The mushroom bodies (MBs), a paired structure in the insect brain, play a major role in storing and retrieving olfactory memories. We tested whether olfactory learning and odor processing is impaired in honeybees in which MB subunits were partially ablated. Using hydroxyurea (HU) to selectively kill proliferating cells, we created honeybees with varying degrees of MB lesions. Three-dimensional reconstructions of brains were generated to analyze the drug-induced morphological changes. These reconstructions show that, with few exceptions, only the MBs were affected by the drug, while other brain areas remained morphometrically intact. Typically, lesions affected only the MB in one hemisphere of the brain. To preclude HU-induced physiologic deficits in the antennal lobe (AL) affecting olfactory learning, we measured the responses to odors in the AL using an in vivo calcium imaging approach. The response patterns did not differ between the AL of intact versus ablated brain sides within respective specimens. We, therefore, carried out side-specific classical discriminative olfactory conditioning of the proboscis extension reflex (PER) with control bees and with HU-treated bees with or without MB ablations. All experimental groups learned equally to discriminate and respond to a rewarded (CS+) but not to an unrewarded (CS-) conditioned stimulus during acquisition and retention tests. Thus, our results indicate that partial MB lesions do not affect this form of elemental olfactory learning.

Les champignons corps (MBs), une structure de paire dans le cerveau des insecte, jouent un r&ocirc;le important dans le stockage et la r&eacute;cup&eacute;ration des souvenirs olfactifs. Nous avons v&eacute;rifi&eacute; si olfactive d&#39;apprentissage et de traitement des odeurs est alt&eacute;r&eacute;e chez les abeilles dans laquelle MB sous-unit&eacute;s ont &eacute;t&eacute; partiellement ablation. &Agrave; l&#39;aide de l&#39;hydroxyur&eacute;e (HU) de tuer s&eacute;lectivement des cellules en prolif&eacute;ration, nous avons cr&eacute;&eacute; des abeilles avec divers degr&eacute;s de l&eacute;sions MB. Reconstructions tridimensionnelles du cerveau ont &eacute;t&eacute; g&eacute;n&eacute;r&eacute;es pour analyser les changements morphologiques induites par les m&eacute;dicaments. Ces reconstitutions montrent que, &agrave; quelques exceptions pr&egrave;s, seuls les MBs &eacute;taient touch&eacute;es par la drogue, tandis que les autres r&eacute;gions du cerveau sont rest&eacute;s intacte de la morphom&eacute;trie. En r&egrave;gle g&eacute;n&eacute;rale, les l&eacute;sions ne touch&eacute;s que la Mo dans un h&eacute;misph&egrave;re du cerveau. Afin d&#39;emp&ecirc;cher les d&eacute;ficits physiologiques induits par HU dans le lobe antennaire (AL) qui affectent l&#39;apprentissage olfactif, nous avons mesur&eacute; les r&eacute;ponses aux odeurs El &agrave; l&#39;aide d&#39;un calcium in vivo imaging approche. Les profils de r&eacute;ponse ne diff&eacute;raient pas de l&#39;AL d&#39;intacte par rapport aux c&ocirc;t&eacute;s du cerveau ablation au sein des &eacute;chantillons respectifs. Nous avons, par cons&eacute;quent, effectu&eacute; ax&eacute;s sur le c&ocirc;t&eacute; classique discriminative olfactif conditionner le r&eacute;flexe d&#39;extension du proboscis (PER) avec abeilles contr&ocirc;le et abeilles HU-trait&eacute;s avec ou sans ablations MB. Les groupes exp&eacute;rimentaux appris aussi &agrave; une discrimination et de r&eacute;pondre &agrave; une r&eacute;compense (CS +) mais pas &agrave; un stimulus de conditionn&eacute; (CS-) r&eacute;compens&eacute; lors d&#39;acquisition et de r&eacute;tention des tests. Ainsi, nos r&eacute;sultats indiquent que les l&eacute;sions partielles MB n&#39;affectent pas cette forme d&#39;apprentissage olfactif &eacute;l&eacute;mentaire.

I corpi dei funghi (MBs), una struttura appaiato nel cervello degli insetti, svolgono un ruolo importante nella conservazione e recupero delle memorie olfattive. Abbiamo testato se olfattiva odore elaborazione e apprendimento &egrave; alterata in honeybees in cui MB subunit&agrave; sono stati parzialmente ablazione. Utilizzando l&#39;idrossiurea (HU) di uccidere selettivamente le cellule in proliferazione, abbiamo creato le API con vari gradi di lesioni MB. Ricostruzioni tridimensionali del cervello sono stati generati per analizzare i cambiamenti morfologici farmaco-indotta. Queste ricostruzioni mostrano che, con poche eccezioni, solo i MBs sono stati colpiti dalla droga, mentre altre zone del cervello &egrave; rimasto intatto morfometrico. In genere, le lesioni colpite solo il MB in un emisfero del cervello. Per escludere deficit fisiologici HU-indotta nel lobo antenne (AL) che interessano apprendimento olfattivo, abbiamo misurato le risposte agli odori in AL utilizzando un calcio in vivo imaging approccio. I modelli di risposta non differiscono tra la di intatto contro i lati del cervello ablato all&#39;interno dei rispettivi esemplari. Abbiamo, quindi, effettuata lato specifiche classica discriminatorio olfattiva condizionata proboscide estensione reflex (PER) con le API di controllo e con le API di HU-trattati con o senza ablazioni MB. Tutti i gruppi sperimentali imparato ugualmente a discriminare e rispondere a un premiato (CS +) ma non a un unrewarded stimolo condizionato (CS-) durante l&#39;acquisizione e il test di ritenzione. Cos&igrave;, i nostri risultati indicano che le lesioni parziali MB non influenzano questa forma di apprendimento olfattivo Elementale.

Ablazione indotta da idrossiurea corpo parziale dei funghi non influisce acquisizione e ritenzione di condizionamento olfattivo differenziale nelle API.

Complex external stimuli such as odorants are believed to be internally represented in the brain by spatiotemporal activity patterns of extensive neuronal ensembles. These activity patterns can be recorded by optical imaging techniques. However, optical imaging with conventional fluorescence dyes usually does not allow for resolving the activity of biologically defined groups of neurons. Therefore, specifically targeting reporter molecules to neuron populations of common genetic identity is an important goal. We report the use of the genetically encoded calcium-sensitive fluorescence protein cameleon 2.1 in the Drosophila brain. We visualized odorant-evoked intracellular calcium concentration changes in selectively labeled olfactory projection neurons both postsynaptically in the antennal lobe, the primary olfactory neuropil, and presynaptically in the mushroom body calyx, a structure involved in olfactory learning and memory. As a technical achievement, we show that calcium imaging with a genetically encoded fluorescence probe is feasible in a brain in vivo. This will allow one to combine Drosophila's advanced genetic tools with the physiological analysis of brain function. Moreover, we report for the first time optical imaging recordings in synaptic regions of the Drosophila mushroom body calyx and antennal lobe. This provides an important step for the use of Drosophila as a model system in olfaction.

Complejos est&iacute;mulos externos tales como olores se creen que representarse internamente en el cerebro por los patrones de actividad espacio-temporal de conjuntos neuronales extensas. Estos patrones de actividad pueden grabarse por t&eacute;cnicas de imagen &oacute;pticas. Sin embargo, im&aacute;genes &oacute;pticas con tintes de fluorescencia convencional generalmente no permiten resolver la actividad de grupos biol&oacute;gicamente definidos de neuronas. Por lo tanto, dirigidos espec&iacute;ficamente contra las mol&eacute;culas de reportero a poblaciones de neuronas de identidad gen&eacute;tica com&uacute;n es un objetivo importante. Divulgamos el uso de la fluorescencia gen&eacute;ticamente codificados de sensibles al calcio prote&iacute;na cameleon 2.1 en el cerebro de la Drosophila. Visualizamos cambios de concentraci&oacute;n de calcio intracelular odorante-evocado en las neuronas de proyecci&oacute;n olfativa selectivamente etiquetados postsynaptically l&oacute;bulo antenal, el neuropil olfativo primario tanto en presynaptically en el c&aacute;liz del cuerpo de la seta, una estructura en la memoria y el aprendizaje olfativo. Como un logro t&eacute;cnico, demostramos que la proyecci&oacute;n de imagen de calcio con una sonda de fluorescencia gen&eacute;ticamente codificados es factible en un cerebro en vivo. Esto permitir&aacute; a avanzadas herramientas gen&eacute;ticas de Drosophila se combinan con el an&aacute;lisis fisiol&oacute;gico de la funci&oacute;n cerebral. Por otra parte, nos informan de las primeras grabaciones imagen &oacute;ptica de tiempo en regiones sin&aacute;pticas del c&aacute;liz del cuerpo de la seta de Drosophila y l&oacute;bulo antenal. Esto proporciona un paso importante para el uso de Drosophila como modelo en el olfato.

Gen&eacute;tico expresaron cameleon en Drosophila melanogaster se utiliza para visualizar la informaci&oacute;n olfativa en las neuronas de proyecci&oacute;n.
