Adipocyte differentiation is a key process implicated in the pathogenesis of obesity and insulin resistance. Its regulation is triggered by a cascade of transcription factors, including the CCAAT/enhancer binding proteins (C/EBPs) and peroxisome proliferator-activated receptor-gamma (PPARgamma). Growth factors such as transforming growth factor-beta1 (TGF-beta1) are known to inhibit adipocyte differentiation in vitro, via the C/EBP pathway, and in vivo, but whether a downstream mediator of TGF-beta1, connective tissue growth factor (CTGF), also known as CCN2, has a similar role is unknown. Mouse 3T3-L1 cells were differentiated into adipocytes by using standard methods, and effects and regulation of CTGF were studied. Intervention with recombinant human CTGF during differing stages of differentiation caused an inhibition in the development of the adipocyte phenotype, according to the gene expression of the differentiation markers adiponectin and PPARgamma, as well as suppression of lipid accumulation and expression of the lipogenic enzyme glycerol-3-phosphate dehydrogenase. Whereas CTGF gene expression promptly fell by 90% as 3T3-L1 preadipocytes differentiated into mature adipocytes, CTGF mRNA expression was induced by added TGF-beta1. CTGF applied to cells early in the course of differentiation inhibited total cell protein levels and nuclear localization of the beta-isoform of C/EBP (C/EBP-beta) and, subsequently, total cell C/EBP-alpha levels. CTGF also inhibited the adipocyte differentiation program in primary cultures of mouse preadipocytes. Expression of CTGF mRNA was twofold higher in the central fat depots of mice compared with subcutaneous fat, suggesting a potential role for CTGF in vivo. In summary, these data show that CTGF inhibits the adipocyte differentiation program.

Connective tissue growth factor inhibits adipocyte differentiation.

CTGF inibisce anche il programma di differenziazione degli adipociti in colture primarie di preadipociti del mouse. Espressione di mRNA CTGF era duplice superiore nei depositi di grassi centrali dei topi rispetto al grasso sottocutaneo, suggerendo un ruolo potenziale per CTGF in vivo. La diferenciaci&oacute;n del adipocito es un proceso clave implicado en la patogenia de la obesidad y resistencia a la insulina. Su regulaci&oacute;n es accionada por una cascada de factores de transcripci&oacute;n, como las prote&iacute;nas de uni&oacute;n a CCAAT/enhancer (C/EBPs) y peroxisome proliferator-activado del receptor-gamma (PPARgamma). Factores de crecimiento como transforming growth factor-beta1 (TGF-beta1) se sabe que inhiben la diferenciaci&oacute;n del adipocito in vitro, v&iacute;a el camino de la C/EBP e in vivo, pero si un mediador aguas abajo de TGF-beta1, factor de crecimiento de tejido conectivo (CTGF), tambi&eacute;n conocido como CCN2, tiene un papel similar es desconocido. Las c&eacute;lulas 3T3-L1 de rat&oacute;n fueron distinguidas en los adipocitos mediante el uso de m&eacute;todos est&aacute;ndar, y se estudiaron los efectos y la regulaci&oacute;n del CTGF. Intervenci&oacute;n con recombinante humana CTGF durante diferentes etapas de la diferenciaci&oacute;n caus&oacute; una inhibici&oacute;n en el desarrollo del fenotipo adipocito, seg&uacute;n la expresi&oacute;n del gen de la adiponectina de marcadores de diferenciaci&oacute;n y PPARgamma, as&iacute; como la supresi&oacute;n de la acumulaci&oacute;n de l&iacute;pidos y la expresi&oacute;n de la lipog&eacute;nicos enzima glicerol-3-fosfato deshidrogenasa. Mientras que la expresi&oacute;n del gen CTGF r&aacute;pidamente cay&oacute; en un 90% como 3T3-L1 preadipocytes distinguido en adipocitos maduros, la expresi&oacute;n del mRNA CTGF fue inducida por TGF-beta1 a&ntilde;adido. MPO (myeloperoxidase) catalyses the oxidation of chloride, bromide and thiocyanate by hydrogen peroxide to HOCl (hypochlorous acid), HOBr (hypobromous acid) and HOSCN (hypothiocyanous acid) respectively. Specificity constants indicate that SCN- is a major substrate for MPO. HOSCN is also a major oxidant generated by other peroxidases including salivary, gastric and eosinophil peroxidases. While HOCl and HOBr are powerful oxidizing agents, HOSCN is a less reactive, but more specific, oxidant which targets thiols and especially low pKa species. In the present study we show that HOSCN targets cysteine residues present in PTPs (protein tyrosine phosphatases) with this resulting in a loss of PTP activity for the isolated enzyme, in cell lysates and intact J774A.1 macrophage-like cells. Inhibition also occurs with MPO-generated HOCl and HOBr, but is more marked with MPO-generated HOSCN, particularly at longer incubation times. This inhibition is reversed by dithiothreitol, particularly at early time points, consistent with the reversible oxidation of the active site cysteine residue to give either a cysteine-SCN adduct or a sulfenic acid. Inhibition of PTP activity is associated with increased phosphorylation of p38a and ERK2 (extracellular-signal-regulated kinase 2) as detected by Western blot analysis and phosphoprotein arrays, and results in altered MAPK (mitogen-activated protein kinase) signalling. These data indicate that the highly selective targeting of some protein thiols by HOSCN can result in perturbation of cellular phosphorylation and altered cell signalling. These changes occur with (patho)physiological concentrations of SCN- ions, and implicate HOSCN as an important mediator of inflammation-induced oxidative damage, particularly in smokers who have elevated plasma levels of SCN-.

The myeloperoxidase-derived oxidant HOSCN inhibits protein tyrosine phosphatases and modulates cell signalling via the mitogen-activated protein kinase (MAPK) pathway in macrophages.

Inhibition de l&#39;activit&eacute; PTP est associ&eacute;e &agrave; la phosphorylation accrue de p38a et ERK2 (extracellulaire-signal-regulated kinase 2) tel que d&eacute;tect&eacute; par Western blot analyse et phosphoprot&eacute;ine tableaux et r&eacute;sultats en alt&eacute;r&eacute;e MAPK (mitogen-activated protein kinase) signalisation. Ces donn&eacute;es indiquent que le ciblage hautement s&eacute;lective de certains thiols prot&eacute;iques par HOSCN peut entra&icirc;ner dans la perturbation de la phosphorylation cellulaire et signalisation cellulaire alt&eacute;r&eacute;e. L&#39;oxydant d&eacute;riv&eacute; de my&eacute;loperoxydase HOSCN inhibe la prot&eacute;ine tyrosine phosphatases et module de cellule signalisation par la voie de mitogen-activated protein kinases (MAPK) dans les macrophages.

Catalizza MPO (mieloperossidasi) l&#39;ossidazione del cloruro, bromuro e tiocianato di idrogeno perossido a HOCl (acido ipocloroso), HBrO (acido ipobromoso) e HOSCN (acido hypothiocyanous) rispettivamente. Costanti di specificit&agrave; indicano che SCN - &egrave; un importante substrato per MPO. During atherosclerosis monocyte-derived macrophages accumulate cholesteryl esters from low-density lipoproteins (LDLs) via lectin-like oxidised LDL receptor-1 (LOX-1) and class AI and AII (SR-AI, SR-AII) and class B (SR-BI, CD36) scavenger receptors. Here we examined the hypothesis that hyperglycaemia may modulate receptor expression and hence lipid accumulation in macrophages. Human monocytes were matured into macrophages in 30 versus 5 mM glucose and receptor expression and lipid accumulation quantified. High glucose elevated LOX1 mRNA, but decreased SR-AI, SR-BI, LDLR, and CD36 mRNA. SR-BI and CD36 protein levels were decreased. Normo- and hyperglycaemic cells accumulated cholesteryl esters from modified LDL to a greater extent than control LDL, but total and individual cholesteryl ester accumulation was not affected by glucose levels. It is concluded that, whilst macrophage scavenger receptor mRNA and protein levels can be modulated by high glucose, these are not key factors in lipid accumulation by human macrophages under the conditions examined.

Effect of exposure of human monocyte-derived macrophages to high, versus normal, glucose on subsequent lipid accumulation from glycated and acetylated low-density lipoproteins.

Au cours de l&#39;ath&eacute;roscl&eacute;rose, macrophages d&eacute;riv&eacute;s de monocytes s&#39;accumulent des esters de cholest&eacute;rol des lipoprot&eacute;ines de basse densit&eacute; (LDL) via les LDL oxyd&eacute;es de type lectine r&eacute;cepteur-1 (LOX-1) et classe AI et AII (SR-AI, AII-SR), r&eacute;cepteurs de classe B (SR-BI, CD36) au Tr&eacute;sor. Ici, nous avons examin&eacute; l&#39;hypoth&egrave;se que l&#39;hyperglyc&eacute;mie peut moduler l&#39;expression des r&eacute;cepteurs et donc accumulation de lipides dans les macrophages. Qui abbiamo esaminato l&#39;ipotesi che iperglicemia pu&ograve; modulano l&#39;espressione dei recettori e quindi accumulo di lipidi nei macrofagi. Connective tissue growth factor (CTGF), also known as CCN-2, is a cysteine-rich secreted protein that is involved in a range of biological processes including regulation of cell growth and differentiation. Our previous in vitro studies have shown that CCN-2 inhibits adipocyte differentiation, although whether CCN-2 is regulated in vivo in adipogenesis is undetermined and was investigated in this study. C57BL/6 male mice were fed either standard laboratory chow (ND) or diet high in fat (HFD, 45% fat) for 15 or 24 weeks. HFD animals which gained >5 g in weight (termed HFD-Fat) were insulin resistant and were compared to HFD fed animals which failed to gain weight (termed HFD-Lean). HFD-Fat mice had significantly increased CCN-2 mRNA levels in both the subcutaneous and epididymal fat pads, whereas CCN-2 mRNA was not induced in the epididymal site in HFD-Lean mice. Also in HFD-fed animals, epididymal CCN-2 mRNA correlated positively with key genes involved in adipocyte differentiation, adiponectin and PPARÎ³ (p<0.001 and p<0.002, respectively). Additionally, epididymal CCN-2 mRNA correlated positively with two markers of tissue turnover, PAI-1 in HFD-Fat mice only, and TIMP-1 but only in the HFD-Lean mice. Collectively, these findings suggest that CCN-2 plays a role in adipocyte differentiation in vivo and thus in the pathogenesis of obesity linked with insulin resistance.

Connective tissue growth factor/CCN-2 is upregulated in epididymal and subcutaneous fat depots in a dietary-induced obesity model.

Tissu conjonctif facteur de croissance/CCN-2 est augment&eacute;e dans les d&eacute;p&ocirc;ts de graisses &eacute;pididymaires et sous-cutan&eacute;e dans un mod&egrave;le de l&#39;ob&eacute;sit&eacute; induite par l&#39;alimentation.

Collettivamente, questi risultati suggeriscono che CCN-2 Gioca un ruolo nella differenziazione degli adipociti in vivo e quindi nella patogenesi dell&#39;obesit&agrave; legata con insulino-resistenza.
