引言

1. O-糖基化反应的操作步骤

  1. 化合物的合成 α/β-12(表1中第12项)
    注:第1至13项 表1 采用类似的方法进行。
    1. 核糖核苷2',3'-二醇的临时保护40
      1. 在10 mL梨形烧瓶(烧瓶1)中,溶解甘露糖基供体 α-9 (28.4 mg, 0.0486 mmol)52,尿苷 10 (7.9 毫克,0.0324 毫摩尔)和 4-(三氟甲基)苯硼酸 11c (9.3 mg,0.0490 mmol)溶于无水吡啶(0.40 mL)。
        注意:建议使用10 mL梨形烧瓶,因为在步骤1.1.3.1中,反应混合物将被转移至含有分子筛粉末的烧瓶2(一个带有隔垫的10 mL双颈圆底烧瓶)。

      2. 在常压下用微波加热分子筛,随后在真空泵减压条件下冷却(重复3次),然后在减压下用热风枪干燥,同时多次用氩气置换空气。
    3. 糖基化
      1. 将步骤1.1.1.4所得残余物溶解于1号烧瓶中的丙腈(0.64 mL)或其他溶剂中,并将该溶液转移至2号烧瓶。

2. β-28的去保护(图2)

  1. 在5 mL小瓶中,加入β-28(25.2 mg,0.0300 mmol)和10 M甲胺甲醇溶液(2.0 mL)58。
  2. 在0 °C下搅拌反应混合物2小时,随后升温至室温。
  3. 继续搅拌13小时后,使用氯仿/甲醇[10/1 (v/v)]体系通过薄层色谱(TLC)监测反应进度[Rf (β-35) = 0.20]。
  4. 使用旋转蒸发仪浓缩反应混合物。
  5. 将所得残余物溶于水(15 mL)中,用二氯甲烷(15 mL,3次)在50 mL分液漏斗中萃取水相。

  7. 通过制备型高效液相色谱(HPLC)纯化剩余残余物[色谱柱:ODS(十八烷基硅烷)色谱柱(20Φ × 250 mm),流动相:含0.1% (v/v) 三氟乙酸的水溶液,流速:8.0 mL/min,检测波长:266 nm,温度:25 °C,保留时间:20 min],得到β-35(7.9 mg,62%,无色无定形固体)59。

3. 环状硼酸酯的核磁共振研究(图3 和 4)

  1. 36的制备与测定
    1. 在10 mL梨形烧瓶中,溶解尿苷 10 (34.3 毫克,0.140 毫摩尔)和 4-(三氟甲基)苯硼酸 11c (40.0 毫克,0.211 毫摩尔)溶于无水吡啶(1.00 毫升)中。

结果
