoria celular y molecular en la fase post aguda despu&eacute;s del trasplante. En conclusi&oacute;n, la respuesta inflamatoria celular y molecular severa en injertos al&oacute;genos trasplantados sin terapia inmunosupresiva es mejorada por todos tres reg&iacute;menes inmunosupresores, pero tacrolimus fue encontrado para ser m&aacute;s eficiente que sirolimus en 14 d&iacute;as despu&eacute;s del trasplante. Nuestros resultados no descartan el uso de sirolimus como &uacute;nico tratamiento inmunosupresor, pero indican y confirmar la potencia y efectividad de tacrolimus como inmunosupresor de base en el campo del trasplante intestinal.

CLINICAL BACKGROUND: In all patients undergoing abdominal surgery, a transient phase of interruption of bowel motility, named postoperative ileus (POI) occurs. POI is often accepted as an unavoidable "physiological" response and a self-limiting complication after surgery although it has a significant impact on patient morbidity with prolonged hospitalization and increased costs. Annual economic burden has been estimated as much as US $1.47 billion in the USA (Iyer et al. in J Manag Care Pharm 15(6):485-494, 2009). PATHOPHYSIOLOGY: The pathophysiology has been elucidated within the last decades, demonstrating that both, neurogenic and inflammatory mechanisms are involved in response to the surgical trauma. It is now generally accepted that POI pathogenesis processes in two phases: a first neurogenic phase is accountable for the immediate postoperative impairment of bowel motility. This is followed by a second immunological phase that can last for days and mainly affects strength and length of POI. More recent findings demonstrate a bidirectional interaction between the nervous and the immune system, and this interaction significantly contributed to our present understanding of POI pathophysiology. Although nerval mechanisms have a significant impact in the early phase of POI, the contribution of the immune system and subsequently its manipulation has risen as the most promising strategy in prevention or treatment of the clinically relevant prolonged form of POI. AIMS: The present manuscript will give an update on the inflammatory responses, the involved cell types, and participating immune mediators in POI.

POI est souvent accept&eacute; comme une r&eacute;ponse &laquo; physiologique &raquo; in&eacute;vitables et une complication spontan&eacute;ment vers la gu&eacute;rison apr&egrave;s la chirurgie, bien qu&#39;il ait un impact significatif sur la morbidit&eacute; patiente avec une hospitalisation prolong&eacute;e et une augmentation des co&ucirc;ts. Elle est suivie d&#39;une deuxi&egrave;me phase immunologique qui peut durer des jours et touche principalement la force et la longueur de POI. R&eacute;sultats plus r&eacute;cents montrent une interaction bidirectionnelle entre le syst&egrave;me nerveux et le syst&egrave;me immunitaire et cette interaction consid&eacute;rablement contribu&eacute; &agrave; notre compr&eacute;hension actuelle de la pathophysiologie POI. POI wird oft als eine unvermeidbare &quot;physiologisch&quot; und eine selbstbegrenzend Komplikation nach der Operation akzeptiert, obwohl es erhebliche Auswirkungen auf Patienten Morbidit&auml;t mit l&auml;ngerem Krankenhausaufenthalt und erh&ouml;hte Kosten hat. Es folgt eine zweite Phase der immunologischen, das kann Tage dauern und betrifft haupts&auml;chlich St&auml;rke und L&auml;nge des POI. Neuere Ergebnisse zeigen eine bidirektionale Interaktion zwischen Nerven und das Immunsystem und diese Interaktion zu unserem heutigen Verst&auml;ndnis der POI Pathophysiologie wesentlich beigetragen. Obwohl nervale Mechanismen in der fr&uuml;hen Phase der POI erhebliche Auswirkungen haben, ist der Beitrag des Immunsystems und sp&auml;ter seiner Manipulation als die vielversprechendsten Strategie in der Vorbeugung oder Behandlung von klinisch relevanten verl&auml;ngerte Form von POI gestiegen. PATOFISIOLOGIA: La fisiopatologia &egrave; stata chiarita all&#39;interno degli ultimi decenni, dimostrando che entrambi, neurogeno e infiammatori meccanismi sono coinvolti in risposta al trauma chirurgico. Oggi &egrave; generalmente accettato che la patogenesi POI i processi in due fasi: una prima fase neurogena &egrave; responsabile per il danno immediato postoperatorio della motilit&agrave; intestinale. ANTECEDENTES cl&iacute;nicos: En los pacientes sometidos a cirug&iacute;a abdominal, ocurre una fase transitoria de interrupci&oacute;n de la motilidad intestinal, llamada &iacute;leo postoperatorio (POI). POI se acepta como una respuesta &quot;fisiol&oacute;gica&quot; inevitable y una complicaci&oacute;n autolimitada a menudo despu&eacute;s de la cirug&iacute;a aunque tiene un impacto significativo sobre la morbilidad de paciente con hospitalizaci&oacute;n prolongada y el aumento de los costos. Carga econ&oacute;mica anual se ha estimado tanto como US $1,47 billones en los E.e.u.u. (Iyer et en J Manag Care Pharm 15 (6): 485-494, 2009). Fisiopatolog&iacute;a: La patofisiolog&iacute;a ha sido dilucidada en las &uacute;ltimas d&eacute;cadas, demostrando que est&aacute;n involucrados mecanismos tanto neurog&eacute;nicos e inflamatorios en respuesta al trauma quir&uacute;rgico. Ahora est&aacute; generalmente aceptado que la patogenesia POI procesos en dos fases: una primera fase neurog&eacute;nica es responsable por el deterioro postoperatorio inmediato de la motilidad intestinal. Aunque nerval mecanismos tienen un impacto significativo en la fase temprana de POI, la contribuci&oacute;n del sistema inmune y posteriormente su manipulaci&oacute;n ha aumentado como la estrategia m&aacute;s prometedora en la prevenci&oacute;n o el tratamiento de la forma prolongada cl&iacute;nicamente relevante de POI. OBJETIVOS: El presente manuscrito dar&aacute; una actualizaci&oacute;n en las respuestas inflamatorias, tipos celulares implicados y participantes mediadores inmunes en POI.

Ileus is caused by the initiation of a complex cascade of molecular and cellular inflammatory responses within the intestinal muscularis, which might be species specific. Our objective was to investigate a possible immunological divergence in the mechanisms of postoperative- and endotoxin-induced ileus in C57BL/6 mice and Sprague-Dawley rats. Gastrointestinal transit (GIT) was measured at 24 h after the injurious stimulus. MPO-staining and F4/80 immunohistochemistry were used to quantify polymorphonuclear and monocyte infiltration of jejunal muscularis whole-mounts, and intestinal muscularis MCP-1, ICAM-1 and iNOS gene expression was assessed by RT-PCR. Intestinal muscularis subjected to in vivo surgical manipulation (SM) or LPS treatment was cultured for 24 h, and the liberation of nitric oxide and chemokines/cytokines into the culture medium was analyzed by Griess reaction and Luminex multiplex assay. Intestinal SM and lipopolysaccharide (LPS) (15 mg/kg) caused a significant delay in gastrointestinal transit, which was more severe in mice compared to rats in both injury models. Both SM- and LPS-triggered neutrophil and monocytic extravasation into the rat jejunal muscularis exceeded the cellular infiltration seen in mice. These results correlated with significantly greater increases in rat muscularis MCP-1 (syn. CCL2), ICAM-1 and iNOS message with more subsequent NO production after SM or LPS compared to mouse. The cultured muscularis obtained from SM mice released significantly more inflammatory proteins such as TNF-Î±, IL-1-Î±, IL-4 and GM-CSF compared to the manipulated rat muscularis. In contrast, LPS initiated the secretion of significantly more IL-1Î² by the inflamed rat muscularis compared to the mouse, but GM-CSF (syn. CSF2) liberation from mouse muscularis was markedly higher compared to LPS-treated rat muscularis. The data indicate that mechanistically the development of ileus in rat is mediated predominately through a leukocytic pathway consisting of chemotaxis, cellular extravasation and NO liberation. Whereas, the more intense mouse ileus evolves via a potent but injury-specific local cytokine response.

Differential molecular and cellular immune mechanisms of postoperative and LPS-induced ileus in mice and rats.

Notre objectif &eacute;tait d&#39;&eacute;tudier une &eacute;ventuelle divergence immunologique dans les m&eacute;canismes de l&#39;il&eacute;us postop&eacute;ratoire endotoxine induite et C57BL/6 souris et rats Sprague-Dawley. Ileus wird durch die Einleitung eine komplexe Kaskade von molekularen und zellul&auml;ren entz&uuml;ndlichen Reaktionen innerhalb der intestinalen Muscularis, denen m&ouml;glicherweise bestimmte Arten verursacht. Unser Ziel war es, eine m&ouml;gliche immunologische Divergenz in die Mechanismen der postoperativen - und Endotoxin-induzierte Ileus in Sprague-Dawley Ratten und C57BL/6 M&auml;usen zu untersuchen. Magen-Darm-Transit (GIT) wurde nach den sch&auml;digenden Reiz an 24 h gemessen. MPO-F&auml;rbung und F4/80 Immunhistochemie wurden zur Polymorphonuclear und Monozyte Infiltration Jejunal Muscularis ganz-Halterungen zu quantifizieren und intestinale Muscularis MCP-1, ICAM-1 und iNOS Genexpression wurde ermittelt, indem die RT-PCR. Intestinale Muscularis einer in-vivo chirurgische Manipulation (SM) oder LPS-Behandlung unterzogen wurde f&uuml;r 24 Stunden kultiviert, und die Befreiung von Stickstoffmonoxid und Chemokinen/Zytokine in das Kulturmedium wurde von Griess-Reaktion und Luminex-multiplex-Probe analysiert. Intestinale SM und Lipopolysaccharid (LPS) (15 mg/kg) verursacht eine erhebliche Verz&ouml;gerung im Magen-Darm-Transit, die schwerere bei M&auml;usen, Ratten in beide Verletzung-Modelle gegen&uuml;ber war. Beide SM-LPS-ausgel&ouml;st und neutrophile und monozyt&auml;ren Paravasation in der Ratte Jejunal Muscularis &uuml;berschritten die zellul&auml;re Infiltration in M&auml;use gesehen. Diese Ergebnisse korreliert mit deutlich h&ouml;here Erh&ouml;hungen der Ratte Muscularis MCP-1 (SYN. CCL2), ICAM-1 und iNOS message mit mehr nachfolgende Nr. Produktion nach SM oder LPS im Vergleich zu Maus. Die kultivierten Muscularis gewonnenen SM M&auml;use ver&ouml;ffentlicht deutlich mehr entz&uuml;ndliche Proteine wie TNF-&alpha;, IL-1-&alpha;, IL-4 und GM-CSF im Vergleich zu den manipulierten Ratte Muscularis. Im Gegensatz dazu LPS initiierten die entz&uuml;ndeten Ratte-Muscularis, verglichen mit der Maus die Sekretion von deutlich mehr IL-1&beta;, aber GM-CSF (SYN. CSF2) Befreiung von Maus Muscularis war deutlich h&ouml;her im Vergleich zu LPS behandelten Ratten Muscularis. Die Daten zeigen, dass mechanistisch die Entwicklung von Ileus bei Ratte vermittelten &uuml;berwiegend durch eine leukocytic Pathway bestehend aus Chemotaxis, zellul&auml;re Paravasation und keine Befreiung. In der Erw&auml;gung, dass die intensiver Maus-Ileus &uuml;ber eine potente aber Verletzungen-spezifische lokale Zytokin-Antwort entwickelt.

Il nostro obiettivo era quello di indagare su una possibile divergenza immunologica nei meccanismi dell&#39;ileo postoperatorio ed endotossina-indotta in ratti Sprague-Dawley e topi C57BL/6. Transito gastrointestinale (GIT) &egrave; stata misurata a 24 ore dopo lo stimolo nocivo. MPO-colorazione e immunoistochimica F4/80 sono stati utilizzati per quantificare l&#39;infiltrazione di polimorfonucleati e monociti di digiunale muscolare della intero-monta, ed espressione di gene di MCP-1, ICAM-1 e iNOS muscolare intestinale &egrave; stata valutata mediante RT-PCR. Muscolare intestinale sottoposto a trattamento LPS o manipolazione chirurgica in vivo (SM) &egrave; stata colta per 24 h, e la liberazione di ossido nitrico e citochine/chemochine nel terreno di coltura &egrave; stata analizzata da reazione di Griess e dosaggio multiplex Luminex. Intestinale SM e lipopolisaccaride (LPS) (15 mg/kg) ha causato un notevole ritardo in transito gastrointestinale, che era pi&ugrave; grave nei topi rispetto ai ratti di entrambi i modelli lesioni. Entrambi stravaso SM e LPS innescato dal neutrofili e monocitica nel ratto digiunale muscolare ha superato l&#39;infiltrazione cellulare visto nei topi. La colta muscolare ottenuti da topi SM rilasciato proteine significativamente pi&ugrave; infiammatorie come TNF-&alpha;, IL-1-&alpha;, IL-4 e GM-CSF rispetto al muscolare ratto manipolati. Al contrario, LPS ha avviato la secrezione di significativamente pi&ugrave; IL-1 &beta; dal ratto infiammata muscolare rispetto al mouse, ma liberazione di GM-CSF (SYN. QCS2) da mouse muscolare era nettamente superiore rispetto alle muscolare di ratto trattate con LPS. I dati indicano che meccanicistico lo sviluppo dell&#39;ileo nel ratto &egrave; mediata prevalentemente attraverso un pathway leucocitari composto di chemiotassi, stravaso cellulare e nessuna liberazione. Considerando che, il pi&ugrave; intenso ileo del mouse si evolve attraverso una risposta potente ma lesioni specifiche citochine locali.

&Iacute;leo es causado por la iniciaci&oacute;n de una compleja cascada de respuestas inflamatorias moleculares y celulares dentro de los muscularis intestinales, que pueden ser espec&iacute;fico de especie. Nuestro objetivo era investigar una posible divergencia inmunol&oacute;gica en los mecanismos del &iacute;leo postoperatorio y endotoxina-inducida en ratones C57BL/6 y ratas Sprague-Dawley. Tr&aacute;nsito gastrointestinal (TGI) fue medido a las 24 h despu&eacute;s del est&iacute;mulo perjudicial. Coloraci&oacute;n de MPO y F4/80 immunohistochemistry fueron utilizados para cuantificar la infiltraci&oacute;n de polimorfonucleares y monocitos de yeyunal muscularis conjunto-montajes y muscularis intestinal MCP-1, ICAM-1 y iNOS expresi&oacute;n g&eacute;nica se evalu&oacute; mediante RT-PCR. Muscularis intestinal sometido a manipulaci&oacute;n quir&uacute;rgica in vivo (SM) ni LPS fue cultivado durante 24 h, y la liberaci&oacute;n de &oacute;xido n&iacute;trico y quimiocinas/citoquinas en el medio de cultivo fue analizada por reacci&oacute;n de Griess y an&aacute;lisis multiplex Luminex. Intestinal SM y lipopolisac&aacute;rido (LPS) (15 mg/kg) causaron una demora significativa en tr&aacute;nsito gastrointestinal, que era m&aacute;s grave en ratones en comparaci&oacute;n con ratas en ambos modelos de lesiones. Ambos extravasaci&oacute;n neutr&oacute;filos y monoc&iacute;tica SM y LPS-activadas en las rata yeyunal muscularis excedi&oacute; la infiltraci&oacute;n celular en ratones. Estos resultados correlacionaron con aumenta significativamente mayor en rata muscularis MCP-1 (Syn. CCL2), ICAM-1 y iNOS mensaje con NO m&aacute;s posterior producci&oacute;n despu&eacute;s de SM o LPS en comparaci&oacute;n con el rat&oacute;n. Los muscularis cultivados obtenidos de ratones SM lanz&oacute; prote&iacute;nas significativamente m&aacute;s inflamatorias como TNF-&alpha;, IL-1-&alpha;, IL-4 y GM-CSF en comparaci&oacute;n con los muscularis ratas manipuladas. Por el contrario, LPS iniciaron la secreci&oacute;n de significativamente m&aacute;s IL-1&beta; por los muscularis rata inflamado en comparaci&oacute;n con el rat&oacute;n, pero liberaci&oacute;n de GM-CSF (Syn. CSF2) de rat&oacute;n muscularis fue notablemente superior comparado con muscularis de rata tratada con LPS. Los datos indican que mec&aacute;nico el desarrollo del &iacute;leo en rata es predominantemente mediado a trav&eacute;s de una v&iacute;a leucoc&iacute;tica formado por quimiotaxis, extravasaci&oacute;n celular y ninguna liberaci&oacute;n.
