利用酶标仪通过钙流测定法表征蛋白酶激活受体调节剂

摘要

已开发出一种改良的内皮细胞钙流测定实验方案,用于鉴定蛋白酶激活受体(PARs)的配体。新方案将总检测时间缩短了90至120分钟,并可获得可重复的浓度-反应曲线。

摘要

利用细胞内荧光钙结合染料以及可同时检测多达1536个孔荧光发射的成像仪器,可高通量地快速监测细胞内钙离子浓度的变化。然而,这些仪器成本高昂,且相较于广泛使用的、逐孔扫描的酶标仪,维护难度更高。本文介绍了一种优化的酶标仪检测方法,适用于内皮细胞系(EA.hy926),通过钙结合染料Fluo-4检测蛋白酶激活受体(PAR)介导的Gαq信号通路激活及其后续的钙动员过程。该方法已用于表征多种PAR配体,包括Dockendorff实验室发现的靶向PAR1的变构抗炎“parmodulin”配体。本方案无需使用自动化液体处理系统,可在96孔板中实现对PAR配体的中等通量筛选,且应可推广应用于其他引发钙动员的受体研究。

引言

蛋白酶激活受体(PARs)1,2,3 是一类A类G蛋白偶联受体(GPCRs)的亚家族,表达于多种细胞类型,包括血小板和内皮细胞4,5,6,7与大多数G蛋白偶联受体(GPCRs)不同,蛋白酶激活受体(PARs)具有独特的分子内激活机制。大多数GPCRs由可溶性配体与特定的结合口袋相互作用而被激活,而PARs则通过其N端胞外域的蛋白水解切割被激活。 N-末端,从而形成一个新的锚定配体,该配体可与细胞表面的胞外环2结构域相互作用6,8,9这种相互作用会激活受体,并可启动多条信号通路,促进炎症和血小板活化等效应4,10,11,12不同的蛋白酶可通过在PARs上特异位点的切割来激活它们 N-末端,暴露出不同的拴系配体(TL),这些配体可稳定受体构象,从而启动不同的信号通路9,13,14,15例如,在该亚家族中研究最为深入的成员PAR1中,凝血酶对其的切割作用可支持多种生物学过程,包括血小板活化和白细胞向内皮的募集;但当该受体过度表达或过度激活时,则可能引发有害效应。4,16,17,18,19,20,21相反,活化蛋白C(aPC)的切割可促进抗炎效应并维持内皮屏障的完整性。15,22,23,24,25,26,27,28,29PARs 也可通过跨分子方式被 TLs 的肽类似物激活13,30,31这些肽段通常用于替代靶向PAR的蛋白酶,以检测PAR的抑制(调节)作用,本方案中亦采用这些肽段。

多种疾病与病理性PAR1信号传导相关,包括脓毒症22,32、心血管疾病33,34,35,36,37,38、肾脏疾病39,40,41,42、镰状细胞病43、纤维化44、骨质疏松症和骨关节炎45,46、神经退行性疾病47,48,49,50,51以及癌症52,53,54,55,56,57,58,59。自20世纪90年代以来,PAR1拮抗剂已被研究作为治疗心血管疾病的抗血小板药物,而与该受体相关的疾病谱不断扩展,迫切需要鉴定新型配体,用作生物学探针(工具化合物)或潜在治疗剂。目前,仅有一种经美国食品药品监督管理局(FDA)批准的PAR1拮抗剂——沃拉帕沙(vorapaxar),用于高危患者的冠状动脉疾病治疗34,36,37,60。另一种PAR1拮抗剂——肽类类似物PZ-128,已在心脏导管插入术患者中成功完成II期临床试验,用于预防血栓形成38。Dockendorff研究团队专注于一类独立的小分子药物化学与药理学研究,即被称为parmodulin的PAR1配体61,62。与已报道的PAR1拮抗剂(如沃拉帕沙)不同,parmodulin是PAR1的别构性、偏向性调节剂,可选择性阻断Gαq通路,同时促进类似于活化蛋白C(aPC)的细胞保护效应。与沃拉帕沙等强效正位PAR1拮抗剂不同,已发表的parmodulin还具有可逆性63,64,65。

最初,Flaumenhaft 及其同事鉴定出 parmodulins 抑制血小板中P-选择素表达或颗粒分泌的能力61,66然而,需要一种替代方法来研究parmodulins对内皮细胞的作用。一种常见的监测GPCR相关信号转导的方法是检测细胞内Ca2+ 动员,一种重要的第二信使,可通过合适的细胞内钙离子结合染料进行检测67,68大量证据表明,PAR1诱导的钙动员是通过激活G蛋白实现的αq69,70. 一旦被其锚定配体(或合适的外源性配体)激活,PAR1 会发生构象改变,导致与 G 蛋白结合的鸟苷二磷酸(GDP)αq 由三磷酸鸟苷(GTP)替换的亚基68. Gαq 亚基随后激活磷脂酶Cβ(PLC-β),后者催化磷脂酰肌醇4,5-二磷酸(PIP2)水解,生成1,4,5-肌醇三磷酸(IP3)和二酰甘油(DAG)。最后,IP3 与IP结合3-敏感的 Ca2+ 内质网膜上的通道,允许 Ca2+ 释放到细胞质中,在此与 Ca 结合2+依赖性荧光染料(如 Fluo-4),添加至细胞中71. 这一过程在数秒内发生,可提高 Ca 浓度2+ 100 倍,导致钙结合染料的量发生显著变化,并产生强烈的荧光信号。

2018年,Dockendorff课题组报道了一种中等通量的Ca2+ 可用于鉴定G蛋白偶联受体拮抗剂的迁移实验αq PAR1信号通路72所使用的检测方法 EA.hy92673一种杂交人源内皮细胞系,可用于多次传代而PAR1表达无明显变化,且已建立用于 体外 细胞保护作用的测定 

原始方案使用96孔板中的EA.hy926细胞,并用Fluo-4/AM染料进行负载,该染料因其在488 nm处具有强烈的荧光信号以及高细胞通透性而被选用。染料进入细胞后,使用自动化的8通道液体处理仪进行了耗时较长的洗涤步骤(无法获得更快速的液体处理方法,例如96通道洗板机)。与未采用精细、自动化机器人换液的方法相比,该检测方法的可重复性更优。随后将拮抗剂与细胞孵育,通过依次加入选择性激动剂(每次处理16个孔)激活PAR1,通过检测钙离子动员及染料结合所引起的荧光变化来确定其活性。

尽管该实验方案可用于检测PAR1介导的钙动员,但其受限于每块96孔板检测所需的时间。实验时间过长不仅限制了每日可筛选的化合物数量,而且随着时间推移,染料会逐渐外排,导致基础荧光信号升高,从而缩小检测窗口。造成实验耗时较长的原因之一是使用8通道液体处理仪进行洗板,这使每次实验额外增加30分钟以上。此外,所需移液吸头也因供应链问题而难以获取。本文报道了一种更新的PAR介导钙动员检测方案,该方案无需使用液体处理仪,因而可实现更高通量的检测。本方案也适用于检测其他可引发细胞内钙动员的G蛋白偶联受体(GPCR)信号通路。 这种改进后的酶标仪检测方案 特别适合资源有限、无法配备昂贵细胞成像设备但需要快速筛选大量化合物的学术实验室和小型工业实验室。关于使用酶标成像仪进行钙动员检测的示例,可参见Caers等人的研究74。

方案

以下方案的步骤1和步骤2中进行的所有培养基更换/添加操作均须在无菌操作台内完成。除非另有说明,所有在无菌操作台内使用的塑料器皿必须购买时已灭菌并密封,或通过高压灭菌器适当灭菌。

1. EA.hy926 细胞系的启动

  1. 获取 EA.hy926 细胞。
  2. 将细胞冻存管置于液氮罐的气相中保存。
  3. 通过将 DMEM、FBS 和青霉素-链霉素溶液(具体信息见材料表)在 37 °C 水浴中预热 15–30 分钟,制备完全 DMEM 培养基。向 500 mL DMEM 中加入 50 mL FBS 和 1 mL 青霉素-链霉素溶液,轻轻颠倒混匀。
    注意:剧烈混匀会导致产生过多气泡。
  4. 从液氮中取出一支细胞冻存管,在 37 °C 水浴中温育 5 分钟。
  5. 用酒精喷洒细胞冻存管和完全 DMEM 培养基瓶进行消毒,然后转移至无菌操作台。
  6. 使用 1,000 µL 移液器小心地将细胞转移至一支 15 mL 无菌离心管中。向冻存管中加入 1 mL DMEM 漂洗,并将液体一并转移至离心管中。
  7. 使用离心机(150 × g,5 分钟,22 °C)离心形成细胞沉淀。用抽吸法去除离心管中的上清液,注意不要扰动细胞沉淀。
  8. 向离心管中加入 3 mL 完全 DMEM 培养基,使用 1,000 µL 移液器轻轻吹打重悬细胞,使沉淀充分分散。
    注意:避免剧烈吹打产生气泡,以免激活或损伤细胞。
  9. 使用 20 µL 或 200 µL 吸头的移液器,取 10 µL 细胞悬液与 10 µL 台盼蓝在微量离心管中混合。取 10 µL 细胞悬液与台盼蓝的混合液加入血细胞计数板,进行细胞计数。
    注意:使用 0.1–10 µL 吸头可能导致细胞剪切损伤。处理细胞悬液时不应使用此类吸头。
  10. 使用 10 或 25 mL 移液管向细胞悬液中添加完全 DMEM 培养基,调整细胞密度至 66,000 个细胞/mL。将 15 mL 细胞悬液接种至 T-75 培养瓶中。通过南北、东西方向轻晃培养瓶,确保细胞悬液均匀分布。
  11. 将细胞置于培养箱中培养,直至达到汇合状态(通常为 4–6 天),通过显微镜观察评估。此时可将细胞用于实验(从第 2 节开始),或以推荐接种密度(每支 T-75 培养瓶中加入 1,000,000 个细胞于 15 mL 完全培养基中)接种至新培养瓶进行扩增,以制备冻存细胞。
    注意:细胞在培养瓶中可连续培养长达 3 周,其形态、健康状态及检测性能无明显变化。应每隔 3–4 天更换一次完全培养基。

2. 将 EA.hy926 细胞接种至 96 孔板

注意:一个汇合的 T-75 培养瓶中的细胞可用于制备两到三个检测板,也可选择性扩增至多达 15 个新的 T-75 培养瓶。在正常工作日内,使用第 4 和第 5 部分的方案可筛选至少五个检测板。以下说明描述的是制备和测试一个检测板的操作步骤,若需制备更多检测板,可重复步骤 2.2、2.7、2.12 和 3.2 以完成所需数量的检测板制备。通常情况下,每天制备四个检测板,用于检测最多 16 种化合物的浓度-反应关系,这需要两个长满细胞的 T-75 培养瓶。

  1. 将DMEM完全培养基置于37 °C水浴中预热15-30分钟。用酒精喷洒DMEM完全培养基瓶身进行消毒,然后将其转移至无菌操作台。
  2. 向带盖的黑色侧壁、透明底96孔板的每个孔中加入100 µL灭菌的0.4%明胶溶液。将该板连同明胶溶液一起放入培养箱(37 °C,5% CO2)中孵育30分钟。
  3. 使用倒置显微镜确认T-75培养瓶中的细胞融合度达到80-100%。通过抽吸法移除T-75培养瓶中的DMEM完全培养基。
  4. 用10 mL PBS清洗培养瓶中的细胞,并轻轻将培养瓶从北向南、从东向西移动。通过抽吸法移除PBS。
  5. 向培养瓶中加入5 mL细胞解离液,并将培养瓶 放入培养箱(37 °C,5% CO2)中孵育约12分钟。6分钟后轻敲培养瓶以促进细胞解离。
    注意:使用所述细胞解离试剂孵育细胞超过15分钟会导致细胞开始聚集,从而增加准确计数的难度,并可能干扰后续检测。
  6. 向培养瓶中加入5 mL预热的DMEM完全培养基,并轻轻混匀以中和细胞解离液/细胞悬液。使用10 mL移液管将细胞悬液转移至无菌50 mL离心管中。通过离心(150 × g,5分钟,22 °C)形成细胞沉淀。
  7. 在离心形成沉淀的同时,使用连接真空泵的巴斯德移液管抽吸去除96孔板中的明胶溶液。从去除明胶起至准备加样前,保持移除板盖状态。
    注意:可选择性地使用多通道移液器或连接真空泵的无菌分流管。
  8. 使用连接真空泵的巴斯德移液管或移液管抽吸去除离心管中的上清液;注意不要扰动细胞沉淀。
  9. 使用5或10 mL移液管向离心管中加入新鲜DMEM完全培养基。通过使用1,000 µL移液器或5 mL移液管反复吹打,轻轻打散细胞沉淀。
    注意:通常使用8 mL DMEM,但可加入更大体积以便于细胞计数。
  10. 使用20 µL或200 µL吸头的移液器,在微量离心管中将10 µL细胞悬液与10 µL台盼蓝混合。取10 µL细胞悬液与台盼蓝的混合液加入血细胞计数板,进行细胞计数。
  11. 使用10或25 mL移液管向细胞悬液中加入DMEM完全培养基,调整细胞密度至600,000个细胞/mL。此浓度可使96孔板每孔最终细胞密度为60,000个细胞/孔(经实验确定为最佳密度)。
    注意:一次成功的常规细胞培养通常可获得12,000,000至15,000,000个细胞,足以接种两到三个96孔板。
  12. 轻轻颠倒离心管以混匀细胞悬液,然后将其加入50 mL多通道移液器储液槽中。每30秒轻轻左右摇动储液槽以保持细胞均匀悬浮,使用多通道移液器向每个孔加入100 µL细胞悬液。盖上透明板盖后,在无菌操作台表面将板从北向南、从东向西轻轻滑动振荡,以确保细胞分布均匀。将板放入培养箱(37 °C,5% CO2)中孵育16-24小时。
    注意:可使用额外的细胞悬液制备更多培养板,或重复步骤1.10-1.11以继续传代细胞系。

3. 钙流测定实验准备

  1. 准备检测试剂。
    1. 通过将36 mg丙磺舒加入微量离心管中,并溶于0.6 M NaOH(500 µL)中,配制丙磺舒溶液(250 mM,水溶液)。涡旋混匀溶液。
      注意:丙磺舒通过抑制有机阴离子转运体,提高染料在细胞内的滞留75,76,77。
    2. 配制HBSS-HEPES检测缓冲液:将HEPES(1.19 g)加入500 mL无钙、无镁、无酚红的HBSS瓶中(形成10 mM HEPES溶液)。向缓冲液中补充MgCl2(1 M水溶液,500 µL)和CaCl2(1 M水溶液,500 µL)。混匀溶液,不使用时置于5 °C冰箱中保存。
      1. 每块96孔板,取50 mL HBSS-HEPES检测缓冲液加入50 mL离心管中,加入500 µL丙磺舒溶液,充分混匀。使用前将缓冲液恢复至室温。
    3. 通过将Pluronic F-127(20 mg)加入微量离心管或HPLC 小瓶中,并用200 µL DMSO溶解,配制10% Pluronic F-127的DMSO溶液。
      注意:该溶液可在室温下保存,适用于约30块板。Pluronic F-127在室温下溶解缓慢,可轻微加热以促进溶解。
    4. 首先将24 µL DMSO加入含Fluo-4/AM(50 µg)的小瓶中以溶解染料,配制染料负载缓冲液。在用铝箔包裹的15 mL离心管中,加入6 mL HBSS-HEPES检测缓冲液,并补充6 µL 10% Pluronic F-127和6 µL Fluo-4/AM溶液。短暂涡旋混匀,避光静置于室温10分钟。
      注意:为防止Fluo-4光漂白,应用铝箔包裹含染料溶液的离心管及检测板盖,避免光照。
  2. 准备检测板。
    1. 从培养箱中取出检测板,使用倒置显微镜确认细胞融合度。通过轻弹将完全DMEM培养基弃入水槽,并用纸巾轻拭板面。
      注意:将板水平持握并轻弹,然后旋转180°重复操作,即可充分去除培养基。
    2. 将含丙磺舒的HBSS-HEPES检测缓冲液加入50 mL多通道移液器储液槽中。
      注意:该溶液将用于后续洗涤步骤,应暂时放置并用铝箔覆盖,直至使用。
    3. 使用多通道移液器向每孔加入100 µL 含丙磺舒的HBSS-HEPES检测缓冲液,使细胞在丙磺舒中孵育5分钟。以与步骤3.2.1相同的方式轻弹去除HBSS-HEPES检测缓冲液。
    4. 将染料负载缓冲液加入另一个50 mL多通道移液器储液槽中。使用多通道移液器向检测板每孔加入50 µL 染料负载缓冲液。将板在37 °C、5% CO2条件下孵育45分钟。
    5. 在板孵育期间,配制激动剂/拮抗剂溶液。
      1. 拮抗剂溶液以31.6 mM浓度保存于DMSO中。在PCR管中,将2 µL原液与38 µL 0.1% BSA/水混合,得到1.58 mM溶液。取该溶液2 µL加入检测板每孔,终浓度为31.6 µM。将4 µL 1.58 mM拮抗剂溶液与36 µL 5% DMSO/水(不加BSA)混合,并通过连续稀释法配制其余浓度。
      2. 在HBSS-HEPES检测缓冲液中配制108.3 µM(16.7倍)的TFLLRN-NH2溶液,当每孔加入6 µL时,终浓度为6.5 µM。例如,将2 mg TFLLRN-NH2溶于5.244 mL HBSS-HEPES检测缓冲液中,得到0.5 mM储备液。取1.083 mL该溶液与3.917 mL HBSS-HEPES检测缓冲液混合,得到108.3 µM溶液。
        注意:不使用时,TFLLRN-NH2溶液应保存于-20 °C冰箱中。
    6. 从培养箱中取出板,使用荧光显微镜在适当条件下观察染料摄取情况(通常使用绿色荧光蛋白通道,参见图1以了解Fluo-4成功加载至细胞的示例)。以与步骤3.2.1相同的方式轻弹去除染料负载缓冲液。
    7. 每孔用50 µL 含丙磺舒的HBSS-HEPES检测缓冲液(来自步骤3.2.3)洗涤细胞两次(通过轻弹将液体弃入水槽)。
    8. 向每孔加入92 µL HBSS-HEPES检测缓冲液,再次使用荧光显微镜观察细胞,确保过量染料已完全去除,且细胞仍贴壁良好。
    9. 使用多通道移液器向相应孔中加入2 µL拮抗剂溶液或溶剂(参见表1中的代表性板图)。

4. 进行钙流测定实验

  1. 设置酶标仪参数如下:温度:37 °C;激发波长:485 nm;发射波长:525 nm;检测高度:7.8 mm(已优化);闪光次数:100;重复次数:20。
  2. 将酶标板置于酶标仪中,在37 °C下孵育15分钟。检测第1和第2列的背景荧光(每孔扫描5次)。
  3. 将酶标板从酶标仪中取出,使用多通道移液器从8联PCR管条中快速向第1和第2列的每孔加入6 µL激动剂溶液。
    注意:此步骤中,我们使用带有0.1-10 µL吸头的电子8通道移液器。
  4. 在细胞发生钙离子动员的过程中,检测荧光信号的变化。根据上述设置对每孔进行20次扫描。
    注意:对两列中的每孔各扫描20次约需5分钟(即每孔两次扫描间隔约15秒)。
  5. 以两列为一组,依次对第3至第12列重复执行步骤4.2–4.4。

5. 数据分析

  1. 将检测数据按每组两列分别导出为独立的电子表格。
  2. 使用函数 AVG(第一行扫描:最后一行扫描) 计算基础荧光读数的平均值。对两列中的每个孔均执行此操作。
  3. 使用函数 MAX(第一行扫描:最后一行扫描) 确定加入激动剂后的最大荧光值。
  4. 通过从最大荧光值中减去基础荧光值,计算荧光变化量。
  5. 从同一列中每个孔的荧光变化量中减去阴性对照(仅加入溶媒而无激动剂)所计算出的荧光变化量。
  6. 将每个孔的荧光变化量除以溶媒(0.1% DMSO/水)+ 6.5 µM TFLLRN-NH2 孔的荧光变化量,得到相对(归一化)荧光变化量。
  7. 将非对照组数值以百分比形式复制到统计与绘图软件程序中。若使用所引用的软件,请选择 XY 表格选项,并将 数值 设为 X 轴,并列子列中的重复值 设为 Y 轴。
  8. 使用该程序根据数据绘制浓度-反应曲线(CRCs)。若使用所引用的软件,请选择 log(抑制剂) vs. 反应 — 可变斜率(四参数) 选项。

结果

该实验的主要目的是为三到四种新的parmodulin生成浓度-反应曲线(CRC)。在每块实验板上,通常还需同时生成一个已知化合物(如NRD-21)的CRC,作为实验质量控制,因其具有已知的IC50值。为生成CRC,应设计如表1所示的板图。若仅需单点浓度反应数据,则应将化合物以10 µM(或其他选定浓度)的终浓度加入至少三个孔中。

进行该检测时,酶标仪应显示荧光信号迅速上升(上升应始于前3-4次扫描内)(参见图2A中的示例输出,与图2B中的失败输出对比)。需要注意的是,激动剂必须在15秒内快速加入,以便酶标仪能够准确捕捉荧光信号的上升。在正常工作的实验中,空白对照孔的荧光信号应保持平稳,而阳性对照孔(空白 + 6.5 µM TFLLRN-NH2)的荧光信号应在三到四次扫描后开始上升。当达到最大值(约10次扫描后),荧光信号应开始缓慢下降。在高效拮抗剂的实验中,最高浓度拮抗剂孔的信号应保持相...

讨论

尽管先前报道的方案72总体上可靠,并使我们能够鉴定出一种新的先导型parmodulin NRD-2162,但仍希望获得一种更高效的方案。在新冠疫情期间因物资短缺,该检测方法进一步受到限制。自动液体处理仪所用吸头的采购变得困难,而尝试对吸头进行清洗、灭菌并重复使用则导致CRC结果出现显著变异。这促使我们紧急开展了一系列实验,旨在优化并简化基于PAR的钙动员检测方法。

在本研究中,确定一种可用于洗涤步骤的自动化液体处理仪的有效替代方法是主要优先事项。在先前的检测方法开发过程中,使用机器人液体处理仪可减少因人工换液所导致的误差。然而,这会显著延长检测时间,因为使用8通道液体处理仪进行换液会使每块检测板的总耗时增加30至45分钟。尽管此前已尝试过一种称为" flicking"(弹甩)的人工换液方法,但这些实验并未在细胞接种前对检测板进行明胶包被处理。通过明胶预处理,可确保细胞在手动去除培养基时仍牢固附着于板上,不受操作力度影响。使用经明胶包被并采用弹甩法换液的检测板所生成的CRC...
