Ovarian cancer is a gynecological malignant tumor with a high mortality rate among women, owing to metastatic progression and recurrence. Acquisition of invasiveness is accompanied by the loss of epithelial features and a gain of a mesenchymal phenotype, a process known as epithelial-mesenchymal transition (EMT). Transforming growth factor-β (TGF-β) has been implicated in the regulation of EMT. In the present study, we aimed to investigate the role of long noncoding RNA H19 and microRNA-370 (miR-370-3p) in TGF-β-induced EMT. Ovarian cancer cell lines SKOV-3 and OVCAR3 were incubated with different concentrations of TGF-β, and the results showed that TGF-β treatment upregulated H19 and downregulated miR-370-3p. In addition, an H19 knockdown or miR-370-3p overexpression suppressed TGF-β-induced EMT, while H19 overexpression or a miR-370-3p knockdown promoted TGF-β-induced EMT. Mechanistically, H19 could directly bind to miR-370-3p and effectively act as its competing endogenous RNA. Furthermore, we demonstrated that this activity of H19 was involved in its promotion of TGF-β-induced EMT. Thus, our results may provide novel insights into the process of TGF-β-induced EMT.

Long noncoding RNA H19 promotes transforming growth factor-β-induced epithelial-mesenchymal transition by acting as a competing endogenous RNA of miR-370-3p in ovarian cancer cells.

Antimicrobial photodynamic inactivation (aPDI) employs photosensitizing dyes activated by visible light to produce reactive oxygen species. aPDI is independent of the antibiotic resistance status of the target cells, and is thought unlikely to produce resistance itself. Among many PS that have been investigated, tetracyclines occupy a unique niche. They are potentially dual-action compounds that can both kill bacteria under illumination, and prevent bacterial regrowth by inhibiting ribosomes. Tetracycline antibiotics are regarded as bacteriostatic rather than bactericidal. Doxycycline (DOTC) is excited best by UVA light (365 nm) while demeclocycline (DMCT) can be efficiently activated by blue light (415 nm) as well as UVA. Both compounds were able to eradicate Gram-positive (methicillin-resistant Staphylococcus aureus) and Gram-negative (Escherichia coli) bacteria (>6 log(10) steps of killing) at concentrations (10-50μM) and fluences (10-20J/cm2). In contrast to methylene blue, MB plus red light, tetracyclines photoinactivated bacteria in rich growth medium. When ~3 logs of bacteria were killed with DMCT/DOTC+light and the surviving cells were added to growth medium, further bacterial killing was observed, while the same experiment with MB allowed complete regrowth. MIC studies were carried out either in the dark or exposed to 0.5mW/cm2 blue light. Up to three extra steps (8-fold) increased antibiotic activity was found with light compared to dark, with MRSA and tetracycline-resistant strains of E. coli. Tetracyclines can accumulate in bacterial ribosomes, where they could be photoactivated with blue/UVA light producing microbial killing via ROS generation.

Tetracyclines function as dual-action light-activated antibiotics.

Tetracyclines (including demeclocycline, DMCT, or doxycycline, DOTC) represent a class of dual-action antibacterial compounds, which can act as antibiotics in the dark, and also as photosensitizers under illumination with blue or UVA light. It is known that tetracyclines are taken up inside bacterial cells where they bind to ribosomes. In the present study, we investigated the photochemical mechanism: Type 1 (hydroxyl radicals); Type 2 (singlet oxygen); or Type 3 (oxygen independent). Moreover, we asked whether addition of potassium iodide (KI) could potentiate the aPDI activity of tetracyclines. High concentrations of KI (200-400 mM) strongly potentiated (up to 5 logs of extra killing) light-mediated killing of Gram-negative Escherichia coli or Gram-positive MRSA (although the latter was somewhat less susceptible). KI potentiation was still apparent after a washing step showing that the iodide could penetrate the E. coli cells where the tetracycline had bound. When cells were added to the tetracycline + KI mixture after light, killing was observed in the case of E. coli showing formation of free molecular iodine. Addition of azide quenched the formation of iodine but not hydrogen peroxide. DMCT but not DOTC iodinated tyrosine. Both E. coli and MRSA could be killed by tetracyclines plus light in the absence of oxygen and this killing was not quenched by azide. A mouse model of a superficial wound infection caused by bioluminescent E. coli could be treated by topical application of DMCT and blue light and bacterial regrowth did not occur owing to the continued anti biotic activity of the tetracycline.

Successful treatment of polymorphic light eruption with UVA rush hardening: A report of 5 cases.

Sepsis is a systemic inflammatory reaction caused by various infectious or noninfectious factors, which can lead to shock, multiple organ dysfunction syndrome, and death. It is one of the common complications and a main cause of death in critically ill patients. At present, the treatments of sepsis are mainly focused on the controlling of inflammatory response and reduction of various organ function damage, including anti-infection, hormones, mechanical ventilation, nutritional support, and traditional Chinese medicine (TCM). Among them, Xuebijing injection (XBJI) is an important derivative of TCM, which is widely used in clinical research. However, the molecular mechanism of XBJI on sepsis is still not clear. The mechanism of treatment of "bacteria, poison and inflammation" and the effects of multi-ingredient, multi-target, and multi-pathway have still not been clarified. For solving this issue, we designed a new systems pharmacology strategy which combines target genes of XBJI and the pathogenetic genes of sepsis to construct functional response space (FRS). The key response proteins in the FRS were determined by using a novel node importance calculation method and were condensed by a dynamic programming strategy to conduct the critical functional ingredients group (CFIG). The results showed that enriched pathways of key response proteins selected from FRS could cover 95.83% of the enriched pathways of reference targets, which were defined as the intersections of ingredient targets and pathogenetic genes. The targets of the optimized CFIG with 60 ingredients could be enriched into 182 pathways which covered 81.58% of 152 pathways of 1,606 pathogenetic genes. The prediction of CFIG targets showed that the CFIG of XBJI could affect sepsis synergistically through genes such as TAK1, TNF-α, IL-1β, and MEK1 in the pathways of MAPK, NF-κB, PI3K-AKT, Toll-like receptor, and tumor necrosis factor signaling. Finally, the effects of apigenin, baicalein, and luteolin were evaluated by  experiments and were proved to be effective in reducing the production of intracellular reactive oxygen species in lipopolysaccharide-stimulated RAW264.7 cells, significantly. These results indicate that the novel integrative model can promote reliability and accuracy on depicting the CFIGs in XBJI and figure out a methodological coordinate for simplicity, mechanism analysis, and secondary development of formulas in TCM.

Detecting Critical Functional Ingredients Group and Mechanism of Xuebijing Injection in Treating Sepsis.

Anxiety and depression in nasopharyngeal carcinoma patients and network analysis to identify central symptoms: A cross-sectional study from a high-incidence area.

Patients with residual nasopharyngeal carcinoma after receiving definitive treatment have poor prognoses. Although immune checkpoint therapies have achieved breakthroughs for treating recurrent and metastatic nasopharyngeal carcinoma, none of these strategies have been assessed for treating residual nasopharyngeal carcinoma. In this single-arm, phase 2 trial, we aimed to evaluate the antitumor efficacy and safety of toripalimab (anti-PD1 antibody) plus capecitabine in patients with residual nasopharyngeal carcinoma after definitive treatment (ChiCTR1900023710). Primary endpoint of this trial was the objective response rate assessed according to RECIST (version 1.1). Secondary endpoints included complete response rate, disease control rate, duration of response, progression-free survival, safety profile, and treatment compliance. Between June 1, 2020, and May 31, 2021, 23 patients were recruited and received six cycles of toripalimab plus capecitabine every 3 weeks. In efficacy analyses, 13 patients (56.5%) had complete response, and 9 patients (39.1%) had partial response, with an objective response rate of 95.7% (95% CI 78.1-99.9). The trial met its prespecified primary endpoint. In safety analyses, 21 of (91.3%) 23 patients had treatment-related adverse events. The most frequently reported adverse event was hand-foot syndrome (11 patients [47.8%]). The most common grade 3 adverse event was hand-foot syndrome (two patients [8.7%]). No grades 4-5 treatment-related adverse events were recorded. This phase 2 trial shows that combining toripalimab with capecitabine has promising antitumour activity and a manageable safety profile for patients with residual nasopharyngeal carcinoma.

Capecitabine Maintenance Therapy in Patients with Residual Nasopharyngeal Carcinoma: A Single-Arm, Phase II Trial.

Bst polymerase is a large fragment of DNA polymerase I with strong strand displacement activity and broad substrate adaptability. It is one of the most commonly used enzymes in developing isothermal DNA amplification reactions for trace nucleic acid detection. Although many of these methods are ultrasensitive and specific, they often suffer from complex primer design, multienzyme dependency, and complicated amplification systems relating to unexpected side reactions and false reported signals. In this work, we designed a simple, rapid, and accurate Bst polymerase-catalyzed isothermal exponential amplification reaction (named BIEAR) for multiplex detection of microRNAs and tried it in diabetic retinopathy (DR) screening and progression risk assessment. The BIEAR system relies solely on one DNA hairpin triggering probe, one double-stranded DNA signal probe, and one type of Bst polymerase to drive bidirectional strand extension and displacement reaction cycles, which produce exponentially amplified fluorescence signals quantitatively dependent on the target microRNAs. The system has achieved pM-graded detection limits in individual and multiplex detection of miR-21 and miR-93, and the reaction time for detecting miR-93 is only 10 min. It exhibited high accuracy with 99.20-105.6% recovery rates in measuring miR-21 in human plasma samples. We have also applied BIEAR to detect concentrations of blood circulating miR-21 and miR-93 and combined them as an indicator for screening DR, achieving a diagnostic sensitivity of 93.80%, a specificity of 84.80%, and an area under the curve of 0.952. This novel method provides a simple, convenient, and accurate approach for potential molecular diagnosis such as early screening of DR.

A Simple and Rapid Bst Polymerase-Catalyzed Isothermal Exponential Amplification Reaction for Multiplex Detection of MicroRNAs.

In nasopharyngeal carcinoma, cisplatin is known to be associated with poor treatment compliance and notable side effects. More effective and safer platinum drugs are needed for the treatment of patients with nasopharyngeal carcinoma. In 2021, our multicenter, randomized, phase 3 trial reported that lobaplatin and fluorouracil induction chemotherapy plus concurrent chemoradiotherapy resulted in non-inferior survival and fewer toxic effects than did cisplatin-based therapy in nasopharyngeal carcinoma. Data from the 10-year survival analysis are updated here. With a median follow-up of 10.6 years in the intention-to-treat population, 10-year progression-free survival is 70.7% in the lobaplatin-based therapy group vs. 71.9% in the cisplatin-based therapy group (HR 1.02, 95% CI 0.72-1.43; log-rank p = 0.885). The difference between the groups is 1.2% (95% CI -6.7-9.1, p = 0.015), which is lower than the prespecified non-inferiority margin of 10%. The results are similar when we analyze patients in the per-protocol population. In the univariable and multivariable analyses, stage is an independent prognostic factor for progression-free survival (p = 0.001). The subgroup analyses suggest that the non-inferiority of lobaplatin-based therapy did not differ among specific populations. The incidence of late toxic effects is similar between the therapy groups, except for grades 1-2 peripheral neuropathy (p = 0.033), grades 1-2 deafness/otitis (p = 0.021), and grades 1-2/3 nephrotoxicity (p = 0.005; p = 0.021), the incidence of which is greater in the cisplatin-based therapy group than in the lobaplatin-based therapy group. Our findings suggest that lobaplatin and fluorouracil induction chemotherapy plus lobaplatin-based concurrent chemoradiotherapy is an alternative doublet treatment strategy to cisplatin-based concurrent chemoradiotherapy in patients with locoregionally advanced nasopharyngeal carcinoma.
