We describe a scheme for tagging an alanine-based peptide with a Cu(II) and a nitroxide to measure unfolding transitions. The enhancement in longitudinal relaxation rate of the nitroxide due to the presence of Cu(II) was measured at physiological temperatures by pulsed electron spin resonance (ESR). The change in relaxation rate provided the average interspin distance between the Cu(II) and the nitroxide. Control experiments on a proline-based peptide verify the robustness of the method. The change in interspin distances with temperature for the alanine-based peptide is in accord with the change in helicity measured by circular dichroism. The data provide an opportunity to examine the unfolding process in polyalanine peptides. The distance in the folded state is in concordance with molecular dynamics. However, the ESR experiment measures an average distance of 17 A in the unfolded state, whereas molecular dynamics indicates a distance of 42 A if the unfolded geometry was a polyproline type II helix. Therefore, ESR demonstrates that the unfolded state of this alanine-based peptide is not an ideal extended polyproline type II helix.

The overall morphology and Cu(II) ion coordination for the aggregated amyloid-beta(1-40) [Abeta(1-40)] in N-ethylmorpholine (NEM) buffer are affected by Cu(II) ion concentration. This effect is investigated by transmission electron microscopy (TEM), atomic force microscopy (AFM), and electron spin echo envelope modulation (ESEEM) spectroscopy. At lower than equimolar concentrations of Cu(II) ions, fibrillar aggregates of Abeta(1-40) are observed. At these concentrations of Cu(II), the monomeric and fibrillar Abeta(1-40) ESEEM data indicate that the Cu(II) ion is coordinated by histidine residues. For aggregated Abeta(1-40) at a Cu(II):Abeta molar ratio of 2:1, TEM and AFM images show both linear fibrils and granular amorphous aggregates. The ESEEM spectra show that the multi-histidine coordination for Cu(II) ion partially breaks up and becomes exposed to water or exchangeable protons of the peptide at a higher Cu(II) concentration. Since the continuous-wave electron spin resonance results also suggest two copper-binding sites in Abeta(1-40), the proton ESEEM peak may arise from the second copper-binding site, which may be significantly involved in the formation of granular amorphous aggregates. Thioflavin T fluorescence and circular dichroism experiments also show that Cu(II) inhibits the formation of fibrils and induces a nonfibrillar beta-sheet conformation. Therefore, we propose that Abeta(1-40) has a second copper-binding site in a proton-rich environment and the second binding Cu(II) ion interferes with a conformational transition into amyloid fibrils, inducing the formation of granular amorphous aggregates.

Tuttavia, dettagli strutturali dell&#39;infezione HIV-1 non sono state osservate, a causa di sfide tecnologiche nella lavorazione con rare e dinamici particelle di HIV-1 nelle cellule umane. Riportiamo l&#39;analisi strutturale delle interazioni dell&#39;HIV-1 e cellula ospite mediante un correlativo ad alta velocit&agrave; 3D live-cella-imaging e cryoET Metodo. Utilizzando questo metodo, vi abbiamo mostrato in condizioni quasi nativa che hyperstable intatta mutante nuclei di HIV-1 sono rilasciati nel citoplasma delle cellule ospiti. Abbiamo ottenuto ulteriori prove dirette a suggerire che un hyperstable mutante capside, E45A, ha mostrato lo smontaggio del capside ritardato rispetto al capside del selvaggio-tipo. Insieme, questi risultati dimostrano i vantaggi del nostro vivere-cella correlativo e cryoET approccio per processi dinamici, quali infezioni virali di imaging.

Cryo-electron tomography (cryoET) allows 3D visualization of cellular structures at molecular resolution in a close-to-native state and therefore has the potential to help elucidate early events of HIV-1 infection in host cells. However, structural details of infecting HIV-1 have not been observed, due to technological challenges in working with rare and dynamic HIV-1 particles in human cells. Here, we report structural analysis of HIV-1 and host-cell interactions by means of a correlative high-speed 3D live-cell-imaging and cryoET method. Using this method, we showed under near-native conditions that intact hyperstable mutant HIV-1 cores are released into the cytoplasm of host cells. We further obtained direct evidence to suggest that a hyperstable mutant capsid, E45A, showed delayed capsid disassembly compared to the wild-type capsid. Together, these results demonstrate the advantages of our correlative live-cell and cryoET approach for imaging dynamic processes, such as viral infection.

Cryo-tomographie &#xE9;lectronique (cryoET) permet la visualisation 3D des structures cellulaires &#xE0; la r&#xE9;solution mol&#xE9;culaire dans un &#xE9;tat proche de l&#x26;#39;origine et a donc le potentiel pour aider &#xE0; &#xE9;lucider les &#xE9;v&#xE9;nements pr&#xE9;coces de l&#x26;#39;infection VIH-1 dans les cellules h&#xF4;tes. Cependant, les d&#xE9;tails structurels de l&#x26;#39;infection VIH-1 n&#x26;#39;ont pas &#xE9;t&#xE9; observ&#xE9;es, en raison de probl&#xE8;mes technologiques dans le travail avec de rares et dynamique du VIH-1 particules dans les cellules humaines. Ici, nous rapportons l&#x26;#39;analyse structurale du VIH-1 et la cellule h&#xF4;te interactions au moyen d&#x26;#39;un corr&#xE9;lative &#xE0; haute vitesse 3D des cellules vivantes d&#x26;#39;imagerie et de la m&#xE9;thode cryoET. En utilisant cette m&#xE9;thode, nous avons montr&#xE9; dans des conditions proches des conditions qui indig&#xE8;nes intactes hyperstable mutantes du VIH-1 sont des noyaux lib&#xE9;r&#xE9; dans le cytoplasme des cellules h&#xF4;tes. Nous avons en outre obtenu des preuves directes permettant de penser qu&#x26;#39;une capside hyperstable mutant, E45A, a montr&#xE9; le d&#xE9;montage retard&#xE9; capside par rapport &#xE0; la capside de type sauvage. Cryo-Elektron Tomographie (CryoET) erm&ouml;glicht die 3D Visualisierung von Zellstrukturen Molekulare Aufl&ouml;sung in einem Ende-zu-nativ-Zustand und hat somit das Potenzial, zur fr&uuml;hen Ereignisse von HIV-1 Infektion in Wirtszellen zu erhellen. Jedoch wurden strukturelle Details f&uuml;r die HIV-1 Infektion nicht, aufgrund der technologischen Herausforderungen im Umgang mit seltenen und dynamische HIV-1-Partikel in den menschlichen Zellen beobachtet. Hier berichten wir Strukturanalyse von HIV-1 und Host-Zell-Interaktionen durch eine Korrelative Hochgeschwindigkeits-3D live-Cell-Imaging und CryoET Methode. Mit dieser Methode, unter fast Native Bedingungen zeigten wir, dass intakte Hyperstable werden mutant HIV-1-Kerne in das Zytoplasma der Wirtszellen abgegeben. Wir erhielten weitere direkte Hinweise darauf, dass ein Hyperstable mutant Kapsid, E45A, zeigte im Vergleich zu Wildtyp-Kapsid verz&ouml;gerte Kapsid-Demontage. Diese Ergebnisse werden gemeinsam die Vorteile unserer Korrelative Leben-Zelle und CryoET Ansatz f&uuml;r imaging-dynamische Prozesse, wie virale Infektion.

Cryo-tomograf&#xED;a electr&#xF3;nica (cryoET) permite la visualizaci&#xF3;n en 3D de las estructuras celulares con resoluci&#xF3;n molecular en un estado cercano al nativo y por lo tanto, tiene el potencial para ayudar a esclarecer los acontecimientos iniciales de la infecci&#xF3;n VIH-1 en las c&#xE9;lulas hu&#xE9;sped. Sin embargo, los detalles estructurales de infectar el VIH-1 no se han observado, debido a problemas tecnol&#xF3;gicos en el trabajo con raras y din&#xE1;mica del VIH-1 part&#xED;culas en las c&#xE9;lulas humanas. Aqu&#xED;, se presenta el an&#xE1;lisis estructural del VIH-1 y las interacciones c&#xE9;lula hu&#xE9;sped por medio de un correlativo de alta velocidad 3D en vivo de c&#xE9;lulas de im&#xE1;genes y m&#xE9;todo cryoET. El uso de este m&#xE9;todo, se mostr&#xF3; en condiciones pr&#xF3;ximas a las condiciones que nativos intactos hiperestable mutantes del VIH-1 n&#xFA;cleos son liberados en el citoplasma de las c&#xE9;lulas hu&#xE9;sped. Hemos obtenido m&#xE1;s evidencia directa que sugiera que un hiperestable c&#xE1;pside mutante, E45A, mostr&#xF3; retrasa el desmontaje c&#xE1;pside en comparaci&#xF3;n con la c&#xE1;pside de tipo salvaje. En conjunto, estos resultados ponen de manifiesto las ventajas de nuestro enfoque correlato en vivo de las c&#xE9;lulas y cryoET de im&#xE1;genes de procesos din&#xE1;micos, como la infecci&#xF3;n viral.
