The terminal modification of glycans by Î²4 addition of N-acetylgalactosamine to N-acetylglucosamine with formation of the N,N-diacetyllactosediamine (LacdiNAc) moiety has been well documented for a number of N-linked glycoproteins and peptides, like neurohormones. Much less is known about O-glycoproteins in this regard because only human zona pellucida glycoprotein 3 (ZP3) and bovine proopiomelanocortin were reported to be LacdiNAc-modified. In searching for mammalian proteins modified with O-linked LacdiNAc we identified six positive species among nine endogenous and recombinant O-glycoproteins, which were extracellular matrix, or matrix-related proteins. These are ZP3 and the five novel LacdiNAc-positive species ECM1, AMACO, nidogen-1, Î±-dystroglycan, and neurofascin. The mass spectrometric analyses revealed a core 2-based tetrasaccharide as the common structural basis of O-linked LacdiNAc that could be further modified, similar to the type 2 LacNAc termini, with fucose, sialic acid, or sulfate. Here, we provide structural evidence for a novel type of mucin-type O-glycans that is strictly specific for LacdiNAc termini: sugar phosphorylation with formation of GalNAcÎ²1-4(phospho-)GlcNAc. The structural details of the phosphatase-labile compound were elucidated by MS(2) analysis of tetralysine complexes and by MS(n) measurements of the permethylated glycan alditols. Phospho-LacdiNAc was detected in human HEK-293 as well as in mouse myoblast cells and in bovine brain tissue.

O-linked N,N'-diacetyllactosamine (LacdiNAc)-modified glycans in extracellular matrix glycoproteins are specifically phosphorylated at subterminal N-acetylglucosamine.

En la b&uacute;squeda de prote&iacute;nas mam&iacute;feras modificadas con LacdiNAc O-ligado se identificaron seis especies positivas entre nueve end&oacute;genas y recombinantes O-glicoprote&iacute;nas, matriz extracelular, o prote&iacute;nas relacionadas con la matriz. Se trata de ZP3 y las cinco especies nuevas del LacdiNAc-positivo ECM1, AMACO, nidogen-1, &alpha;-dystroglycan y neurofascin. Los an&aacute;lisis de espectrometr&iacute;a de masa revelan un tetrasac&aacute;rido basadas en 2 de base como la base estructural com&uacute;n de LacdiNAc vinculados O que pod&iacute;a ser m&aacute;s modificada, similares a los t&eacute;rminos de la LacNAc del tipo 2, con fucosa, &aacute;cido si&aacute;lico o sulfato. Aqu&iacute;, nos proporcionan la evidencia estructural para un tipo nuevo de tipo mucina O-glicanos estrictamente espec&iacute;fico para LacdiNAc termini: az&uacute;car fosforilaci&oacute;n con formaci&oacute;n de GlcNAc GalNAc&beta;1-4(phospho-). The accumulation of an autofluorescent pigment called lipofuscin in neurons is an invariable hallmark of brain aging. So far, this material has been considered to be waste material without particular relevance for cellular pathology. However, two lines of evidence argue that lipofuscin may play a yet unidentified role for pathological cellular functions: (i) Genetic forms of premature accumulation of similar autofluorescent material in neuronal ceroid lipofuscinosis indicate a direct disease-associated link to lipofuscin; (ii) Retinal pigment epithelium cell lipofuscin is mechanistically linked to age-associated macular degeneration. Here, we purified autofluorescent material from the temporal and hippocampal cortices of three different human individuals by a two-step ultracentrifugation on sucrose gradients. For human brain lipofuscin, we could identify a common set of 49 (among > 200 total) proteins that are mainly derived from mitochondria, cytoskeleton, and cell membrane. This brain lipofuscin proteome was validated in an interspecies comparison with whole brain rat lipofuscin (total > 300 proteins), purified by the same procedure, yielding an overlap of 32 proteins (64%) between lipofuscins of both species. Our study is the first to characterize human and rat brain lipofuscin and identifies high homology, pointing to common cellular pathomechanisms of age-associated lipofuscin accumulation despite the huge (40-fold) difference in the lifespan of these species. Our identification of these distinct proteins will now allow research in disturbed molecular pathways during age-associated dysfunctional lysosomal degradation.

La acumulaci&oacute;n de un pigmento de autofluorescent llamado lipofuscina en las neuronas es un sello invariable del envejecimiento del cerebro. Hasta ahora, este material se ha considerado que el material de desecho sin relevancia particular para la patolog&iacute;a celular. Sin embargo, dos l&iacute;neas de evidencia argumentan que lipofuscina puede desempe&ntilde;ar un papel a&uacute;n no identificado para las funciones celulares patol&oacute;gicos: formas (i) gen&eacute;tica de la acumulaci&oacute;n de material autofluorescent similar en lipofuscinosis ceroide neuronal indican una relaci&oacute;n directa asociada a la enfermedad a la lipofuscina; (ii) pigmentario epitelio celular lipofuscin es mec&aacute;nico vinculada a asociados con la edad degeneraci&oacute;n macular. Aqu&iacute;, nos purifica autofluorescent material de las cortezas temporales e hipocampales de tres diferentes individuos humanos una ultracentrifugaci&oacute;n de dos etapas en gradientes de sacarosa. Para el cerebro humano lipofucsina, podr&iacute;amos identificar un conjunto com&uacute;n de 49 (entre &gt; 200 total) las prote&iacute;nas que se derivan principalmente de las mitocondrias, citoesqueleto y membrana de la c&eacute;lula. Este cerebro lipofuscina proteoma fue validado en una comparaci&oacute;n entre las especies con todo el cerebro rata lipofuscina (total &gt; 300 prote&iacute;nas), mediante el mismo procedimiento, que rinde una superposici&oacute;n de 32 prote&iacute;nas (64%) entre los lipofuscins de ambas especies. Nuestro estudio es el primero en caracterizar humana y lipofuscina del cerebro de la rata e identifica elevada homolog&iacute;a, se&ntilde;alando (aumentar&aacute;) pathomechanisms celular com&uacute;n de acumulaci&oacute;n de lipofuscina asociados con la edad a pesar de la enorme diferencia en la vida &uacute;til de estas especies. Nuestra identificaci&oacute;n de estas prote&iacute;nas distintas ahora permitir&aacute; investigaci&oacute;n en v&iacute;as moleculares perturbadas durante la degradaci&oacute;n lisosomal disfuncional asociados con la edad.
