Protein-protein interactions play an important role in many virus-encoded functions and in virus-host interactions. While a "classical" yeast two-hybrid system (Y2H) is one of the most common techniques to detect such interactions, it has a number of limitations, including a requirement for the proteins of interest to be relocated to the nucleus. Modified Y2H, such as the Sos recruitment system (SRS), which detect interactions occurring in the cytoplasm rather than the nucleus, allow proteins from viruses replicating in the cytoplasm to be tested in a more natural context. In this study, a SRS was used to detect interactions involving proteins from vesicular stomatitis virus (VSV), a prototypic non-segmented negative strand RNA (NNS) virus. All five full-length VSV proteins, as well as several truncated proteins, were screened against each other. Using the SRS, most interactions demonstrated previously involving VSV phosphoprotein, nucleocapsid (N) and large polymerase proteins were confirmed independently, while difficulties were encountered using the membrane associated matrix and glycoproteins. A human cDNA library was also screened against VSV N protein and one cellular protein, SFRS18, was identified which interacted with N in this context. The system presented can be redesigned easily for studies in other less tractable NNS viruses.

Detecting protein-protein interactions in vesicular stomatitis virus using a cytoplasmic yeast two hybrid system.

Interacciones prote&iacute;na-prote&iacute;na juegan un papel importante en muchas funciones de virus-codificado y en virus-hu&eacute;sped interacciones. Mientras que un sistema de dos h&iacute;bridos de levadura &quot;cl&aacute;sica&quot; (Y2H) es una de las t&eacute;cnicas m&aacute;s comunes para detectar tales interacciones, tiene una serie de limitaciones, incluyendo un requisito para las prote&iacute;nas de inter&eacute;s a ser reasentados en el n&uacute;cleo. Y2H modificado, como el sistema de reclutamiento de Sos (SRS), que detecta las interacciones que ocurren en el citoplasma m&aacute;s el n&uacute;cleo, permiten a las prote&iacute;nas de virus que se replican en el citoplasma para ser probado en un contexto m&aacute;s natural. En este estudio, se utiliz&oacute; un SRS para detectar interacciones con las prote&iacute;nas del virus de la estomatitis vesicular (VSV), un filamento negativo no segmentado un virus ARN (NNS). Se revisaron las cinco prote&iacute;nas VSV integrales, as&iacute; como varias prote&iacute;nas truncadas, cara a cara. Usando el SRS, mayor&iacute;a de las interacciones hab&iacute;an demostrado previamente que fosfoprote&iacute;na VSV, nucleoc&aacute;pside (N) y prote&iacute;nas grandes polimerasa fueron confirmadas independientemente, mientras eran dificultades usando la membrana asociada matriz y glicoprote&iacute;nas. Una biblioteca de cDNA humano tambi&eacute;n se proyect&oacute; contra la prote&iacute;na VSV N y una prote&iacute;na celular, SFRS18, fue identificada que interactuaron con N en este contexto. El sistema presentado puede ser redise&ntilde;ado f&aacute;cilmente para estudios en otros virus NNS menos manejables.
