&#x989E;&#x53F6;&#x766B;&#x75EB;&#x60A3;&#x8005;&#x7684;&#x8EAF;&#x4F53;&#x53D8;&#x5F62;&#x795E;&#x7ECF;&#x5143;&#x3002;

We studied morphologic characteristics of dysmorphic neurons in the hippocampus of seven patients with medically intractable TLE and compare histological, clinical, and imaging features with ten TLE patients with classical hippocampal sclerosis without abnormal cells. Such dysmorphic neurons were observed in the hilus of the dentate gyrus and were characterized by giant or misshapen cells with abnormal cytoskeletal structure and atypical dendritic processes that resembled the dysmorphic neurons from cortical dysplasias. Specimens with dysmorphic cells also contained other cytoarchitectural abnormalities including bilamination of the dentate granular cell layer (four out seven cases), and the presence of Cajal-Retzius cells in the dentate gyrus or Ammon's horn (five out seven cases). There were no statistically significant differences regarding the age at onset, duration of epilepsy, and hippocampal asymmetry ratio between patients with or without dysmorphic cells. Nevertheless, it is interesting to note that a higher proportion of patients with dysmorphic neurons continued to present auras after surgery, when compared with patients without those cells.

Nous avons &eacute;tudi&eacute; les caract&eacute;ristiques morphologiques de dysmorphic de neurones dans l&#39;hippocampe des sept patients avec des fonctionnalit&eacute;s m&eacute;dicalement insolubles TLE et comparez histologique, cliniques et d&#39;imagerie pour des patients de la TLE dix avec la scl&eacute;rose hippocampique classique sans la pr&eacute;sence de cellules anormales. Sp&eacute;cimens avec cellules dysmorphiques contenaient aussi d&#39;autres anomalies cytoarchitecturaux dont bilamellaire de la couche de cellules granuleuses gyrus (quatre sur sept cas) et la pr&eacute;sence de cellules de Cajal-Retzius dans le gyrus dent&eacute; ou corne d&#39;Ammon &eacute;prouve (cinq des sept cas). Il n&#39;y a aucune diff&eacute;rence statistiquement significative concernant l&#39;&acirc;ge &agrave; l&#39;apparition, la dur&eacute;e de l&#39;&eacute;pilepsie et le rapport de l&#39;hippocampe de l&#39;asym&eacute;trie entre les patients avec ou sans cellules dysmorphiques. N&eacute;anmoins, il est int&eacute;ressant de noter qu&#39;une proportion plus &eacute;lev&eacute;e de patients atteints de neurones dysmorphiques poursuit en pr&eacute;sentant des auras apr&egrave;s la chirurgie, en comparaison avec les patients sans ces cellules.

Wir studierte morphologische Merkmale der dysmorphe Neuronen im Hippocampus von sieben Patienten mit medizinisch therapieresistente TLE und vergleichen histologischen, klinischen und bildgebenden Funktionen mit zehn TLE-Patienten mit klassischen hippocampale Sklerose ohne abnormen Zellen. Solche dysmorphe Neuronen in den Hilus der Gyrus dentatus beobachtet wurden und zeichneten sich durch riesige oder deformierte Zellen mit abnormen Zytoskelett Struktur und atypische dendritische Prozesse, die die dysmorphe Neuronen aus kortikale Dysplasien &auml;hnelte. Exemplare mit dysmorphe Zellen enthielt auch andere Cytoarchitectural-Anomalien einschlie&szlig;lich Bilamination der Gyrus granulare Zelle Schicht (vier von sieben F&auml;llen) und das Vorhandensein von Cajal-Retzius-Zellen in den Gyrus dentatus oder Ammonsformation (f&uuml;nf von sieben F&auml;llen). Es gab keinerlei statistisch signifikante Unterschiede in Bezug auf das Alter bei Beginn, Dauer der Epilepsie und hippocampal Asymmetrie Verh&auml;ltnis zwischen Patienten mit oder ohne dysmorphe Zellen. Dennoch ist es interessant festzustellen, dass ein h&ouml;herer Anteil der Patienten mit dysmorphe Neuronen weiterhin Auren nach Operation im Vergleich zu Patienten ohne diese Zellen zu pr&auml;sentieren.

Abbiamo studiato caratteristiche morfologiche di dismorfico neuroni nell&#39;ippocampo di sette pazienti con caratteristiche clinici e imaging TLE e confronto istologico, medicamente intrattabile con i pazienti dieci TLE con Sclerosi ippocampale classica senza cellule anormali. Tali neuroni dismorfici sono stati osservati in hilus del giro dentato e sono stati caratterizzati da cellule gigante o deforme con anomala struttura citoscheletriche e processi dendritiche atipici che assomigliavano ai neuroni dismorfici da displasie corticali. Campioni con cellule dismorfico contenevano anche altre anomalie citoarchitettonica incluso bilamellare dello strato delle cellule granulari dentato (quattro sette casi) e la presenza di cellule di Cajal-Retzius il giro dentato o corno di Ammon (cinque sette casi). Non c&#39;erano differenze statisticamente significative per quanto riguarda l&#39;et&agrave; di insorgenza, la durata dell&#39;epilessia e rapporto di asimmetria hippocampal tra pazienti con o senza cellule dismorfico. Tuttavia, &egrave; interessante notare che una proporzione maggiore di pazienti con neuroni dismorfici ha continuato a presentare Aure dopo l&#39;intervento chirurgico, rispetto ai pazienti senza quelle cellule.

Estudiamos las caracter&iacute;sticas morfol&oacute;gicas de dysmorphic neuronas en el hipocampo de siete pacientes con m&eacute;dicamente insuperables TLE y comparar histol&oacute;gica, caracter&iacute;sticas cl&iacute;nicas y proyecci&oacute;n de imagen con diez TLE padecen esclerosis hipocampal cl&aacute;sica sin c&eacute;lulas anormales. Estas neuronas dysmorphic fueron observadas en el hilio del giro dentado y se caracterizaron por c&eacute;lulas gigantes o deformes con estructura anormal del citoesqueleto y procesos dendr&iacute;ticos anormales que se asemejaba a las neuronas dysmorphic de displasias corticales. Las muestras con c&eacute;lulas dysmorphic tambi&eacute;n conten&iacute;an otras anormalidades citoarquitect&oacute;nicos incluyendo bilaminar de la capa de c&eacute;lulas granulares dentado (cuatro a siete casos) y la presencia de c&eacute;lulas de Cajal-Retzius en el giro dentado o cuerno de Ammon (cinco a siete casos). No hubo diferencias estad&iacute;sticamente significativas con respecto a la edad en el inicio, la duraci&oacute;n de la epilepsia y relaci&oacute;n de asimetr&iacute;a hippocampal entre pacientes con o sin c&eacute;lulas dysmorphic. Sin embargo, es interesante notar que una mayor proporci&oacute;n de pacientes con neuronas dysmorphic contin&uacute;a presentes auras despu&eacute;s de la cirug&iacute;a, en comparaci&oacute;n con pacientes sin esas c&eacute;lulas.

Protein aggregation and amyloid accumulation in different tissues are associated with cellular dysfunction and toxicity in important human pathologies, including Alzheimer's disease and various forms of systemic amyloidosis. Soluble oligomers formed at the early stages of protein aggregation have been increasingly recognized as the main toxic species in amyloid diseases. To gain insight into the mechanisms of toxicity instigated by soluble protein oligomers, we have investigated the aggregation of hen egg white lysozyme (HEWL), a normally harmless protein. HEWL initially aggregates into beta-sheet rich, roughly spherical oligomers which appear to convert with time into protofibrils and mature amyloid fibrils. HEWL oligomers are potently neurotoxic to rat cortical neurons in culture, while mature amyloid fibrils are little or non-toxic. Interestingly, when added to cortical neuronal cultures HEWL oligomers induce tau hyperphosphorylation at epitopes that are characteristically phosphorylated in neurons exposed to soluble oligomers of the amyloid-beta peptide. Furthermore, injection of HEWL oligomers in the cerebral cortices of adult rats induces extensive neurodegeneration in different brain areas. These results show that soluble oligomers from a non-disease related protein can mimic specific neuronal pathologies thought to be induced by soluble amyloid-beta peptide oligomers in Alzheimer's disease and support the notion that amyloid oligomers from different proteins may share common structural determinants that would explain their generic cytotoxicities.

Agr&eacute;gation de la prot&eacute;ine et l&#39;accumulation d&#39;amylo&iuml;de dans diff&eacute;rents tissus sont associ&eacute;es &agrave; la toxicit&eacute; dans des pathologies humaines importantes, y compris la maladie d&#39;Alzheimer et diverses formes d&#39;amylose syst&eacute;mique et dysfonctionnement cellulaire. LYSOZYME agr&eacute;gats initialement en oligom&egrave;res &agrave; peu pr&egrave;s sph&eacute;riques, riches de b&ecirc;ta-feuille qui apparaissent pour convertir les protofibrilles et fibrilles amylo&iuml;des matures avec le temps. LYSOZYME oligom&egrave;res sont puissamment neurotoxiques de neurones corticaux de rat en culture, tandis que les fibrilles amylo&iuml;des matures sont peu ou non toxiques. Fait int&eacute;ressant, lorsque ajout&eacute; &agrave; corticale cultures de neurones des oligom&egrave;res de lysozyme induisent tau hyperphosphoryl&eacute;e ep&iacute;topes differents qui sont caract&eacute;ristiquement phosphoryl&eacute;e dans les neurones expos&eacute;s &agrave; des oligom&egrave;res solubles du peptide b&ecirc;ta-amylo&iuml;de. Aggregazione proteica e l&#39;accumulo di amiloide nei tessuti diversi sono associati a disfunzione cellulare e tossicit&agrave; in importanti patologie umane, incluso il morbo di Alzheimer e varie forme di amiloidosi sistemica. Oligomeri solubili formati nelle fasi iniziali di aggregazione proteica sono stati sempre pi&ugrave; riconosciuti come la principale specie tossiche nelle malattie amiloide. Per guadagnare la comprensione dei meccanismi di tossicit&agrave; istigato da oligomeri di proteina solubile, abbiamo studiato l&#39;aggregazione di gallina albume lisozima (HEWL), una proteina normalmente innocuo. Questi risultati mostrano che oligomeri solubili da una non-malattia correlata della proteina pu&ograve; mimare specifiche patologie neuronali, pensati per essere indotta da oligomeri solubile del peptide beta-amiloide nella malattia di Alzheimer e supporta la nozione che oligomeri amiloide da proteine differenti possono condividere comuni determinanti strutturali che spiegherebbero la loro cytotoxicities generico.

Cytoarchitectural modifications of the dentate gyrus are among the most obvious abnormalities observed in the hippocampal sclerosis associated with refractory epilepsy. Here, we examined the morphological changes of granular cells (dispersion, bilamination and cell loss) in sclerotic hippocampi from nine TLE patients, comparing abnormal and preserved areas. A total of 2,577 granular cells were analyzed with respect to four different histopathological patterns: areas with bilamination (n = 936), areas with dispersion (n = 905), areas with neuronal loss (n = 279), and preserved areas (n = 457). Quantitative parameters included somatic perimeter (P), area (A) and form factor (ff). Although different patterns were often observed in the same patient, highly significant differences were observed (p < 0.0001) when patterns were compared to one another. Since granular cell dispersion and bilamination have different morphological aspects in sclerotic hippocampi from TLE patients, we suggest that these patterns should be considered separately. Future studies are needed to determine the frequency with which these patterns occur in the general population and whether each one can interfere with seizure susceptibility.

The isolectin Vicia villosa B(4) (VV) selectively recognizes N-acetyl-galactosamine-terminal glycoconjugates that form perineuronal nets (PNNs) around a subset of neurons in the cerebral cortex. PNNs are thought to participate in the guidance of incoming thalamic axons and in the posterior stabilization and maintenance of synaptic contacts. Here we examine the spatial and temporal distribution of biotinylated VV in tangential sections through layer IV of the posteromedial barrel subfield in the primary somatosensory cortex (PMBSF) of rats ranging from postnatal day (P)3 to P60, which underwent unilateral deafferentation of whiskers at birth. In the afferented hemisphere, labeling first appears at P5, with a diffuse distribution, probably associated with neuropil, inside PMBSF barrels. VV distribution remains diffuse during the following week, and declines around P17. From P24 onward, however, proteoglycans form PNNs around cell bodies preferentially localized in septal regions of the PMBSF. In the contralateral, deafferented PMBSF the diffuse labeling also appears on P5, but first develops into elongated, homogeneous stripes, which disappear after P24, leaving only scattered cell bodies along layer IV. Our results indicate that proteoglycans appear simultaneous to barrel formation in the developing rat while segregation of PNNs to septal cells might be driven by afferent activity.

D-serine is a co-agonist of NMDA receptor (NMDAR) and plays important roles in synaptic plasticity mechanisms. Serine racemase (SR) is a brain-enriched enzyme that converts L-serine to D-serine. SR interacts with the protein interacting with C-kinase 1 (PICK1), which is known to direct protein kinase C (PKC) to its targets in cells. Here, we investigated whether PKC activity regulates SR activity and D-serine availability in the brain. In vitro, PKC phosphorylated SR and decreased its activity. PKC activation increased SR phosphorylation in serine residues and reduced D-serine levels in astrocyte and neuronal cultures. Conversely, PKC inhibition decreased basal SR phosphorylation and increased cellular D-serine levels. In vivo modulation of PKC activity regulated both SR phosphorylation and D-serine levels in rat frontal cortex. Finally, rats that completed an object recognition task showed decreased SR phosphorylation and increased D-serine/total serine ratios, which was markedly correlated with decreased PKC activity in both cortex and hippocampus. Results indicate that PKC phosphorylates SR in serine residues and regulates D-serine availability in the brain. Protein kinase C activity regulates D-serine availability in the brain.

D-s&eacute;rine est un co-agoniste du r&eacute;cepteur NMDA (NMDAR) et joue un r&ocirc;le important dans les m&eacute;canismes de la plasticit&eacute; synaptique. S&eacute;rine rac&eacute;mase (SR) est une enzyme qui convertit la L-s&eacute;rine &agrave; la D-s&eacute;rine enrichi de cerveau. SR interagit avec la prot&eacute;ine interagissant avec C-kinase 1 (PICK1), qui est appel&eacute; &agrave; diriger la prot&eacute;ine kinase C (PKC) &agrave; ses cibles dans les cellules. Serina Racemasa (SR) es una enzima cerebral enriquecida que convierte la L-serina a la D-serina. SR interact&uacute;a con la prote&iacute;na interactuando con C-quinasa 1 (PICK1), que es conocida por dirigir la prote&iacute;na quinasa C (PKC) para sus objetivos en las c&eacute;lulas. Aqu&iacute;, hemos investigado si la actividad de la PKC regula actividad de SR y la disponibilidad de la D-serina en el cerebro. In vitro, PKC fosforila SR y hab&iacute;a disminuido su actividad. Los resultados indican que la PKC fosforila SR en residuos de serina y regula la disponibilidad de la D-serina en el cerebro.
