1. 用洗涤剂为基础的方法去细胞植物组织

  1. 使用新鲜或冻结的F. 刚毛, 叶样本。冻结未使用的新鲜样品在一个-20 °c 冷藏库和存储, 以供将来使用 (最多一年)。
    注: 使用几乎任何所需植物的茎或叶组织。延长贮存时间会对组织造成损害。
    1. 根据样品的预期用途确定要处理的样品的尺寸和形状 (切成条的样品非常适合于机械测试应用, 同时8毫米圆盘样品在多井培养应用中很有用。).在室温下 (20-25 °c) 去离子 H2O, 用锋利的、干净的活检冲床将叶子切成8毫米的圆盘。

  4. 将非离子表面活性剂/漂白剂溶液每24小时更换一次, 直到完全清除样品 (参照图 1A进行可视比较)。然后在2分钟的摇动板上用去离子水孵化样品, 冲洗掉多余的非离子表面活性剂/漂白剂溶液。将摇板设置为低设置以防止损坏样品。

  1. 准备 5% (v/v) 漂白剂 (次氯酸钠) 和 3% (瓦特/v) 碳酸氢钠 (NaHCO 3) 解决方案.将溶液在通风罩中加热至60-70 摄氏度, 用于F. 刚毛叶片样品在热板上搅拌时切成8毫米的圆盘。
