Entwicklung von Methoden zum Schutz von Organismen in Metall-verschmutzten Umgebungen ist von unserem Verst&auml;ndnis der Mechanismen der zellul&auml;ren Entgiftung abh&auml;ngig. In diesem Zusammenhang sind halb-Molek&uuml;l ATP verbindliche Kassette (ABC) Transporter der HMT-1-Unterfamilie f&uuml;r Kadmium (Cd) Entgiftung erforderlich. HMTs haben strukturelle Architektur konserviert, das unterscheidet sie von anderen ABC-Transporter und erm&ouml;glicht die Identifizierung von UCP1 in Genome verschiedener Arten, einschlie&szlig;lich des Menschen. Vor kurzem entdeckten wir, dass HMT-1 aus einfachen, einzelligen Organismus, Schizosaccharomyces Pombe, SpHMT1, unabh&auml;ngig von der Phytochelatine-Synthase (PCS agiert) und Cd, aber nicht andere Schwermetalle entgiftet. Ob HMTs aus vielzelligen Organismen Toleranz nur auf Cd oder auch andere Schwermetalle verleihen, ist nicht bekannt.

Developing methods for protecting organisms in metal-polluted environments is contingent upon our understanding of cellular detoxification mechanisms. In this regard, half-molecule ATP-binding cassette (ABC) transporters of the HMT-1 subfamily are required for cadmium (Cd) detoxification. HMTs have conserved structural architecture that distinguishes them from other ABC transporters and allows the identification of homologs in genomes of different species including humans. We recently discovered that HMT-1 from the simple, unicellular organism, Schizosaccharomyces pombe, SpHMT1, acts independently of phytochelatin synthase (PCS) and detoxifies Cd, but not other heavy metals. Whether HMTs from multicellular organisms confer tolerance only to Cd or also to other heavy metals is not known.

Preclinical studies of cranial radiation therapy (RT) using animal brain tumor models have been hampered by technical limitations in the delivery of clinically relevant RT. We established a bioimageable mouse model of glioblastoma multiforme (GBM) and an image-guided radiation delivery system that facilitated precise tumor localization and treatment and which closely resembled clinical RT. Our novel radiation system makes use of magnetic resonance imaging (MRI) and bioluminescent imaging (BLI) to define tumor volumes, computed tomographic (CT) imaging for accurate treatment planning, a novel mouse immobilization system, and precise treatments delivered with the Small Animal Radiation Research Platform. We demonstrated that, in vivo, BLI correlated well with MRI for defining tumor volumes. Our novel restraint system enhanced setup reproducibility and precision, was atraumatic, and minimized artifacts on CT imaging used for treatment planning. We confirmed precise radiation delivery through immunofluorescent analysis of the phosphorylation of histone H2AX in irradiated brains and brain tumors. Assays with an intravenous near-infrared fluorescent probe confirmed that radiation of orthografts increased disruption of the tumor blood-brain barrier (BBB). This integrated model system, which facilitated delivery of precise, reproducible, stereotactic cranial RT in mice and confirmed RT's resultant histologic and BBB changes, may aid future brain tumor research.

Estudios precl&iacute;nicos de radioterapia craneal (RT) utilizando modelos de tumores de cerebro de animales se han visto obstaculizados por las limitaciones t&eacute;cnicas en la entrega de RT cl&iacute;nicamente relevante. Hemos establecido un modelo de rat&oacute;n de bioimageable de glioblastoma multiforme (GBM) y un sistema de entrega de radiaci&oacute;n guiada por la imagen que facilit&oacute; la localizaci&oacute;n exacta del tumor y tratamiento y que cerca se asemej&oacute; cl&iacute;nico RT Nuestro sistema de radiaci&oacute;n novela hace uso de resonancia magn&eacute;tica (MRI) y bioluminiscente im&aacute;genes (BLI) para definir vol&uacute;menes de tumor, proyecci&oacute;n de imagen (CT) tomogr&aacute;fica computada para la planificaci&oacute;n del tratamiento exacto, un sistema de inmovilizaci&oacute;n de rat&oacute;n novela y tratamientos precisos entregan con la plataforma de investigaci&oacute;n de radiaci&oacute;n de Animal peque&ntilde;o. Hemos demostrado que, en vivo, BLI correlacionaron bien con MRI para definir vol&uacute;menes de tumor. Nuestro sistema de sujeci&oacute;n novela mejorado configuraci&oacute;n reproducibilidad y precisi&oacute;n, fue atraum&aacute;tica y minimiza artefactos en proyecci&oacute;n de imagen de CT utilizado para la planificaci&oacute;n del tratamiento. Se confirm&oacute; la entrega de radiaci&oacute;n precisas a trav&eacute;s de an&aacute;lisis inmunofluorescente de la fosforilaci&oacute;n de la histona H2AX en cerebro irradiado y tumores cerebrales. Ensayos con una sonda fluorescente del infrarrojo cercano intravenosa confirman que la radiaci&oacute;n de orthografts mayor interrupci&oacute;n de la barrera tumor blood - brain (BBB). Este sistema modelo integrado, lo que facilit&oacute; la entrega de RT craneal estereot&aacute;ctica, precisa y reproducible en ratones y confirm&oacute; el resultado histol&oacute;gico de RT y los cambios BBB, puede ayudar a investigaci&oacute;n de tumor cerebral futuro.

Un m&eacute;todo integrado para reproducible y preciso guiados por im&aacute;genes estereot&aacute;ctica irradiaci&oacute;n craneal de tumores cerebrales mediante la plataforma de investigaci&oacute;n de la radiaci&oacute;n de animales peque&ntilde;os.
