The interplay between peritoneal mesothelial cells and ovarian cancer cells is critical for the initiation and peritoneal dissemination of, and ascites formation in, ovarian cancer. The production of lysophosphatidic acid (LPA) by both peritoneal mesothelial cells and ovarian cancer cells has been shown to promote metastatic phenotype in ovarian cancer. Herein, we report that exogenous addition or ectopic overexpression of the matricellular protein SPARC (secreted protein acidic and rich in cysteine) significantly attenuated LPA-induced proliferation, chemotaxis, and invasion in both highly metastatic SKOV3 and less metastatic OVCAR3 ovarian cancer cell lines. SPARC appears to modulate these functions, at least in part, through the regulation of LPA receptor levels and the attenuation of extracellular signal-regulated kinase (ERK) 1/2 and protein kinase B/AKT signaling. Moreover, our results show that SPARC not only significantly inhibited both basal and LPA-induced interleukin (IL) 6 production in both cell lines but also attenuated IL-6-induced mitogenic, chemotactic, and proinvasive effects, in part, through significant suppression of ERK1/2 and, to a lesser extent, of signal transducers and activators of transcription 3 signaling pathways. Our results strongly suggest that SPARC exerts a dual inhibitory effect on LPA-induced mesothelial-ovarian cancer cell crosstalk through the regulation of both LPA-induced IL-6 production and function. Taken together, our findings underscore the use of SPARC as a potential therapeutic candidate in peritoneal ovarian carcinomatosis.

SPARC inhibits LPA-mediated mesothelial-ovarian cancer cell crosstalk.

The matricellular protein secreted protein acidic and rich in cysteine (SPARC) modulates cell adhesion, proliferation, matrix deposition, and tissue remodeling. SPARC has been shown to regulate the expression of collagen type I and transforming growth factor-beta1 in mesangial cells and to be highly expressed during tubulointerstitial fibrosis in rat angiotensin (ANG) II infusion models. We hypothesized that SPARC is a downstream effector of ANG II and that loss of host SPARC function provides a protective effect on renal damage and fibrosis associated with ANG II hypertension. Our results revealed that cultured primary mesangial cells displayed a concentration-dependent increase in SPARC expression in response to ANG II. After a 14-day chronic infusion of ANG II, hypertensive SPARC-null mice exhibited significantly attenuated levels of urinary and renal indicators of oxidative stress and inflammation and decreased renal perivascular and tubulointerstitial fibrosis relative to wild-type hypertensive controls. Moreover, the observed renal protective changes in SPARC-null mice were found to be independent of blood pressure. These results identify SPARC as an effector of ANG II signaling and suggest an important role for SPARC in mediating ANG II-induced oxidative stress, inflammation, and fibrosis.

Secreted protein acidic and rich in cysteine deficiency ameliorates renal inflammation and fibrosis in angiotensin hypertension.

Malignant ascites is a major source of morbidity and mortality in ovarian cancer patients. It functions as a permissive reactive tumor-host microenvironment and provides sustenance for the floating tumor cells through a plethora of survival/metastasis-associated molecules. Using a syngeneic, immunocompetent model of peritoneal ovarian carcinomatosis in SP(-/-) mice, we investigated the molecular mechanisms implicated in the interplay between host secreted protein acidic and rich in cysteine (SPARC) and ascitic fluid prosurvival/prometastasis factors that result in the significantly augmented levels of vascular endothelial growth factor (VEGF) and matrix metalloproteinases (MMP). Ascitic fluid-enhanced ID8 invasiveness was mediated through VEGF via a positive feedback loop with MMP-2 and MMP-9 and through activation of alpha(v) and beta(1) integrins. Host SPARC down-regulated the VEGF-MMP axis at the transcriptional and posttranscriptional levels. In vitro, SPARC attenuated the basal as well as VEGF-induced integrin activation in tumor cells. SPARC inhibited the VEGF- and integrin-mediated ID8 proliferation in vitro and significantly suppressed their tumorigenicity in vivo. Relative to SP(+/+), SP(-/-) ascitic fluid contained significantly higher levels of bioactive lipids and exerted stronger chemotactic, proinvasive, and mitogenic effects on ID8 cells in vitro. SP(-/-) ascites also contained high levels of interleukin-6, macrophage chemoattractant protein-1, and 8-isoprostane (prostaglandin F(2)alpha) that were positively correlated with extensive infiltration of SP(-/-) ovarian tumors and ascites with macrophages. In summary, our findings strongly suggest that host SPARC normalizes the microenvironment of ovarian cancer malignant ascites through down-regulation of the VEGF-integrin-MMP axis, decreases the levels and activity of bioactive lipids, and ameliorates downstream inflammation.

Normalization of the ovarian cancer microenvironment by SPARC.

Epigenetic silencing of tumor suppressor genes is a new focus of investigation in the generation and proliferation of carcinomas. Secreted protein acidic and rich in cysteine (SPARC) is reportedly detrimental to the growth of ovarian cancer cells and has been shown to be epigenetically silenced in several cancers. We hypothesized that SPARC is downregulated in ovarian cancer through aberrant promoter hypermethylation. To that end, we analyzed SPARC expression in ovarian cancer cell lines and investigated the methylation status of the Sparc promoter using methylation-specific polymerase chain reaction. Our results show that SPARC mRNA expression is decreased in three (33%) and absent in four (44%) of the nine ovarian cancer cell lines studied, which correlated with hypermethylation of the Sparc promoter. Treatment with the demethylating agent 5-aza-2'-deoxycytidine rescued SPARC mRNA and protein expression. Addition of exogenous SPARC, as well as ectopic expression by an adenoviral vector, resulted in decreased proliferation of ovarian cancer cell lines. Investigation of primary tumors revealed that the Sparc promoter is methylated in 68% of primary ovarian tumors and that the levels of SPARC protein decrease as the disease progresses from low to high grade. Lastly, de novo methylation of Sparc promoter was shown to be mediated by DNA methyltransferase 3a. These results implicate Sparc promoter methylation as an important factor in the genesis and survival of ovarian carcinomas and provide new insights into the potential use of SPARC as a novel biomarker and/or treatment modality for this disease.

To study Ca(2+) signaling in the endothelium of murine feed arteries, we determined the in vitro stability of endothelial cell (EC) tubes freshly isolated from abdominal muscle feed arteries of male and female C57BL/6 mice (5-9 mo, 25-35 g). We tested the hypothesis that intracellular Ca(2+) concentration ([Ca(2+)](i)) responses to muscarinic receptor activation would increase with temperature. Intact EC tubes (length: 1-2 mm, width: 65-80 Î¼m) were isolated using gentle enzymatic digestion with trituration to remove smooth muscle cells. A freshly isolated EC tube was secured in a chamber and superfused at 24 (room temperature), 32, or 37Â°C. Using fura-2 dye, [Ca(2+)](i) was monitored (ratio of fluorescence at 340- to 380-nm wavelength) at rest and in response to bolus doses of ACh (20 nmol to 200 Î¼mol). The morphological integrity of EC tubes was preserved at 24 and 32Â°C. Based on the Ca(2+) K(d) values we determined for fura-2 (174 nM at 24Â°C and 146 nM at 32Â°C), resting [Ca(2+)](i) remained stable for 180 min at both 24 and 32Â°C (27 Â± 4 and 34 Â± 2 nM, respectively), with peak responses to ACh (20 Î¼mol) increasing from âˆ¼220 nM at 24Â°C to âˆ¼500 nM at 32Â°C (P < 0.05). There was no difference in responses to ACh between EC tubes from male versus female mice. When EC tubes were maintained at 37Â°C (typical in vivo temperature), resting [Ca(2+)](i) increased by âˆ¼30% within 15 min, and gaps formed between individual ECs as they retracted and extruded dye, precluding further study. We conclude that EC tubes enable Ca(2+) signaling to be evaluated in the freshly isolated endothelium of murine feed arteries. While Ca(2+) responses are enhanced by approximately twofold at 32 versus 24Â°C, the instability of EC tubes at 37Â°C precludes their study at typical body temperature.

Non non c&#39;era alcuna differenza nelle risposte di ACh tra tubi CE da maschio contro topi femminili. Quando CE tubi sono stati mantenuti a 37 &deg; C (temperatura tipica in vivo), riposo [Ca(2+)](i) aumentato di &sim;30% entro 15 min e le lacune formate tra singoli ECs poich&egrave; retratto ed estruso colorante, precludendo ulteriore studio. Para el estudio de Ca (2 +) de se&#xF1;alizaci&#xF3;n en el endotelio de las arterias de alimentaci&#xF3;n de rat&#xF3;n, se determin&#xF3; la estabilidad in vitro de c&#xE9;lulas endoteliales (CE) tubos reci&#xE9;n aisladas de las arterias musculares abdominales de alimentaci&#xF3;n de machos y hembras C57BL / 6 ratones (5-9 meses, 25-35 g). Pusimos a prueba la hip&#xF3;tesis de que la intracelular de Ca (2 +) de concentraci&#xF3;n ([Ca (2 +)] (i)) las respuestas a la activaci&#xF3;n de los receptores muscar&#xED;nicos que aumentan con la temperatura. Intactos los tubos de la CE (longitud: 2.1 mm, ancho: 65 a 80 micras) se aislaron mediante digesti&#xF3;n enzim&#xE1;tica suave con la trituraci&#xF3;n para eliminar las c&#xE9;lulas musculares lisas. Un tubo CE reci&#xE9;n aislada fue asegurado en una c&#xE1;mara y superfused al 24 (temperatura ambiente), 32, &#xF3; 37 &#xB0; C. Usando fura-2 colorante, [Ca (2 +)] (i) se control&#xF3; (relaci&#xF3;n de fluorescencia a 340 - a 380-nm de longitud de onda) en reposo y en respuesta a las dosis de bolo de ACh (20 nmol a 200 micromoles). No hubo diferencia en las respuestas a ACh entre los tubos de la CE de ratones machos frente hembra. Cuando los tubos de la CE se mantuvieron a 37 &#xB0; C (t&#xED;pico de la temperatura in vivo), en reposo [Ca (2 +)] (i) aumento en un 30% en los 15 minutos, y los huecos formados entre el individuo AE, ya que se retract&#xF3; y el tinte extruido, lo que impide estudio adicional. This MiniReview is focused on the nature of intercellular signalling along the endothelium that helps to co-ordinate blood flow control in vascular resistance networks. Vasodilation initiated by contracting skeletal muscle ascends from arterioles within the tissue to encompass resistance arteries upstream and thereby increase blood flow during exercise. In resistance vessels, acetylcholine microiontophoresis or intracellular current injection initiates hyperpolarization that conducts through gap junction channels (GJCs) along the vessel wall resulting in conducted vasodilation (CVD). Both ascending vasodilation and CVD are eliminated with endothelial cell (EC) disruption, pointing to common signalling events and mutual dependence upon EC integrity. As demonstrated by electrical coupling and dye transfer during intracellular recording, their longitudinal orientation and robust expression of GJCs enable ECs to play a predominant role in CVD. Once conduction is initiated, a major interest centres on whether CVD is purely passive or involves additional 'active' signalling events. Here, we discuss components for Ca(2+) and electrical signalling with an emphasis on intercellular coupling through endothelial GJCs. We stress the importance of understanding relationships between intracellular Ca(2+) dynamics, EC hyperpolarization and CVD while integrating findings from isolated ECs into more complex interactions in vivo. Whereas endothelial dysfunction accompanies cardiovascular disease and the components of intra- and inter-cellular signalling are increasingly well defined, little is known of how Ca(2+) signalling and electrical conduction along microvascular endothelium are altered in diseased states. Thus, greater insight into how these relationships are governed and interact is a key goal for continued research efforts.

Background and purpose:â€ƒ Electrical conduction along endothelium of resistance vessels has not been determined independent from the influence of smooth muscle, surrounding tissue or blood. Two interrelated hypotheses were tested: (1) Intercellular conduction of electrical signals is manifest in endothelial cell (EC) tubes; (2) Inhibitors of gap junction channels (GJCs) have confounding actions on EC electrical and Ca(2+) signaling. Experimental approach:â€ƒ Intact EC tubes were isolated from abdominal muscle feed arteries of C57BL/6 mice. Hyperpolarization was initiated with indirect (acetylcholine) and direct (NS309) stimulation of intermediate- and small-conductance Ca(2+) -activated K(+) channels (IK(Ca) /SK(Ca) ). Remote membrane potential (V(m) ) responses to intracellular current injection defined the length constant (Î») for electrical conduction. Dye coupling was evaluated following intracellular microinjection of propidium iodide. Intracellular Ca(2+) dynamics were determined using Fura-2 photometry. Carbenoxolone (CBX) or Î²-glycyrrhetinic acid (Î²GA) were used to investigate the role of GJCs. Key results:â€ƒ Steady-state V(m) of ECs was âˆ¼-25 mV. Acetylcholine and NS309 hyperpolarized ECs by âˆ¼-40 mV and âˆ¼-60 mV, respectively. Electrical conduction decayed monoexponentially with distance (Î»âˆ¼1.4 mm). Propidium iodide injected into one EC spread into surrounding ECs. CBX or Î²GA inhibited dye transfer, electrical conduction and EC hyperpolarization reversibly. Both agents elevated resting Ca(2+) while Î²GA inhibited responses to ACh. Conclusions and implications:â€ƒ Individual cells are well-coupled to each other within EC tubes. Inhibiting GJCs with glycyrrhetinic acid derivatives coincides with loss of hyperpolarization mediated by IK(Ca) /SK(Ca) irrespective of Ca(2+) signaling, obviating use of these agents in resolving key determinants of electrical conduction along the endothelium.

Antecedentes y finalidad de la conducci&#xF3;n el&#xE9;ctrica a lo largo de endotelio de los vasos de resistencia no ha sido determinada independiente de la influencia de m&#xFA;sculo liso, el tejido circundante o de sangre. Dos hip&#xF3;tesis relacionadas entre s&#xED; se pusieron a prueba: (1) la conducci&#xF3;n intercelular de las se&#xF1;ales el&#xE9;ctricas se manifiesta en las c&#xE9;lulas endoteliales (CE) tubos, (2) Los inhibidores de los canales de derivaci&#xF3;n brecha (GJCs) tienen acciones de confusi&#xF3;n en CE el&#xE9;ctrica y Ca (2 +) de se&#xF1;alizaci&#xF3;n. Enfoque experimental: intactos los tubos de la CE fueron aisladas de las arterias musculares abdominales de alimentaci&#xF3;n de ratones C57BL / 6. Carbenoxolona (CBX) o &#x3B2;-glycyrrhetinic &#xE1;cido (&#x3B2;GA) fueron utilizados para investigar el papel de GJCs. Principales resultados: El estado de equilibrio V (m) de EC fue de ~ -25 mV. La acetilcolina y la NS309 hiperpolarizado EC by ~ -40 mV y -60 mV ~, respectivamente. La conducci&#xF3;n el&#xE9;ctrica decay&#xF3; monoexponentially con la distancia (lambda ~ 1.4 mm). Yoduro de propidio se inyecta en una extensi&#xF3;n que rodea la CE en EC. CBX o &#x3B2;GA inhibe la transferencia de tintes, la conducci&#xF3;n el&#xE9;ctrica e hiperpolarizaci&#xF3;n CE reversible. Ambos agentes elevado reposo Ca (2 +) mientras que &#x3B2;GA inhibe las respuestas a la ACh. Conclusiones y consecuencias: las c&#xE9;lulas individuales est&#xE1;n bien acoplados entre s&#xED; dentro de los tubos de la CE. La inhibici&#xF3;n de GJCs con derivados del &#xE1;cido glicirret&#xED;nico coincide con la p&#xE9;rdida de hiperpolarizaci&#xF3;n mediada por IK (Ca) / SK (Ca), con independencia de Ca (2 +) de se&#xF1;alizaci&#xF3;n, obviando el uso de estos agentes en la soluci&#xF3;n de los determinantes clave de la conducci&#xF3;n el&#xE9;ctrica a lo largo del endotelio.

Intercellular conduction of electrical signals underlies spreading vasodilation of resistance arteries. Small- and intermediate-conductance Ca(2+)-activated K(+) channels of endothelial cells serve a dual function by initiating hyperpolarization and modulating electrical conduction. We tested the hypothesis that regulation of electrical signaling by small- and intermediate-conductance Ca(2+)-activated K(+) channels is altered with advancing age.
