HSV-1 infection of the cornea leads to a potentially blinding immunoinflammatory lesion of the cornea, termed herpetic stromal keratitis. It has also been shown that one of the factors limiting inflammation of the cornea is the presence of Fas ligand (FasL) on corneal epithelium and endothelium. In this study, the role played by FasL expression in the cornea following acute infection with HSV-1 was determined. Both BALB/c and C57BL/6 (B6) mice with HSV-1 infection were compared with their lpr and gld counterparts. Results indicated that mice bearing mutations in the Fas Ag (lpr) displayed the most severe disease, whereas the FasL-defective gld mouse displayed an intermediate phenotype. It was further demonstrated that increased disease was due to lack of Fas expression on bone marrow-derived cells. Of interest, although virus persisted slightly longer in the corneas of mice bearing lpr and gld mutations, the persistence of infectious virus in the trigeminal ganglia was the same for all strains infected. Further, B6 mice bearing lpr and gld mutations were also more resistant to virus-induced mortality than were wild-type B6 mice. Thus, neither disease nor mortality correlated with viral replication in these mice. Collectively, the findings indicate that the presence of FasL on the cornea restricts the entry of Fas(+) bone marrow-derived inflammatory cells and thus reduces the severity of HSK.

HSV-1 infection de la corn&eacute;e conduit &agrave; une l&eacute;sion immunoinflammatoires potentiellement aveuglante de la corn&eacute;e, appel&eacute;e k&eacute;ratite stromale herp&eacute;tique. Il a &eacute;galement &eacute;t&eacute; d&eacute;montr&eacute; que l&#39;un des facteurs limitant l&#39;inflammation de la corn&eacute;e est la pr&eacute;sence du ligand de Fas (FasL) sur l&#39;&eacute;pith&eacute;lium de la corn&eacute;e et l&#39;endoth&eacute;lium. Dans cette &eacute;tude, le r&ocirc;le jou&eacute; par l&#39;expression de FasL dans la corn&eacute;e suite &agrave; une infection aigu&euml; par HSV-1 a &eacute;t&eacute; d&eacute;termin&eacute;. HSV-1 Infektion der Hornhaut f&uuml;hrt zu einer potenziell blendenden Immunoinflammatory L&auml;sion der Hornhaut, stromale Keratitis herpetica bezeichnet. Es hat sich auch gezeigt, dass einer der Faktoren, die Begrenzung der Entz&uuml;ndung der Hornhaut das Vorhandensein von Fas-Ligand (FasL) auf Hornhaut-Epithel und Endothel ist. In dieser Studie wurde die Rolle der FasL-Expression in der Hornhaut, die nach der akuten Infektion mit HSV-1 bestimmt. BALB/c und C57BL/6 (B6) M&auml;use mit HSV-1-Infektion wurden mit ihren Lpr und Gld-Pendants verglichen. Ergebnisse gekennzeichnet, dass M&auml;use mit Mutationen in der Fas-Ag (Lpr) die schwerste Krankheit angezeigt, w&auml;hrend die FasL-Genen Gld-Maus eine fortgeschrittene Ph&auml;notyp angezeigt. Es wurde weiter gezeigt, dass erh&ouml;hte Krankheit wegen des Mangels der Fas Ausdruck auf Knochenmark gewonnenen Zellen. Von Interesse obwohl Virus etwas l&auml;nger in der Kornea M&auml;use mit Mutationen mit Lpr und Gld beibehalten wurde das Fortbestehen des ansteckenden Virus in den Trigeminalganglien das gleiche f&uuml;r alle St&auml;mme infiziert. Dar&uuml;ber hinaus waren B6 M&auml;use mit Lpr und Gld-Mutationen auch widerstandsf&auml;higer gegen Virus-induzierte Sterblichkeit als Wildtyp B6 M&auml;use waren. Also weder die Krankheit als auch die Sterblichkeit mit Virusreplikation in diesen M&auml;usen korreliert. Zusammenfassend zufolge die Ergebnisse das Vorhandensein von FasL auf der Hornhaut schr&auml;nkt den Eintrag Fas(+) Knochenmark-abgeleitete entz&uuml;ndliche Zellen und reduziert so die Schwere der HSK.

HSV-1 infezione della cornea conduce ad una lesione potenzialmente accecante immunoinflammatory della cornea, chiamata cheratite stromale erpetica. Inoltre &egrave; stato dimostrato che uno dei fattori limitanti l&#39;infiammazione della cornea &egrave; la presenza di ligando Fas (FasL) sull&#39;epitelio corneale e dell&#39;endotelio. In questo studio, il ruolo svolto dall&#39;espressione FasL nella cornea dopo infezione acuta con HSV-1 &egrave; stato determinato. BALB/c sia i topi C57BL/6 (B6) con infezione da HSV-1 sono stati confrontati con le loro controparti lpr e gld. Ulteriormente, B6 topi recanti mutazioni lpr e gld erano anche pi&ugrave; resistenti alla mortalit&agrave; indotta da virus che erano topi B6 wild-type. Cos&igrave;, n&eacute; malattia n&eacute; mortalit&agrave; correlata con replicazione virale in questi topi. Collettivamente, i risultati indicano che la presenza di FasL sulla cornea limita l&#39;ingresso di Fas(+) derivate da midollo osseo di cellule infiammatorie e quindi riduce la gravit&agrave; dell&#39;HSK.

&#xC950; Fas &#xBC0F; Fas&#xC5D0; &#xB3CC;&#xC5F0;&#xBCC0;&#xC774;&#xC640; &#xB9AC;&#xAC04;&#xB4DC; &#xC99D;&#xAC00; herpetic stromal &#xC5FC; &#xAC01; &#xB9C9; &#xAC10;&#xC5FC; HSV-1&#xC5D0; &#xB530;&#xB77C; &#xBCF4;&#xC5EC; &#xC90D;&#xB2C8;&#xB2E4;.

Infecci&oacute;n por HSV-1 de la c&oacute;rnea conduce a una lesi&oacute;n inmunoinflamatorias potencialmente cegador de la c&oacute;rnea, denominada Queratitis herp&eacute;tica estromal. Tambi&eacute;n se ha demostrado que uno de los factores que limitan la inflamaci&oacute;n de la c&oacute;rnea es la presencia de Fas ligand (FasL) en epitelio corneal y el endotelio. En este estudio, se determin&oacute; el papel desempe&ntilde;ado por la expresi&oacute;n de FasL en la c&oacute;rnea despu&eacute;s de la infecci&oacute;n aguda con HSV-1. BALB/c y ratones C57BL/6 (B6) con infecci&oacute;n por HSV-1 fueron comparados con sus contrapartes de lpr y gld. Los resultados indicaron que los ratones con mutaciones en el Fas Ag (lpr) muestran la enfermedad m&aacute;s severa, mientras que el rat&oacute;n de gld FasL defectuoso muestra un fenotipo intermedio. Qued&oacute; demostrado adem&aacute;s que la enfermedad aumentada era debida a la falta de expresi&oacute;n de Fas en las c&eacute;lulas de m&eacute;dula &oacute;sea. De inter&eacute;s, aunque el virus persiste un poco m&aacute;s largo en las c&oacute;rneas de los ratones con mutaciones de lpr y gld, la persistencia del virus infeccioso en los ganglios del trig&eacute;mino fue la misma para todas las cepas infectadas. Adem&aacute;s, B6 ratones mutaciones lpr y gld tambi&eacute;n eran m&aacute;s resistentes a la mortalidad inducida por virus que eran ratones de tipo salvaje B6. As&iacute;, la enfermedad ni mortalidad correlacionada con la replicaci&oacute;n viral en estos ratones. Herpetic stromal keratitis (HSK) is characterized by an inflammatory response that includes neutrophils, macrophages, NK cells, and T cells. The factors that are responsible for this inflammation are proinflammatory cytokines and chemokines. Many of these factors have been defined for primary disease, but relatively few have been investigated during recurrent HSK. The present study was designed to determine the role that two of these factors, IL-6 and CXCL1, play during recurrent HSK. Results clearly indicate that unlike primary disease, IL-6 plays no role in recurrent HSK. However, the presence of CXCL1 is required for recurrent HSK as evidenced by the lack of corneal disease in mice treated with anti-CXCL1 Ab. This was confirmed using mice lacking the primary receptor for CXCL1, CXCR2. Corneal disease in this strain was significantly reduced compared with wild-type C57BL/6 controls. Unexpectedly, lack of disease occurs even though CXCL1 knockout mice display increased viral shedding at the cornea. The primary mechanism that CXCL1 plays during disease is its ability to stimulate neutrophils to infiltrate the cornea following reactivation. This paper provides further evidence that primary HSK and recurrent HSK possess overlapping yet distinct disease mechanisms.
