Immunization with naked plasmid DNA elicits strong cell-mediated immune responses. In the present study, we examine strategies to enhance epitope-specific cytotoxic T lymphocyte (CTL) responses using DNA constructs, expressing a minimal class I epitope of the gp120 of HIV-IIIB. Here, we evaluate the effect of CD4+ T cell (T(H)) epitope affinity for the MHC II molecule on the immunogenicity of our DNA vaccines. Our data indicate that a low-affinity T(H) epitope decreased the magnitude of the CTL responses. In addition, we observed decreased numbers of epitope-specific T helper cells and CTLs, as well as diminished cytokine secretion and proliferative responses. Thus, the immunogenicity of a DNA epitope vaccine can be modulated by altering the affinity of the T(H) epitope.

CD4+ T cell epitope affinity to MHC II influences the magnitude of CTL responses elicited by DNA epitope vaccines.

Immunisation avec l&#39;ADN de plasmide nu suscite de fortes r&eacute;ponses &agrave; m&eacute;diation cellulaire immunitaire. Dans la pr&eacute;sente &eacute;tude, nous examinons les strat&eacute;gies visant &agrave; am&eacute;liorer l&#39;&eacute;pitope sp&eacute;cifique des lymphocytes T cytotoxiques (CTL) r&eacute;ponses en utilisant des constructions de l&#39;ADN, exprimant un nombre minimal de classe I &eacute;pitope de la gp120 du VIH-IIIB. Ici, nous &eacute;valuons l&#39;effet des cellules T CD4 + (T(H)) &eacute;pitope affinit&eacute; pour la mol&eacute;cule MHC II sur l&#39;immunog&eacute;nicit&eacute; des vaccins de notre ADN. Nos donn&eacute;es indiquent qu&#39;un faible affinit&eacute; T(H) &eacute;pitope a diminu&eacute; l&#39;ampleur des r&eacute;ponses CTL. In der vorliegenden Studie untersuchen wir Strategien zur Verbesserung der Epitop-spezifische zytotoxische T-Lymphozyten (CTL) Antworten Konstrukte der DNA, zum Ausdruck bringen eine minimale Klasse I Epitope von gp120 des HIV-IIIB. Hier bewerten wir die Wirkung der CD4 + T-Zellen (T(H))-Epitop-Affinit&auml;t f&uuml;r das MHC-II-Molek&uuml;l auf der Immunogenit&auml;t von unserer DNA-Impfstoffen. Unsere Daten zeigen, dass eine niedrige Affinit&auml;t JF Epitop das Ausma&szlig; der CTL-Antworten verringert. Dar&uuml;ber hinaus wir verringerte Anzahl an Epitop-spezifische T-Helferzellen und CTLs zu beobachten sowie Zytokin-Sekretion und proliferative Reaktionen vermindert. Somit kann die Immunogenit&auml;t eines DNA-Epitop-Impfstoffs moduliert werden, durch die Ver&auml;nderung der Affinit&auml;t des JF-Epitop.

CD4 + T Zelle Epitope Affinit&auml;t MHC II beeinflusst das Ausma&szlig; der CTL-Antworten von DNA Epitop Impfstoffe hervorgerufen.

Immunizzazione con DNA del plasmide nudo suscita forte cella &ndash; mediata risposta immunitaria. Nel presente studio, esaminiamo le strategie per migliorare l&#39;epitopo specifico dei linfociti T citotossici (CTL) risposte utilizzando costrutti di DNA, esprimendo un minimo classe epitopo di gp120 di HIV-IIIB. Qui, noi di valutare l&#39;effetto delle cellule T CD4 + (T(H)) epitopo affinit&agrave; per la molecola di MHC II sulla immunogenicit&agrave; dei vaccini il nostro DNA. I nostri dati indicano che un epitopo T(H) bassa affinit&agrave; diminuita la grandezza delle risposte CTL. Inmunizaci&oacute;n con ADN del pl&aacute;smido desnudo provoca fuertes celular &ndash; transmitida por inmunorespuestas. En el presente estudio, se examinan estrategias para mejorar espec&iacute;ficos del ep&iacute;topo respuestas de linfocitos de T citot&oacute;xicos (CTL) usando ADN construcciones, expresando un m&iacute;nimo clase I del ep&iacute;topo de la gp120 del VIH-IIIB. Unlike activated T cells, quiescent CD4+ T cells have shown resistance to human immunodeficiency virus (HIV) infection due to a block in the early events of the viral life cycle. To further investigate the nature of this block, we infected quiescent CD4+ T cells with HIV-1(NL4-3) and immediately stimulated them. Compared to activated (prestimulated) cells, these poststimulated cells showed slightly decreased viral entry and delays in the completion of reverse transcription. However, the relative efficiency of integration was similar to that of prestimulated cells. Together, this resulted in decreased expression of tat/rev mRNA and synthesis of viral protein. Quiescent T lymphocytes containing latent human immunodeficiency virus (HIV) provide a long-lived viral reservoir. This reservoir may be the source of active infection that is reinitiated following the cessation of antiretroviral therapy. Therefore, it is important to understand the mechanisms involved in latent infection to develop new strategies to eliminate the latent HIV reservoir. We have previously demonstrated that latently infected quiescent lymphocytes can be generated during thymopoiesis in vivo in the SCID-hu mouse system. However, there is still a pressing need for an in vitro model of HIV latency in primary human cells. Here, we present a novel in vitro model that recapitulates key aspects of dormant HIV infection. Using an enhanced green fluorescent protein-luciferase fusion protein-containing reporter virus, we have generated a stable infection in primary human CD4(+) CD8(+) thymocytes in the absence of viral gene expression. T-cell activation induces a >200-fold induction of reporter activity. The induced reporter activity originates from a fully reverse-transcribed and integrated genome. We further demonstrate that this model can be useful to study long terminal repeat regulation, as previously characterized NF-kappaB response element mutations decrease the activation of viral gene expression. This model can therefore be used to study intricate molecular aspects of activation-inducible HIV infection in primary cells.

Multiple infection of cells is common, and cells that are productively infected with an integrated provirus frequently also contain unintegrated HIV-1 DNA. Here we examine the influence of an integrated provirus on unintegrated HIV-1 DNA (uDNA).

Until very recently, quiescent CD4(+) T cells were thought to be resistant to human immunodeficiency virus (HIV) infection. Subsequent studies, attempting to fully elucidate the mechanisms of resistance, showed that quiescent cells could become infected by HIV at low efficiency and form a latently infected population. In this study, we set out to identify the sites of viral integration and to assess the efficiency of the overall integration process in quiescent cells. Based on our results, HIV integration in quiescent CD4(+) T cells occurs in sites similar to those of their prestimulated counterparts. While site selections are similar, the integration process in quiescent cells is plagued by the formation of high levels of incorrectly processed viral ends and abortive two-long-terminal-repeat circles.

The ability of HIV to infect quiescent CD4+ T cells has been a topic of intense debate. While early studies suggested that the virus could not infect this particular T cell subset, subsequent studies using more sensitive protocols demonstrated that these cells could inefficiently support HIV infection. Additional studies showed that the kinetics of infection in quiescent cells was delayed and multiple stages of the viral life cycle were marred by inefficiencies. Despite that, proviral DNA has been found in these cells presenting them as a potential viral reservoir. The goal of cancer immunotherapy is the generation of an effective, stable, and self-renewing antitumor T-cell population. One such approach involves the use of high-affinity cancer-specific T-cell receptors in gene-therapy protocols. Here, we present the generation of functional tumor-specific human T cells in vivo from genetically modified human hematopoietic stem cells (hHSC) using a human/mouse chimera model. Transduced hHSC expressing an HLA-A*0201-restricted melanoma-specific T-cell receptor were introduced into humanized mice, resulting in the generation of a sizeable melanoma-specific naÃ¯ve CD8(+) T-cell population. Following tumor challenge, these transgenic CD8(+) T cells, in the absence of additional manipulation, limited and cleared human melanoma tumors in vivo. Furthermore, the genetically enhanced T cells underwent proper thymic selection, because we did not observe any responses against non-HLA-matched tumors, and no killing of any kind occurred in the absence of a human thymus. Finally, the transduced hHSC established long-term bone marrow engraftment. These studies present a potential therapeutic approach and an important tool to understand better and to optimize the human immune response to melanoma and, potentially, to other types of cancer.

Antitumor activity from antigen-specific CD8 T cells generated in vivo from genetically engineered human hematopoietic stem cells.

El objetivo de inmunoterapia del c&aacute;ncer es la generaci&oacute;n de una poblaci&oacute;n de c&eacute;lulas T antitumoral eficaz, estable y renueva a s&iacute; misma. Uno de los enfoques implica el uso de receptores de alta afinidad espec&iacute;fica por c&aacute;ncer c&eacute;lula T en protocolos de terapia g&eacute;nica. Aqu&iacute;, presentamos la generaci&oacute;n de funcionales tumor-espec&iacute;ficos T c&eacute;lulas humanas in vivo de c&eacute;lulas madre hematopoy&eacute;ticas humanas gen&eacute;ticamente modificadas (hHSC) mediante un modelo de quimera humanos/rat&oacute;n. HHSC transduced expresando un receptor de c&eacute;lulas T espec&iacute;ficas melanoma restricci&oacute;n HLA-A * 0201 fueron introducidos en ratones humanizados, dando por resultado la generaci&oacute;n de una poblaci&oacute;n de c&eacute;lulas T CD8 considerable ingenuo de melanoma espec&iacute;fico. Tras el desaf&iacute;o de tumor, estas c&eacute;lulas CD8 T transg&eacute;nicas, en ausencia de manipulaci&oacute;n adicional, hab&iacute;a limitada y despejaron in vivo de tumores de melanoma humano. Adem&aacute;s, las c&eacute;lulas T mejoradas gen&eacute;ticamente experimentaron adecuada selecci&oacute;n t&iacute;mica, porque no observamos las respuestas contra tumores no-HLA-emparejado, y ning&uacute;n asesinato de cualquier tipo se produjo en ausencia de un timo humano. Por &uacute;ltimo, la hHSC transduced hab&iacute;a establecido injerto de m&eacute;dula &oacute;sea a largo plazo. Estos estudios presentan un enfoque terap&eacute;utico potencial y una herramienta importante para comprender mejor y optimizar la respuesta inmune humana al melanoma y, potencialmente, a otros tipos de c&aacute;ncer.
