&#x591A;&#x80FD;&#x5E72;&#x7684;&#x7315;&#x7334;&#x9AA8;&#x9AD3;&#x95F4;&#x5145;&#x8D28;&#x5E72;&#x7EC6;&#x80DE;&#x7684;&#x8868;&#x5F81;&#x3002;

The isolation and characterization of embryonic and adult stem cells from higher-order mammalian species will enhance the understanding of the biology and therapeutic application of stem cells. The aim of this study was to purify rhesus mesenchymal stem cells (MSCs) from adult bone marrow and to characterize functionally their abilities to differentiate along diverse lineages. Adherent cells from adult rhesus macaque bone marrow were characterized for their growth characteristics, lineage differentiation, cell-surface antigen expression, telomere length, chromosome content, and transcription factor gene expression. Rhesus bone marrow MSCs (BMSCs) are very heterogeneous, composed of primarily long, thin cells and some smaller, round cells. The cells are capable of differentiating along osteogenic, chondrogenic, and adipogenic lineages in vitro. The cell morphology and multipotential differentiation capabilities are maintained throughout extended culture. They express CD59, CD90 (Thy-1), CD105, and HLA-1 and were negative for hematopoietic markers such as CD3, CD4, CD8, CD11b, CD13, CD34, and platelet endothelial cell adhesion molecule-1 (CD31). BMSCs were also demonstrated to express the mRNA for important stem cell-related transcription factors such as Oct-4, Sox-2, Rex-1, and Nanog. Rhesus BMSCs have a normal chromosome content, and the shortening of telomeres is minimal during early passages. These data demonstrate that BMSCs isolated from rhesus macaques have a high degree of commonality with MSCs isolated from other species. Therefore, isolation of these cells provides an effective and convenient method for rapid expansion of pluripotent rhesus MSCs.

Characterization of multipotent mesenchymal stem cells from the bone marrow of rhesus macaques.

L&#39;isolement et la caract&eacute;risation des cellules souches embryonnaires et adultes d&#39;esp&egrave;ces de mammif&egrave;res ordre sup&eacute;rieur permettra d&#39;am&eacute;liorer la compr&eacute;hension de la biologie et de l&#39;utilisation th&eacute;rapeutique des cellules souches. Le but de cette &eacute;tude &eacute;tait de purifier les cellules souches de le rh&eacute;sus m&eacute;senchymateuses (CSM) de la moelle osseuse adulte et de caract&eacute;riser fonctionnellement leurs capacit&eacute;s &agrave; se diff&eacute;rencier selon des lign&eacute;es diff&eacute;rentes. Les cellules adh&eacute;rentes de la moelle &eacute;pini&egrave;re macaque rh&eacute;sus adultes ont &eacute;t&eacute; caract&eacute;ris&eacute;s pour leurs caract&eacute;ristiques de croissance, diff&eacute;renciation de la lign&eacute;e, expression de l&#39;antig&egrave;ne de surface de la cellule, la longueur des t&eacute;lom&egrave;res, contenu chromosomique et expression de g&egrave;ne de facteur de transcription. Moelle osseuse de rh&eacute;sus MSCs (BMSC) sont tr&egrave;s h&eacute;t&eacute;rog&egrave;ne, compos&eacute;e de cellules principalement longs, minces et quelques plus petits, autour des cellules. Les cellules sont capables de diff&eacute;rencier le long ost&eacute;og&eacute;nique, chondrog&eacute;niques et les lign&eacute;es adipocytaire in vitro. Les capacit&eacute;s de diff&eacute;renciation cellulaire morphologie et multipotential sont maintenues tout au long de la culture prolong&eacute;e. Ils expriment CD59, CD90 (Thy-1), CD105 et HLA-1 et ont &eacute;t&eacute; n&eacute;gatifs pour les marqueurs h&eacute;matopo&iuml;&eacute;tiques tels que CD3, CD4, CD8, CD11b, CD13, CD34 et plaquettes endothelial cell la mol&eacute;cule-1 adh&eacute;rence (CD31). Isolamento e caratterizzazione di cellule staminali embrionali e adulte da specie di mammiferi di ordine superiore miglioreranno la comprensione della biologia e applicazione terapeutica delle cellule staminali. Lo scopo di questo studio era di purificare rhesus le cellule staminali mesenchimali (MSCs) da adulta del midollo osseo e per caratterizzare funzionalmente le loro abilit&agrave; di differenziare lungo diversi lignaggi. Cellule aderenti da midollo osseo di macaco rhesus adulti furono caratterizzate per le loro caratteristiche di crescita, differenziazione di lignaggio, espressione dell&#39;antigene di superficie cellulare, lunghezza dei telomeri, contenuto del cromosoma ed espressione del gene transcription factor. Rhesus midollo osseo (BMSCs) MSCs sono molto eterogeneo, composto di cellule principalmente lunghe, sottile e alcuni pi&ugrave; piccoli, rotondi cellule. Le cellule sono in grado di differenziare lungo osteogeniche, chondrogenic e adipogenic lignaggi in vitro. La capacit&agrave; di differenziazione delle cellule morfologia e multipotential sono mantenute in tutta estesa cultura. Mesenchymal stem cells (MSC) derived from bone marrow stem cells (BMSC) and adipose tissue stem cells (ASC) of humans and rhesus macaques were evaluated for their cell cycle properties during protracted culture in vitro. Human ASCs (hASC) and rhesus BMSCs (rBMSC) underwent significantly more total population doublings than human BMSCs (hBMSC) and rhesus ASCs (rASC). The cell cycle profile of all MSCs was altered as cultures aged. hMSCs underwent an increase in the frequency of cells in the S phase at P20 and P30. However, rhesus MSCs from both sources developed a distinct polyploid population of cells at P20, which progressed to aneuploidy by P30. Karyotype analysis of MSCs revealed the development of tetraploid or aneuploid karyotypes in the rhesus cells at P20 or P30. Analysis of the transcriptome of the MSCs from early and late passages revealed significant alterations in the patterns of gene expression (8.8% of the genes were differentially expressed in hBMSCs versus hASCs, and 5.5% in rBMSCs versus rASCs). Gene expression changes were much less evident within the same cell type as aging occurred (0.7% in hMSCs and 0.9% in rMSC). Gene ontology analysis showed that functions involved in protein catabolism and regulation of pol II transcription were overrepresented in rASCs, whereas the regulation of I kappa B/nuclear factor-kappaB cascade were overrepresented in hBMSCs. Functional analysis of genes that were differentially expressed in rASCs and hBMSCs revealed that pathways involved in cell cycle, cell cycle checkpoints, protein-ubiquitination, and apoptosis were altered.

Long-term in vitro expansion alters the biology of adult mesenchymal stem cells.

Cellules souches m&eacute;senchymateuses (CSM) d&eacute;riv&eacute;s de cellules souches de la moelle osseuse (BMSC) et le tissu adipeux cellules souches (ASC) des &ecirc;tres humains et les macaques rh&eacute;sus ont &eacute;t&eacute; &eacute;valu&eacute;s pour leurs propri&eacute;t&eacute;s du cycle cellulaire au cours de la longue culture in vitro. ASCs humaines (hASC) et rh&eacute;sus BMSC (rBMSC) subit des doublements de population totale significativement plus que BMSC humaine (hBMSC) et rh&eacute;sus ASCs (SRAC). Gen Ontologie Analyse zeigte, dass Funktionen Protein Katabolismus und Regulierung der Pol II Transkription beteiligt &uuml;berrepr&auml;sentiert in rASCs waren, w&auml;hrend die Verordnung ich Kappa B/nuclear Factor-KappaB Kaskade in hBMSCs &uuml;berrepr&auml;sentiert waren. Funktionelle Analyse von Genen, die differentiell in rASCs und hBMSCs ausgedr&uuml;ckt wurden ergab, dass Wege Zellzyklus, Zellzyklus Checkpoints, Protein-Ubiquitination und Apoptose beteiligt ver&auml;ndert wurden.

Cellule staminali mesenchimali (MSC) derivate da cellule staminali del midollo osseo (BMSC) e tessuto adiposo cellule staminali (ASC) degli esseri umani e macachi rhesus sono stati valutati per le loro propriet&agrave; ciclo cellulare durante prolungata coltura in vitro. Umano ASCs (hASC) e BMSCs rhesus (rBMSC) sub&igrave; notevolmente pi&ugrave; totale popolazione raddoppi rispetto umano BMSCs (hBMSC) e rhesus ASCs (rASC). Il profilo di ciclo cellulare di tutte le MSCs &egrave; stato alterato come le culture di et&agrave;. hMSCs ha subito un aumento della frequenza di cellule in fase S a P20 e P30. Tuttavia, rhesus MSCs da entrambe le fonti sviluppato una popolazione poliploidi distinta di cellule a P20, che ha progredito per aneuploidia da P30. Analisi del cariotipo di MSCs ha rivelato lo sviluppo di cariotipi tetraploidi o aneuploidi nelle cellule rhesus P20 o P30. Gene ontology analisi ha mostrato che funzioni coinvolte nel catabolismo proteico e regolazione della trascrizione di pol II erano sovrarappresentate nel rASCs, considerando che il regolamento di I cascade kappa B/nuclear factor-kappaB erano sovrarappresentati nel hBMSCs. Analisi funzionale di geni differenzialmente espresse in rASCs e hBMSCs hanno rivelato che vie coinvolte nel ciclo cellulare, Checkpoint del ciclo cellulare, la proteina-ubiquitination e apoptosi sono stati alterati.

Las c&eacute;lulas madre mesenquimales (MSC) derivadas de c&eacute;lulas madre de m&eacute;dula &oacute;sea (CMMo) y tejido adiposo c&eacute;lulas (ASC) de seres humanos y macacos rhesus fueron evaluados por sus propiedades de ciclo celular durante prolongada cultivo in vitro. ASCs humanas (hASC) y BMSCs (rBMSC) del macaco de la India experimentaron significativamente m&aacute;s doblajes de la poblaci&oacute;n total de BMSCs humanas (hBMSC) y rhesus ASCs (rASC).
