The mechanisms governing the kinetics of climbing fibre-mediated Ca2+ transients in spiny dendrites of cerebellar Purkinje cells (PCs) were quantified with high-resolution confocal Ca2+ imaging. Ca2+ dynamics in parvalbumin (PV-/-) and parvalbumin/calbindin D28k null-mutant (PV/CB-/-) mice were compared with responses in wild-type (WT) animals. In the WT, Ca2+ transients in dendritic shafts were characterised by double exponential decay kinetics that were not due to buffered Ca2+ diffusion or saturation of the indicator dye. Ca2+ transients in PV-/- PCs reached the same peak amplitude as in the WT but the biphasic nature of the decay was less pronounced, an effect that could be attributed to PV's slow binding kinetics. In contrast, peak amplitudes in PV/CB-/- PCs were about two times higher than in the WT and the decay became nearly monophasic. Numerical simulations indicate that the residual deviation from a single exponential decay in PV/CB-/- is due to saturation of the Ca2+ indicator dye. Furthermore, the simulations imply that the effect of uncharacterised endogenous Ca2+ binding proteins is negligible, that buffered diffusion and dye saturation significantly affects spineous Ca2+ transients but not those in the dendritic shafts, and that neither CB nor PV undergoes saturation in spines or dendrites during climbing fibre-evoked Ca2+ transients. Calbindin's medium-affinity binding sites are fast enough to reduce the peak amplitude of the Ca2+ signal. However, similar to PV, delayed binding by CB leads to biphasic Ca2+ decay kinetics. Our results suggest that the distinct kinetics of PV and CB underlie the biphasic kinetics of synaptically evoked Ca2+ transients in dendritic shafts of PCs.

Mutational analysis of dendritic Ca2+ kinetics in rodent Purkinje cells: role of parvalbumin and calbindin D28k.

Ca2 + dinamiche in parvalbumin (PV-/-) e parvalbumin/calbindin D28k null-mutante (PV/CB-/-) topi sono stati confrontati con le risposte in animali di wild-type (WT). Use-dependent changes of synaptic transmission are thought to depend on the evaluation of temporal correlations between pre- and postsynaptic excitatory activity. Previous studies have demonstrated long-term potentiation (LTP) in layer V pyramidal cells after coincident pairing of presynaptic excitatory input with postsynaptic depolarizations, evoking backpropagating action potentials (BAPs). Here we paired excitatory layer II input with somatic hyperpolarization, which blocked BAPs in layer V pyramidal cells of rat visual cortex and induced reliable long-term depression (LTD). Upon cholinergic receptor activation, this BAP-independent protocol also induced LTP, which was not dependent on N-methyl-d-aspartate receptor activation, but blocked by metabotropic glutamate receptor (mGluR) antagonists.

Inhibitory interactions play a crucial role in the synchronization of neuronal activity. Here we investigate the effect of GABAergic PSPs on spike timing in cortical neurons that exhibit an oscillatory modulation of their membrane potential. To this end we combined numerical simulations with in-vitro patch-clamp recordings from layer II/III pyramidal cells of the rat visual cortex. Special emphasis was placed on exploring how the reversal potential of the GABAergic synaptic currents (EGABA) and the phase relations of the PSPs relative to the oscillation cycles affect the timing of spikes riding on the depolarizing peaks of the oscillations. The simulations predicted: (1) With EGABA more negative than the oscillation minima PSPs are hyperpolarizing at all phases and thus delay or prevent spikes. (2) With EGABA being more positive than the oscillation maxima PSPs are depolarizing in a phase-independent way and lead to a phase advance of spikes. (3) In the intermediate case where EGABA lies within oscillation maxima and minima PSPs are either hyper- or depolarizing depending on their phase relations to the V(m) oscillations and can therefore either delay or advance spikes. Experiments conducted in this most interesting last configuration with biphasic PSPs agreed with the model predictions. Additional theoretical investigations revealed the effect of these PSP induced shifts in spike timing on synchronization in neuronal circuits. The results suggest that GABAergic mechanisms can assume highly specific timing functions in oscillatory networks.

Interactions inhibitrices jouent un r&#xF4;le crucial dans la synchronisation de l&#x26;#39;activit&#xE9; neuronale. Nous &#xE9;tudions ici l&#x26;#39;effet de la PSP GABAergiques sur le calendrier pic dans les neurones corticaux qui pr&#xE9;sentent une modulation oscillatoire de leur potentiel de membrane. &#xC0; cette fin, nous avons combin&#xE9; des simulations num&#xE9;riques avec in-vitro de patch-clamp enregistrements de la couche II / III cellules pyramidales du cortex de rat visuelle. Des exp&#xE9;riences men&#xE9;es dans cette configuration la plus int&#xE9;ressante dernier avec PSP biphasiques d&#x26;#39;accord avec les pr&#xE9;dictions du mod&#xE8;le. D&#x26;#39;autres investigations th&#xE9;oriques ont r&#xE9;v&#xE9;l&#xE9; l&#x26;#39;effet de ces changements induits PSP dans le calendrier pic sur la synchronisation dans les circuits neuronaux. Interazioni inibitorie giocano un ruolo cruciale nella sincronizzazione dell&#39;attivit&agrave; neuronale. Qui si indaga l&#39;effetto di GABAergic PSP sulla tempistica di spike in neuroni corticali che esibiscono una modulazione oscillatoria del loro potenziale di membrana. A tal fine abbiamo combinato simulazioni numeriche con le registrazioni di patch-clamp in vitro di cellule piramidali di livello II/III della corteccia visiva del ratto. Particolare enfasi era posta sull&#39;esplorazione come il potenziale di inversione delle correnti sinaptiche GABAergico (EGABA) e le relazioni di fase della PSP rispetto i cicli di oscillazione possono influenzare la tempistica dei picchi cavalcando le cime depolarizing delle oscillazioni. Le simulazioni predette: (1) con EGABA pi&ugrave; negativo rispetto la minima oscillazione PSP &egrave; hyperpolarizing in tutte le fasi e quindi ritarda o prevenzione spikes. (2) Con EGABA essere pi&ugrave; positivo rispetto la maxima oscillazione PSP sono depolarizzante in modo indipendente dalla fase e portare ad un avanzamento di fase dei picchi. (3) Nel caso intermedio dove EGABA si trova all&#39;interno di oscillazione massimi e minimi PSP sono iper- o depolarizzante a seconda delle loro relazioni di fase con le oscillazioni di V(m) e pu&ograve; pertanto o ritardo o anticipo picchi. Esperimenti condotti in questa ultima configurazione pi&ugrave; interessante con bifasico che PSP concordato con le previsioni del modello. Ulteriori indagini teoriche ha rivelato che l&#39;effetto di queste PSP indotto variazioni nel timing spike su sincronizzazione nei circuiti neuronali. I risultati suggeriscono che meccanismi di GABAergic possono assumere funzioni di temporizzazione altamente specifiche in reti oscillatori.

Interacciones inhibitorias juegan un papel crucial en la sincronizaci&#xF3;n de la actividad neuronal. En este trabajo investigamos el efecto de la PSP GABA&#xE9;rgicas sobre el momento pico de las neuronas corticales que muestran una modulaci&#xF3;n oscilatoria de su potencial de membrana. Con este fin hemos combinado simulaciones num&#xE9;ricas con las in-vitro de patch-clamp grabaciones de la capa II / III de las c&#xE9;lulas piramidales de la corteza cerebral de rata visual. Se hizo especial hincapi&#xE9; en la exploraci&#xF3;n de c&#xF3;mo el potencial de inversi&#xF3;n de las corrientes sin&#xE1;pticas GABA&#xE9;rgicas (EGABA) y las relaciones de fase de las PSP en relaci&#xF3;n con los ciclos de oscilaci&#xF3;n afectar&#xE1; el tiempo de los picos de montar a caballo en los picos de despolarizaci&#xF3;n de las oscilaciones. Neurons have a wide range of dendritic morphologies the functions of which are largely unknown. We used an optimization procedure to find neuronal morphological structures for two computational tasks: first, neuronal morphologies were selected for linearly summing excitatory synaptic potentials (EPSPs); second, structures were selected that distinguished the temporal order of EPSPs. The solutions resembled the morphology of real neurons. In particular the neurons optimized for linear summation electrotonically separated their synapses, as found in avian nucleus laminaris neurons, and neurons optimized for spike-order detection had primary dendrites of significantly different diameter, as found in the basal and apical dendrites of cortical pyramidal neurons. This similarity makes an experimentally testable prediction of our theoretical approach, which is that pyramidal neurons can act as spike-order detectors for basal and apical inputs. The automated mapping between neuronal function and structure introduced here could allow a large catalog of computational functions to be built indexed by morphological structure.

Pharmacologically isolated GABAergic irregular spiking and stuttering interneurons in the mouse visual cortex display highly irregular spike times, with high coefficients of variation approximately 0.9-3, in response to a depolarizing, constant current input. This is in marked contrast to cortical pyramidal cells, which spike quite regularly in response to the same current injection. We applied time-series analysis methods to show that the irregular behavior of the interneurons was not a consequence of low-dimensional, deterministic processes. These methods were also applied to the Hindmarsh and Rose neuronal model to confirm that the methods are adequate for the types of data under investigation. This result has important consequences for the origin of fluctuations observed in the cortex in vivo.

The response of an oscillator to perturbations is described by its phase-response curve (PRC), which is related to the type of bifurcation leading from rest to tonic spiking. In a recent experimental study, we have shown that the type of PRC in cortical pyramidal neurons can be switched by cholinergic neuromodulation from type II (biphasic) to type I (monophasic). We explored how intrinsic mechanisms affected by acetylcholine influence the PRC using three different types of neuronal models: a theta neuron, single-compartment neurons and a multi-compartment neuron. In all of these models a decrease in the amount of a spike-frequency adaptation current was a necessary and sufficient condition for the shape of the PRC to change from biphasic (type II) to purely positive (type I).

Spike generation in cortical neurons depends on the interplay between diverse intrinsic conductances. The phase response curve (PRC) is a measure of the spike time shift caused by perturbations of the membrane potential as a function of the phase of the spike cycle of a neuron. Near the rheobase, purely positive (type I) phase-response curves are associated with an onset of repetitive firing through a saddle-node bifurcation, whereas biphasic (type II) phase-response curves point towards a transition based on a Hopf-Andronov bifurcation. In recordings from layer 2/3 pyramidal neurons in cortical slices, cholinergic action, consistent with down-regulation of slow voltage-dependent potassium currents such as the M-current, switched the PRC from type II to type I. This is the first report showing that cholinergic neuromodulation may cause a qualitative switch in the PRCs type implying a change in the fundamental dynamical mechanism of spike generation.

For many classes of neurons, the relationship between computational function and dendritic morphology remains unclear. To gain insights into this relationship, we utilize an inverse approach in which we optimize model neurons with realistic morphologies and ion channel distributions (of I(KA) and I(CaT)) to perform a computational function. In this study, the desired function is input-order detection: neurons have to respond differentially to the arrival of two inputs in a different temporal order. There is a single free parameter in this function, namely, the time lag between the arrivals of the two inputs. Systematically varying this parameter allowed us to map one axis of function space to structure space. Because the function of the optimized model neurons is known with certainty, their thorough analysis provides insights into the relationship between the neurons' functions, morphologies, ion channel distributions, and electrophysiological dynamics. Finally, we discuss issues of optimality in nervous systems.

Fast inhibitory synaptic inputs, which cause conductance changes that typically last for 10-100 ms, participate in the generation and maintenance of cortical rhythms. We show here that these fast events can have influences that outlast the duration of the synaptic potentials by interacting with subthreshold membrane potential oscillations. Inhibitory postsynaptic potentials (IPSPs) in cortical neurons in vitro shifted the oscillatory phase for several seconds. The phase shift caused by two IPSPs or two current pulses summed non-linearly. Cholinergic neuromodulation increased the power of the oscillations and decreased the magnitude of the phase shifts. These results show that the intrinsic conductances of cortical pyramidal neurons can carry information about inhibitory inputs and can extend the integration window for synaptic input.

We discuss the significance of our results and provide guidelines to choose the best method based on the available data.

Fly lobula plate tangential cells are known to perform wide-field motion integration. It is assumed that the shape of these neurons, and in particular the shape of the subclass of VS cells, is responsible for this type of computation. We employed an inverse approach to investigate the morphology-function relationship underlying wide-field motion integration in VS cells. In the inverse approach detailed, model neurons are optimized to perform a predefined computation: here, wide-field motion integration. We embedded the model neurons to be optimized in a biologically plausible model of fly motion detection to provide realistic inputs, and subsequently optimized model neuron with and without active conductances (g(Na), g(K), g(K(Na))) along their dendrites to perform this computation. We found that both passive and active optimized model neurons perform well as wide-field motion integrators. In addition, all optimized morphologies share the same blueprint as real VS cells. In addition, we also found a recurring blueprint for the distribution of g(K) and g(Na) in the active models. Moreover, we demonstrate how this morphology and distribution of conductances contribute to wide-field motion integration. As such, by using the inverse approach we can predict the still unknown distribution of g(K) and g(Na) and their role in motion integration in VS cells.

We outline an inverse approach for investigating dendritic function-structure relationships by optimizing dendritic trees for a priori chosen computational functions. The inverse approach can be applied in two different ways. First, we can use it as a "hypothesis generator" in which we optimize dendrites for a function of general interest. The optimization yields an artificial dendrite that is subsequently compared to real neurons. This comparison potentially allows us to propose hypotheses about the function of real neurons. In this way, we investigated dendrites that optimally perform input-order detection. Second, we can use it as a "function confirmation" by optimizing dendrites for functions hypothesized to be performed by classes of neurons. If the optimized, artificial, dendrites resemble the dendrites of real neurons the artificial dendrites corroborate the hypothesized function of the real neuron. Moreover, properties of the artificial dendrites can lead to predictions about yet unmeasured properties. In this way, we investigated wide-field motion integration performed by the VS cells of the fly visual system. In outlining the inverse approach and two applications, we also elaborate on the nature of dendritic function. We furthermore discuss the role of optimality in assigning functions to dendrites and point out interesting future directions.

This letter discusses temporal order coding and detection in nervous systems. Detection of temporal order in the external world is an adaptive function of nervous systems. In addition, coding based on the temporal order of signals can be used as an internal code. Such temporal order coding is a subset of temporal coding. We discuss two examples of processing the temporal order of external events: the auditory location detection system in birds and the visual direction detection system in flies. We then discuss how somatosensory stimulus intensities are translated into a temporal order code in the human peripheral nervous system. We next turn our attention to input order coding in the mammalian cortex. We review work demonstrating the capabilities of cortical neurons for detecting input order. We then discuss research refuting and demonstrating the representation of stimulus features in the cortex by means of input order. After some general theoretical considerations on input order detection and coding, we conclude by discussing the existing and potential use of input order coding in neuromorphic engineering.

We present an FPGA implementation of a re-configurable, polychronous spiking neural network with a large capacity for spatial-temporal patterns. The proposed neural network generates delay paths de novo, so that only connections that actually appear in the training patterns will be created. This allows the proposed network to use all the axons (variables) to store information. Spike Timing Dependent Delay Plasticity is used to fine-tune and add dynamics to the network. We use a time multiplexing approach allowing us to achieve 4096 (4k) neurons and up to 1.15 million programmable delay axons on a Virtex 6 FPGA. Test results show that the proposed neural network is capable of successfully recalling more than 95% of all spikes for 96% of the stored patterns. The tests also show that the neural network is robust to noise from random input spikes.

Cortical spike trains are highly irregular both during ongoing, spontaneous activity and when driven at high firing rates. There is uncertainty about the source of this irregularity, ranging from intrinsic noise sources in neurons to collective effects in large-scale cortical networks. Cortical interneurons display highly irregular spike times (coefficient of variation of the interspike intervals >1) in response to dc-current injection in vitro. This is in marked contrast to cortical pyramidal cells, which spike highly irregularly in vivo, but regularly in vitro. We show with in vitro recordings and computational models that this is due to the fast activation kinetics of interneuronal K(+) currents. This explanation holds over a wide parameter range and with Gaussian white, power-law, and Ornstein-Uhlenbeck noise. The intrinsically irregular spiking of interneurons could contribute to the irregularity of the cortical network.
