The Mitotic Exit Network (MEN) proteins - including the protein kinase Cdc15 and the protein phosphatase Cdc14 - are essential for exit from mitosis in Saccharomyces cerevisiae. To identify downstream targets of the MEN, we sought telophase arrest bypassed (tab) mutations that bypassed the essential requirement for CDC15. Previous studies identified net1(tab2-1) and CDC14(TAB6-1) as mutations in the RENT complex subunits Net1 and Cdc14, respectively, and revealed that the MEN acts by promoting release of Cdc14 from its nucleolar Net1 anchor during anaphase. However, the remaining tab mutants were not characterized.

Multiple telophase arrest bypassed (tab) mutants alleviate the essential requirement for Cdc15 in exit from mitosis in S. cerevisiae.

Pour identifier les cibles en aval des hommes, nous avons cherch&eacute; des mutations d&#39;arrestation contourn&eacute;e (onglet) t&eacute;lophase non soumis &agrave; l&#39;exigence essentielle pour la CDC15. Des &eacute;tudes ant&eacute;rieures identifi&eacute; net1(tab2-1) et CDC14(TAB6-1) comme des mutations dans les sous-unit&eacute;s complexes louer Net1 et Cdc14, respectivement et a r&eacute;v&eacute;l&eacute; que les hommes agit en favorisant la lib&eacute;ration de Cdc14 de son ancre Net1 nucl&eacute;olaire durant l&#39;anaphase. Plusieurs mutants d&#39;arrestation contourn&eacute;e (onglet) t&eacute;lophase soulager la condition essentielle pour Cdc15 en sortie de la mitose chez S. cerevisiae.

Um nachgelagerte Ziele der M&auml;nner zu identifizieren, suchten wir Telophase Verhaftung umgangen (Registerkarte) Mutationen, die die Grundvoraussetzung f&uuml;r CDC15 umgangen. Fr&uuml;here Studien identifiziert net1(tab2-1) und CDC14(TAB6-1) als Mutationen in die Miete komplexen Untereinheiten Net1 und Cdc14, und ergab, dass die M&auml;nner durch die F&ouml;rderung der Freisetzung von Cdc14 aus seiner nukleol&auml;res Net1-Anker w&auml;hrend Anaphase fungiert. Allerdings wurden die verbleibenden Registerkarte Mutanten nicht gekennzeichnet.

When cells are primed to exit mitosis, the RENT complex is disassembled and Cdc14 is released from the nucleolus.

Prior to anaphase in Saccharomyces cerevisiae, Cdc14 protein phosphatase is sequestered within the nucleolus and inhibited by Net1, a component of the RENT complex in budding yeast. During anaphase the RENT complex disassembles, allowing Cdc14 to migrate to the nucleus and cytoplasm where it catalyzes exit from mitosis. The mechanism of Cdc14 release appears to involve the polo-like kinase Cdc5, which is capable of promoting the dissociation of a recombinant Net1.Cdc14 complex in vitro by phosphorylation of Net1. We report here the phosphorylation site mapping of recombinant Net1 (Net1N) and a mutant Net1N allele (Net1N-19m) with 19 serines or threonines mutated to alanine. A variety of chromatographic and mass spectrometric-based strategies were used, including immobilized metal-affinity chromatography, alkaline phosphatase treatment, matrix-assisted laser-desorption post-source decay, and a multidimensional electrospray mass spectrometry-based approach. No one approach was able to identify all phosphopeptides in the tryptic digests of these proteins. Most notably, the presence of a basic residue near the phosphorylated residue significantly hampered the ability of alkaline phosphatase to hydrolyze the phosphate moiety. A major goal of research in proteomics is to identify all proteins and their interactions and post-translational modification states. The failure of any single method to identify all sites in highly phosphorylated Net1N, however, raises significant concerns about how feasible it is to map phosphorylation sites throughout the proteome using existing technologies.

In many organisms, polo kinases appear to play multiple roles during M-phase progression. To provide new insights into the function of budding yeast polo kinase Cdc5p, we generated novel temperature-sensitive cdc5 mutants by mutagenizing the C-terminal domain. Here we show that, at a semipermissive temperature, the cdc5-3 mutant exhibited a synergistic bud elongation and growth defect with loss of HSL1, a component important for normal G(2)/M transition. Loss of SWE1, which phosphorylates and inactivates the budding yeast Cdk1 homolog Cdc28p, suppressed the cdc5-3 hsl1Delta defect, suggesting that Cdc5p functions at a point upstream of Swe1p. In addition, the cdc5-4 and cdc5-7 mutants exhibited chained cell morphologies with shared cytoplasms between the connected cell bodies, indicating a cytokinetic defect. Close examination of these mutants revealed delayed septin assembly at the incipient bud site and loosely organized septin rings at the mother-bud neck. Components in the mitotic exit network (MEN) play important roles in normal cytokinesis. However, loss of BFA1 or BUB2, negative regulators of the MEN, failed to remedy the cytokinetic defect of these mutants, indicating that Cdc5p promotes cytokinesis independently of Bfa1p and Bub2p. Thus, Cdc5p contributes to the activation of the Swe1p-dependent Cdc28p/Clb pathway, normal septin function, and cytokinesis.

Loss of CDC5 function in Saccharomyces cerevisiae leads to defects in Swe1p regulation and Bfa1p/Bub2p-independent cytokinesis.

Cooperative interactions are key to diverse biological phenomena ranging from multicellularity to mutualism. Such diversity makes the ability to create and control cooperation desirable for potential applications in areas as varied as agriculture, pollutant treatment, and medicine. Here we show that persistent cooperation can be engineered by introducing a small set of genetic modifications into previously noninteracting cell populations. Specifically, we report the construction of a synthetic obligatory cooperative system, termed CoSMO (cooperation that is synthetic and mutually obligatory), which consists of a pair of nonmating yeast strains, each supplying an essential metabolite to the other strain. The behavior of the two strains in isolation, however, revealed unintended constraints that restrict cooperation, such as asymmetry in starvation tolerance and delays in nutrient release until near cell death. However, the joint system is shown mathematically and experimentally to be viable over a wide range of initial conditions, with oscillating population ratio settling to a value predicted by nutrient supply and consumption. Unexpectedly, even in the absence of explicitly engineered mechanisms to stabilize cooperation, the cooperative system can consistently develop increased ability to survive reductions in population density. Extending synthetic biology from the design of genetic circuits to the engineering of ecological interactions, CoSMO provides a quantitative system for linking processes at the cellular level to the collective behavior at the system level, as well as a genetically tractable system for studying the evolution of cooperation.

What affects the ecology and evolution of symbioses? How do symbiotic interactions generate ecological patterns? How do symbiotic partners evolve and coevolve? Many of these questions are difficult to address in natural systems. Artificial systems, from abstract to living, have been constructed to capture essential features of natural symbioses and to address these key questions. With reduced complexity and increased controllability, artificial systems can serve as useful models for natural systems. We review how artificial systems have contributed to our understanding of symbioses.

Cooperation via production of common goods is found in diverse life forms ranging from viruses to social animals. However, natural selection predicts a "tragedy of the commons": Cheaters, benefiting from without producing costly common goods, are more fit than cooperators and should destroy cooperation. In an attempt to discover novel mechanisms of cheater control, we eliminated known ones using a yeast cooperator-cheater system engineered to supply or exploit essential nutrients. Surprisingly, although less fit than cheaters, cooperators quickly dominated a fraction of cocultures. Cooperators isolated from these cocultures were superior to the cheater isolates they had been cocultured with, even though these cheaters were superior to ancestral cooperators. Resequencing and phenotypic analyses revealed that evolved cooperators and cheaters all harbored mutations adaptive to the nutrient-limited cooperative environment, allowing growth at a much lower concentration of nutrient than their ancestors. Even after the initial round of adaptation, evolved cooperators still stochastically dominated cheaters derived from them. We propose the "adaptive race" model: If during adaptation to an environment, the fitness gain of cooperators exceeds that of cheaters by at least the fitness cost of cooperation, the tragedy of the commons can be averted. Although cooperators and cheaters sample from the same pool of adaptive mutations, this symmetry is soon broken: The best cooperators purge cheaters and continue to grow, whereas the best cheaters cause rapid self-extinction. We speculate that adaptation to changing environments may contribute to the persistence of cooperative systems before the appearance of more sophisticated mechanisms of cheater control.

Cooperaci&oacute;n mediante la producci&oacute;n de los bienes comunes se encuentra en diversas formas de vida que van desde virus hasta animales sociales. Sin embargo, la selecci&oacute;n natural predice una &quot;tragedia de los comunes&quot;: los tramposos, benefician sin producir costosos bienes comunes, son m&aacute;s ajuste que cooperadores y deben destruir la cooperaci&oacute;n. En un intento de descubrir nuevos mecanismos de control de tramposo, eliminamos conocidas, utilizando un sistema de cooperador-tramposo de levadura dise&ntilde;ado para suministrar o explotar los nutrientes esenciales. Sorprendentemente, aunque menos ajuste que los tramposos, cooperadores r&aacute;pidamente dominaban una fracci&oacute;n de cocultures. Cooperadores aislados de estos cocultures fueron superiores a los aislamientos de tramposo hab&iacute;an sido morfolog&iacute;a, aunque estos tramposos fueron superiores a los cooperadores ancestrales. An&aacute;lisis fenot&iacute;picos y resequencing revelaron que evolucionaron los tramposos y sus colaboradores todo abrigaron mutaciones adaptables al entorno cooperativo limitada nutrientes, permitiendo el crecimiento en una concentraci&oacute;n mucho m&aacute;s baja de nutrientes que sus antepasados. Incluso despu&eacute;s de la ronda inicial de adaptaci&oacute;n, evolucionado cooperadores tramposos a&uacute;n estoc&aacute;stico dominados derivados de los mismos. Se propone el modelo de &quot;raza adaptativa&quot;: Si durante la adaptaci&oacute;n a un entorno, el aumento de la aptitud de cooperadores supera de los tramposos por al menos el costo de la adecuaci&oacute;n de la cooperaci&oacute;n, la tragedia de los comunes puede evitarse.
