方案

1. 发育期暴露(或类似 binge 的乙醇暴露模型)

  1. 在出生后第3天(PD3),确定每只动物的性别,必要时进行交叉抚养,以确保每窝仔鼠数量(8只)和性别分布(4雄:4雌)保持一致。
    注意:为避免实验干扰因素,应尽可能保持每窝仔鼠数量和性别分布的一致性。尽管本方案采用每窝8只仔鼠(4雄4雌),但可根据实验设计需求调整窝仔数量或性别分布。
  2. 在每窝动物的足垫皮下注射少量黑色印度墨水,以进行个体识别。
  3. 将窝仔鼠伪随机分配为实验组(包含50%酒精暴露(AE)和50%假插管对照(SI)仔鼠)或哺乳对照组(SC)(在出生后第4至9天期间不进行任何插管、尾部剪切或分离操作,仅进行每日称重和耳标打孔)。
    1. 为保持组间规模一致,实验组的窝数应设为对照组(SC)的两倍。
  4. 称重每只动物,然后将其放回其原代笼具中。在插管期间(出生后第4至9天),应每天进行动物称重。
    1. 将整窝幼崽从母鼠身上移开。
    2. 将幼崽置于加热垫上。
    3. 记录每只幼崽的体重。
  5. 在第4天,称量每只动物后,根据每只幼鼠的体重(来自步骤1.4)计算所需酒精量,使每只动物的总摄入量为5.25 g/kg/天8.

  11. 在第二次酒精给药后一个半小时(即每日血液酒精浓度达到峰值的时间点),采集酒精暴露组(AE)和分离对照组(SI)仔鼠的血液并进行离心处理 通过 用于后续血液酒精浓度分析的尾部取样35.
    1. 采集60 µL血液。

3. 跑轮运动

  1. 在出生后第30天(PD30),将一半带动物的笼子分配至跑轮组(WR)。将这些动物饲养在配有可自由进入的不锈钢跑轮的笼子中。

  2. 在出生后第30天(PD30)和第36天(PD36)称量所有动物的体重。

  4. 让动物在其相应的饲养条件下继续饲养12天。

4. 环境复杂性

  1. 在实验动物出生后第42天(PD 42)当天上午9点前准备好丰富环境(EC)笼具。
    1. 准备一个30 " x 18 " x 36 " 的镀锌钢制笼子。
      注:笼具应具有多个层级,能够承受多只大鼠的重量,内含标准垫料,并设有多个可安装饮水瓶和饲料分配器的位置。
    2. 在笼内放置大小、形状各异的新奇彩色物体。
      1. 在EC笼内放置6个大型玩具。确保每个玩具足够大,可供3只或更多大鼠同时互动。

      4. 使用颜色、形状、大小等各不相同的玩具。etc. 新颖性对此干预措施至关重要(见讨论部分)。

5. 收集组织

注意:组织采集(例如,用多聚甲醛灌注)和储存(例如,冷冻、石蜡包埋)可通过多种方法进行。以下将说明使用0.1 M磷酸盐缓冲液(PBS)中含4%多聚甲醛(4% paraformaldehyde in PBS)溶液进行灌注的过程8

注意:多聚甲醛具有致癌性,还可能引起皮肤刺激、过敏性皮肤反应或眼部损伤。请使用适当的眼部/皮肤防护措施。

  1. 每次将一只大鼠暴露于异氟烷中,使其轻度麻醉。
  2. 向大鼠腹腔注射 2 mL/kg 的氯胺酮/赛拉嗪混合液(1.5 mL 赛拉嗪与 10 mL 氯胺酮混合)。
    注意:氯胺酮和赛拉嗪在配制注射混合液前的储存浓度均为 100 mg/mL。
