Biomedical research has undergone a major shift in emphasis over the past decade from characterizing the genomes of organisms to characterizing their proteomes. The high-throughput approaches that were successfully applied to sequencing of genomes, such as miniaturization and automation, have been adapted for high-throughput cloning and protein production. High-throughput platforms allow for a multi-construct, multi-parallel approach to expression optimization and construct evaluation. We describe here a series of baculovirus transfer and expression vectors that contain ligation-independent cloning regions originally designed for use in high-throughput Escherichia coli expression evaluation. These new vectors allow for parallel cloning of the same gene construct into a variety of baculovirus or E. coli expression vectors. A high-throughput platform for construct expression evaluation in baculovirus-infected insect cells was developed to utilize these vectors. Data from baculovirus infection expression trials for multiple constructs of two target protein systems relevant to the study of human diseases are presented. The target proteins exhibit a wide variation in behavior and illustrate the benefit of investigating multiple cell types, fusion partners and secretion signals in optimization of constructs and conditions for eukaryotic protein production.

La recherche biom&#xE9;dicale a subi un changement majeur dans l&#x26;#39;accent sur la derni&#xE8;re d&#xE9;cennie de caract&#xE9;riser les g&#xE9;nomes des organismes &#xE0; la caract&#xE9;risation de leurs prot&#xE9;omes. Les approches &#xE0; haut d&#xE9;bit qui ont &#xE9;t&#xE9; appliqu&#xE9;es avec succ&#xE8;s au s&#xE9;quen&#xE7;age des g&#xE9;nomes, tels que la miniaturisation et l&#x26;#39;automatisation ont &#xE9;t&#xE9; adapt&#xE9;es pour les haut-d&#xE9;bit de clonage et la production de prot&#xE9;ines. Plates-formes &#xE0; haut d&#xE9;bit permettent une construction multi-multi-parall&#xE8;le approche de l&#x26;#39;optimisation d&#x26;#39;expression et l&#x26;#39;&#xE9;valuation construction. Nouveaux Vecteurs de clonage ligature-Independent Compatible avec une plate-forme &#xE0; haut d&#xE9;bit pour l&#x26;#39;&#xE9;valuation d&#x26;#39;expression parall&#xE8;le Construct Utilisation des cellules d&#x26;#39;insectes infect&#xE9;es par le baculovirus

Biomedizinische Forschung hat eine gro&szlig;e Verschiebung der Schwerpunkte im letzten Jahrzehnt von Charakterisierung der Genome Organismen zu charakterisieren ihre Proteome unterzogen. Die Hochdurchsatz-Ans&auml;tze, die zur Sequenzierung von Genomen, wie Miniaturisierung und Automation, erfolgreich angewendet wurden, wurden f&uuml;r Hochdurchsatz-Klonen und Protein-Produktion angepasst. Hochdurchsatz-Plattformen erlauben einen multi-construct, multi-parallel Ansatz f&uuml;r Expression-Optimierung und Bewertung zu konstruieren. Wir beschreiben hier eine Reihe von Baculovirus-Transfer und Ausdruck Vektoren, die enthalten Ligaturen-unabh&auml;ngige Klonen Regionen urspr&uuml;nglich f&uuml;r Hochdurchsatz-Escherichia coli Auswertung verwenden. Diese neuen Vektoren erm&ouml;glichen parallel Klonen von dem gleichen Gen-Konstrukt in einer Vielzahl von Baculovirus oder E. Coli Expressionsvektoren. Die Zielproteinen weisen eine gro&szlig;e Unterschiede im Verhalten und den Nutzen der Untersuchung mehrerer Zelltypen, Fusion Partner und Sekretion Signale in der Optimierung von Konstrukten und eukaryotic Protein Produktionsbedingungen zu veranschaulichen.

Ricerca biomedica ha subito un cambiamento importante nell&#39;enfasi nell&#39;ultimo decennio da caratterizzare i genomi di organismi che caratterizzano la loro ai proteomi. Gli approcci di alto-rendimento che sono stati applicati correttamente al sequenziamento dei genomi, come la miniaturizzazione e l&#39;automazione, sono stati adattati per la produzione di clonazione e proteina di alto-rendimento. Piattaforme high-throughput consentono un approccio multi-construct, 640P per ottimizzazione di espressione e costruire valutazione. Vengono presentati dati da baculovirus infezione espressione prove per i costrutti pi&ugrave; di due sistemi della proteina target rilevanti per lo studio delle malattie umane. Le proteine bersaglio che esibiscono un&#39;ampia variazione nel comportamento e illustrano il beneficio di indagare pi&ugrave; tipi di cellule, partner di fusione e segnali secrezione nell&#39;ottimizzazione dei costrutti e delle condizioni per la produzione di proteina eucariotica.
