当前 JoVE 方法集合的目的是提供一套公正且多样化的方案,以便全面研究心律失常发生的各个方面。

钙稳态改变是心律失常发生的标志之一,但研究仍然具有挑战性,因为需要考虑多个方面,每个方面都需要特定的技术来研究 1,2。在这个方法集合中,Tomsits 等人提供了一种协议,允许从同一只小鼠中分离心房和心室心肌细胞,并同时研究 L 型钙电流和钙瞬变3。因此,该协议使研究人员能够在全球范围内更全面地研究钙紊乱,同时减少动物数量,这代表了一项重大的科学和伦理改进。

除心肌细胞外,其他细胞类型(例如成纤维细胞或巨噬细胞)也在电生理学中发挥重要作用6,7,8。为了研究传导系统中的细胞类型特异性影响,Xia等人提供了一个分步指南,以识别小鼠的窦房结(SAN)和房室结(AVN)9,10,然后进行显微解剖和荧光激活细胞分选(FACS)以分离和研究巨噬细胞9或通过全安装免疫荧光成像。这允许鉴定不同的细胞类型,评估三维结构,并研究SAN / AVN10内的细胞类型特异性蛋白质表达。

此外,可以在Kumar等人提出的全安装制剂上进行多电极阵列(MEA)记录11。这种方法是对当前使用的技术的一个很好的补充,因为它相对快速和简单,并且还提供了卓越的空间分辨率。它允许药物干预,并且可以很容易地应用于心脏的其他特定区域(例如,AVN)。

自主神经系统在电生理学中起着至关重要的作用 12,13,14,并且存在治疗策略,例如交感神经切除术 15161718 或心神经节消融术19。然而,机制基础在很大程度上仍然未知,特别是因为自主神经系统的解剖结构在小鼠中不容易接近。在本期中,Scherschel 等人提供了一种协议,允许识别和解剖小鼠的星状神经节,以研究各种细胞类型中的基因和蛋白质表达20。这将允许研究自主神经重塑的新方面。

在细胞水平上研究电生理学对于研究基本机制至关重要。然而,仍然需要在动物模型中进行体内研究来验证细胞发现并确认已识别靶标结构的(病理)生理相关性21。小鼠的应用最为广泛,但转基因小鼠的生成尤其复杂。在本期中,Shimura等人提出了一种使用CRISPR-Cas9基因编辑的方案,专门研究截短蛋白的复杂调控作用及其在体内的影响。为此,他们生成了一个转基因小鼠模型,该模型在编码连接蛋白-43的GJA1基因中存在单点突变。这导致全长 Cx43 的正则表达,但截短的 Cx4322 的表达减少。

体内研究的主要局限性之一是需要在实验期间麻醉动物,这可能会影响各种电生理参数,因此可能不代表“真实”情况23,24。另一方面,遥测设备允许在数周至数月的长时间内研究清醒和自由移动的小鼠25,26。Rötzer 等人描述了如何植入遥测设备以同时记录小鼠的心率和血压,这特别允许研究自主神经系统和压力反射敏感性27

尽管遥测设备是非常强大的工具,但仍然存在一个主要挑战:如何分析这些大型数据集。为了解决这个问题,Tomsits 等人开发了一种协议,以标准化的半自动方式分析遥测心电图记录28。按照他们的方案,研究人员可以获得基本的心电图参数(例如,心率或 PR 间期)并筛查单独定义的心律失常(例如,停顿、心动过缓/心动过速)。重要的是,他们还讨论了记录和分析遥测心电图的挑战和陷阱,包括一些提高信号质量的指导。

新开发的医疗设备,如人工瓣膜,在应用于人类患者之前,在动物模型中进行了全面测试29。然而,对健康动物的研究可能无法完全转化为人类患者,尤其是那些以前接受过手术的患者30,31。为了克服这些限制,Grab等人提供了一种用于3D打印个体患者特定心脏模型的方案32。他们的模型具有弹性,类似于生物力学特性以及与人类患者相似的血流动力学特征;这些模型允许对医生进行教学、测试新的介入程序或设备以及规划个体化治疗策略,特别是对于解剖结构复杂的患者。
