一种用于在无菌条件下评估植物对小分子响应的灵活低成本水培系统

摘要

一种简单、通用且低成本的体外水培系统已成功优化,可在无菌条件下进行大规模实验。该系统有助于将化学物质以溶液形式施加,并被根系高效吸收,适用于分子、生物化学和生理学研究。

摘要

植物生物学的大量研究采用水培培养方法进行。在此工作中,一种 体外 介绍了一种用于评估植物对化学物质及其他感兴趣物质反应的水培生长系统。该系统在获得均一且健康的C3 和 C4 模式物种 拟南芥Setaria viridis分别。无菌培养可避免藻类和微生物污染,而这些污染是已知的限制水培植物正常生长与发育的因素。此外,该系统具有可扩展性,既可大规模收获植物材料并尽量减少机械损伤,也可根据需要收获植物的特定部位。本文提供了一个详细的操作方案,表明该系统组装简便且成本低廉,主要利用移液器架作为植物生长的主要平台。该系统的可行性已通过以下实验验证: 拟南芥 幼苗用于评估药物AZD-8055(一种雷帕霉素靶蛋白激酶(TOR)的化学抑制剂)的作用。在根和茎中,AZD-8055处理30分钟后即可有效检测到TOR活性的抑制。此外,经AZD-8055处理的植株表现出预期的淀粉过量表型。我们建议将该水培系统作为植物研究人员监测植物诱导剂或抑制剂作用的理想方法,同时也可用于利用同位素标记化合物评估代谢通量,后者通常需要使用昂贵的试剂。

引言

利用水培法种植植物的优势在培育大型且均一的植株方面已得到广泛认可,有助于开展可重复的实验1,2,3。在此系统中,营养液的成分可在植物生长发育的各个阶段得到精确控制并实现循环利用。此外,与土壤栽培植物可能遭遇的养分匮乏和水分不足等非生物胁迫不同,水培植物的根系不会受到此类胁迫的影响4。由于水培植物所表现出的形态学和生理学特征与土壤培养的植物高度相似,该系统已被广泛应用于科研领域,因为它能够无损伤地监测根系与地上部分的生长,并实现无损采收2,5

由于可以调节营养液的成分和浓度,大多数利用水培条件的研究都用于表征微量元素和大量元素的功能1,3,6,7,8。然而,该系统已被证明在植物生物学的广泛应用中极为有用,例如阐明植物激素和化学物质的功能。例如,独脚金内酯被鉴定为一类新型激素9,以及油菜素内酯处理诱导的加速生长表型10,均是在水培条件下完成的。此外,该系统还支持使用标记同位素(例如14N/15N 和 13CO211,12 进行实验,通过质谱技术评估这些同位素在蛋白质和代谢物中的掺入情况。

鉴于该系统在植物研究中的重要性,近年来已设计出大量水培培养系统,包括使用(i)将幼苗从培养板转移到水培容器的方法3,13;(ii)岩棉材料,但其限制了对根系早期发育阶段的观察2,14,15;(iii)聚乙烯颗粒作为漂浮载体,导致小分子或处理试剂难以均匀施加16;或(iv)仅能培养少量植株的系统9,17。许多此类方案中描述的水培罐体积通常较大(小体积范围为1–5 L,最大可达32 L)18,这使得化学试剂的应用成本极为高昂。尽管已有少数研究描述了在无菌条件下进行的水培培养8,19,但系统的组装通常非常繁琐,需要将尼龙网精确地安装到塑料或玻璃容器中5,8,17,20

由于其重要性 拟南芥 作为一种模式植物,大多数水培系统都是为该物种设计的1,2,8,14,18,19,20然而,已有少数研究报道了通过对种子进行预处理以提高其萌发率和同步化程度,从而实现其他植物物种的水培生长特性 体外8,16为了进行大规模操作,我们开发了一种建立简易且低成本维持型水培系统的方法,该系统可在无菌条件下培养植物,包括 A. thaliana 以及其他物种,例如禾草 Setaria viridis该方法适用于多种实验,可最大限度地促进幼苗生长,实现同步化,并便于监测。此外,该系统具有多项优势:(i)组装简便,组件可重复使用;(ii)可轻松向液体培养基中添加不同化学物质;(iii)幼苗直接在培养基中萌发和生长,无需转移至水培系统;(iv)可密切观察地上部分和根系的发育/生长过程,且收获幼苗时不会造成损伤;(v)可实现大规模操作,同时维持生理条件。

方案

1. 液体和固体培养基的制备

  1. 使用含维生素的半强度 Murashige 和 Skoog (MS) 培养基配制液体培养基 [氯化钴(II)五水合物 0.0125 mg/L、硫酸铜(II)五水合物 0.0125 mg/L、乙二胺四乙酸铁钠 18.35 mg/L、硼酸 3.10 mg/L、碘化钾 0.415 mg/L、硫酸锰一水合物 8.45 mg/L、钼酸钠二水合物 0.125 mg/L、硫酸锌七水合物 4.30 mg/L、氯化钙 166.01 mg/L、磷酸二氢钾 85 mg/L、硝酸钾 950 mg/L、硫酸镁 90.27 mg/L、硝酸铵 825 mg/L、甘氨酸 1 mg/L、肌醇 50 mg/L、烟酸 0.25 mg/L、盐酸吡哆醇 0.25 mg/L 和盐酸硫胺素 0.05 mg/L],添加 0.25 g/L MES,并用 10 M KOH 将 pH 值调节至 5.8。
  2. 加入 10 g/L 琼脂,制成半强度 MS 固体培养基。使用前在 121 °C 下高压灭菌 20 分钟。

2. 水培系统的组装

注意:应严格遵循这些步骤以构建水培系统。

  1. 材料灭菌
    1. 将用作小型培养罐的移液器吸头架(不带盖子)装入高压灭菌袋中。在121 °C、15 psi条件下高压灭菌20分钟。
      注意:我们使用的聚丙烯移液器吸头架尺寸如下:120 mm(长)× 89 mm(宽)× 55 mm(高)。移液器吸头的平面部分应具有可添加培养基的面积。也可使用其他类型的吸头架(见材料表)。
      注意:在整个水培系统组装过程中,必须使用超净工作台,使用前需用70%乙醇清洁并消毒。实验人员应穿戴实验服,清洗双手及任何暴露的皮肤,并用70%乙醇进行消毒。除药物处理外,佩戴手套为可选操作。
    2. 在进入超净工作台前,使用70%乙醇清洁上述所有配件(一次性塑料盒、胶带、移液器、剪刀和镊子)。若超净工作台具备该功能,可在组装水培系统前开启紫外灯照射10分钟,以保持工作区域无污染。
  2. 小型培养罐组装
    1. 用胶带密封移液器吸头平面的上表面(图1B)。如条件允许,可将其置于紫外灯下照射10分钟。
    2. 使用多通道移液器,向每个孔中加入180 µL已融化的温热固体MS培养基(图1C)。
      注意:当制备多个培养罐时,可使用加热板以防止MS培养基凝固。
    3. 让培养基完全凝固(约30分钟)。
      注意:在培养基凝固期间,可开启紫外灯。
    4. 用液体MS培养基完全填满移液器吸头架(图1D),并确保固体培养基与液体培养基之间紧密接触。
    5. 移除移液器吸头平面上表面的胶带,小心地将其安装到吸头架上。此时水培系统已准备就绪,可接种灭菌后的种子。

3. 种子灭菌

  1. 放置 500 拟南芥 将种子放入1.5 mL离心管中。根据实验所需植物数量,使用相应数量的离心管。
  2. 用70%乙醇轻轻振荡清洗种子2分钟。待种子沉降后,小心移除乙醇。
  3. 加入1 mL含2 µL吐温20(polysorbate 20)的10%次氯酸钠溶液,振荡5分钟,小心移除溶液。
  4. 用无菌蒸馏水冲洗种子,直至完全去除所有次氯酸钠残留(约5次)。
    注意:表面灭菌后,将种子浸入无菌蒸馏水中,并在4 °C避光条件下层积处理5天,以同步萌发。
    注意:种子的 Setaria viridis (登录号 A10.1)在浓硫酸中预孵育 15 分钟(以打破物理休眠),用无菌蒸馏水充分冲洗,然后在含 0.1% 吐温 20 的 5% 次氯酸钠溶液中轻轻振荡消毒 5 分钟21。其余灭菌步骤与前述步骤相同 拟南芥 种子

4. 种子处理

  1. 使用无菌手术刀略微剪切200 µL吸头的尖端。
  2. 移液管吸取 拟南芥 将种子接种到移液管尖端平面上方的固体培养基表面。注意避免培养基从平面上松脱,否则种子会受到遮蔽,导致幼苗无法正常生长。图1E).
    注意:使用无菌镊子 Setaria 种子(胚朝上放置)。
  3. 尽可能将多个小型培养罐存放在一次性塑料盒内,以维持高湿度环境并防止微生物污染(图1F).
  4. 使用胶带将一次性塑料盒完全密封,以避免污染。
  5. 将水培系统放入为特定植物提供适宜生长条件的生长室中。
    注意:本研究中采用以下条件:75% 相对湿度,以及 150 µmol m⁻² s⁻¹ 的光照强度。-2 s-1 光照强度及春分/秋分条件:12小时光照(21 °C)/12小时黑暗(19 °C) 拟南芥,或 300 µmol m-2 s-1 光照强度和12小时光照(28 °C)/12小时黑暗(25 °C)条件 Setaria (图 1G1H).

5. 验证该水培系统在抑制雷帕霉素靶蛋白激酶中的应用

注意:该水培系统最初是为了便于向植物施加化学物质而开发的,而这些化学物质通常在大规模实验中应用时成本非常高。作为概念验证,使用了已知可特异性靶向TOR蛋白激酶ATP结合位点的ATP竞争性抑制剂AZD-805522,以追踪其对拟南芥(A. thaliana)Columbia-0生态型(诺丁汉拟南芥种质中心,NASC编号:N22681)幼苗中TOR活性的抑制作用。此处简要描述所用实验方案。

  1. 根据BBCH标准,将种子水培至1.04期23 (约11天)置于上述气候条件下培养。在夜间结束时(EN),更换营养液,分别加入含0.05% DMSO(对照)、2 µM AZD-8055(TOR抑制剂,溶于DMSO)的新鲜培养基,或不作处理(mock)。
  2. 在处理后的不同时间点收获一些幼苗,并将其分离为根和地上部分。将样品置于液氮中速冻,然后在机器人研磨仪中研磨成细粉(参见 材料表),并将粉末在 -80 °C 保存至使用。
  3. 根据Dobrenel的方法对40S核糖体蛋白S6(RPS6)的磷酸化与非磷酸化形式进行免疫印迹分析 24.
  4. 对完整幼苗进行漂白以去除样品色素,用蒸馏水洗涤,然后浸入碘溶液中5分钟25,然后在体视显微镜下(0.63倍物镜,20倍近似,7.5倍放大)拍摄幼苗照片,以定性评估淀粉含量。
  5. 通过将淀粉酶解后释放的葡萄糖与NADP的还原反应偶联,采用分光光度法测定葡萄糖含量,从而定量淀粉+ 至 NADPH26,27.
  6. 按照Caldana所述方法进行总RNA提取、cDNA合成以及定量RT-PCR检测 28 评估与不同类型胁迫相关基因的表达水平。
  7. 可选地,在类似气候条件下(60% 湿度,150 µmol m⁻² s⁻¹ 光照强度,16 小时光照/8 小时黑暗光周期,22 °C),将幼苗种植于容积为 0.1 L 的塑料盆中的园艺基质上生长 [60 % 湿度,150 µmol m⁻² s⁻¹,16 小时光照/8 小时黑暗光周期,22 °C]-2 s-1 光照强度以及12小时光照(21 °C)/12小时黑暗(19 °C)的春分条件],以便与水培条件下生长的幼苗进行比较。
    注意:基因表达检测所用的目标基因为 ABF3 (At4g34000) ASN1 (At3g47340),以及 TPS5 (At4g17770),并采用ΔCt法对其表达水平进行标准化处理29 使用 ACT2 (At3g18780)或 PDF2 (At4g04890)作为内参基因,假设所有样本的PCR扩增效率均为100%。用于定量PCR的寡核苷酸引物对为: ABF3 (GTTCTCAACCTGCAACACAGTGC;TCCAGGAGATACTGCTGCAACC), ASN1 (AGGTGCGGACGAGATCTTTG;GTGAAGAGCCTTGATCTTGC), TPS5 (CTGCTCTGATGCTCCTTCTTCC;AAGCTGGTTTCCAACGATGATG), ACT2 (CGTACAACCGGTATTGTGCTGG;CTCTCTCTGTAAGGATCTTCATG),以及 PDF2 (TAACGTGGCCAAAATGATGC;GTTCTCCACAACCGCTTGGT)

结果

TOR激酶是一种主要的调控因子,在所有真核生物中整合营养和能量信号,促进细胞增殖和生长。为阐明TOR激酶在植物中的功能,已开展了包括构建突变体在内的多项研究 拟南芥 通过RNA干扰或人工微小RNA实现TOR条件性抑制的转基因品系28,30,31,鉴于TOR基因敲除植株表现出胚胎致死的表型32,33,34,35大多数条件性转基因品系受雌二醇、地塞米松或乙醇诱导型启动子调控,这些品系也可利用该水培系统。

讨论

这种优化的水培结构能够成功实现植物的体外培养。种子在移液枪头平坦表面的固体培养基上萌发良好,相较于将种子浸泡在营养液中的系统,具有显著优势。该系统的一大优点是,在幼苗发育过程中,根系可直接与液体培养基接触,无需进行转移操作。此外,可在较小体积的液体培养基中方便地施加化学处理。系统内湿度保持较高,避免了营养液的蒸发及其补充需求。同时,在幼苗建立阶段可轻松实现均匀的生长与发育;当使用小型容器及处于该发育阶段的幼苗时,无需额外供气11018。为确保系统完全无污染,关键步骤是对所用材料进行彻底灭菌,并在组装过程中进行严格操作。由于部分组件无法在高压灭菌器中灭菌(例如,一次性塑料盒),强烈建议先用70%乙醇清洁,再在使用前进行短时间紫外照射。根据我们的经验,在用胶带密封平坦表面并进行培养基凝固期间使用紫外光照射,也有助于避免细菌和真菌污染。此外,操作时应避免触碰培养基,移动移液枪架时应始终握住其侧面。
