所有动物实验均按照德黑兰大学医学研究所动物护理和使用委员会(IACUC)进行。通过锐化断头刀刃和采用果断和迅速的叶片运动,尽最大努力尽量减少对大鼠的痛苦和有害影响。

1. 大脑均质化和线粒体隔离

注:所有线粒体隔离试剂均根据西姆斯和安德森14号制备。

  1. 为线粒体隔离准备缓冲液

  2. 大鼠大脑的隔离
    注:根据西姆斯和安德森14,对雄性大鼠进行斩首和脑切除(150-200克)。
    1. 在冰桶中放入 30 mL 烧杯,并在烧杯中加入 10 mL 的冷 IB。
    2. 用小动物断头台将大鼠斩首,并在斩首后1分钟内从头骨中取出大脑,以限制线粒体特性的恶化。
    3. 迅速将大脑转移到含有冷IB的烧杯。
  3. 线粒体的脑均质化和制备
    注:线粒体分数根据西姆斯和安德森14所述的协议被隔离,并作了一些修改,如前15所述。重要的是快速工作,并在整个过程中保持所有工作。
    1. 用30 mL的IB清洗组织2x,转移到装有冷IB的烧杯,用剪刀细切大脑。

    8. 使用慢速加速(45 s 从 0 rpm 到 500 rpm,然后是正常的加速)和减速(无制动器),在 4°C 下以 30,700x g的固定角度转子离心 5 分钟。这将产生两个不同的材料带(图1A,左图)。

    13. 在4°C下在6,900x g下离心10分钟,产生坚紧的颗粒(图1A,右图)。

2. 蛋白质浓度测定

注:蛋白质浓度使用Lowry等人16的方法测量。

  1. 为劳里测定准备解决方案
    1. 制备溶液A:重量为0.4g的NaOH和2克的Na2CO3,溶解在80 mL的DW中,然后将溶液转移到100 mL体积烧瓶中,并通过添加DW将体积增加到100 mL。
    2. 制备溶液B:称量0.1克酸钾和0.05克CuSO4,溶解在8 mL DW中,然后将溶液转移到10 mL体积烧瓶中,通过添加DW将体积增加到10 mL。
      注:两种溶液A和B可在4°C下保持稳定长达6个月。
    3. 制备溶液C:将0.5 mL的叶林溶液加入7.5 mL的DW。
      注:准备溶液C新鲜,并保持远离光,直到使用。
    4. 通过组合 0、20、40、60、100 和 120 μg/mL,分别结合 0、20、40、60、80、100 和 120 μL 1mg/mL BSA,制备最终浓度为 0、20、40、100 和 120 μL 的 BSA 标准,并结合足够的 DW,使溶液达到 1000 μl。
  2. 蛋白质浓度测量
    注: 为了获得更高的精度,在三元中运行此步骤。
    1. 在 96 孔板的每个孔中加入 50 μL 的标准溶液和线粒体均质,然后添加 45 μL 溶液 A。然后,在50°C的温水浴中孵育10分钟。
    2. 加入5μL溶液B,在室温(RT)下在黑暗中孵育10分钟。
    3. 加入150 μL溶液C,在50°C的温水浴中孵育10分钟。
    4. 将板加载到板读取器中,并在 650 nm 处记录标准和线粒体悬浮液的吸光值。然后,使用校准曲线计算线粒体的蛋白质含量。

3. 线粒体膜完整性测定

注:通过测量Triton X-100在膜中断前后的孤立线粒体中的马精酸脱氢酶(MDH)活性,可以确认线粒体膜的完整性。

  1. 用冷IB稀释线粒体均质至1mg/mL,并放入两个1.5 mL管中(通常每管线粒体195 μL)。
  2. 将 5 μL 20% (v/v) Triton X-100 (与 DW 一起稀释) 添加到一个管中(作为最大酶活性的正控制),将 5 μL 的 DW 添加到另一个管(用于控制),然后与搅拌器混合。
  3. 在30°C的温水浴中孵育管子10分钟。
  4. 在16,000 x g和4°C的固定角度转子中,管离心,15分钟。
  5. 使用下一节中描述的标准分光光度测定法,仔细收集生成的上生物,以测定线粒体 MDH 的活性。

4. MDH活动确定

注:MDH活性是按索托卡萨等人17所述的分光光度测量的。

  1. 为MDH活性测定准备解决方案
    1. 准备50 mM Tris-HCl溶液(pH = 7.5):使用Tris-HCl在DW中制备7.9mg/mL溶液,并在25°C时将pH值调整为7.5,使用1.0 M NaOH。
    2. 制备50 mM牛沙酸酯溶液:在Tris-HCl中使用奥洛醋酸盐制备6.6mg/mL溶液。
      注意:此解决方案在解决方案中一次不稳定,应在使用前立即准备好。
    3. 准备10 mM β-NADH溶液:在Tris-HCl中使用β-NADH制备7.81mg/mL溶液。
  2. MDH 活动确定
    注:1.0 mL反应混合物中的最终测定浓度为50 mM Tris-HCl、5 mM牛酸酯和0.1 mM β-NADH。
    1. 将分光光度计设置为 25°C 和 340 nm。移液器 890 μL Tris-HCl 缓冲液、100 μL 牛油酸盐溶液和 10 μL NADH 溶液,用于空白比色剂和 880 μL Tris-HCl 缓冲液、100 μL 的牛油酸溶液和 10 μL 的 NADH 溶液。
    2. 在分光光度计中孵育3⁄4分钟,参考空白。
    3. 然后向样品比色皿中加入10μL线粒体均质(1mg/mL),立即通过反转混合,并记录在340nm下NADH氧化导致的吸收率降低1分钟。

5. 体外β-核糖核酸、牛胰岛素和HEWL纤维蛋白形成

  1. 蛋白质制备
    1. β-核糖核酸:
      注:重组β-核糖核酸的表达和纯化由Hoyer等人18号进行,并进行了一些修改,SDS-PAGE确认了β-核糖核酸的纯度。
      1. 对磷酸盐缓冲盐水(PBS)进行透析纯化+-核素过夜。
      2. 在275nm19时,使用5600M-1 cm-1的消光系数确定蛋白质浓度。
      3. 在1.5 mL管中分量蛋白质,储存在-80°C,直到使用。
    2. 牛胰岛素和HEWL:
      1. 同时提供牛胰岛素和HEWL。
      2. 将每种蛋白质溶解在50 mM甘氨酸缓冲液中(pH值 = 1.6;用HCl调节pH值)。
      3. 在276纳米和280纳米的1.0mg/mL中,使用1.0和2.63的灭绝系数确定牛胰岛素和HEWL浓度,分别为20、21。
  2. 淀粉样颤动诱导
    1. 淀粉样颤动解决方案的准备:
      1. 制备皮洛文T(ThT)缓冲液:重量为0.3克NaH2PO4,将其溶解在80 mL的DW中,将pH调整为6.5,并通过添加DW将体积增加到100 mL。
      2. 制备ThT库存溶液(5 mM):重1.6毫克ThT,将其溶解在1mL的ThT缓冲液中,通过0.22 μm滤纸,在4°C时远离光线,直到使用。
        注:此溶液可在4°C下储存长达4周。
      3. 制备 ThT 溶液 (1 mM):将 200 μL 的 ThT 库存溶液 (5 mM) 添加到 800 μL 的 ThT 缓冲液中。

    2. 体外β-核糖核酸淀粉样纤维蛋白形成:
      1. 将蛋白质溶液(200 μM)和6μL的ThT溶液(1 mM)添加到1.5 mL管中,然后搅拌。
      2. 在37°C的温混剂中孵育管子,在800rpm的恒定搅拌下孵育4天。
      3. 在固定时间间隔后,将孵育样品的等分(10 μL)进行,加入490μL的ThT缓冲液,彻底混合,在RT孵育5分钟。
      4. 将激发和发射狭缝宽度分别设置为 5 nm 和 10 nm,并使用荧光分光光度计在 440 nm 处通过激发测量 ThT 荧光,在 485 nm 处发射 ThT 荧光。
    3. 体外牛胰岛素淀粉样纤维蛋白形成:
      1. 将637μL的蛋白质溶液(250μM)加入1.5 mL管。然后,加入13 μL的ThT溶液(1 mM),然后搅拌。
      2. 在清底96孔板的每个孔中加入含有20μM ThT的蛋白溶液(200μM)等分(200μL),用水晶般清晰的密封胶带密封板。
      3. 将板加载到荧光板读取器中,并在 57°C 下孵育,无搅拌。
      4. 以 30 分钟间隔测量 ThT 荧光,在 440 nm 处进行激发,在 485 nm 处发射,为 12 小时。

    4. 体外HEWL淀粉样纤维蛋白形成:
      1. 在清底96孔板的每口孔中加入含有20μM ThT的含20μM ThT的等分(200μL)蛋白溶液,用水晶般清晰的密封胶带密封板。
      2. 将板加载到荧光板读取器中,并在 57°C 下孵育,无搅拌。
      3. 以 2 小时间隔测量 ThT 荧光,440 nm 的激发和 485 nm 的排放,4 天。
        注:对于所有三种蛋白质,淀粉样纤维蛋白的形成由原子力显微镜确认(图2B)。

6. 用淀粉样纤维、MDH释放测定和ROS测量治疗线粒体

  1. 用淀粉样纤维的孤立线粒体的孵育
    1. 打开离心机和温水浴,分别设置为 4°C 和 30°C。
    2. 使用IF,将线粒体均匀化至1mg/mL的最终浓度。
    3. 准备两个系列1.5 mL管,含有线粒体均质(一个系列用于MDH释放测定,另一个系列用于线粒体ROS测量)。
    4. 向线粒体均质(最终体积 = 200 μL)中加入新鲜或淀粉样蛋白纤维蛋白(最终浓度为5μM、10μM、20μM和25μM;使用PBS或甘氨酸缓冲液作为对照)的二恶客(最终体积=200μL)(见表1,表2),和表3)然后用移液器轻轻搅拌溶液。
      注:对于MDH释放测定,使用Triton X-100(最终浓度为0.5%[v/v])作为最大酶释放的正控制。
    5. 在设置为 30°C 的温水浴中,孵育含有线粒体悬浮液 30 分钟的孵育管。
    6. 测量线粒体 MDH 版本和 ROS 内容,如第 6.2 和 6.3 节所述。
  2. 线粒体 MDH 释放测定
    1. 将孵育的线粒体在16,000 x g下分离15分钟,然后仔细收集产生的上生子,以根据第4节所述对线粒体MDH的活性进行清点。

7. 统计分析
