The method of analysis features the evaluation of the effect of a gene by averaging the effects of the SNPs covered by that gene. Marginal methods that test one gene at a time fail to show any effect. That may be related to the fact that each of these 18 genes adds very little to the risk of cancer. This study determined the levels of serum soluble intercellular adhesion molecule-1 (sICAM-1) and soluble vascular cell adhesion molecular-1 (sVCAM-1) in patients with different types of Keshan disease (KD), examined the relationship between Coxsackie B virus-specific IgM antibody (CBV-IgM) and sICAM-1 or sVCAM-1 in KD patients, and investigated the role of these adhesion molecules in the pathogenesis of KD and their clinical implications. The levels of serum sICAM-1, sVCAM-1 and CBV-IgM were measured by using enzyme-linked immunosorbent assay in 22 patients with chronic Keshan disease (CKD), 27 with latent Keshan disease (LKD) and 28 healthy controls. The subjects in different groups were adjusted for sex and age. Echocardiography was adopted to determine left ventricular ejection fraction (LVEF) in 22 patients with CKD. The results showed that CKD patients had significantly higher levels of sICAM-1 and sVCAM-1 than LKD patients and healthy controls (P<0.01 for all). And there was significant difference in the levels of the 2 adhesion molecules between LKD patients and healthy controls (P<0.05). A negative correlation was found between LVEF and sICAM-1 or sVCAM-1 in CKD patients. The percentage of CBV-specific IgM positive individuals in KD patients was significantly higher than that of healthy controls. In CVB-specific IgM positive patients, the levels of serum sICAM-1 and sVCAM-1 were significantly greater than those in CBV-specific IgM negative counterpart. It was concluded that the increase in the levels of sICAM-1 and sVCAM-1 suggests the progression of inflammation in KD. sICAM-1 and sVCAM-1 can promote the development of myocardial pathology and lead to poor myocardial function. The increased serum sICAM-1 and sVCAM-1 in KD patients may be related to CBV infection.

Circulating adhesion molecules in patients with Keshan disease and their relationship with Coxsackie B virus infection.

Cette &eacute;tude a d&eacute;termin&eacute; les niveaux de s&eacute;rum mol&eacute;cule d&#39;adh&eacute;sion intercellulaire soluble-1 (sICAM-1) et les cellules vasculaires soluble adh&eacute;rence mol&eacute;culaire-1 (sVCAM-1) chez les patients atteints de diff&eacute;rents types de la maladie de Keshan (KD), a examin&eacute; la relation entre les anticorps de IgM sp&eacute;cifiques du virus Coxsackie B (CBV-IgM) et de sICAM-1 ou sVCAM-1 chez les patients de KD et examin&eacute; le r&ocirc;le de ces mol&eacute;cules d&#39;adh&eacute;rence dans la pathogen&egrave;se du KD et leurs implications cliniques. Les niveaux de s&eacute;rum sICAM-1, sVCAM-1 et CBV-IgM ont &eacute;t&eacute; mesur&eacute;s &agrave; l&#39;aide d&#39;enzyme-linked immunosorbent assay chez les 22 patients avec la maladie de Keshan chronique (IRC), 27 avec la maladie de Keshan latente (PFC) et 28 t&eacute;moins sains. Les sujets dans les diff&eacute;rents groupes ont &eacute;t&eacute; ajust&eacute;s pour l&#39;&acirc;ge et le sexe. Diese Studie ermittelt im Serum l&ouml;slich der Adh&auml;sion Molek&uuml;l-1 (sICAM-1) und l&ouml;slichen vaskul&auml;re Zelle Adh&auml;sion Molekulare-1 (vorgestellt-1) bei Patienten mit verschiedenen Arten von Keshan-Krankheit (KD), untersucht die Beziehung zwischen Coxsackie B Virus-spezifische IgM-Antik&ouml;rper (CBV-IgM) und sICAM-1 oder vorgestellt-1 KD-Patienten, und untersucht die Rolle dieser Adh&auml;sion Molek&uuml;le in der Pathogenese von KD und ihre klinischen Auswirkungen. Questo studio ha determinato i livelli del siero molecola di adesione intercellulare solubile-1 (sICAM-1) e solubili delle cellule vascolari adesione molecolare-1 (sVCAM-1) in pazienti con diversi tipi di malattia di Keshan (KD), ha esaminato la relazione tra anticorpo IgM virus-specifico di Coxsackie B (CBV-IgM) e sICAM-1 o sVCAM-1 in pazienti KD e studiato il ruolo di queste molecole di adesione nella patogenesi della KD e le loro implicazioni cliniche. Este estudio se determin&oacute; los niveles de suero soluble adherencia intercelular molecule-1 (sICAM-1) y soluble de la c&eacute;lula vascular adhesi&oacute;n molecular-1 (sVCAM-1) en pacientes con diferentes tipos de enfermedad de Keshan (KD), examin&oacute; la relaci&oacute;n entre los anticuerpos de IgM espec&iacute;ficos virus de Coxsackie B (CBV-IgM) y sICAM-1 o sVCAM-1 en pacientes de KD e investigado el papel de estas mol&eacute;culas de adhesi&oacute;n en la patogenesia de KD y sus implicaciones cl&iacute;nicas. Se midieron los niveles de suero sICAM-1, sVCAM-1 y CBV-IgM usando enzyme-linked immunosorbent assay en 22 pacientes con enfermedad de Keshan cr&oacute;nica (CKD), 27 con la enfermedad de Keshan latente (LKD) y 28 controles sanos. Los temas en los diferentes grupos se ajustaron por sexo y edad. Ecocardiograf&iacute;a fue adoptada para determinar la fracci&oacute;n de eyecci&oacute;n ventricular izquierda (FEVI) en 22 pacientes con nefropat&iacute;as cr&oacute;nicas. Los resultados mostraron que los pacientes ten&iacute;an niveles significativamente m&aacute;s altos de sICAM-1 y sVCAM-1 que LKD pacientes y controles sanos (P &lt; 0.01 para todos). Y no hubo diferencias significativas en los niveles de las mol&eacute;culas de 2 adherencia entre LKD pacientes y controles sanos (P &lt; 0.05). Se encontr&oacute; una correlaci&oacute;n negativa entre la FEVI y sICAM-1 o sVCAM-1 en pacientes con nefropat&iacute;a cr&oacute;nica. El porcentaje de individuos en pacientes KD CBV espec&iacute;ficos IgM fue significativamente mayor que el de controles sanos. En los pacientes positivos de IgM espec&iacute;fica CVB, los niveles de suero sICAM-1 y sVCAM-1 fueron significativamente mayores que en contraparte negativa CBV espec&iacute;ficos IgM. Se concluy&oacute; que el aumento en los niveles de sICAM-1 y sVCAM-1 sugiere la progresi&oacute;n de la inflamaci&oacute;n en KD. sICAM-1 y sVCAM-1 pueden promover el desarrollo de la patolog&iacute;a mioc&aacute;rdica y conducir a insuficiencia mioc&aacute;rdica. La creciente del suero sICAM-1 y sVCAM-1 en pacientes de KD puede deberse a la infecci&oacute;n de la CBV.

ABSTRACT : The genes PTPN22 and HLA-DRB1 have been found by a number of studies to confer an increased risk for rheumatoid arthritis (RA), which indicates that both genes play an important role in RA etiology. It is believed that they not only have strong association with RA individually, but also interact with other related genes that have not been found to have predisposing RA mutations. In this paper, we conduct genome-wide searches for RA-associated gene-gene interactions that involve PTPN22 or HLA-DRB1 using the Genetic Analysis Workshop 16 Problem 1 data from the North American Rheumatoid Arthritis Consortium. MGC13017, HSPCAL3, MIA, PTPNS1L, and IGLVI-70, which showed association with RA in previous studies, have been confirmed in our analysis.

R&eacute;sum&eacute; : Les g&egrave;nes HLA-DRB1 et PTPN22 ont &eacute;t&eacute; trouv&eacute;s par un certain nombre d&#39;&eacute;tudes visant &agrave; conf&eacute;rer &agrave; un risque accru de polyarthrite rhumato&iuml;de (PR), qui indique que les deux g&egrave;nes jouent un r&ocirc;le important dans l&#39;&eacute;tiologie de la RA. Resumen: Los genes PTPN22 y HLA-DRB1 han sido encontrados por una serie de estudios para conferir un mayor riesgo para la artritis reumatoide (RA), que indica que ambos genes juegan un papel importante en la etiolog&iacute;a de la RA. Se cree que no s&oacute;lo tienen una asociaci&oacute;n fuerte con el RA individualmente, pero tambi&eacute;n interactuar con otros genes relacionados que no se han encontrado para tener la predisposici&oacute;n de las mutaciones de la RA. ABSTRACT : Rheumatoid arthritis (RA, MIM 180300) is a chronic and complex autoimmune disease. Using the North American Rheumatoid Arthritis Consortium (NARAC) data set provided in Genetic Analysis Workshop 16 (GAW16), we used the genotype-trait distortion (GTD) scores and proposed analysis procedures to capture the gene-gene interaction effects of multiple susceptibility gene regions on RA. In this paper, we focused on 27 RA candidate gene regions (531 SNPs) based on a literature search. Statistical significance was evaluated using 1000 permutations. HLADRB1 was found to have strong marginal association with RA. We identified 14 significant interactions (p < 0.01), which were aggregated into an association network among 12 selected candidate genes PADI4, FCGR3, TNFRSF1B, ITGAV, BTLA, SLC22A4, IL3, VEGF, TNF, NFKBIL1, TRAF1-C5, and MIF. Based on our and other contributors' findings during the GAW16 conference, we further studied 24 candidate regions with 336 SNPs. We found 23 significant interactions (p-value < 0.01), nine interactions in addition to our initial findings, and the association network was extended to include candidate genes HLA-A, HLA-B, HLA-C, CTLA4, and IL6. As we will discuss in this paper, the reported possible interactions between genes may suggest potential biological activities of RA.

Resumen: la artritis reumatoide (RA, MIM 180300) es una enfermedad autoinmune cr&oacute;nica y compleja. Utilizando el conjunto de datos del consorcio norteamericano de la artritis reumatoide (NARAC) en 16 de taller de an&aacute;lisis gen&eacute;ticos (GAW16), utilizamos las puntuaciones de la distorsi&oacute;n (GTD) genotipo-trait y procedimientos de an&aacute;lisis propuesto para capturar los efectos de la interacci&oacute;n del gen de varias regiones del gen de susceptibilidad en RA En este trabajo, nos enfocamos en 27 RA candidato gene regiones (531 SNPs) basado en una b&uacute;squeda de literatura. Significaci&oacute;n estad&iacute;stica se evalu&oacute; mediante 1000 permutaciones. HLADRB1 fue encontrado para tener fuerte asociaci&oacute;n marginal con ar. Keshan disease (KD) is a dilated cardiomyopathy closely related with a diet deficient in the mineral selenium. It is named for the northeastern Chinese county Keshan, where the disease prevalence is high because of selenium-deficient soil. KD is a gene-environment interaction disease. Here, we used stepwise multiple regression analysis to analyze the risk factors of the disease and the main clinical features in 71 KD patients and 290 controls. The variables analyzed included age, sex, family history of KD, blood selenium level, glutathione peroxidase-1 (GPx-1) activity, variance at codon198 in GPx-1 gene, residence in an endemic area, abnormal electrocardiography (ECG) findings, and cardiothoracic (CT) ratio. The main risk factors found were low GPx-1 activity, family history of KD and living in an endemic area. The main clinical features were increased cardiac load on ECG and increased CT ratio and Tei index. Public health and clinical prevention efforts could focus on increasing GPx-1 activity to address KD. Is selenium deficiency really the certain cause of KD? This is not at all a settled question. And further study is promptly required to investigate the etiology of KD.

Normal cellular behavior can be described as a complex, regulated network of interaction between genes and proteins. Targeted cancer therapies aim to neutralize specific proteins that are necessary for the cancer cell to remain viable in vivo. Ideally, the proteins targeted should be such that their downregulation has a major impact on the survival/fitness of the tumor cells and, at the same time, has a smaller effect on normal cells. It is difficult to use standard analysis methods on gene or protein expression levels to identify these targets because the level thresholds for tumorigenic behavior are different for different genes/proteins. We have developed a novel methodology to identify therapeutic targets by using a new paradigm called "gene centrality." The main idea is that, in addition to being overexpressed, good therapeutic targets should have a high degree of connectivity in the tumor network because one expects that suppression of its expression would affect many other genes. We propose a mathematical quantity called "centrality," which measures the degree of connectivity of genes in a network in which each edge is weighted by the expression level of the target gene. Using our method, we found that several SRC proto-oncogenes LYN, YES1, HCK, FYN, and LCK have high centrality in identifiable subsets of basal-like and HER2+ breast cancers. To experimentally validate the clinical value of this finding, we evaluated the effect of YES1 knockdown in basal-like breast cancer cell lines that overexpress this gene. We found that YES1 downregulation has a significant effect on the survival of these cell lines. Our results identify YES1 as a target for therapeutics in a subset of basal-like breast cancers.
