线虫在促进生态位适应的行为中表现出种内和种间变异,为研究行为可塑性和神经分化的遗传基础提供了极好的范式 28,29,30。Ackley等人31描述了一种稳健的测定方法,用于研究不同Caenorhabditis幼虫的dauer幼虫中的重力趋向性,这可能是一种扩散机制。由于引力行为可能受到其他感觉输入的影响32,Ackley等人31提供了一种简单且具有成本效益的方法来保护动物免受光和电磁场的影响,从而防止这些环境影响对引力行为的已知干扰。该测定将有助于研究线虫感觉统合的分子机制和传播策略的进化。

过去十年单细胞RNA测序技术的发展彻底改变了转录组学分析,使研究人员能够以高分辨率研究多细胞生物体的细胞异质性。在 秀丽隐杆线虫中,不同细胞类型的分离依赖于组织特异性荧光标记物和 FACS33 的表达,这限制了其对转基因工具不可用的其他线虫物种的应用。Woronik等人34 成功地部署了激光显微切割,在没有细胞标记的情况下从蠕虫中分离出组织。他们从秀 丽隐杆线虫 雄性和雌雄同体中分离出尾尖,并使用 CEL-Seq2 进行 RNA-seq 以研究尾部形态发生的机制;然而,值得注意的是,所描述的方法可以很容易地用于不同线虫物种中的其他组织类型,从而允许在组织特异性背景下进行比较转录组分析。

秀丽隐杆线虫和许多寄生线虫含有相似的结构。此外,许多主要的信号系统和发育途径在物种之间是保守的35,36。茎和球茎线虫 Ditylenchus dipsaci 是一种植物寄生线虫,影响全球 500 多种植物物种,造成粮食作物的毁灭性损失。Cammalleri 等人 37 描述了一种在豌豆植株中繁殖大量 D. dipsaci 种群的有效方法。作者还提供了一个详细的方案,用于识别可能通过损害蠕虫的流动性而作为杀线虫剂的小分子候选者。实验室培养的便利性加上最近可用的基因组序列38 使 D. dipsaci 成为与 C. elegans 进行比较研究的有用模型,从而有可能发现其生物学的新方面并制定有效的生物防治策略。
