&#x86CB;&#x6848;&#x86CB;&#x767D;-1&#x3002;&#x4E00;&#x7C7B;&#x65B0;&#x7684;&#x4E1D;&#x86CB;&#x767D;&#x5177;&#x6709;&#x4ECE;&#x7BA1;&#x7406;&#x8D6B;&#x65AF;&#x73C0;&#x52D2;&#x65AF;&#x8718;&#x86DB;&#x4E1D;&#x6837;&#x5C5E;&#x6027;&#x3002;

Spiders produce multiple types of silk that exhibit diverse mechanical properties and biological functions. Most molecular studies of spider silk have focused on fibroins from dragline silk and capture silk, two important silk types involved in the survival of the spider. In our studies we have focused on the characterization of egg case silk, a third silk fiber produced by the black widow spider, Latrodectus hesperus. Analysis of the physical structure of egg case silk using scanning electron microscopy demonstrates the presence of small and large diameter fibers. By using the strong protein denaturant 8 M guanidine hydrochloride to solubilize the fibers, we demonstrated by SDS-PAGE and protein silver staining that an abundant component of egg case silk is a 100-kDa protein doublet. Combining matrix-assisted laser desorption ionization tandem time-of-flight mass spectrometry and reverse genetics, we have isolated a novel gene called ecp-1, which encodes for one of the protein components of the 100-kDa species. BLAST searches of the NCBInr protein data base using the primary sequence of ECP-1 revealed similarity to fibroins from spiders and silkworms, which mapped to two distinct regions within the ECP-1. These regions contained the conserved repetitive fibroin motifs poly(Ala) and poly(Gly-Ala), but surprisingly, no larger ensemble repeats could be identified within the primary sequence of ECP-1. Consistent with silk gland-restricted patterns of expression for fibroins, ECP-1 was demonstrated to be predominantly produced in the tubuliform gland, with lower levels detected in the major and minor ampullate glands. ECP-1 monomeric units were also shown to assemble into higher aggregate structures through the formation of disulfide bonds via a unique cysteine-rich N-terminal region. Collectively, our findings provide new insight into the components of egg case silk and identify a new class of silk proteins with distinctive molecular features relative to traditional members of the spider silk gene family.

Les araign&#xE9;es produisent plusieurs types de soie qui pr&#xE9;sentent diverses propri&#xE9;t&#xE9;s m&#xE9;caniques et les fonctions biologiques. La plupart des &#xE9;tudes mol&#xE9;culaires de la soie d&#x26;#39;araign&#xE9;e ont port&#xE9; sur de la soie fibroins pelle &#xE0; benne tra&#xEE;nante et de la soie de capture, deux types de soie importantes impliqu&#xE9;es dans la survie de l&#x26;#39;araign&#xE9;e. Dans nos &#xE9;tudes nous nous sommes concentr&#xE9;s sur la caract&#xE9;risation de la soie cas oeuf, une fibre de soie produites dans le tiers par l&#x26;#39;araign&#xE9;e veuve noire, Latrodectus hesperus. Analyse de la structure physique de la soie cas oeuf en utilisant la microscopie &#xE9;lectronique &#xE0; balayage montre la pr&#xE9;sence de fibres d&#x26;#39;un diam&#xE8;tre de petites et grandes. En utilisant la prot&#xE9;ine forte d&#xE9;naturant 8 M de chlorhydrate de guanidine pour solubiliser les fibres, on a d&#xE9;montr&#xE9; par SDS-PAGE et coloration &#xE0; l&#x26;#39;argent prot&#xE9;ine qu &#x26;#39;un composant riche en soie cas oeuf est un doublet de prot&#xE9;ine de 100 kDa. La combinaison de laser assist&#xE9;e par matrice d&#x26;#39;ionisation d&#xE9;sorption spectrom&#xE9;trie de masse tandem temps de vol et de la g&#xE9;n&#xE9;tique inverse, nous avons isol&#xE9; un nouveau g&#xE8;ne appel&#xE9; ecp-1, qui code pour l&#x26;#39;un des composants prot&#xE9;iques des esp&#xE8;ces de 100 kDa. Des recherches BLAST de la base de donn&#xE9;es de prot&#xE9;ines en utilisant NCBInr la s&#xE9;quence primaire de l&#x26;#39;ECP-1 a r&#xE9;v&#xE9;l&#xE9; une similitude &#xE0; fibroins des araign&#xE9;es et des vers &#xE0; soie, qui mapp&#xE9;s &#xE0; deux r&#xE9;gions distinctes au sein de l&#x26;#39;ECP-1. Ces r&#xE9;gions figurent l&#x26;#39;conserv&#xE9; r&#xE9;p&#xE9;titif fibro&#xEF;ne motifs poly (Ala) et le poly (Gly-Ala), mais &#xE9;tonnamment, pas de r&#xE9;p&#xE9;titions d&#x26;#39;ensemble plus importants pourraient &#xEA;tre identifi&#xE9;s au sein de la s&#xE9;quence primaire de l&#x26;#39;ECP-1. Conform&#xE9;ment &#xE0; la soie glande restriction des modes d&#x26;#39;expression pour fibroins, ECP-1 a &#xE9;t&#xE9; d&#xE9;montr&#xE9; &#xEA;tre principalement produite par la glande tubuliform, avec des niveaux inf&#xE9;rieurs d&#xE9;tect&#xE9;es dans les glandes ampullaires majeures et mineures. ECP-1 unit&#xE9;s monom&#xE8;res ont &#xE9;t&#xE9; &#xE9;galement montr&#xE9; &#xE0; assembler en structures sup&#xE9;rieures agr&#xE9;g&#xE9;s &#xE0; travers la formation de ponts disulfures par l&#x26;#39;interm&#xE9;diaire d&#x26;#39;un unique riche en cyst&#xE9;ine r&#xE9;gion N-terminale. Collectivement, nos r&#xE9;sultats fournissent un nouvel &#xE9;clairage sur les composantes de la soie cas d&#x26;#39;&#x153;ufs et d&#x26;#39;identifier une nouvelle classe de prot&#xE9;ines de soie avec distinctifs caract&#xE9;ristiques mol&#xE9;culaires relatives aux membres traditionnels de la famille araign&#xE9;e g&#xE8;ne de la soie.

Ragni producono pi&ugrave; tipi di seta che presentano diverse caratteristiche meccaniche e funzioni biologiche. Studi molecolari pi&ugrave; di seta di ragno sono concentrati su fibroins da dragline seta e seta di cattura, due importanti tipi di seta coinvolti nella sopravvivenza del ragno. Nei nostri studi ci hanno concentrato sulla caratterizzazione di uovo caso seta, un terzo di seta fibra prodotta dal ragno vedova nera, Latrodectus hesperus. Analisi della struttura fisica dell&#39;uovo caso seta mediante scanning electron microscopy dimostra la presenza di fibre di piccolo e grande diametro. Utilizzando il cloridrato della guanidina forte proteina denaturante 8 M per solubilizzare le fibre, abbiamo dimostrato da SDS-PAGE e proteina d&#39;argento colorazione che un&#39;abbondante componente dell&#39;uovo caso seta &egrave; un doppietto di proteina di 100 kDa. Ricerche abbattitori di NCBInr della proteina base-dati utilizzando la sequenza primaria di ECP-1 ha rivelato la somiglianza a fibroins da ragni e bachi da seta, che associati a due regioni distinte all&#39;interno dell&#39;ECP-1. Queste regioni contenevano il conservato fibroin ripetitivi motivi poly(Ala) e poly(Gly-Ala), ma sorprendentemente, ripetizioni di ensemble pi&ugrave; grande non potrebbero essere identificati all&#39;interno della sequenza primaria di ECP-1. Coerente con i modelli limitati da seta ghiandola dell&#39;espressione per fibroins, ECP-1 &egrave; stato dimostrato essere prevalentemente prodotto nella ghiandola tubuliform, con i livelli pi&ugrave; bassi rilevati nelle ghiandole ampullate maggiori e minori. ECP-1 unit&agrave; monomeriche sono stati mostrati anche da assemblare in strutture di aggregazione superiore attraverso la formazione di legami bisolfurico tramite un&#39;unica regione N-terminale ricca di cisteina. Collettivamente, i nostri risultati forniscono nuove nei componenti dell&#39;uovo caso seta e identificano una nuova classe di proteine della seta con caratteristiche distintive molecolare rispetto al tradizionali membri della famiglia genica seta ragno.

Las ara&#xF1;as producen varios tipos de seda que presentan diversas propiedades mec&#xE1;nicas y funciones biol&#xF3;gicas. Mayor&#xED;a de los estudios moleculares de la seda de ara&#xF1;a se han centrado en fibroins de la seda y la seda Dragline de captura, dos tipos de seda importantes que intervienen en la supervivencia de la ara&#xF1;a. En nuestros estudios nos hemos centrado en la caracterizaci&#xF3;n de la seda el caso del huevo, una fibra de seda tercera producida por la ara&#xF1;a viuda negro, Latrodectus hesperus. An&#xE1;lisis de la estructura f&#xED;sica de seda huevo caso mediante microscop&#xED;a electr&#xF3;nica de barrido demuestra la presencia de fibras de di&#xE1;metro peque&#xF1;o y grande. Mediante el uso de la prote&#xED;na fuerte desnaturalizante 8 M de hidrocloruro de guanidina para solubilizar las fibras, hemos demostrado por SDS-PAGE y tinci&#xF3;n con plata prote&#xED;na que un componente abundante de seda huevo caso es un doblete prote&#xED;na 100-kDa. La combinaci&#xF3;n de matriz asistida por l&#xE1;ser de ionizaci&#xF3;n de desorci&#xF3;n t&#xE1;ndem tiempo de vuelo espectrometr&#xED;a de masas y la gen&#xE9;tica inversa, hemos aislado un nuevo gen llamado ECP-1, que codifica para uno de los componentes proteicos de la especie 100-kDa. BLAST b&#xFA;squedas de la prote&#xED;na NCBInr base de datos usando la secuencia primaria de la PAC-1 revel&#xF3; similitud con fibroins de las ara&#xF1;as y los gusanos de seda, que se asignan a dos regiones distintas dentro de la PAC-1. Estas regiones conservadas conten&#xED;a el repetitivo fibro&#xED;na motivos poli (Ala) y poli (Gly-Ala), pero, sorprendentemente, no se repite conjunto m&#xE1;s amplio podr&#xED;a ser identificado dentro de la secuencia primaria de la PAC-1. De acuerdo con seda restringidas gl&#xE1;ndula patrones de expresi&#xF3;n de fibroins, ECP-1 se ha demostrado que se produce principalmente en la gl&#xE1;ndula tubuliform, con niveles m&#xE1;s bajos detectados en las gl&#xE1;ndulas ampullate mayores y menores. ECP-1 unidades monom&#xE9;ricas se muestra tambi&#xE9;n para montar en mayores estructuras agregadas a trav&#xE9;s de la formaci&#xF3;n de enlaces disulfuro a trav&#xE9;s de una &#xFA;nica rica en ciste&#xED;na regi&#xF3;n N-terminal. En conjunto, nuestros resultados proporcionan una nueva visi&#xF3;n de los componentes de la seda de huevo caso e identificar una nueva clase de prote&#xED;nas de la seda, con distintas caracter&#xED;sticas moleculares en relaci&#xF3;n con los miembros tradicionales de la familia de genes de seda de ara&#xF1;a.

Members of the basic helix-loop-helix (bHLH) family are required for a number of different developmental pathways, including lymphopoiesis, myogenesis, neurogenesis, and sex determination. Screening a cDNA library prepared from silk-producing glands of the black widow spider, we have identified a new bHLH transcription factor named SGSF. Within the bHLH region, SGSF showed considerable conservation with other HLH proteins, including Drosophila melanogaster achaete and scute, as well as three HLH proteins identified by gene prediction programs. The expression pattern of SGSF was restricted to a subset of silk-producing glands, which include the tubuliform and major ampullate glands. SGSF was capable of binding an E-box element as a heterodimer with the E protein, E47, but was unable to bind this motif as a homodimer. SGSF was demonstrated to be a nuclear transcription factor capable of attenuating the transactivation of E47 homodimers in mammalian cells. SGSF represents the first example of a silk gland-restricted bHLH protein, and its expression pattern suggests that SGSF plays a role in regulating differentiation of cells in the spider that control silk gland formation or egg case silk gene expression.

La detecci&#xF3;n de una biblioteca de cDNA preparada a partir de las gl&#xE1;ndulas productoras de seda de la ara&#xF1;a viuda negro, hemos identificado un factor de transcripci&#xF3;n bHLH nuevo llamado SGSF. Dentro de la regi&#xF3;n bHLH, SGSF mostr&#xF3; conservaci&#xF3;n considerable con prote&#xED;nas HLH otros, incluyendo Drosophila melanogaster achaete y escudo, as&#xED; como tres prote&#xED;nas HLH identificados por los programas de predicci&#xF3;n de genes. El patr&#xF3;n de expresi&#xF3;n de SGSF estaba restringido a un subconjunto de las gl&#xE1;ndulas productoras de seda, que incluyen la tubuliform y las gl&#xE1;ndulas principales ampullate. Araneoid spiders use specialized abdominal glands to manufacture up to seven different protein-based silks/glues that have diverse physical properties. The fibroin sequences that encode egg case fibers (cover silk for the egg case sac) and the secondary structure of these threads have not been previously determined. In this study, MALDI tandem TOF mass spectrometry (MS/MS) and reverse genetics were used to isolate the first egg case fibroin, named tubuliform spidroin 1 (TuSp1), from the black widow spider, Latrodectus hesperus. Real-time quantitative PCR analysis demonstrates TuSp1 is selectively expressed in the tubuliform gland. Analysis of the amino acid composition of raw egg case silk closely aligns with the predicted amino acid composition from the primary sequence of TuSp1, which supports the assertion that TuSp1 represents a major component of egg case fibers. TuSp1 is composed of highly homogeneous repeats that are 184 amino acids in length. The long stretches of polyalanine and glycine-alanine subrepeats, which account for the crystalline regions of minor ampullate and major ampullate fibers, are very poorly represented in TuSp1. However, polyserine blocks and short polyalanine stretches were highly iterated within the primary sequence, and (13)C NMR spectroscopy demonstrated that the majority of alanine was found in a beta-sheet structure in post-spun egg case silk. The TuSp1 repeat unit does not display substantial sequence similarity to any previously described fibroin genes or proteins, suggesting that TuSp1 is a highly divergent member of the spider silk gene family.

Araneoid ragni utilizzano speciali ghiandole addominali per produrre fino a sette differenti basati su proteine sete/colle che hanno propriet&agrave; fisiche diverse. Le sequenze fibroin che codificano uovo caso fibre (coperchio seta per il sac dell&#39;uovo caso) e la struttura secondaria di queste discussioni non sono state determinate in precedenza. Spider silk proteins are well-known for their extraordinary mechanical properties, displaying remarkable strength and toughness. In this study, matrix-assisted laser desorption ionization (MALDI) tandem time-of-flight (TOF) mass spectrometry (MS/MS) and reverse genetics were used to isolate a new cDNA sequence that encodes for a protein assembled into egg case silk from the black widow spider, Latrodectus hesperus. Analysis of the primary sequence of this protein reveals approximately 52% identity to the egg case protein 1 (ECP-1) fibroin-like family member. On the basis of the similarity in the primary sequence and expression pattern, we have named this factor egg case protein 2 (ECP-2). Alignments of ECP-1 and ECP-2 demonstrate highly conserved N termini, with 16 Cys residues found within the first 153 amino acids. Traditional ensemble repeats found within reported fibroins were poorly represented in the primary sequence of ECP-2, but scattered blocks of polyalanine were present, along with a C terminus rich in GA repeats. Reverse transcription quantitative PCR analysis showed that ECP-2 is predominantly expressed in the tubuliform gland. Relative to ECP-1, ECP-2 mRNA levels were determined to be >2-fold higher. MALDI MS/MS analysis of peptide fragments generated from the large-diameter core fiber after enzymatic digestion and acid hydrolysis demonstrated the presence of a fiber that is trimeric in nature, containing tubuliform spidroin 1 (TuSp1), ECP-1, and ECP-2. We also report an additional primary sequence for TuSp1, demonstrating that TuSp1 contains two Cys residues within a nonrepetitive N-terminal region. In combination with the distinctive protein architectures of ECP-1 and ECP-2, along with their co-localization with TuSp1 in the core fiber, our findings suggest that ECP-1 and ECP-2 play important structural roles in the egg case silk fiber.

Prote&#xED;nas de seda de ara&#xF1;a son bien conocidos por sus propiedades mec&#xE1;nicas extraordinarias, mostrando una notable fortaleza y tenacidad. En este estudio, l&#xE1;ser asistida por matriz de ionizaci&#xF3;n de desorci&#xF3;n (MALDI) t&#xE1;ndem de tiempo de vuelo (TOF) espectrometr&#xED;a de masas (MS / MS) y la gen&#xE9;tica inversa se utiliza para aislar una nueva secuencia de ADNc que codifica para una prote&#xED;na de huevo montada en seda caso de la ara&#xF1;a viuda negro, Latrodectus hesperus. An&#xE1;lisis de la secuencia primaria de esta prote&#xED;na revela aproximadamente 52% de identidad con el caso de huevo prote&#xED;na 1 (ECP-1)-fibro&#xED;na como miembro de la familia. Sobre la base de la similitud de la secuencia primaria y el patr&#xF3;n de expresi&#xF3;n, que hemos denominado este huevo factor proteico caso 2 (ECP-2). En relaci&#xF3;n con ECP-1, ECP-2 niveles de mRNA se determin&#xF3; que eran&#x26;gt; 2 veces mayor. MALDI MS / MS an&#xE1;lisis de fragmentos de p&#xE9;ptidos generados a partir de la fibra de n&#xFA;cleo de gran di&#xE1;metro despu&#xE9;s de la digesti&#xF3;n enzim&#xE1;tica y la hidr&#xF3;lisis &#xE1;cida demostrado la presencia de una fibra que es trim&#xE9;rica en la naturaleza, que contiene tubuliform spidroin 1 (TuSp1), FEC-1, y ECP 2- .
