2型固有淋巴细胞(ILC2s)参与2型炎症反应,主要在应对蠕虫感染、过敏性疾病、代谢稳态以及组织修复中发挥作用。本研究展示了一种从小鼠鼻黏膜中分离ILC2s并检测其CD226表达水平的方法。

摘要

多年来,关于2型固有淋巴细胞(ILC2s)的研究大量发表,ILC2s被广泛认为参与调控多种病理过程,包括抗蠕虫免疫、组织修复、产热作用以及哮喘和变应性鼻炎(AR)等自身免疫性疾病。ILC2s长期定位于皮肤、肠道、肺部和鼻腔等外周组织;然而,其在鼻黏膜免疫中的确切功能尚缺乏明确信息。CD226是一种活化性共刺激分子,主要表达于自然杀伤(NK)细胞、T细胞和炎性单核细胞。然而,ILC2s是否表达CD226,以及其在ILC2s相关疾病发病机制中是否发挥作用,目前仍不清楚。本文中,我们建立了一种从鼻黏膜分离和鉴定ILC2s的方法,并检测了来自健康小鼠和AR小鼠的ILC2s上CD226的表达情况。本文详细描述了从小鼠鼻黏膜分离和鉴定ILC2s的技术流程,该方法将有助于深入探究鼻黏膜免疫性疾病内部的病理机制。

引言

2型固有淋巴细胞(ILC2s)最初在小鼠的腹膜腔组织中被发现,随后证实其存在于血液及其他外周组织中,例如肺、皮肤和鼻腔1,2,3。作为组织常驻细胞,ILC2s主要通过局部维持和增殖,并通过产生大量2型细胞因子、诱导2型免疫应答,发挥抵御外来有害刺激的第一道防线作用4,5,6。ILC2s还可通过迁移至感染组织发挥作用7,8

与辅助性T细胞2(Th2)类似,ILC2细胞复杂的调控网络确保了其在多种2型炎症性疾病(包括气道过敏性疾病)发展过程中发挥重要作用8,9。在哮喘中,上皮细胞来源的警报素可激活ILC2细胞,后者通过分泌白细胞介素(IL)-4、IL-5和IL-13进一步促进肺部炎症10。临床研究还表明,重度哮喘患者痰液和血液中的ILC2水平显著升高,提示ILC2与哮喘严重程度相关,并可作为哮喘进展的预测指标11

变应性鼻炎(AR)是一种常见的慢性炎症性疾病,每年影响数百万人,而针对该疾病的治疗手段有限12,13。ILC2在AR的病理生理过程中发挥关键作用,无论是在致敏阶段,还是在症状产生和炎症阶段14。在AR患者中,外周血中ILC2的水平已被报道在局部和全身均升高15。然而,ILC2对AR病理生理学及疾病进展的具体作用及其潜在机制仍需进一步研究。

CD226 是一种跨膜糖蛋白,作为共刺激分子发挥作用,主要表达于自然杀伤(NK)细胞、T细胞以及其他炎性单核细胞上16,17。CD226 与其配体(CD155 和/或 CD112)或竞争分子(TIGIT)之间的相互作用使其能够参与多种免疫细胞的生物学功能18。抗原呈递细胞上的配体与细胞毒性淋巴细胞(CTL)表面的 CD226 结合,可同时促进两种细胞的活化;而 CD226 的竞争分子 TIGIT(含 Ig 和 ITIM 结构域的 T 细胞免疫受体)则可进一步抑制 CTL 的活化19,20。一项人类体外(ex vivo)研究表明,T 细胞上的 CD226 和 CD155 通过差异性调控 Th 亚群,调节 Th1/Th17 与 Th2 之间的平衡21。CD226 同样可介导血小板黏附及 NK 细胞的肿瘤杀伤活性22,23。同时,CD226 在多种感染性疾病、自身免疫性疾病及肿瘤发病机制中的作用已被广泛研究18,24,25。目前,CD226 已成为免疫治疗领域的一个新热点。研究发现,细胞外囊泡可通过逆转 NK 细胞上 CD226 的表达,恢复其细胞毒性活性,从而干预肺癌的进展26。最近一项研究通过单细胞 RNA 测序发现,胎儿肠道第 3 组固有淋巴样细胞(ILC3)中存在一个高表达 CD226 的亚群27,提示 CD226 可能在固有淋巴样细胞介导的免疫反应中发挥重要作用。

目前关于2型固有淋巴细胞(ILC2s)在气道炎症中的认知主要基于哮喘研究,然而其在鼻黏膜免疫中的功能尚不清楚。因此,本研究建立了一种从鼻黏膜中分离和鉴定ILC2s的实验方案。该研究聚焦于鼻组织中ILC2s表面CD226的表达情况,并比较健康小鼠与变应性鼻炎(AR)小鼠之间CD226表达的差异。这可能为ILC2介导的局部免疫调节机制提供新的见解,并为开发AR治疗新策略奠定基础。

方案

所有实验均按照《实验动物护理和使用指南》进行。所有实验程序和方案均经中国人民解放军第四军医大学科学研究伦理委员会批准(编号:20211008)。

1. 小鼠雄激素性脱发模型的建立

  1. 在无特定病原体条件下饲养8至12周龄的雄性和雌性野生型(WT)C57BL/6小鼠,并提供标准实验室饲料和饮水。
  2. 在洁净台中将50 µg卵清蛋白(OVA)乳化于含有2 mg氢氧化铝的0.2 mL无菌PBS中以维持无菌状态。于第0、7和14天,通过腹腔注射方式向雄性或雌性小鼠分别注射50 µg乳化后的OVA。
  3. 于第21、22、23、24和25天,在吸入麻醉(2-3%异氟烷,氧气流速0.5 L/min)条件下,将50 µg溶于30 µL无菌PBS中的OVA经鼻内滴注给小鼠(每侧鼻孔15 µL)。
  4. 末次激发后24小时(第26天)处死 小鼠。

2. 从鼻黏膜中分离单个核细胞(MNCs)

  1. 在深度麻醉下通过颈椎脱位处死小鼠。将小鼠头部朝上浸入75%乙醇中5分钟,避免乙醇进入外鼻孔。将小鼠腹部向下置于手术台上。
  2. 剪除门齿。沿头部中线做切口,用剪刀剪开皮肤。
  3. 去除下颌骨,沿腭部末端完整切除整个鼻部组织,并将其放入含有5 mL冰上预冷的磷酸盐缓冲液(PBS)的60 mm培养皿中。使用剪刀和镊子去除附着在骨组织上的软组织和肌肉。
  4. 将小鼠鼻部转移至另一含有5 mL冰上预冷PBS的60 mm培养皿中,用5 mL冰上预冷PBS洗涤骨组织两次。
  5. 将鼻部组织转移至1.5 mL微量离心管中。
  6. 充分研磨后,将鼻部组织转移至含有2 mL预热消化缓冲液(RPMI 1640培养基添加1 mg/mL IV型胶原酶和10 U/mL DNase I)的15 mL离心管中。
  7. 拧紧管盖,将离心管垂直放置于37 °C恒温摇床中,以120–150 rpm持续振荡40分钟。
    注意:为获得最高酶活性,需将消化缓冲液预热至37 °C。
  8. 加入5 mL含10%胎牛血清(FBS)的冰上预冷RPMI 1640培养基,以终止消化过程。
  9. 通过70 µm细胞滤筛过滤,去除固体碎片。
  10. 在500 × g条件下离心5分钟,然后轻轻弃去上清液。
  11. 将沉淀用冰上预冷的RPMI 1640培养基重悬,再次在500 × g条件下离心5分钟,轻轻弃去上清液。
  12. 将细胞沉淀重悬于4 mL 40%密度梯度介质中(160 µL 10× PBS + 1.44 mL密度梯度介质原液 + 2.4 mL RPMI 1640培养基)。
  13. 将巴斯德吸管轻轻插入管底,缓慢加入2.5 mL 80%密度梯度介质(200 µL 10× PBS + 1.8 mL密度梯度介质原液 + 0.5 mL RPMI 1640培养基)。
  14. 将离心机的加速度和减速度设置为低于三档,在室温(RT)下以400 × g离心15分钟。
  15. 在收集界面细胞前,先去除上层杂质,以避免潜在污染。使用移液器小心吸取40%/80%密度梯度介质界面处的单个核细胞(MNCs)层,转移至含有2 mL冰上预冷PBS的15 mL离心管中。用冰上预冷PBS洗涤细胞两次。

3. 用于流式细胞术分析的表面染色

  1. 收集细胞并在 500 x g 4 °C下离心5分钟。将细胞沉淀重悬于染色缓冲液(含2% FBS和0.1% NaN₃的PBS)中3)并在 500 x g 4 °C下离心5分钟。弃去上清液。
    警告:配制染色缓冲液时需谨慎。NaN3 毒性很强,若吞食、吸入或与皮肤接触,可能造成器官损伤。同时,避免其释放到环境中。
  2. 将细胞重悬于每管 80 µL 封闭液中(抗 CD16/32 抗体按 1:100 比例稀释于染色缓冲液中),4 °C 避光孵育 30 分钟。
  3. 不洗涤,直接加入20 µL适当稀释的表面染色抗体混合液(表1).
  4. 在向样本中加入抗体混合物之前,每项检测加入1 µL可固定活力染料520(FVD)。4 °C避光孵育30分钟。设置相应的同型对照抗体和荧光减一(FMO)作为阴性对照。
  5. 用500 µL染色缓冲液重悬细胞,500 x g离心洗涤 g 4 °C下离心5分钟。
  6. 将细胞沉淀重悬于200 µL染色缓冲液中。向染色后的细胞中加入50 µL涡旋混匀的绝对计数微球,混匀后进行流式细胞术(FCM)分析。
    注:FCM 数据使用光谱细胞分析仪获得,并采用流式细胞术(FCM)分析软件进行分析。

结果

建立卵清蛋白(OVA)诱导的小鼠模型以探讨2型固有淋巴细胞(ILC2s)在变应性鼻炎(AR)中的作用。AR小鼠模型的构建基于既往研究,并进行了轻微修改28,29,30,31。在最后一次鼻腔激发后,记录10分钟的视频以统计喷嚏和鼻搔抓的频率。OVA诱导的AR小鼠的过敏症状见图1。AR小鼠组鼻摩擦和喷嚏的频率显著高于对照组。

接下来,我们描述了一种从小鼠鼻黏膜中分离单个核细胞(MNCs)并利用流式细胞术(FCM)对其进行表征的实验步骤。每只小鼠鼻腔可获得约 2-3 × 106 个淋巴细胞。通过FVD染色排除死细胞。分离细胞中2型固有淋巴细胞(ILC2s)的设门策略为:Lin(CD11b、CD11c、CD3 和 B220) CD4...

讨论

ILC2s 与 2 型炎症及炎性疾病密切相关,这一点已通过越来越多的研究得到证实。小鼠模型和人类观察研究共同促进了对其在上呼吸道中功能的深入理解。在哮喘的病理生理过程中,ILC2s 通过胸腺基质淋巴生成素(thymic stromal lymphopoietin)、IL-25 和 IL-33 被激活,这些因子主要由上皮细胞产生。随后,ILC2s 类似 Th2 细胞,分泌 IL-4、IL-5 和 IL-13,从而加剧 2 型炎症32。此外,不同亚群的 ILC2s 还可产生 IL-10 和 IL-1733,34,35。变应性鼻炎(AR)也是一种由 2 型细胞介导的气道过敏性炎症。近年来已有研究证实 ILC2s 参与了 AR 的病理生理过程14。然而,ILC2s 在 AR 中的潜在调控机制仍 largely 未知。
