(-)-Epigallocatechin gallate (EGCG), abundant in green tea, is a potent anti-microbial and anti-tumor compound. This investigation used immunoblot, flow cytometry, microarray, and indirect immunofluorescence analyses to show that at concentrations exceeding 50 microM, EGCG inhibits the expression of Epstein-Barr virus (EBV) lytic proteins, including Rta, Zta, and EA-D, but does not affect the expression of EBNA-1. Moreover, DNA microarray and transient transfection analyses demonstrated that EGCG blocks EBV lytic cycle by inhibiting the transcription of immediate-early genes, thus inhibiting the initiation of EBV lytic cascade.

A mutant library of 249 mutants with mutations that span the entire Epstein-Barr virus (EBV) genome was generated by transposition with EZ : : TN <KAN-2> and insertion with an apramycin resistance gene by a PCR-targeting method. This study also demonstrates the feasibility of generating deletions and site-specific mutations in the BRLF1 promoter on the EBV genome to determine the regions in the promoter that are crucial to transcription. Analysing BZLF1 and BRLF1 mutants by microarray analysis revealed that these two genes regulate the transcription of EBV lytic genes differently. A BZLF1 mutation affects global expression of EBV lytic genes; almost no lytic gene is expressed by the mutant after lytic induction. However, although a BRLF1 mutant still transcribes most lytic genes, the expression of these lytic genes is inefficient. Furthermore, this study shows that the proximal Zta-response element in the BRLF1 promoter is crucial to BRLF1 transcription from the EBV genome, despite the fact that another work demonstrated that this site was unimportant in transient transfection analysis. Furthermore, mutants with a mutation in BDLF1 and BORF1 cannot assemble viral capsids. Results of this study demonstrate the usefulness of a comprehensive mutant library in genetic analyses of EBV.

BRCA1-associated protein 2 (BRAP2) is known to interact with the kinase suppressor of Ras 1 (KSR1), inhibiting the ERK signal transduction cascade. This study found that an Epstein-Barr virus (EBV) immediate-early protein, Rta, is a binding partner of BRAP2 in yeast and confirmed the binding in vitro by a glutathione S-transferase pull-down assay and in vivo by co-immunoprecipitation in 293(maxi-EBV) cells. Binding studies also showed that Rta and KSR1 interacted with the C-terminal 202 aa region in BRAP2. Additionally, Rta appeared to prevent the binding of KSR1 to BRAP2, activating the ERK signal transduction pathway and the transcription of an EBV immediate-early gene, BZLF1. Activation of the ERK signal transduction pathway by Rta may be critical for the maintenance of the lytic state of EBV.

Reactivation of Epstein-Barr virus (EBV) from latency to the lytic cycle is required for the production of viral particles. Here, we examine the capacity of resveratrol to inhibit the EBV lytic cycle. Our results show that resveratrol inhibits the transcription of EBV immediate early genes, the expression of EBV lytic proteins, including Rta, Zta, and EA-D and reduces viron production, suggesting that this compound may be useful for preventing the proliferation of the virus.

This study demonstrates that BBLF1 is a myristoylated and palmitoylated protein, as are UL11 and UL99. Myristoylation of BBLF1 both facilitates its membrane anchoring and stabilizes it. BBLF1 is shown to localize to the trans-Golgi network (TGN) along with gp350/220, a site where final envelopment of EBV particles takes place. The localization of BBLF1 at the TGN requires myristoylation and two acidic clusters, which interact with PACS-1, a cytosolic protein, to mediate retrograde transport from the endosomes to the TGN. Knockdown of the expression of BBLF1 during EBV lytic replication reduces the production of virus particles, demonstrating the requirement of BBLF1 to achieve optimal production of virus particles. Virus Epstein - Barr es un virus de herpes gamma causal asociado a varios carcinomas y linfomas. Este virus codifica por lo menos 25 pre-miRNAs, que se expresan en c&eacute;lulas infectadas que rendir miRNAs maduros detectados m&aacute;s de 50. miRNAs son peque&ntilde;os, no codificante RNAs que inhiben la expresi&oacute;n g&eacute;nica mediante la promoci&oacute;n de la inhibici&oacute;n de la traducci&oacute;n o de degradaci&oacute;n de los mRNAs. Actualmente, la funci&oacute;n de estos miRNAs virales y la contribuci&oacute;n que aportan al ciclo de vida de EBV todav&iacute;a siguen sin conocerse, debido a las dificultades en la identificaci&oacute;n de genes celulares y virales regulados por estas miRNAs. Hemos comparado y contrastado dos m&eacute;todos para identificar los objetivos de miRNAs viral para identificar las ventajas y limitaciones de cada m&eacute;todo para ayudar a descubrir las funciones de miRNAs de EBV.

Epstein-Barr virus is a gamma-herpes virus that is causally associated with several lymphomas and carcinomas. This virus encodes at least 25 pre-miRNAs, which are expressed in infected cells to yield more than 50 detected mature miRNAs. miRNAs are small, non-coding RNAs that inhibit gene expression by promoting the inhibition of translation or of degradation of mRNAs. Currently, the function of these viral miRNAs and the contribution they provide to EBV's life-cycle remain largely unknown, due to difficulties in identifying cellular and viral genes regulated by these miRNAs. We have compared and contrasted two methods to identify targets of viral miRNAs in order to identify the advantages and limitations of each method to aid in uncovering the functions of EBV's miRNAs.

Actuellement, la fonction de ces miARN viraux et la contribution qu&#39;ils fournissent au cycle de vie du EBV demeurent largement inconnus, en raison de difficult&eacute;s dans l&#39;identification des g&egrave;nes virus et cellulaires r&eacute;gul&eacute;s par ces miARN. Nous avons compar&eacute; et compar&eacute; deux m&eacute;thodes pour identifier les cibles des miARN viraux afin d&#39;identifier les avantages et les limites de chaque m&eacute;thode pour aider &agrave; d&eacute;couvrir les fonctions des miARN de l&#39;EBV.

Epstein - Barr-Virus ist ein Gamma-Herpes-Virus, der mehrere Lymphome und Karzinome kausal zugeordnet ist. Dieser Virus kodiert mindestens 25 Pre-MiRNAs, die in infizierten Zellen zu mehr als 50 gefundenen MiRNAs Reife f&uuml;hren ausgedr&uuml;ckt werden. MiRNAs sind klein, nicht-kodierenden RNAs, die Genexpression hemmen durch die Hemmung der &Uuml;bersetzung oder der Abbau von mRNAs zu f&ouml;rdern. Derzeit Funktion diese virale MiRNAs und den Beitrag, den sie der EBV-Lebenszyklus zur Verf&uuml;gung stellen, bleiben weitgehend unbekannt, wegen Schwierigkeiten bei der Ermittlung der zellul&auml;ren und virale Gene, die durch diese MiRNAs reguliert. Wir haben verglichen und kontrastiert zwei Methoden, um die Ziele des viralen MiRNAs zu identifizieren, um die vor- und Nachteile der einzelnen Methoden zur Unterst&uuml;tzung der Aufdeckung der Funktionen der EBV die MiRNAs zu identifizieren.

Questo virus codifica per almeno 25 pre-miRNAs, che sono espressi in cellule infettate a produrre pi&ugrave; di 50 riconosciuto mature miRNAs. miRNAs sono piccole, non-coding RNAs che inibiscono l&#39;espressione genica promuovendo l&#39;inibizione della traduzione o della degradazione del mRNA. Attualmente, la funzione di questi miRNAs virale e il contributo che essi forniscono al ciclo di vita di EBV rimangono in gran parte sconosciuto, a causa delle difficolt&agrave; nell&#39;identificazione di geni virali e cellulari regolati da questi miRNAs.
