The success of a biological control agent depends on key traits, particularly reproductive potential, environmental tolerance, and ability to be cultured. These traits can deteriorate rapidly when the biological control agent is reared in culture. Trait deterioration under laboratory conditions has been widely documented in the entomopathogenic nematode (EPN) Heterorhabditis bacteriophora (Hb) but the specific mechanisms behind these genetic processes remain unclear. This research investigates the molecular mechanisms of trait deterioration of two experimental lines of Hb, an inbred line (L5M) and its original parental line (OHB). We generated transcriptional profiles of two experimental lines of Hb, identified the differentially expressed genes (DEGs) and validated their differential expression in the deteriorated line.

Der Erfolg eines biologischen Kontrolle-Agenten h&auml;ngt wichtige Merkmale, insbesondere reproduktive Potential, &ouml;kologische Toleranz und F&auml;higkeit, kultiviert werden. Diese Z&uuml;ge k&ouml;nnen schnell verschlechtern, wenn der biologische Kontrolle-Agent in Kultur aufgezogen ist. Mutually beneficial interactions between microorganisms and animals are a conserved and ubiquitous feature of biotic systems. In many instances animals, including humans, are dependent on their microbial associates for nutrition, defense, or development. To maintain these vital relationships, animals have evolved processes that ensure faithful transmission of specific microbial symbionts between generations. Elucidating mechanisms of transmission and symbiont specificity has been aided by the study of experimentally tractable invertebrate animals with diverse and highly evolved associations with microorganisms. Here, we review several invertebrate model systems that contribute to our current understanding of symbiont transmission, recognition, and specificity. Although the details of transmission and symbiont selection vary among associations, comparisons of diverse mutualistic associations are revealing a number of common themes, including restriction of symbiont diversity during transmission and glycan-lectin interactions during partner selection and recruitment.

En muchos casos animales, incluidos a los seres humanos, dependen de sus asociados microbianas para nutrici&oacute;n, defensa y desarrollo. The nematode Steinernema carpocapsae infects and kills many pest insects in agro-ecosystems and is commonly used in biocontrol of these pests. Growth of the nematodes prior to distribution for biocontrol commonly results in deterioration of traits that are essential for nematode persistence in field applications. To better understand the mechanisms underlying trait deterioration of the efficacy of natural parasitism in entomopathogenic nematodes, we explored the maintenance of fitness related traits including reproductive capacity, heat tolerance, virulence to insects and 'tail standing' (formerly called nictation) among laboratory-cultured lines derived from natural, randomly mating populations of S. carpocapsae. Laboratory cultured nematode lines with fitness-related trait values below wild-type levels regained wild-type levels of reproductive and heat tolerance traits when outcrossed with a non-deteriorated line, while virulence and 'tail standing' did not deteriorate in our experiments. Crossbreeding two trait-deteriorated lines with each other also resulted in restoration of trait means to wild-type levels in most crossbred lines. Our results implicate inbreeding depression as the primary cause of trait deterioration in the laboratory cultured S. carpocapsae. We further suggest the possibility of creating inbred lines purged of deleterious alleles as founders in commercial nematode growth.

Croissance des n&eacute;matodes avant la distribution pour la lutte biologique se traduit commun&eacute;ment par d&eacute;t&eacute;rioration des traits qui sont essentielles pour la persistance de n&eacute;matode dans les applications de terrain. Pour mieux comprendre les m&eacute;canismes qui sous-tendent la d&eacute;t&eacute;rioration de la caract&eacute;ristique de l&#39;efficacit&eacute; du parasitisme naturel dans les n&eacute;matodes entomopathog&egrave;nes, nous avons explor&eacute; le maintien d&#39;aptitude li&eacute;e caract&eacute;ristiques, y compris la capacit&eacute; de reproduction, tol&eacute;rance &agrave; la chaleur, la virulence aux insectes et aux &#39;queue debout&quot;(anciennement appel&eacute; nictation) entre les lign&eacute;es cultiv&eacute;es en laboratoire d&eacute;riv&eacute; naturel, accouplement au hasard des populations de S. carpocapsae. Labor kultiviert Fadenwurm Linien mit Fitness-&auml;hnliche Merkmal-Werte unter Wildtyp Ebenen wiedererlangt Wildtyp Ebenen der Fortpflanzungsmedizin und Hitze Toleranz-Merkmale bei outcrossed mit einer Linie nicht verschlechtert, w&auml;hrend Virulenz und &#39;Schwanz stehend&#39; in unseren Experimenten nicht verschlechtert. Kreuzung von zwei Merkmal verschlechterte sich Linien miteinander f&uuml;hrte auch Restaurierung Merkmal bedeutet Wildtyp Ebenen der meisten gekreuzte Linien. Per comprendere meglio i meccanismi alla base di deterioramento tratto dell&#39;efficacia di parassitismo naturale in nematodi entomopatogeni, abbiamo esplorato il mantenimento dell&#39;idoneit&agrave; relativi tratti tra cui la capacit&agrave; riproduttiva, tolleranza di calore, la virulenza di insetti e &#39;coda in piedi&#39; (precedentemente chiamato nictation) tra le linee coltivate in laboratorio derivata dalla naturale, casualmente l&#39;accoppiamento delle popolazioni di S. carpocapsae. Members of the genus Xenorhabdus are entomopathogenic bacteria that associate with nematodes. The nematode-bacteria pair infects and kills insects, with both partners contributing to insect pathogenesis and the bacteria providing nutrition to the nematode from available insect-derived nutrients. The nematode provides the bacteria with protection from predators, access to nutrients, and a mechanism of dispersal. Members of the bacterial genus Photorhabdus also associate with nematodes to kill insects, and both genera of bacteria provide similar services to their different nematode hosts through unique physiological and metabolic mechanisms. We posited that these differences would be reflected in their respective genomes. To test this, we sequenced to completion the genomes of Xenorhabdus nematophila ATCC 19061 and Xenorhabdus bovienii SS-2004. As expected, both Xenorhabdus genomes encode many anti-insecticidal compounds, commensurate with their entomopathogenic lifestyle. Despite the similarities in lifestyle between Xenorhabdus and Photorhabdus bacteria, a comparative analysis of the Xenorhabdus, Photorhabdus luminescens, and P. asymbiotica genomes suggests genomic divergence. These findings indicate that evolutionary changes shaped by symbiotic interactions can follow different routes to achieve similar end points.

Xenorhabdus bovienii (SS-2004) bacteria reside in the intestine of the infective-juvenile (IJ) stage of the entomopathogenic nematode, Steinernema jollieti. The recent sequencing of the X.â€ƒbovienii genome facilitates its use as a model to understand host - symbiont interactions. To provide a biological foundation for such studies, we characterized X.â€ƒbovienii in vitro and host interaction phenotypes. Within the nematode host X.â€ƒbovienii was contained within a membrane bound envelope that also enclosed the nematode-derived intravesicular structure. Steinernema jollieti nematodes cultivated on mixed lawns of X.â€ƒbovienii expressing green or DsRed fluorescent proteins were predominantly colonized by one or the other strain, suggesting the colonizing population is founded by a few cells. Xenorhabdus bovienii exhibits phenotypic variation between orange-pigmented primary form and cream-pigmented secondary form. Each form can colonize IJ nematodes when cultured in vitro on agar. However, IJs did not develop or emerge from Galleria mellonella insects infected with secondary form. Unlike primary-form infected insects that were soft and flexible, secondary-form infected insects retained a rigid exoskeleton structure. Xenorhabdus bovienii primary and secondary form isolates are virulent towards Manduca sexta and several other insects. However, primary form stocks present attenuated virulence, suggesting that X.â€ƒbovienii, like Xenorhabdus nematophila may undergo virulence modulation.

The gammaproteobacterium Xenorhabdus nematophila is a mutualistic symbiont that colonizes the intestine of the nematode Steinernema carpocapsae. nilB (nematode intestine localization) is essential for X. nematophila colonization of nematodes and is predicted to encode an integral outer membrane beta-barrel protein, but evidence supporting this prediction has not been reported. The function of NilB is not known, but when expressed with two other factors encoded by nilA and nilC, it confers upon noncognate Xenorhabdus spp. the ability to colonize S. carpocapsae nematodes. We present evidence that NilB is a surface-exposed outer membrane protein whose expression is repressed by NilR and growth in nutrient-rich medium. Bioinformatic analyses reveal that NilB is the only characterized member of a family of proteins distinguished by N-terminal region tetratricopeptide repeats (TPR) and a conserved C-terminal domain of unknown function (DUF560). Members of this family occur in diverse bacteria and are prevalent in the genomes of mucosal pathogens. Insertion and deletion mutational analyses support a beta-barrel structure model with an N-terminal globular domain, 14 transmembrane strands, and seven extracellular surface loops and reveal critical roles for the globular domain and surface loop 6 in nematode colonization. Epifluorescence microscopy of these mutants demonstrates that NilB is necessary at early stages of colonization. These findings are an important step in understanding the function of NilB and, by extension, its homologs in mucosal pathogens.

Mutational analyses reveal overall topology and functional regions of NilB, a bacterial outer membrane protein required for host association in a model of animal-microbe mutualism.

El gammaproteobacterium Xenorhabdus nematophila es un simbionte mutualista que coloniza el intestino del nematodo Steinernema carpocapsae. nilB (localizaci&oacute;n del intestino nematodo) es esencial para la colonizaci&oacute;n de X. nematophila de nematodos y se prev&eacute; que codifica una prote&iacute;na de membrana externa integral barril beta, pero no se ha divulgado la evidencia que apoya esta predicci&oacute;n. Se desconoce la funci&oacute;n de NilB, pero cuando se expresan con otros dos factores codificados por nilA y nilC, confiere noncognate Xenorhabdus spp. la capacidad para colonizar nematodos de S. carpocapsae. Presentamos evidencia que NilB es una prote&iacute;na de membrana externa superficie expuesta cuya expresi&oacute;n se reprime por NilR y crecimiento en medio rico en nutrientes. An&aacute;lisis bioinform&aacute;tico revelan que NilB es el &uacute;nico miembro caracterizado de una familia de prote&iacute;nas que se caracteriza por repeticiones de tetratricopeptide de regi&oacute;n N-terminal (TPR) y un dominio c-terminal conservado de funci&oacute;n desconocida (DUF560). Miembros de esta familia ocurren en diversas bacterias y son frecuentes en los genomas de mucosas pat&oacute;genos. Inserci&oacute;n y eliminaci&oacute;n apoyo de an&aacute;lisis mutacional, un modelo de estructura de barril beta con un dominio globular de N-terminal, 14 filamentos transmembrana y superficie extracelular siete lazos y revelar papeles cr&iacute;ticos para el dominio globular y la superficie del lazo 6 en nematodos colonizaci&oacute;n. Microscop&iacute;a de epifluorescencia de estos mutantes demuestra que NilB es necesario en las primeras etapas de la colonizaci&oacute;n. Estos resultados son un paso importante en la comprensi&oacute;n de la funci&oacute;n de NilB y, por extensi&oacute;n, sus hom&oacute;logos en mucosas pat&oacute;genos.
