为这些研究确定一个经过转化的动物模型时,应考虑到被动抗体转移的胎盘类型和机制,以及通过母乳转移抗体。哺乳动物的胎盘主要有三种类型,基于母体-胎儿界面的组织类型和层,包括血细胞(灵长类动物、啮齿动物和兔子)、内皮体(食肉动物)和表皮(马、猪和反胃动物)。血吸盘是最具有侵入性的类型,允许产妇血液供应和胆汁或最外层胎儿膜之间的直接通信。根据足部细胞层的数量,有几种异质胎盘的变化,包括灵长类动物体内的异质胎盘、兔子的异质胎盘,以及在大鼠和小鼠中观察到的止痛胎盘。产妇血液供应和胆汁之间的这种直接接触允许在妊娠期间被动地将抗体转移至胎盘。IgG是唯一显著穿过人类胎盘8的抗体类,而IgA是人类母乳9中主要的Ig类。在科学相关的模型中,只有灵长类动物(包括人类)、兔子和豚鼠在牛奶10、11中将IgG转移到子宫和IgA中。因此,兔子模型包含的因素可与人类中控制IgG的异位转移和IgA的哺乳转移的因素相媲美。

注:所需材料在 材料表中提供。

1. 兔子安定和麻醉

  1. 通过以1毫克/公斤的剂量在肌肉内施用丙丙氨酸(IM),使雌性兔子(性成熟;大约5-30个月大)。根据动物的大小,使用1或3 mL注射器与25 G针。埃帕西拉肌肉是肌肉内注射的首选部位。
    注意:丙丙氨酸也可以在皮下施用,但 IM 是实验室首选的,因为它的作用更快,降低了皮肤病变的发生率。
  2. 等待 10-15 分钟,让丙氨酸生效。
  3. 将相连的鼻锥放在动物鼻子上,用异氟烷对兔子进行麻醉。将蒸发器调整至高达 4% 异氟素,并结合高达 4 升/分钟氧气。兔子对异丙酮有很高的厌恶感,所以在遮蔽动物时需要足够的克制。
  4. 一旦完全麻醉,经pinna、踏板和/或心绞痛反射评估,将眼润滑剂涂在每个眼睛上,以防止眼睛干燥和随后的角膜溃疡。
  5. 持续监测麻醉期间的反应和呼吸,一旦达到足够的麻醉平面,将异氟兰率降低到1-2%。

2. 鼻内免疫

  1. 在动物处理之前准备免疫解决方案。
  2. 如上所述,让兔子吃了。
  3. 一旦实验室成员准备好接种疫苗,兔子处于麻醉的足够平面,关闭异氟素和氧气,并取出鼻锥。
  4. 将兔子放在背上重新任职,并以大约 45° 的角度支撑颈部和头部,以便于管理疫苗的实验室成员访问和可视化两个鼻孔。
  5. 将移液器装载不超过 100μL 的疫苗溶液,并快速管理每个鼻孔中的溶液。移液器应以大约 45° 的角度保持,向鼻通道的中间侧倾斜。
    注意:免疫的目的是解决接触鼻腔粘膜的问题,因此不应将尖端放在鼻腔内,因为这可能会导致粘膜组织的磨擦或刺激,并可能影响鼻腔管理疫苗的免疫原性。疫苗应快速施用,并以同样的方式在另一个纳雷进行。
  6. 在两个鼻腔进行管理后,将兔子在后部重新任职30秒,以尽量减少疫苗溶液的泄漏。
    注:实验室通常一次管理不超过100μL的鼻孔。如果要管理更大的量,最多500μL,疫苗可以在100μL的报价中给予,免疫接种和重复的疫苗额外给药之间有30秒的休息时间,每次接种之间休息30秒,直到疫苗总量交付。
  7. 免疫后,将兔子放在腹腔进行恢复,并密切监测动物,直到它能够维持粪骨再生。

3. 牛奶收集

  1. 如上所述,让哺乳的兔子受宠为一体。
  2. 用酒精拭子/擦拭清洁边缘耳静脉的皮肤。
  3. 使用1mL注射器和25克针头,通过边缘耳静脉静脉注射约1-2IU催产素,诱导牛奶放气。
