Coronary artery disease (CAD) poses a worldwide health threat. Compelling evidence shows that pericardial adipose tissue (PAT), a brown-like adipose adjacent to the external surface of the pericardium, is associated with CAD. However, the specific molecular mechanisms of PAT in CAD are elusive. This study aims to characterize human PAT and explore its association with CAD.

[F]AlF-NOTA-ADH-1: A new PET molecular radiotracer for imaging of N-cadherin-positive tumors.

Bats, recognized as considerable reservoirs for coronaviruses (CoVs), serve as natural hosts for several highly pathogenic CoVs, including SARS-CoV and SARS-CoV-2. Investigating the bat CoV community provides insights into the origin for highly pathogenic CoVs and highlights bat CoVs with potential spillover risks. This study probes the evolution, recombination, host range, geographical distribution, and cross-species transmission characteristics of bat CoVs across China and its associated CoVs in other regions. Through detailed research on 13,064 bat samples from 14 provinces of China, 1141 CoV strains are found across 10 subgenera and one unclassified Alpha-CoV, generating 399 complete genome sequences. Within bat CoVs, 11 new CoV species are identified and 425 recombination events are detected. Bats in southern China, particularly in Yunnan province, exhibit a pronounced diversity of CoVs. Limited sampling and low detection rates exist for CoVs in Myotacovirus, Nyctacovirus, Hibecovirus, Nobecovirus in China. The genus Myotis is highlighted as a potential ancestral host for Alpha-CoV, with the genus Hipposideros suggested as a likely progenitor host for bat-associated Beta-CoV, indicating the complexity of cross-species transmission dynamics. Through the comprehensive analysis, this study enriches the understanding of bat CoVs and offers a valuable resource for future research.

Panoramic analysis of coronaviruses carried by representative bat species in Southern China to better understand the coronavirus sphere.

Bats (Order Chiroptera) are an important reservoir of emerging zoonotic microbes, including viruses of public health concern such as henipaviruses, lyssaviruses, and SARS-related coronaviruses. Despite the continued discovery of new viruses in bat populations, a significant proportion of these viral agents remain uncharacterized, highlighting the imperative for additional research aimed at elucidating their evolutionary relationship and taxonomic classification.

Unveiling bat-borne viruses: a comprehensive classification and analysis of virome evolution.

Transforming growth factor β (TGF-β) is well known to play paradoxical roles in tumorigenesis as it has both growth-inhibitory and pro-metastatic effects. However, the underlying mechanisms of how TGF-β drives the opposing responses remain largely unknown. Here, we report that ERBB4, a member of the ERBB receptor tyrosine kinase family, specifically promotes TGF-β's metastatic response but not its anti-growth response. ERBB4 directly phosphorylates Tyr162 in the linker region of SMAD4, which enables SMAD4 to achieve a higher DNA-binding ability and potentiates TGF-β-induced gene transcription associated with epithelial-to-mesenchymal transition (EMT), cell migration, and invasion without affecting the genes involved in growth inhibition. These selective effects facilitate lung cancer metastasis in mouse models. This discovery sheds light on the previously unrecognized role of SMAD4 as a substrate of ERBB4 and highlights the selective involvement of the ERBB4-SMAD4 regulatory axis in tumor metastasis.

ERBB4 selectively amplifies TGF-β pro-metastatic responses.

Fatty acid metabolism plays a critical role in cancer development. In this study, a novel medium-chain fatty acid (MCFA) PET tracer, 2-[F]fluorohexanoic acid (F-FHA), was successfully synthesized with high radiochemical yield (65 ± 5.4%) and purity (>99%). Tumor radiotracer uptake at 60 min in A431, Hek293, Kpc, 4T1, Hep3b, and Lm3 xenograft models was 7.1 ± 0.4, 8.8 ± 0.6, 7.7 ± 0.5, 7.2 ± 0.4, 5.1 ± 0.3, and 4.9 ± 0.3 percentage injected dose per gram (% ID/g), respectively. Pretreatment with salicylic acid (SA), an inhibitor of medium-chain acyl-CoA synthetase (ACSM), significantly enhanced tumor uptake ( < 0.05) but showed no effect on blood uptake ( = 0.115). The biodistribution analysis demonstrated that F-FHA exhibited rapid hepatic and cardiac accumulation followed by gradual clearance, whereas tumor uptake exhibited gradual elevation within the initial 20 min and remained relatively stable through the 90 min observation period. F-FHA warranted further investigation as a potential PET imaging agent for tumors.
