Patients with thymic malignancy have high rates of autoimmunity leading to a variety of autoimmune diseases, most commonly myasthenia gravis caused by anti-acetylcholine receptor autoantibodies. High rates of autoantibodies to cytokines have also been described, although prevalence, spectrum, and functionality of these anti-cytokine autoantibodies are poorly defined. To better understand the presence and function of anti-cytokine autoantibodies, we created a luciferase immunoprecipitation system panel to search for autoantibodies against 39 different cytokines and examined plasma from controls (n = 30) and patients with thymic neoplasia (n = 17). In this screen, our patients showed statistically elevated, but highly heterogeneous immunoreactivity against 16 of the 39 cytokines. Some patients showed autoantibodies to multiple cytokines. Functional testing proved that autoantibodies directed against interferon-Î±, interferon-Î², interleukin-1Î± (IL-1Î±), IL-12p35, IL-12p40, and IL-17A had biologic blocking activity in vitro. All patients with opportunistic infection showed multiple anti-cytokine autoantibodies (range 3-11), suggesting that anti-cytokine autoantibodies may be important in the pathogenesis of opportunistic infections in patients with thymic malignancy. Les patients atteints de tumeurs malignes du thymus ont des taux &#xE9;lev&#xE9;s d&#x26;#39;auto-immunit&#xE9; conduisant &#xE0; une vari&#xE9;t&#xE9; de maladies auto-immunes, le plus souvent caus&#xE9;es par la myasth&#xE9;nie auto-anticorps anti-r&#xE9;cepteurs de l&#x26;#39;ac&#xE9;tylcholine. Les taux &#xE9;lev&#xE9;s d&#x26;#39;auto-anticorps &#xE0; des cytokines ont &#xE9;galement &#xE9;t&#xE9; d&#xE9;crites, bien que la pr&#xE9;valence, le spectre, et la fonctionnalit&#xE9; de ces auto-anticorps anti-cytokine sont mal d&#xE9;finis. Certains patients ont montr&#xE9; des autoanticorps dirig&#xE9;s contre des cytokines multiples. Les tests fonctionnels prouv&#xE9; que auto-anticorps dirig&#xE9;s contre l&#x26;#39;interf&#xE9;ron &#x3B1;-, &#x3B2;-interf&#xE9;ron, l&#x26;#39;interleukine-1&#x3B1; (IL-1&#x3B1;), IL-12p35, l&#x26;#39;IL-12p40 et IL-17A avait une activit&#xE9; biologique in vitro de blocage. Anti-cytokine auto-anticorps sont associ&#xE9;s &#xE0; l&#x26;#39;infection opportuniste chez les patients atteints d&#x26;#39;une n&#xE9;oplasie thymique

Patienten mit Thymom B&ouml;sartigkeit haben hohe Raten von Autoimmunit&auml;t f&uuml;hren zu einer Vielzahl von Autoimmunerkrankungen, am h&auml;ufigsten Myasthenia Gravis durch Acetylcholinrezeptor-Rezeptor-Autoantik&ouml;rper verursacht. Hohe Raten von Autoantik&ouml;rpern gegen Zytokine sind auch beschrieben worden, obwohl die Pr&auml;valenz, Spektrum und Funktionalit&auml;t dieser Anti-Zytokin-Autoantik&ouml;rper schlecht definiert sind. Um das Vorhandensein und die Funktion des Anti-Cytokine Autoantik&ouml;rper besser zu verstehen, wir erstellt ein Luciferase Immunopr&auml;zipitation System Panel Suche nach Autoantik&ouml;rpern gegen 39 verschiedene Zytokine und Plasmas Steuerelemente gepr&uuml;ft (n = 30) und Patienten mit Thymom Neoplasie (n = 17). Auf diesem Bild unserer Patienten zeigte statistisch erh&ouml;hten, aber &auml;u&szlig;erst heterogenen Immunoreactivity gegen 16 von 39 Zytokine. Einige Patienten zeigte Autoantik&ouml;rper gegen mehrere Zytokine. Funktionale Tests bewiesen, dass Autoantik&ouml;rper gegen Interferon-&alpha;, Interferon-&beta;, Interleukin-1&alpha; (IL-1&alpha;), IL - 12 p 35, IL - 12 p 40 und IL-17A biologischen in-vitro-blockierenden T&auml;tigkeit hatte. I pazienti con tumori maligni del timo hanno alti tassi di autoimmunit&agrave; che conduce ad una variet&agrave; di malattie autoimmuni, pi&ugrave; comunemente miastenia grave causata da autoanticorpi anti recettore acetilcolina. Tassi elevati di autoanticorpi di citochine sono stati anche descritti, anche se la funzionalit&agrave; di questi autoanticorpi anti-citochine, spettro e prevalenza sono mal definiti. Per meglio comprendere la presenza e la funzione di autoanticorpi anti-citochine, abbiamo creato un pannello di sistema immunoprecipitazione luciferase per la ricerca di autoanticorpi contro 39 diverse citochine ed esaminato al plasma da controlli (n = 30) e pazienti con neoplasie del timo (n = 17). In questa schermata, i nostri pazienti hanno mostrato immunoreattivit&agrave; statisticamente elevato, ma altamente eterogeneo contro 16 i cytokines 39. Alcuni pazienti hanno mostrato gli autoanticorpi di citochine multiple. Test funzionale ha dimostrato che gli autoanticorpi diretti contro interferone-&alpha;, interferone-&beta;, interleuchina-1&alpha; (IL 1&alpha;), IL-12 p 35, IL-12 p 40 e IL-17A avevano attivit&agrave; blocco biologico in vitro. Synthetic biology involves the engineering of proteins, signaling pathways and even whole organisms using modular designs and formats. A major tool of synthetic biology is artificial gene synthesis, which provides a direct means from a conceptualized DNA sequence to the corresponding physical DNA for the construction of a variety of biological components. To date, synthetic biology has often been used to answer fundamental questions in basic research, but now is poised to greatly enhance translational research. In this review, we discuss several translational applications of synthetic biology including the construction of novel diagnostics and vaccines, development of new synthetic pathways for drug screening and biosynthesis, and the creation of engineered viruses and microbes to fight human disease. Together these and other novel translational applications of artificial gene synthesis and synthetic biology have the opportunity to make major advances for improving human health.
