摘要

引言

昆虫迁徙在全球昆虫种群动态分布中起着至关重要的作用,例如棉铃虫(Helicoverpa armigera)、东方黏虫(Mythimna separate)、芋双线枯叶蛾(Spodoptera litura)和甜菜夜蛾(Spodoptera exigua)等害虫已被报道在中国造成严重危害1,2,3,4。迁徙性害虫的长距离飞行、季节性移动、高繁殖力以及生态因素给这些害虫的预测、预报和防控带来了巨大困难5。为了揭示迁徙性害虫如何根据气候变化或周期调整其适应能力与行为变化,必须开展害虫迁徙监测6。为了维持其生长、繁殖和生存,昆虫在进化过程中获得了连续的适应能力;这一系列适应性生命特征导致生殖系统发生诸多变化,例如迁徙策略可调控长距离迁徙过程中的卵巢发育。

卵巢发育在迁徙性害虫中较为常见,这会影响其种群的增长7因此,卵巢发育长期以来一直是迁徙性害虫研究的热点话题。一系列研究已提出了多种卵巢发育指标和分类策略。迄今为止,已有多种方法被用于分析卵巢发育,例如, Loxostege sticticalis - 草地螟卵巢发育,包括初期羽化阶段、早期产卵期、产卵期和产卵末期8一些研究人员根据迁徙期间卵黄颜色的发育情况对卵巢等级进行划分 鳞翅目昆虫 害虫,例如 S. exigua - 甜菜夜蛾 Pseudaletia unipuncta 真正的行军虫, 和 Cnaphalocrocis medinalis- 水稻卷叶螟等9,10,11,12在以往的研究中,棉铃虫和水稻纵卷叶螟等害虫的卵巢发育水平被划分为五个阶段:卵黄沉积期、卵粒成熟期、成熟待产期、产卵高峰期和产卵末期13,14欧洲玉米螟的卵巢发育分为六个发育阶段:卵黄沉积期、卵子成熟期、产卵前期、产卵高峰期和产卵末期15.

此外,同一属的昆虫具有不同的发育阶段,例如草地贪夜蛾(Spodoptera frugiperda)的卵巢发育可分为四个等级:卵黄沉积期、成熟待产期、产卵高峰期和产卵末期16。另一方面,甜菜夜蛾(Spodoptera exigua)的卵巢发育则分为五个等级:透明期、卵黄发生期、卵成熟期、排卵期和产卵后期17。

以往的研究仅能依据卵黄的颜色成熟度、产卵行为和卵的发育情况,将发育阶段划分为从单个到多个卵巢发育水平,但无法根据生殖系统的解剖结构进行分类。基于形态发生解剖学的卵巢发育研究较少。本文设计了一种解剖方法,通过两种卵巢组织类型来预测种群中的迁徙型雌性个体,进而基于卵巢发育阶段和交配囊的解剖形态发生特征阐明其生殖动态,为区分迁徙型野生雌性个体提供直接证据。

一些研究发现,迁徙性 夜蛾科 昆虫物种经常被探照灯捕获18大多数迁徙性动物的卵巢 夜蛾科 昆虫物种在迁徙初期处于发育的早期阶段,随着迁徙进程的推进,其卵巢发育水平逐渐升高。本研究描述了用于评估卵巢发育等级的解剖方法,以研究通过探照灯捕获的不同雌性害虫种群的两种生殖组织。该方法不仅有助于深入理解昆虫迁徙动态,还为昆虫分类、昆虫生理学研究、害虫预测及雌性害虫种群的预报提供了便利。

方案

注意:在捕获野生迁徙昆虫之前,请注意安全措施,建议佩戴安全装备(手套、长袖衬衫和护目镜)。不使用时应关闭诱捕器,以避免其他安全隐患以及灯光过热。解剖前遵循安全操作规程非常重要,例如佩戴手套、护目镜和实验服,以防止接触体液和化学物质。

1. 迁飞昆虫的捕获

  1. 使用探照灯开始本实验方案以捕捉昆虫。本方案中,测试昆虫来源为中国山东省济南市济阳区(36.977088° N, 116.982747° E)。
  2. 使用由不锈钢制成的探照灯主体,其外壳为矩形结构,并配备 GT75 型卤素灯头,功率为 1000 W。将灯头置于中央作为光源。
  3. 在灯体内部下方放置一个漏斗状昆虫收集通道,在底部放置一个直径为 5 cm 的昆虫收集盒,随后连接一个 60 目的昆虫收集网袋(0.5 m × 0.5 m),用于收集被灯光诱捕的昆虫。已知灯光的投影高度距地面约 500 m。
  4. 本方案重点研究野生型雌虫的迁飞动态及其卵巢发育情况;因此需采集不同种类的昆虫(本研究采集时间为 2021 年至 2022 年的 4 月至 8 月)。避免采集体型较小或受伤的昆虫,应选择体型相近的大型害虫用于本实验。

2. 昆虫的制备

  1. 将从网袋(0.5 m)中采集的所有昆虫转移至网笼(30 cm × 30 cm)中,然后提供一个含有10%灭菌蜂蜜水溶液的培养皿(喂食为可选步骤)。将网笼置于27 ± 2 °C、相对湿度65% ± 12%的环境中,并在黑暗条件下保持8–12小时。
  2. 选取当天飞入网笼的野生型雌虫,小心地将其分别转移至单独的玻璃小瓶中,并用棉塞封口。避免直接用手操作,以防因压力过大而损伤或伤害害虫。
    ​注意:所有野生型雌虫均在夜间捕获,解剖操作在白天进行。因此,每项实验均在一天内完成。

3. 昆虫麻痹方法的准备(图1)

  1. 将选定的雌性害虫单独放入中间的果蝇小管中,使用CO2气体通过吹枪针头使其产生轻度麻痹。为确认害虫是否已麻痹,可用软毛刷轻轻触碰或推动害虫。若对轻微刺激无反应且无法移动,则表明麻痹成功。
    ​注意:低温(-20 °C)也可作为麻痹昆虫的替代方法。

4. 昆虫解剖

  1. 将刚麻痹的雌虫放入含有无水乙醇(10 mL)的解剖培养皿中。为避免翅上的鳞毛和粉末在解剖过程中产生干扰,需先用无水乙醇浸透活体或麻痹的昆虫,再用清水冲洗。
  2. 使用两对镊子,从胸部与腹部连接处分离背翅。
  3. 将腹部转移至一个新的一次性培养皿中,加入适量清水(深度2–5 mm),用解剖镊子从尖端口部向尾部沿背腹中线轻轻剥离腹部外骨骼。另一侧重复相同操作,然后将其放入清水中以分散完整的组织。
  4. 用镊子小心剥离表皮脂肪组织,并轻柔牵拉并释放卵巢。
  5. 用解剖镊子轻轻去除卵巢周围的脂肪颗粒及其他器官。通常害虫的卵巢多向腹部两侧内折叠,操作时应尽量在液体环境中展开卵巢,从中间缓慢剥离交配囊,并挑出附着在卵巢管上的脂肪颗粒。
  6. 从卵巢和交配囊的垂直后端轻轻夹住,小心向下展开。为避免展开过程中造成损伤,可将卵巢转移至含有清水的新培养皿或洁净培养皿中。夹住卵巢顶端,向内侧轻轻展开;此步骤需谨慎操作,避免损伤卵管。

5. 卵巢组织解剖学数据分析

  1. 在此步骤中,根据每只昆虫卵的颜色和大小评估其卵的发育情况,以判断其成熟度。然后,根据卵的发育情况判断卵巢等级。
    注意:不同卵巢发育阶段的划分主要依据产卵前或卵黄沉淀水平的卵发育阶段。昆虫卵粒发育可进一步细分为多个阶段,以便更清晰地描述,例如卵黄出现期、卵黄成熟期和卵黄退化期。卵粒的成熟度取决于卵的饱满程度、颜色和大小来判断。
  2. 解剖后,务必分离雌性交配囊组织与完整的卵巢,并观察其形态以区分物种,因为大多数交配囊的解剖结构在不同物种间存在差异。因此,可利用交配囊特征进行物种鉴别。
  3. 在此步骤中,评估卵巢解剖结构,并分析卵巢发育等级。将卵巢组织分为五个等级(1级至5级)。
    1. 根据以下变化和结构特征对组织进行分类:一级(1级)为发育早期(乳白色透明),腹部饱满,组织柔软,脂肪体蓬松,颜色较浅,不易剥离;二级(2级)为卵黄沉积阶段,若需进一步区分,可依据卵巢管变长变粗的情况对卵巢单独分级;三级(3级)脂肪体减少,仅有少量颗粒附着于卵巢上;四级(4级)组织弹性降低,易断裂,部分卵可能已进入输卵管中部;五级(5级)易于识别,脂肪体极少或缺失,卵巢萎缩且脆弱。
  4. 根据实验需要,使用数码相机拍摄图像。

结果

卵的发育
采用上述方案分析卵巢中卵的发育过程。为此,首先将卵总体分为四个阶段,以区分不同物种(如棉铃虫、黏虫、芋头斜纹夜蛾和甜菜夜蛾)中卵发育的早期与成熟阶段。此处观察到的是羽化早期(乳白色透明阶段)。图2A显示卵巢尚未开始发育,卵巢管呈丝状,具有良好的弹性。由于卵体细小,在日光下无法观察到已形成的卵粒,但通过拉伸卵巢管并透光观察,可见乳白色斑点。

卵巢发育(卵黄沉积阶段)包含大量略显未成熟、呈乳白色的卵粒,卵粒之间紧密接触(图2B)。图2C展示了待产成熟阶段(成熟期),可见卵粒呈珠状,完全成熟且色泽鲜黄。图2D展示了卵黄消退期(产卵末期),此时卵巢管脆弱,大部分活跃的卵粒已排出,仅残留少量或无卵粒,可能尚存不活跃的卵粒。

讨论
