Non-SMC condensin I complex subunit G (NCAPG) is expressed in various human cancers, including gliomas. However, its biological function in glioma remains unclear. The present study was designed to determine the biological functions of NCAPG in glioma and to evaluate the association of NCAPG expression with glioma progression.

NCAPG as a Novel Prognostic Biomarker in Glioma.

Gliomas account for 75% of all primary malignant brain tumors in adults and are associated with high mortality. Emerging evidence has demonstrated that baculoviral inhibitor of apoptosis repeat containing 5 () plays a critical role in cell apoptosis and the progression of diverse cancers. However, no studies have yet focused on the immunological function and mechanisms of upstream  regulation in the progression of low-grade gliomas (LGG). Here, we evaluated  expression and clinical characteristics in people with LGG using the Chinese Glioma Genome Atlas, The Cancer Genome Atlas, Gene Expression Omnibus, Rembrandt, and Gravendeel databases. We used Kaplan-Meier statistics and receiver operating characteristic (ROC) curves to analyze the prognostic value of  in LGG. Kyoto Encyclopedia of Genes and Genomes (KEGG) and Gene Ontology (GO) enrichment terms were also explored to identify functional roles of . The Tumor Immune Estimation Resource (TIMER) and Tumor Immune System Interaction (TISIDB) databases were used to examine the correlation between  expression and immune cell infiltration in LGG. The Genomics of Drug Sensitivity in Cancer (GDSC) and Cancer Therapeutics Response Portal (CTRP) databases were used to examine the potential drugs targeting . We used transwell and wound healing assays to determine the biological functions of  in glioma cell migration. Our results demonstrated that  was highly expressed in LGG and the expression level correlated with tumor grade, prognosis, histological subtype, isocitrate dehydrogenase 1 () mutation, 1p/19q chromosomal co-deletion, chemotherapy status, and O[6]-methylguanine-DNA methyltransferase (MGMT) promoter methylation status. GO and KEGG analysis showed that  is primarily involved in cell proliferation and immune response-related signaling pathways. We also found that  was significantly correlated with m6A modification and diverse drug sensitivity. TIMER and TISIDB database analysis showed that  expression is associated with infiltration of diverse immune cells and immune modulation in LGG.  knockdown inhibited LGG cell migration. Collectively, our results demonstrate that  is correlated with cell migration and immune infiltration in LGG and may be a useful prognostic biomarker.

The Coiled-coil domain-containing proteins (CCDCs) are expressed in many cancers, but the role of Coiled-coil domain-containing protein 103 (CCDC103) in cancers remains unclear. Further investigations are necessary to ascertain its diagnostic significance and understand its biological function in cancers. This study aims to elucidate the biological functionalities of CCDC103 in glioma and evaluate the correlation between CCDC103 expression with glioma progression.

CCDC103 as a Prognostic Biomarker Correlated with Tumor Progression and Immune Infiltration in Glioma.

 Dihydrolipoamide S-acetyltransferase (DLAT) is one of the cuproptosis-related genes (CRGs). Increasing evidence suggests that DLAT plays a critical role in various cancers. However, information about its functions and potential mechanisms in liver hepatocellular carcinoma (HCC) is still limited.  Bioinformatics analysis were used to evaluate the potential association of DLAT expression with N6-methyladenosine (m6A) modification, clinical features, survival prognosis, biological functions, immune infiltration, and immune checkpoint molecules (ICM) in tumor patients. In addition, the expression of DLAT in HCC tissues was verified using immunohistochemistry (IHC).  The aberrant expression of DLAT had a significant impact on the prognosis of patients with various tumors. Importantly, DLAT expression was strongly associated with immune cell infiltration and immune checkpoint molecules (ICM) in tumors. For the first time, we found a significant positive correlation between DLAT expression and m6A regulatory factors in liver cancer, and that DLAT is associated with the PLK1 pathway, PI3K-AKT signaling pathway, Notch signaling pathway, WNT signaling pathway, and Aurora B pathway.  Our results showed a significant increase in DLAT expression in HCC. Furthermore, the prognosis of patients with high DLAT expression was poor. Importantly, DLAT was correlated with immune cell infiltration and immune checkpoint molecules in HCC patients. Together, our results indicate that DLAT represents a promising therapeutic target for HCC patients.
