Cocaine is a major stimulant drug which can have rewarding and locomotor activating effects. It is used by non-addicts to enhance concentration and performance in a work or social setting. It was suggested that cocaine may amplify the impact of mood and sensory stimulation on behaviour. Here we tested whether cocaine can enhance the impact of novel sensory stimulation on exploratory locomotor activity. In this study the effects of cocaine (0, 2.5, 5.0, 10mg/kg, i.p.) on the locomotor response induced by on-off light stimulation with different light intensities (0, 22, 83, 440 lx) was investigated. Visual stimulation increased locomotor activity and grooming behaviour. Cocaine more dramatically increased locomotion and rearing behaviour but suppressed grooming. Also, visual stimulation effects on grooming were reduced by cocaine. The additive relationship between the cocaine and visual stimulation effects on locomotion and rearing combined with their opposing impact upon grooming point to independent mechanisms mediating cocaine- and visual stimulation-induced behavioural activation.

Il est utilis&eacute; par les toxicomanes pour am&eacute;liorer la concentration et la performance dans un milieu social ou de travail. Il a &eacute;t&eacute; sugg&eacute;r&eacute; que la coca&iuml;ne peut amplifier l&#39;impact de l&#39;humeur et de stimulation sensorielle sur le comportement. Es wurde vermutet, dass Kokain die Auswirkungen von Stimmung und sensorische Stimulation Verhalten verst&auml;rken kann. Hier haben wir getestet, ob Kokain die Auswirkungen der neuartige sensorische Stimulation explorative lokomotorische T&auml;tigkeit verbessern kann. Inoltre, gli effetti di stimolazione visiva su toelettatura sono state ridotte da cocaina. Anesthesia describes a complex state composed of immobility, amnesia, hypnosis (sleep or loss of consciousness), analgesia, and muscle relaxation. Bottom-up approaches explain anesthesia by an interaction of the anesthetic with receptor proteins in the brain, whereas top-down approaches consider predominantly cortical and thalamic network activity and connectivity. Both approaches have a number of explanatory gaps and as yet no unifying view has emerged. In addition to a direct interaction with primary target receptor proteins, general anesthetics have massive effects on neurotransmitter activity in the brain. They can change basal transmitter levels by interacting with neuronal activity, transmitter synthesis, release, reuptake and metabolism. By that way, they can affect a great number of neurotransmitter systems and receptors. Here, we review how different general anesthetics affect extracellular activity of neurotransmitters in the brain during induction, maintenance, and emergence from anesthesia and which functional consequences this may have. Commonalities and differences between different groups of anesthetics in their action on neurotransmitter activity are discussed. We also review how general anesthetics affect the response dynamics of the neurotransmitter systems after sensory stimulation. More than 30 years of research have now yielded a complex picture of the effects of general anesthetics on brain neurotransmitter basal activity and response dynamics. It is suggested that analyzing the effects on neurotransmitter activity is the logical next step after protein interactions in a bottom-up analysis of anesthetic action in the brain on the way to a unifying view of anesthesia.

Enfoques Bottom-up explican anestesia por una interacci&oacute;n de la anestesia con las prote&iacute;nas del receptor en el cerebro, mientras que los enfoques top-down consideran conectividad y actividad de la red predominante tal&aacute;micas y corticales. Ambos enfoques tienen una serie de lagunas explicativas y que sin embargo no ha surgido ninguna visi&oacute;n unificadora. Adem&aacute;s de una interacci&oacute;n directa con prote&iacute;nas receptoras de objetivo principal, los anest&eacute;sicos generales tienen enormes efectos sobre la actividad de los neurotransmisores en el cerebro. Pueden cambiar los niveles basales transmisor interactuando con la actividad neuronal, s&iacute;ntesis de transmisor, liberaci&oacute;n, recaptaci&oacute;n y metabolismo. Por ello, que puedan afectar a un gran n&uacute;mero de sistemas de neurotransmisores y receptores. Aqu&iacute;, repasamos c&oacute;mo diferentes anest&eacute;sicos generales afectan actividad extracelular de los neurotransmisores en el cerebro durante la inducci&oacute;n, mantenimiento y aparici&oacute;n de anestesia y funcionales que las consecuencias que esto puede tener. Se discuten las similitudes y diferencias entre los distintos grupos de anest&eacute;sicos en su acci&oacute;n sobre la actividad de los neurotransmisores. Tambi&eacute;n repasamos c&oacute;mo anest&eacute;sicos generales afectan la din&aacute;mica de la respuesta de los sistemas de neurotransmisores despu&eacute;s del est&iacute;mulo sensorial. M&aacute;s de 30 a&ntilde;os de investigaci&oacute;n han dado ahora un cuadro complejo de los efectos de los anest&eacute;sicos generales en actividad basal del neurotransmisor cerebral y respuesta din&aacute;mica. S&#39;agissant de comportement en plein champ, footslips sur une grille horizontale et l&#39;asym&eacute;trie posturale moteur dans un cylindre ont &eacute;t&eacute; mises en recouvrement apr&egrave;s administration de L-DOPA ou v&eacute;hicule avec ou sans, benserazide pr&eacute;traitement. EN L-DOPA sans benserazide pr&eacute;traitement l&eacute;g&egrave;rement diminu&eacute; des tournures ipsilat&eacute;rale et augment&eacute; les tournures controlat&eacute;rales 10-20 min apr&egrave;s le traitement. EN L-DOPA avec pr&eacute;traitement saline r&eacute;duit controlat&eacute;rales membre ant&eacute;rieur-glissades sur la grille alors qu&#39;aucun effets se sont manifest&eacute;s dans le test du cylindre. l-3,4-Dihydroxyphenylalanine (L-DOPA) remains the most effective drug for therapy of Parkinson's disease. However, the current clinical route of L-DOPA administration is variable and unreliable because of problems with drug absorption and first-pass metabolism. Administration of drugs via the nasal passage has been proven an effective alternate route for a number of medicinal substances. Here we examined the acute behavioral and neurochemical effects of intranasally (IN) applied L-DOPA in rats bearing unilateral lesions of the medial forebrain bundle, with severe depletion (97%) of striatal dopamine. Turning behavior in an open field, footslips on a horizontal grid and postural motor asymmetry in a cylinder were assessed following IN L-DOPA or vehicle administration with, or without, benserazide pre-treatment. IN L-DOPA without benserazide pre-treatment mildly decreased ipsilateral turnings and increased contralateral turnings 10-20 min after the treatment. IN L-DOPA with saline pre-treatment reduced contralateral forelimb-slips on the grid while no effects were evident in the cylinder test. These results support the hypothesis that L-DOPA can bypass the blood-brain barrier by the IN route and alleviate behavioral impairments in the hemiparkinsonian animal model.

Hier untersuchten wir die akute Verhaltens- und neurochemische Wirkung von intranasally (IN) angewandte DOPA bei Ratten mit einseitige L&auml;sionen der medialen Stirnhirn-Bundle mit schweren Ersch&ouml;pfung (97 %) der Striatale Dopamin. Drehen Verhalten in ein offenes Feld, waren Footslips auf ein horizontales Raster und posturale motor Asymmetrie in einem Zylinder bewerteten folgende IN DOPA oder Fahrzeug-Administration mit oder ohne, Benserazid Vorbehandlung. IN DOPA ohne Benserazid Vorbehandlung leicht zur&uuml;ckgegangen ipsilateral Drehsp&auml;ne und erh&ouml;ht kontralateralen Drehsp&auml;ne 10-20 min nach der Behandlung. DOPA mit saline Vorbehandlung reduziert kontralateralen Vorderbein-Slips in der Startaufstellung w&auml;hrend keine Wirkungen in den Zylinder-Test offensichtlich waren. Diese Ergebnisse unterst&uuml;tzen die Hypothese, dass DOPA die Blut - Hirn-Schranke zu, indem die Route IN umgehen und verhaltensbasierte Beeintr&auml;chtigungen im Tiermodell Hemiparkinsonian lindern kann.

Intranasally angewendet lindert DOPA Parkinson-Symptome bei Ratten mit einseitigen Nigro-Striatale 6-OHDA L&auml;sionen.

l-3,4-diidrossifenilalanina (l-dopa) rimane il farmaco pi&ugrave; efficace per la terapia della malattia di Parkinson. Tuttavia, l&#39;attuale tragitto clinico della somministrazione di l-dopa &egrave; variabile e inaffidabile a causa di problemi con il metabolismo di assorbimento e di primo passaggio del farmaco. Somministrazione di farmaci attraverso il passaggio nasale ha dimostrato un efficacia percorso alternativo per un certo numero di sostanze medicamentose. Qui abbiamo esaminato gli effetti neurochimici e comportamentali acuti di intranasale (a) l-dopa applicata nei ratti cuscinetto unilaterali lesioni del fascio proencefalo mediale, con grave deplezione (97%) della dopamina striatali. Girando il comportamento in campo aperto, footslips su una griglia orizzontale e asimmetria posturale motore a un cilindro sono stato valutato seguente IN l-dopa o veicolo amministrazione con, o senza, benserazide pretrattamento. IN l-dopa senza benserazide pretrattamento lievemente diminuito torniture omolaterale e controlaterale torniture &egrave; aumentato 10-20 min dopo il trattamento. IN l-dopa con pretrattamento salino ridotto arto controlaterale-scivola sulla griglia di partenza mentre effetti non erano evidente nel test di cilindro. Questi risultati supportano l&#39;ipotesi che l-dopa pu&ograve; bypassare la barriera emato - encefalica dalla rotta IN e alleviare i disturbi comportamentali nel modello animale hemiparkinsonian.

Intranasale applicata l-dopa allevia i sintomi parkinsoniani in ratti con lesioni 6-OHDA unilaterale nigro-striatali.

l-3, 4-dihidroxifenilalanina (l-dopa) sigue siendo la droga m&aacute;s eficaz para el tratamiento de la enfermedad de Parkinson. Sin embargo, la v&iacute;a cl&iacute;nica actual de administraci&oacute;n de l-dopa es variable y poco confiable debido a problemas con el metabolismo de absorci&oacute;n y el primer paso de la droga. Administraci&oacute;n de f&aacute;rmacos a trav&eacute;s de las fosas nasales ha demostrado una ruta alternativa eficaz para un n&uacute;mero de sustancias medicinales. Aqu&iacute; examinamos los efectos agudos de la conducta y neuroqu&iacute;micos de intranasal (en) l-dopa aplicada en ratas con lesiones unilaterales del haz Prosencef&aacute;lico medial, con agotamiento severo (97%) de la dopamina estriatal. Comportamiento que da vuelta en un campo abierto, footslips en una rejilla horizontal y asimetr&iacute;a postural motor en un cilindro fueron evaluados despu&eacute;s en l-dopa o veh&iacute;culo administraci&oacute;n con o sin tratamiento previo de benserazide. EN l-dopa sin benserazide pretratamiento suavemente disminuy&oacute; rodajes ipsolaterales y aument&oacute; rodajes contralaterales 10-20 minutos despu&eacute;s del tratamiento. EN l-dopa con pretratamiento salino hab&iacute;a reducido contralateral miembro anterior-se desliza en la red mientras que ningunos efectos eran evidentes en la prueba de cilindro. Estos resultados apoyan la hip&oacute;tesis que la l-dopa puede saltarse la barrera hemato - encef&aacute;lica por la ruta IN y aliviar problemas conductuales en el modelo animal de hemiparkinsonian.
