Spinal muscular atrophy (SMA) is an autosomal recessive disorder, which is the leading genetic cause of infantile death. SMA is the most common inherited motor neuron disease and occurs in approximately 1:6000 live births. The gene responsible for SMA is called Survival Motor Neuron-1 (SMN1). Interestingly, a human-specific copy gene is present on the same region of chromosome 5q, called SMN2. Motor neurons are the primary tissue affected in SMA. Although it is clear that SMA is a neurodegenerative disease, there are clinical reports that suggest that other tissues contribute to the overall phenotype, especially in the most severe forms of the disease. In severe SMA cases, a growing number of congenital heart defects have been identified upon autopsy. The most common defect is a developmental defect referred to as hypoplastic left heart. The purpose of this report is to determine whether cardiac tissue is altered in SMA models and whether this could contribute to SMA pathogenesis. Here we identified early-stage developmental defects in a severe model of SMA. Additionally, pathological responses including fibrosis and oxidative stress markers were observed shortly after birth in a less severe model of disease. Similarly, functional differences were detected between wild-type and early-stage SMA animals. Collectively, this work demonstrates the importance of cardiac development and function in these severe models of SMA.

Cardiac defects contribute to the pathology of spinal muscular atrophy models.

L&#x26;#39;amyotrophie spinale (SMA) est une maladie autosomique r&#xE9;cessive, qui est la principale cause g&#xE9;n&#xE9;tique de la mortalit&#xE9; infantile. SMA est le plus commun du moteur maladie h&#xE9;r&#xE9;ditaire neurone et se produit dans environ 1:6000 naissances vivantes. Le g&#xE8;ne responsable de la SMA est appel&#xE9; la survie du motoneurone-1 (SMN1). Int&#xE9;ressant, un g&#xE8;ne humain copie sp&#xE9;cifique est pr&#xE9;sent sur la m&#xEA;me r&#xE9;gion du chromosome 5q, appel&#xE9; SMN2. Les neurones moteurs sont le tissu primaire affect&#xE9;e &#xE0; SMA. Ici, nous avons identifi&#xE9; &#xE0; un stade pr&#xE9;coce des anomalies du d&#xE9;veloppement dans un mod&#xE8;le de grave de SMA. En outre, les r&#xE9;ponses pathologiques, y compris la fibrose et les marqueurs de stress oxydatif ont &#xE9;t&#xE9; observ&#xE9;s peu apr&#xE8;s la naissance dans un mod&#xE8;le moins s&#xE9;v&#xE8;re de la maladie. Spinale Muskelatrophie (SMA) ist eine autosomal-rezessiv vererbt, die die genetische Todesursache infantile ist. SMA ist die h&auml;ufigste vererbte Motoneuron-Erkrankung und tritt bei etwa 1:6000 Lebendgeburten. Das Gen, das verantwortlich f&uuml;r SMA hei&szlig;t Survival Motor Neuron-1 (SMN1). Interessant ist, ist ein Mensch-spezifische Kopie-gen auf der gleichen Region des Chromosoms 5q, genannt SMN2 pr&auml;sent. Motoneuronen sind das prim&auml;re Gewebe in SMA betroffen. Der Zweck dieses Berichts ist zu bestimmen, ob Herzgewebe in SMA-Modellen ge&auml;ndert wird und ob dies zu SMA-Pathogenese beitragen k&ouml;nnte. Hier haben wir fr&uuml;he Entwicklungsst&ouml;rungen M&auml;ngel in einem schweren Modell der SMA identifiziert. Dar&uuml;ber hinaus wurden kurz nach der Geburt in einem weniger schweren Krankheit pathologische Reaktionen einschlie&szlig;lich Fibrose und oxidativen Stress Marker beobachtet. Ebenso wurden funktionale Unterschiede zwischen Wildtyp und Fr&uuml;hphasen-SMA Tieren entdeckt. Zusammenfassend zeigt diese Arbeit die Bedeutung der kardialen Entwicklung und Funktion in diesen schweren Modellen von SMA.

SMA &egrave; la pi&ugrave; comune malattia del motoneurone ereditati e si verifica in circa 1:6000 nati vivi. Il gene responsabile della SMA &egrave; chiamato Survival Motor Neuron-1 (SMN1). &Egrave; interessante notare che, un gene umano specifico copia &egrave; presente sulla stessa regione del cromosoma 5q, denominato SMN2. Motoneuroni sono il tessuto primario colpito in SMA. Anche se &egrave; chiaro che la SMA &egrave; una malattia neurodegenerativa, ci sono rapporti clinici che suggeriscono che altri tessuti contribuiscono per il fenotipo complessivo, soprattutto nelle forme pi&ugrave; gravi della malattia. Nei casi gravi di SMA, un numero crescente di difetti cardiaci congeniti &egrave; stato identificato all&#39;autopsia. Il difetto pi&ugrave; comune &egrave; un difetto dello sviluppo denominato cuore sinistro ipoplasico. Lo scopo di questa relazione &egrave; quello di determinare se il tessuto cardiaco &egrave; alterato in SMA modelli e se questo potrebbe contribuire alla patogenesi della SMA. Qui abbiamo identificato nelle prime fasi difetti dello sviluppo in un modello di SMA grave. Inoltre, sono state osservate risposte patologiche tra cui fibrosi e marcatori di stress ossidativo poco dopo la nascita in un modello meno grave della malattia. Allo stesso modo, sono state rilevate differenze funzionali tra animali SMA wild-type ed early stage. La atrofia muscular espinal (AME) es un trastorno autos&#xF3;mico recesivo, lo que es la principal causa gen&#xE9;tica de la muerte infantil. SMA es el m&#xE1;s com&#xFA;n el motor heredado la enfermedad de las neuronas y ocurre en aproximadamente 1:6000 nacidos vivos. El gen responsable de la SMA se llama neurona motora de supervivencia-1 (SMN1). Curiosamente, un gen copia humano espec&#xED;fico est&#xE1; presente en la misma regi&#xF3;n del cromosoma 5q, llamado SMN2. Las neuronas motoras son el tejido principal afectada en la SMA. Aunque est&#xE1; claro que SMA es una enfermedad neurodegenerativa, existen informes cl&#xED;nicos que sugieren que otros tejidos contribuyen al fenotipo general, especialmente en las formas m&#xE1;s graves de la enfermedad. En los casos graves de SMA, un n&#xFA;mero creciente de defectos cong&#xE9;nitos del coraz&#xF3;n se han identificado tras la autopsia. El defecto m&#xE1;s com&#xFA;n es un defecto del desarrollo conocido como coraz&#xF3;n izquierdo hipopl&#xE1;sico. El prop&#xF3;sito de este informe es determinar si el tejido cardiaco se altera en los modelos de SMA, y si esto podr&#xED;a contribuir a la patog&#xE9;nesis de SMA. Aqu&#xED; hemos identificado las primeras etapas defectos en el desarrollo de un modelo grave de SMA. Adem&#xE1;s, las respuestas patol&#xF3;gicas como la fibrosis y los marcadores de estr&#xE9;s oxidativo se observ&#xF3; poco despu&#xE9;s de nacer en un modelo menos grave de la enfermedad. Del mismo modo, las diferencias funcionales se han detectado entre los animales SMA de tipo salvaje y en etapa temprana. En conjunto, este trabajo demuestra la importancia del desarrollo y la funci&#xF3;n card&#xED;aca en estos modelos severas de SMA.
