&#x57FA;&#x5E95;&#x8F6C;&#x5F55;&#x56E0;&#x5B50; TFIIB &#x7684;&#x94FE;&#x63A5;&#x5668;&#x57DF;&#x63A7;&#x5236;&#x4E0D;&#x540C;&#x7684;&#x5F81;&#x8058;&#x548C;&#x8F6C;&#x5F55;&#x523A;&#x6FC0;&#x529F;&#x80FD;&#x3002;

RNA polymerases (RNAPs) require basal transcription factors to assist them during transcription initiation. One of these factors, TFIIB, combines promoter recognition, recruitment of RNAP, promoter melting, start site selection and various post-initiation functions. The ability of 381 site-directed mutants in the TFIIB 'linker domain' to stimulate abortive transcription was systematically quantitated using promoter-independent dinucleotide extension assays. The results revealed two distinct clusters (mjTFIIB E78-R80 and mjTFIIB R90-G94, respectively) that were particularly sensitive to substitutions. In contrast, a short sequence (mjTFIIB A81-K89) between these two clusters tolerated radical single amino acid substitutions; short deletions in that region even caused a marked increase in the ability of TFIIB to stimulate abortive transcription ('superstimulation'). The superstimulating activity did, however, not correlate with increased recruitment of the TFIIB/RNAP complex because substitutions in a particular residue (mjTFIIB K87) increased recruitment by more than 5-fold without affecting the rate of abortive transcript stimulation. Our work demonstrates that highly localized changes within the TFIIB linker have profound, yet surprisingly disconnected, effects on RNAP recruitment, TFIIB/RNAP complex stability and the rate of transcription initiation. The identification of superstimulating TFIIB variants reveals the existence of a previously unknown rate-limiting step acting on the earliest stages of gene expression.

The linker domain of basal transcription factor TFIIB controls distinct recruitment and transcription stimulation functions.

Un de ces facteurs, TFIIB, combine la reconnaissance de promoteur, recrutement de RNAP, promoteur de fusion, commencer la s&eacute;lection du site et des diverses fonctions post-initiation. La capacit&eacute; de 381 mutants dirig&eacute;e dans le TFIIB &laquo; domaine de l&#39;&eacute;diteur de liens &raquo; pour stimuler la transcription avort&eacute;e a &eacute;t&eacute; dos&eacute;e syst&eacute;matiquement utilisant la prolongation dinucl&eacute;otide ind&eacute;pendant du promoteur. RNA-Polymerasen (RNAPs) erfordern basalen Transkriptionsfaktoren, ihnen zu helfen, w&auml;hrend Initiation der Transkription. Einer dieser Faktoren, TFIIB, kombiniert Projekttr&auml;ger Anerkennung, Rekrutierung von Name, Projekttr&auml;ger, die schmelzen, Standortwahl und verschiedenen post-initiation Funktionen zu starten. Die F&auml;higkeit von 381 Site-verwiesene Mutanten in der TFIIB &#39;Linker Dom&auml;ne&#39; F&ouml;rderung der erfolglosen Transkription wurde systematisch quantifiziert mit Projekttr&auml;ger-unabh&auml;ngige-dinucleotid Erweiterung Proben. Die Ergebnisse zeigten zwei verschiedene Cluster (MjTFIIB E78-R80 und MjTFIIB R90-G94, beziehungsweise), die besonders empfindlich auf Auswechslungen waren. Im Gegensatz dazu toleriert eine kurze Sequenz (MjTFIIB A81-K89) zwischen diesen zwei Clustern radikale einzelne Aminos&auml;ure Ersetzungen; kurze Deletionen in dieser Region verursacht sogar eine deutliche Zunahme in der F&auml;higkeit der TFIIB, abortive Transkription (&#39;Superstimulation&#39;) zu f&ouml;rdern. Die superstimulating Aktivit&auml;t, jedoch nicht mit verst&auml;rkte Rekrutierung von komplexen TFIIB/Name korrelieren, weil Ersetzungen in einer bestimmten R&uuml;ckstand (MjTFIIB K87) Rekrutierung von mehr als 5-fach erh&ouml;ht ohne Auswirkungen auf die Rate der erfolglosen Abschrift Stimulation. Unsere Arbeit zeigt, dass hoch lokalisierte &Auml;nderungen innerhalb der TFIIB-Linker tiefgreifende, dennoch &uuml;berraschend getrennt, auf Name Rekrutierung, TFIIB/Name Komplexstabilit&auml;t und die Rate der Initiation der Transkription auswirken. Die Identifizierung von superstimulating TFIIB Varianten zeigt die Existenz einer bisher unbekannten geschwindigkeitsbestimmende Schritt auf den fr&uuml;hesten Stadien der Genexpression.

Uno di questi fattori, TFIIB, unisce il riconoscimento del promotore, reclutamento di RNAP, promotore di fusione, inizio selezione del sito e varie funzioni di post-initiation. La capacit&agrave; di 381 mutanti sito-diretto nel TFIIB &#39;dominio linker&#39; per stimolare la trascrizione abortiva sistematicamente era quantificati utilizzando dosaggi estensione dinucleotide promotore indipendente. I risultati hanno rivelato due ammassi distinti (mjTFIIB E78-R80 e mjTFIIB G94-R90, rispettivamente) che sono stati particolarmente sensibili alle sostituzioni. Al contrario, una breve sequenza (mjTFIIB A81-K89) tra questi due ammassi tollerato radicale singolo aminoacidiche; breve delezioni in quella regione anche causato un marcato aumento nella capacit&agrave; di TFIIB per stimolare la trascrizione abortiva (&#39;superstimulation&#39;). L&#39;attivit&agrave; superstimulating, tuttavia, non correlare con aumentata assunzione del TFIIB/RNAP complesso perch&eacute; le sostituzioni in un particolare residuo (mjTFIIB K87) aumentato reclutamento di pi&ugrave; di 5-fold senza alterare la frequenza di stimolazione di trascrizione abortiva. Il nostro lavoro dimostra che modifiche altamente localizzate all&#39;interno del linker TFIIB hanno effetti profondi, eppure sorprendentemente disconnessi, sul reclutamento RNAP, TFIIB/RNAP complesso stabilit&agrave; e la velocit&agrave; di iniziazione della trascrizione. The TFIIB linker domain stimulates the catalytic activity of archaeal RNAP. By characterising a range of super-stimulating mutants we identified a novel rate-limiting step in transcription initiation. Our results help to interpret structural findings and pave the way towards higher-resolution structures of the RNAP-TFIIB linker interface.
