La malaria cerebral (CM) es m&aacute;s severo s&iacute;ndrome asociado a infecciones de Plasmodium falciparum. La evidencia experimental sugiere que la enfermedad resulta del secuestro de gl&oacute;bulos rojos-parasitadas (pRBCs) junto con leucocitos inflamatorios en los capilares del cerebro. Previamente hemos demostrado que las c&eacute;lulas NK estimularon la migraci&oacute;n de c&eacute;lulas de T de CXCR3(+) en el cerebro de ratones de ANKA-infectados de Plasmodium berghei. Aqu&iacute; Investigamos si las interacciones entre las c&eacute;lulas NK y c&eacute;lulas dendr&iacute;ticas (DCs) se requieren para la inducci&oacute;n de respuestas de c&eacute;lulas T implicadas en la enfermedad. Para ello, NK c&eacute;lula-agotado y ratones de control estaban infectados con par&aacute;sitos transg&eacute;nicas expresando epitopos del modelo T cell. Las c&eacute;lulas de T de ratones transg&eacute;nicos de TCR espec&iacute;ficos para los epitopos eventualmente fueron transferidas y se determin&oacute; la proliferaci&oacute;n. Depleci&oacute;n de c&eacute;lulas NK redujo significativamente CD8 pero no mediada por DC CD4 T cell cebado. Falta de las c&eacute;lulas NK no comprometer las respuestas de c&eacute;lulas T CD8 en ratones IL-12(-/-), sugiriendo que las c&eacute;lulas NK estimularon la producci&oacute;n de IL-12 por DCs necesaria para el cebado de la c&eacute;lula de T &oacute;ptima. Tambi&eacute;n se investig&oacute; la contribuci&oacute;n de DCs para la funci&oacute;n de las c&eacute;lulas NK. Agotamiento de la DC y supresi&oacute;n gen&eacute;tica de IL-12 reducen dram&aacute;ticamente respuestas de IFN-&gamma; mediada por c&eacute;lulas NK a la malaria. As&iacute; las c&eacute;lulas NK y DCs participan en activaci&oacute;n rec&iacute;proca para la inducci&oacute;n de respuestas inflamatorias en la malaria severa.

Cerebral malaria (CM) is the most severe syndrome associated with Plasmodium falciparum infections. Experimental evidence suggests that disease results from the sequestration of parasitized-red blood cells (pRBCs) together with inflammatory leukocytes within brain capillaries. We have previously shown that NK cells stimulate migration of CXCR3(+) T cells to the brain of Plasmodium berghei ANKA-infected mice. Here we investigated whether interactions between NK cells and dendritic cells (DCs) are required for the induction of T cell responses involved in disease. For that, NK cell-depleted and control mice were infected with transgenic parasites expressing model T cell epitopes. T cells from TCR transgenic mice specific for those epitopes were adoptively transferred and proliferation was determined. NK cell depletion significantly reduced CD8(+) but not CD4(+) DC-mediated T cell priming. Lack of NK cells did not compromise CD8(+) T cell responses in IL-12(-/-) mice, suggesting that NK cells stimulate IL-12 output by DCs required for optimal T cell priming. The contribution of DCs to NK cell function was also investigated. DC depletion and genetic deletion of IL-12 dramatically reduced NK cell-mediated IFN-Î³ responses to malaria. Thus NK cells and DCs engage in reciprocal activation for the induction of inflammatory responses involved in severe malaria.

Neuropaludisme (CM) est le plus grave syndrome associ&eacute; aux infections de Plasmodium falciparum. L&#39;&eacute;vidence exp&eacute;rimentale sugg&egrave;re que la maladie r&eacute;sulte de la s&eacute;questration de parasit&eacute;s-culots globulaires (pRBCs) ainsi que les leucocytes inflammatoires dans les capillaires du cerveau. Nous avons d&eacute;j&agrave; montr&eacute; que les cellules NK stimulent la migration des cellules de T CXCR3(+) au cerveau de souris de ANKA infect&eacute;e par Plasmodium berghei. Ici, nous avons examin&eacute; si les interactions entre les cellules NK et les cellules dendritiques (CD) sont n&eacute;cessaires pour l&#39;induction de lymphocytes T impliqu&eacute;s dans la maladie. Absence de cellules NK ne compromet pas les r&eacute;ponses des cellules T CD8 chez la souris IL-12(-/-), sugg&eacute;rant que les cellules NK stimulent l&#39;IL-12 sortie de contr&ocirc;leurs de domaine requis pour amorcer la pompe optimal des lymphocytes T. Ainsi les cellules NK et DCs engagent r&eacute;ciproque d&#39;activation pour l&#39;induction des r&eacute;ponses inflammatoires impliqu&eacute;s dans le paludisme grave.

Zerebrale Malaria (CM) ist die schwerste Syndrom mit Plasmodium Falciparum-Infektionen. Experimentelle Hinweise darauf, dass die Krankheit durch die Beschlagnahme von Einfluss-rote Blutk&ouml;rperchen (pRBCs) zusammen mit entz&uuml;ndlichen Leukozyten im Gehirn-Kapillaren bedingt. Wir haben bereits gezeigt, dass NK-Zellen Migration von CXCR3(+) T-Zellen des Gehirns von Plasmodium Berghei ANKA-infizierten M&auml;usen stimulieren. Hier untersuchten wir, ob Wechselwirkungen zwischen NK-Zellen und dendritischen Zellen (DCs) erforderlich f&uuml;r die Induktion von T-Zell-Antworten Krankheit beteiligt sind. Daf&uuml;r NK-Zelle-ersch&ouml;pft und Kontrolle M&auml;use mit Transgene Ausdruck Modell T-Zelle Epitopes Parasiten infiziert waren. T-Zellen aus TCR transgene M&auml;use speziell f&uuml;r diese Epitopes wurden adoptively verlegt und Verbreitung bestimmt war. NK Zelle Ersch&ouml;pfung deutlich reduziert, CD8(+) aber nicht CD4(+) DC-vermittelte T-Zell-Grundierung. Fehlen von NK-Zellen keine Kompromisse CD8(+) T Zelle Antworten bei IL-12(-/-) M&auml;usen, was darauf hindeutet, dass NK-Zellen IL-12 Ausgabe von DCs ben&ouml;tigt f&uuml;r optimale T-Zell-Grundierung anregen. Der Beitrag des DCs auf NK-Zellfunktion wurde auch untersucht. DC-Ersch&ouml;pfung und genetische L&ouml;schung von IL-12 reduziert drastisch NK-Zell-vermittelten IFN-&gamma;-Antworten zu Malaria. Teilzunehmen NK-Zellen und DCs wechselseitige Aktivierung f&uuml;r die Induktion von entz&uuml;ndlichen Reaktionen bei schwerer Malaria.

Prove sperimentali suggeriscono che la malattia deriva dal sequestro di globuli rossi parassiti (pRBCs) insieme a infiammatori dei leucociti all&#39;interno dei capillari cerebrali. In precedenza abbiamo dimostrato che le cellule NK stimolano la migrazione delle cellule T CXCR3(+) per il cervello di Plasmodium berghei ANKA-infettati topi. Qui abbiamo indagato se le interazioni tra le cellule NK e cellule dendritiche (DCs) sono necessarie per l&#39;induzione di cellule T risposte coinvolte nella malattia. Per questo, NK cell-impoverito e topi di controllo sono stati infettati da parassiti transgenici che esprimono epitopi T cell modello. Le cellule T da topi transgenici TCR specifici per tali epitopi adoptively sono state trasferite e proliferazione &egrave; stato determinato. Deplezione delle cellule NK ridotto significativamente le cellule ma non CD4(+) DC-mediata delle cellule T priming.
